Noninvasive diagnosis of periportal fibrosis in schistosomiasis mansoni:A comprehensive review

Schistosomiasis mansoni is a neglected disease and key public health problem,mainly due to its high prevalence,the scarcity of public policies,and the severity of some clinical *** fibrosis(PPF)is the commonest complication of chronic schistosomiasis mansoni and its diagnosis requires different *** though wedge biopsy of the liver is considered the gold standard,it is not justified in non-surgical patients,and percutaneous liver biopsy may be informative but does not have sufficient *** PPF tests mostly include biological(serum biomarkers or combined scores)or physical assessments(imaging assessment of fibrosis pattern or tissue stiffness).Moreover,imaging techniques,such as ultrasound,computed tomography,magnetic resonance imaging,and elastography are applied not only to support the diagnosis of schistosomiasis,but also to assess and detect signs of portal hypertension and organ damage due to chronic schistosomiasis.A combination between a comprehensive history and physical examination with biomarkers for liver fibrosis and imaging methods seems to offer the best approach for evaluating these *** addition,understanding their strengths and limitations will allow a more accurate interpretation in the clinical context and can lead to greater accuracy in estimating the degree of fibrosis in patients with Schistosomiasis mansoni(***)*** review will discuss the different noninvasive methods that are currently available for the evaluation of PPF in *** infection,and their application,advantages,and limitations in clinical practice.

BACKGROUND The evaluation of periportal fibrosis(PPF)is essential for a prognostic assessment of patients with Schistosomiasis *** WHO Niamey Protocol defines patterns of fibrosis from abdominal ultrasonography,^(1)H-nuclear magnetic resonance(NMR)-based metabonomics has been employed to assess liver fibrosis in some *** To build 1H-NMR-based metabonomics models(MM)to discriminate mild from significant periportal PPF and identify differences in the metabolite *** A prospective cross-sectional study was performed on schistosomiasis patients at a University Hospital in Northeastern *** evaluated 41 serum samples from 10 patients with mild PPF(C Niamey pattern)and 31 patients with significant PPF(D/E/F Niamey patterns).MM were built using partial least squares-discriminant analysis(PlS-DA)and orthogonal projections to latent structures discriminant analysis(OPlS-DA)*** PlS-DA and OPlS-DA resulted in discrimination between mild and significant PPF groups with R2 and Q2 values of 0.80 and 0.38 and 0.72 and 0.42 for each model,*** OPlS-DA model presented accuracy,sensitivity,and specificity values of 92.7%,90.3%,and 100%to discriminate significant *** metabolites identified as responsible by discrimination were:Nacetylglucosamines,alanine,glycolaldehyde,carbohydrates,and *** MMs discriminated mild from significant PPF patterns in patients with Schistosomiasis mansoni through identification of differences in serum metabolites profiles.

Tailoring the partitioning of proteins using ionic liquids as adjuvants in polymer-polymer aqueous biphasic systems

Aqueous biphasic systems(ABS)are promising for proteins purification;however,when dealing with samples comprising several proteins,the selectivity towards a target protein is difficult to *** this work,the addition of ionic liquids(Ils)as adjuvants(5 wt%and 10 wt%)in ABS composed of polyethylene glycol(PEG)and dextran was investigated to tailor proteins partitioning between the coexisting *** liquid-liquid phase diagrams were determined at 298 K,followed by partition studies of three proteins(bovine serum albumin(BSA),immunoglobulin G(IgG),and cytochrome C(Cyt C)).Partition coefficients of IgG and BSA indicate the preference of both proteins to the dextran-rich phase,whereas Cyt C has no preferential partitioning between the *** addition of chloride-based Ils as adjuvants allows to increase or decrease the partition coefficients,thus tailoring the proteins partitioning between the *** partitioning essentially depends on the Il content in each phase,whereas Cyt C and IgG partitioning is ruled by the Ils chemical structure and established *** docking was carried out to address the Ils effect on the proteins partitioning,supporting experimental observations,while identifying the specific interactions *** partition of each protein in polymer-salt ABS with Ils as adjuvants was determined,demonstrating the higher tailoring ability of polymer-polymer ABS when adding ***,the partition of each protein in presence of the remaining ones was determined,as well as the selectivity of the studied systems to separate each pair of proteins,paving the way for their use in liquidliquid chromatography.

Background:Hepatitis C,caused by the Hepatitis C Virus(HCV),is the second most common form of viral *** geographical distribution of HCV genotypes can be quite complex,making it challenging to ascertain the most prevalent genotype in a specific ***:To address this,a review was conducted to determine the prevalence of HCV genotypes across various provinces and as a whole in *** scientific literature regarding the prevalence,distribution,genotyping,and epidemiology of HCV was gathered from published articles spanning the years ***:Genotype 1 accounted for 5.1%of the patients,with its predominant subtype being 1a at 4.38%.The frequencies of its other subtypes,1b and 1c,were observed to be 1.0%and 0.31%*** 2 had a frequency of 2.66%,with the most widely distributed subtype being 2a at 2.11%of the *** other subtypes,2b and 2c,had frequencies of 0.17%and 0.36%*** most prevalent genotype among all isolates was 3(65.35%),with the most frequent subtype being 3a(55.15%),followed by 3b(7.18%).The prevalence of genotypes 4,5,and 6 were scarce in Pakistan,with frequencies of 0.97%,0.08%,and 0.32%*** prevalence of untypeable and mixed genotypes was 21.34%and 3.53%*** genotypes proves to be a productive method in assisting with the duration and selection of antiviral *** HCV genotypes can exhibit variations in their response to specific antiviral *** genotypes may have distinct natural histories,including variations in disease progression and *** genotypes may lead to more rapid liver damage,while others progress more ***:This information can guide screening and testing strategies,helping to identify individuals at higher risk of developing severe *** the distribution of HCV genotypes in a population can provide valuable insights into the transmission dynamics of the virus.

