Purification,characterization and antiproliferative activity of l-asparaginase from Aspergillus oryzae CCT 3940 with no glutaminase activity

Objective:To explore the anti-proliferative activity of purified l-asparaginase from Aspergillus oryzae CCT 3940(***).Methods:l-asparaginase was produced by submerged fermentation and purified to electrophoresis homogeneity by ionic exchanged chromatography in a fast protein liquid chromatographic *** purified enzyme was characterized and used for the antiproliferative assay against nine tumor cell lines and one non-tumor cell ***:The free glutaminase l-asparaginase was purified 28.6 fold.l-asparaginase showed high stability under physiological condition,remaining stable in the p H range7.0–8.0 after 1 h incubation at temperature range 30–45℃.The Km and Vmax values of purified l-asparaginase were estimated as 0.66 mmol/l and 313 IU/ml,*** purified enzyme could inhibit the growth of a broad range of human tumor cell lines at the concentrations ***,the enzyme from *** CCT 3940 could inhibit tumor growth of leukemia cell line(K562) with a total growth inhibition value of(3.2 ± 2.5) IU/ml and did not inhibit the non-carcinogenic human cell line growth at the concentrations ***:The sensitivity of the cells lines to purified l-asparaginase from *** CCT 3940 appeared to be concentration dependent affording a more significant decrease in cell growth than that observed for the commercial l-asparaginase from Escherichia *** l-asparaginase from *** CCT 3940 has a high potential for pharmaceutical exploitation in the treatment of leukemia.

rong>Introduction:rong> The health and assistance crisis generated by the SARS-CoV2 pandemic has been a serious health problem and a major disruption to the general population, specifically to the most vulnerable groups as cancer patients. This situation is forcing health professionals to face and take on major challenges to maintain the necessary care. Aim: The main objective of this study is to maintain specialized psychological care for oncology patients, and in addition, to describe the principal psychological difficulties of our series of cases. Methods: Before the alarm state in Spain, the therapy was changed from face-to-face to telephone attendance and an e-mail account was opened for specific consultations in which a psycho-oncologist was present in GenesisCare Spain. The psychological interventions carried out during this time have had two characteristics: 1) Cognitive-behavioral therapeutic framework 2) Targeted and specific interventions on issues associated with the COVID-19 pandemic. Results: From March 10 to April 30 (8 weeks), patients and families have been attended to by 2.5 senior psycho-oncologists. A total of 369 psychological services have been attended in Spain in 142 patients. The number of services per patient has ranged from 1 to 4. The median of the long consultation has been 45 minutes. 目的:由于技术原理的限制,目前尚不能对所有的HlA等位基因进行严格的区分,特别是以往没有发现的新基因序列只能通过测序的方法解决,然而,当遇到等位基因杂合时,测序给出的结果仍然无法确认新的序列改变发生在等位基因的哪一侧,这时需要用分子生物学方法分离杂合子然后进行测序才能确定新的基因序列。采用基因克隆方法确认HlA新等位基因。方法:实验于2006-01/05在河南省红十字血液中心HlA实验室,美国海军骨髓库HlA实验室完。成造血干细胞血样由中华骨髓库提供。采用荧光微珠HlA分型方法对中华骨髓库捐献者血样进行HlA分型检测,无法给出确切结果的摸棱两可结果标本用基因克隆(TOPO TA Cloning)、DNA测序的方法确认新的HlA基因序列。结果:通过克隆分离杂台等位基因,再进行测序确认发现新的序列与B~*3709相比,出现4个核苷酸改变:1.355nt C>A,2.363nt C>G,3.412ntG>A,4 477nt C>G,而且均发生在HlA-B基因外显子3(exon 3)。4处改变引起氨基酸编码改变:①编码95 CTC>ATC,氨基酸改变l>I(亮氨酸>异亮氨酸)。②97 AGC>AGG,S>r(丝氨酸>精氨酸)。③114 GAC>AAC D>N(天门冬氨酸>天冬酰胺)。④135 GCC>GCG A=A无氨基酸改变。结论:①新的基因序列已经在GenBnak注册,被WHO的HlA因子命名委员会得到正式命名为HlA-B~*3712基因。②基因克隆是确认HlA新基因的根本方法。

Critical analysis of Children’s Drawings as a Diagnostic Tool for Body Schema and Body Image Disorder in Cerebral Palsy

Cerebral palsy is a multiple disability manifested by motor deficits and impairments of cognition, language, and body perception. To assess if body schema and body image distortion in children and youth with cerebral palsy can be represented through draw-a-person-test, outcome measure of conscious and subconscious body awareness, visual perception and cognition in two age-matched groups, one with cerebral palsy and other with developmental disability, were collected. The outcome was compared within the two groups and with reference data (healthy population) and correlated with the outcome of draw-a-person test. Decreased scores in the draw-a-person test in subjects with cerebral palsy compared both to subjects with developmental disorders and to healthy population had strongly correlated with decreased scores in visual perception. This suggests that draw-a-person test may provide a preliminary guidance for the assessment of visual perception. No statistically significant effect of cognitive abilities on performance in the draw-a-person test was found. The influence of both conscious and subconscious body awareness on the scores in the draw-a-person test was not significant, which confirms that this test alone is not fully reliable to detect the body schema and body image disorder in cerebral palsy.

