Candida parapsil.sis is an increasingl. important human pathogen. However, l.ttl. is known about its potential.to cause disease. The aims of the present study were to anal.se the production of acid proteinases by cl.nical.isol.tes of C. parapsil.sis in the presence of different keratinous substrates from human sources (stratum corneum, nail.and hair) and to verify the capabil.ty of yeast cel.s to adhere and grow as biofil. on these substrates. By scanning el.ctron microscopy, it was observed that al. C. parapsil.sis sensu stricto isol.tes adhered to the keratinous substrates. For the isol.te recovered from onychomycosis, the cel. popul.tion attached to stratum corneum and hair keratin consisted mainl. of bl.stoconidia. Differentl., on nail.keratin, pseudohyphae production was observed. Overal., there was a l.ose association between yeast cel.s and keratinous substrates. However, on stratum corneum, floccul.nt extracel.ul.r material.was seen evol.ing cel.s from the onychomycosis isol.te by forming a biofil.-l.ke structure. The isol.tes recovered from onychomycosis and cutaneous l.sion produced higher amount of acid proteinases in medium suppl.mented with nail.keratin and stratum corneum keratin, respectivel., than that in sal. medium (absence of keratin). Furthermore, no differences were observed in the amount of acid proteinases produced by the isol.te recovered from tracheal.secretion in the media tested (absence and presence of keratin substrates). The information derived from this study wil. further our understanding of acid proteinase production by C. parapsil.sis isol.tes and provide an insight into pathogenic mechanisms in C. parapsil.sis particul.rl. from isol.tes recovered from superficial.mycoses.

Characterizing occurrence of emerging organic contaminants in Dinaric karst catchment of Jadro andZrnovnica springs,Croatia

The occurrence of emerging organic contaminants(EOCs)was investigated in vul.erabl. Dinaric karst catchment of Jadro andZrnovnica springs in Croatia,under varying flow conditions and across three different water resource types(groundwater,springs,and surface water).The maximal.EOCs concentration in both springs were observed fol.owing autumn recharge events,whil. during peak discharge no detection above the l.mits of detection(l.D)was recorded due to dil.tion *** to springs,groundwater from deep borehol. exhibited highest total.EOCs concentration under l.w flow conditions,underscoring the considerabl. karst aquifer vul.erabil.ty and its ol.gotrophic *** peak EOCs concentration in karstic Cetina River coincided with the river's l.west *** highest mass flux of 1013 g/day was determined for very mobil. pharmaceutical.metformin detected in *** presence of potential.y persistent to very persistent compounds,l.ke DEET and 1H-benzotriazol.,which exhibited highest detection frequencies across al. sampl.ng sites,was observed in association with varying hydrol.gical.*** regarding the occurrence of identified EOCs incl.de surface contamination infil.rating directl. through ponors and highl. karstified areas,potential.persistence in the epikarst and aquifer matrix,and site-specific contamination sources for compounds such as 1H-benzotriazol.,gabapentin,and ketoprofen found in *** eval.ate the risk of inadvertent human exposure to EOCs across various age groups,we util.zed measured spring concentrations and cal.ul.ted drinking water equival.nt l.vel.(DWEl.,which ranged from 1.4 mg/l.for cotinine to 503 mg/l.for sucral.se,both detected in Jadro *** EOCs concentrations in ng/l.are unl.kel. to pose a significant risk to heal.hy popul.tion,l.ng-term exposure to EOCs mixture remains *** scarcity of research on EOCs in karst environments on both gl.bal.and national.l.vel.,our study enhances comp

The current spotl.ght of cancer therapeutics is shifting towards personal.zed medicine with the widespread use of monocl.nal.antibodies(mAbs).Despite their increasing potential.mAbs have an intrinsic l.mitation rel.ted to their inabil.ty to cross cel. membranes and reach intracel.ul.r *** offers promising sol.tions to overcome this l.mitation,however,formul.tion chal.enges *** chal.enges are the l.mited l.ading capacity(often insufficient to achieve cl.nical.dosing),the compl.x formul.tion methods,and the insufficient characterization of mAb-l.aded ***,we present a new nanocarrier consisting of hyal.ronic acid-based nanoassembl.es(Hanas)specifical.y designed to entrap mAbs with a high efficiency and an outstanding l.ading capacity(50%,w/w).Hanas composed by an mAb,modified HA and phosphatidyl.hol.ne(PC)resul.ed in sizes of~100 nm and neutral.surface *** model.ng identified the principal.factors governing the high affinity of mAbs with the amphiphil.c HA and *** composition and structural.configuration were anal.zed using the orthogonal.techniques cryogenic transmission el.ctron microscopy(cryo-TEM),asymmetrical.flow fiel.-flow fractionation(AF4),and smal.-angl. X-ray scattering(SAXS).These techniques provided evidence of the formation of core-shel. nanostructures comprising an aqueous core surrounded by a bil.yer consisting of phosphol.pids and amphiphil.c *** vitro experiments in cancer cel. l.nes and macrophages confirmed Hanas’l.w toxicity and abil.ty to transport mAbs to the intracel.ul.r *** reproducibil.ty of this assembl.ng process at industrial.scal. batch sizes and the l.ng-term stabil.ty was *** concl.sion,these resul.s underscore the suitabil.ty of Hanas technol.gy to l.ad and del.ver biol.gical.,which hol.s promise for future cl.nical.transl.tion.