Intestinal anti-inflammatory activity of Ground Cherry(Physalis angulata l.)standardized CO2 phytopharmaceutical preparation

XEN^(■) implant and trabeculectomy medium-term quality of life assessment and comparison of results

OBJECTIVE:To evaluate and compare the quality of life of patients submitted to XEN implant or trabeculectomy and the relationship with potentially involved variables.;METHODS:A cross-sectional study of patients with advanced open-angle glaucoma who underwent implantation of XEN (group 1) and trabeculectomy (group 2) between October 2015 and February 2017. The studied variables were: age, gender, follow-up time, need of topical anti-hypertensive therapy, visual acuity and intraocular pressure (IOP). The quantification of the quality of life was attained through the Glaucoma Symptom Scale (GSS) questionnaire.;RESUlTS:Totally 34 eyes (34 patients) were included, 17 in each group. The mean GSS scores for group 1 were 42.6±6.8 (median, 47; p25, 36.5; p75, 48.5) and for group 2 it was 41.6±7.0 (median, 43; p25, 36.5; p75, 47.0; =0.34). There was a strong negative correlation between the need for topical anti-hypertensive drugs and the GSS result in both groups (=-0.88, ly) and a moderate negative correlation with IOP in group 1 (=-0.50, =0.03).;CONClUSIONS:The analysis demonstrates the non-inferiority of medium-term quality of life of one group in relation to the other (XEN implant and trabeculectomy). The number of topical anti-hypertensive drugs and IOP negatively influenced the quality of life.

Marigold(Tagetes erecta): The Potential Value in the Phytoremediation of Chromium

Environmental pollution with chromium(Cr) is harmful to humans, animals and plants, while in plants it causes diminished growth,anatomical alterations and death. In the present study, the potential value of marigold(Tagetes erecta) in the phytoremediation of Cr has been investigated. The randomized experimental design involved the exposure of plants to nutrient solutions containing 0.00,0.04, 0.08, 0.12, 0.16 or 0.24 mmol l^(-1)Cr(Ⅲ). Chromium toxicity was observed at Cr(Ⅲ) concentrations ≥ 0.12 mmol l^(-1) as demonstrated by diminished growth of the aerial parts and reduced density of the root system. Increasing Cr(Ⅲ) concentrations in the nutrient solution resulted in a higher bioaccumulation of total Cr in the tissues, although translocation from roots to aerial parts was not efficient(maximum value of 25% at 0.12 mmol l^(-1)Cr(Ⅲ)). The Cr bioaccumulation was up to 11-fold greater in roots than in the aerial parts. Tagetes erecta exhibited leaf plasticity when exposed to Cr, indicating the existence of a tolerance mechanism to Cr in this species. Chromium caused a reduction in xilem vases, resulting in a plastic effect in T. erecta leaves that increased the metal tolerance in culture solution. Tagetes spp. are potential Cr hyperaccumulators; at Cr(Ⅲ) concentrations up to 0.12 mmol l^(-1), the plants accumulated levels above that proposed for hyperaccumulators and still maintained a considerable growth and even flourished. However, this study was conducted in nutrient solution, and studies on species confirmation as Cr hyperaccumulator should be conducted in soils for further clarification.

Antibacterial activity and inhibition against Staphylococcus aureus NorA efflux pump by ferulic acid and its esterified derivatives

Objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIC)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MICs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and lEU255 were present commonly in stabilizing all *** PCA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

Objective: To evaluate the trypanocidal, leishmanicidal and cytotoxic activity of Eugenia jambolana(E. jambolana) and Eugenia uniflora(E. uniflora) extracts and ***: The products were characterized by lC–MS. Antiparasitic assays were performed and cytotoxicity was evaluated in fibroblastos. In vitro assays were performed using spectrophotometric evaluation. All assays were performed in ***: The results showed that the extracts and the tannic fraction from E. jambolana inhibited 100% of the epimastigote lines. The ethanolic extract was the most efficient in all concentrations tested against the three parasite strains. In the cytotoxicity assay the flavonoid fraction showed low toxicity. All E. uniflora samples showed cytotoxicity at the highest concentration tested, but the extract showed no toxic effect on the fibroblasts at the lowest concentration. The flavonoid and tannic fractions were more efficient against leishmania braziliensis promastigotes compared to the extract. However, the extracts and the tannic fraction were more effective against leishmania infantum strains. The effect on epimastigote cells was observed at all concentrations tested, with all E. uniflora samples. However, the samples were more effective at the highest concentration, where there was inhibition in 100% of the Trypanosoma cruzi ***: The species E. jambolana and E. uniflora presented antiparasitic activity against all tested parasite strains, indicating that these species can serve as an alternative therapy as they were efficient in the tests performed. The E. uniflora extract and the E. jambolana flavonoid fraction presented a low cytotoxicity, opening the floor for new biological studies.