Injection of bone marrow mesenchymal stem cells by intravenous or intraperitoneal routes is a viable alternative to spinal cord injury treatment in mice

In spite of advances in surgical care and rehabilitation, the consequences of spinal cord injury （SCI） are still challenging. Several experimental therapeutic strategies have been studied in the SCI field, and recent advances have led to the development of therapies that may act on the inhibitory microenvironment. Assorted lineages of stem cells are considered a good treatment for SCI. This study investigated the effect of systemic transplantation of mesenchymal stem cells （MSCs） in a compressive SCI model. Here we present results of the intraperitoneal route, which has not been used previously for MSC administration after compressive SCI. We used adult female C57Bl/6 mice that underwent laminectomy at the T9 level, followed by spinal cord compression for 1 minute with a 30-g vascular clip. The animals were divided into five groups： sham （anesthesia and laminectomy but without compression injury induction）, MSC i.p. （intraperitoneal injection of 8×10^5 MSCs in 500 μl of DMEM at 7 days after SCI）, MSC i.v. （intravenous injection of 8 × 10^5 MSCs in 500μl of DMEM at 7 days after SCI）, DMEM i.p. （intraperitoneal injection of 500μl of DMEM at 7 days after SCI）, DMEM i.v. （intravenous injection of 500 μl of DMEM at 7 days after SCI）. The effects of MSCs transplantation in white matter sparing were analyzed by luxol fast blue staining. The number of preserved fibers was counted in semithin sections stained with toluidine blue and the presence of trophic factors was analyzed by immunohistochemistry. In addition, we analyzed the locomotor performance with Basso Mouse Scale and Global Mobility Test. Our results showed white matter preservation and a larger number of preserved fibers in the MSC groups than in the DMEM groups. Furthermore, the MSC groups had higher levels of trophic factors （brain-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and neurotrophin-4） in the spinal cord and improved locomotor performance. Selective ligninolysis of wheat straw and wood chips by the white-rot fungus lentinula edodes and its influence on in vitro rumen degradability

Background： The present work investigated the influence of lignin content and composition in the fungal treatment of lignocellulosic biomass in order to improve rumen degradability. Wheat straw and wood chips,differing in lignin composition, were treated with lentinula edodes for 0, 2, 4, 8 and 12 wk and the changes occurring during fungal degradation were analyzed using pyrolysis-gas chromatography-mass spectrometry and detergent fiber ***： l. edodes preferentially degraded lignin, with only limited cellulose degradation, in wheat straw and wood chips, leaving a substrate enriched in cellulose. Syringyl（S）-lignin units were preferentially degraded than guaiacyl（G）-lignin units, resulting in a decreased S/G ratio. A decreasing S/G ratio（wheat straw： r =-0.72, wood chips： r =-0.75） and selective lignin degradation（wheat straw： r =-0.69, wood chips： r =-0.88） were correlated with in vitro gas production（IVGP）, a good indicator for rumen ***： l. edodes treatment increased the IVGP of wheat straw and wood chips. Effects on IVGP were similar for wheat straw and wood chips indicating that lignin content and 3D-structure of cell walls influence in vitro rumen degradability more than lignin composition.

Esophagogastric reconstruction in Cobra-Head Shape with Toupet-like Partial Anti-reflux Technique for resection of Proximal Gastric Tumors. Experience with Three Cases from a Non-Asian Population

Background: Proximal gastrectomy is a rarely performed procedure but a feasible option in benign tumors and malignant neoplasms in the proximal third of the stomach since novel options of reconstructions are available nowadays with fewer long-term sequels. Methods: report of three cases of proximal gastric gastrointestinal stromal tumors (GIST), with a description of its presentation, histological characteristics, and follow-up after being treated with proximal gastrectomy with cobra head reconstruction. results: Case 1: A 62-year-old woman with epigastric pain of four months with endoscopic evidence of a cardia GIST. The surgery was performed without complications. The histopathological report confirmed a fusiform GIST of 3.2 × 3 × 2.5 cm, stage IA. No adjuvant treatment was considered. An esophagogram showed no evidence of reflux and no stenosis, and no disease recurrence after 40-months follow-up. Case 2: A 66-year-old woman with dyspepsia and a palpable tumor in the epigastrium. The surgery was performed without complications. The pathology report confirmed a proximal gastric GIST of 13 × 8 × 7 cm, staged II. She received adjuvant treatment with imatinib for 36 months without recurrence and no reflux or stenosis. Case 3: A 55-year-old woman with intermittent hematemesis and right subscapular pain. The surgery was performed without complications. The histopathological report concluded that a GIST tumor of the cardia, 1.4 × 1.2 cm, staged IA. She was left in observation. At a 6-month follow-up, she does not report dysphagia, reflux, or stenosis, and no locoregional recurrence. Conclusions: Open and laparoscopic proximal gastrectomy is a safe therapeutic option for GIST. Furthermore, the reconstruction of the esophagus-gastro anastomosis in the cobra head after proximal gastrectomy is feasible and secure in our population, with good functional results in a short follow-up.
