Rhizopus is fungus of the order Mucorales, responsible for invasive diseases called mucormycosis. The correct identification of these fungi is one of the main questions raised in the literature since the diagnosis is not simple, being the necessary detection in biopsies of infected tissues and isolation of the microorganism for identification by morphological and molecular methods. In vitro susceptibility testing results are not fully clear since there are reports showing that in vitro resistance does not always mean therapeutic failure. This paper describes a case of rhino-orbito-cerebral mucormycosis in a patient with alcoholic chronic pancreatitis and secondary mellitus diabetes involving microbiological diagnostic, antifungal susceptibility tests for conidia and hypha and clinical correlation. The causative agent was identified by morphological characteristics and DNA sequencing. Minimal Inhibitory Concentration (MIC) for amphotericin B (AMB), itraconazole (ITZ) and terbinafine (TERB) was determined by broth microdilution method. Standardization of the dynamic growth evaluation patterns using the automated system BCT (BioCell-Tracer?) was obtained allowing to determining hyphae growth control and evaluating the effect of different concentrations of amphotericin B, itraconazole and terbinafine directly on growing hyphae. The fractional inhibitory concentration (FIC) for the combination of the two antifungals that the patient received at the same time, terbinafine and itraconazole, was also determined trying to correlate with clinical outcome. Strains isolated of retro-orbital abscess were identified as Rhizopus oryzae. MIC results (MIC) testing conidia showed high sensibility to amphotericin B and itraconazole in comparison to the obtained for terbinafine. The tests in the automated system BCT using hyphae showed sensibility to all tested antifungal agents and the FIC results for combination in vitro between terbinafine and itraconazole showed synergism consisten

In this work, the localization, density, morphology and ultrastructure of secretory structures in aerial organs of Flourensia campestris (FC) and F. oolepis (FO) (Asteraceae) by means of a combination of light, fluorescence, transmission (TEM) and scanning electron microscopy (SEM) were examined. The possible role of secretory structures in the production and secretion of the phytotoxic sesquiterpene (-)-hamanasic acid A ((-)HAA) in both species was also assessed. Capitate glandular trichomes were found in all reproductive organs of FC and FO, and were being reported for the first time. These glandular trichomes, typically associated to edges and veins, were of the same type as those already described for vegetative organs, and were abundant in involucral bracts and corolla of tubulose and ligulate flowers. Their density in reproductive organs of both species was similar (ca. 30/mm2) and lower than that found in leaves (ca. 100/mm2) and stems (ca. 160/mm2 in FC, and up to 650/mm2 in FO). Glandular trichomes in vegetative organs followed a species-specific pattern of distribution. TEM and SEM observations suggest that each species differs in the way in which secretory materials are released to the outside: through cracks or pores in FC, or through a loose cuticle in FO. Similar inspections of the secretory ducts revealed lipophilic vacuoles localized in subepithelial and epithelial cells, in which secretions accumulated before being transferred to the duct. The presence of wall ingrowths in subepithelial cells suggests that granulocrine secretion operates in these species. Secretory ducts varied in density and diameter among the organs in both species, with the combination being maximal in woody stems. (-)HAA was only detected in surface secreted resins of both species, and its concentration (2D-TLC, GC-FID) was intimately associated with the distribution and density of glandular trichomes in each organ (capitula, leaves, and stems with primary or secondary growth).

The aim of this study was to evaluate the effects of silver nitrate in the in vitro development and conservation of passion fruit plants. Experiments were carried out at the Laboratory of Culture and Tissues, in the National Cassava and Fruits Research Center (EMBRAPA), using microcuttings of Passiflora gibertii N. E. Br., previously cultivated in vitro. The microcuttings were placed in MS media supplemented with silver nitrate at concentrations of 0, 1, 2, 4 and 8 mg·L-1, and, during 30, 60 and 90 days, the following variables were identified: Shooting length (cm), number of roots, number and coloration of leaves. A completely randomized design with 30 replications in a split-plot scheme was employed. Polynomial regression equations were adjusted in analysis of variance to compare averages of treatments. The obtained results support the assumption of high sensitivity of passion fruit tissues to ethylene, which reflects in the lost of vigor and in the induction of culture senescence by its accumulation. Based on obtained results, and in order to mitigate this problem, it is possible to indicate an addition of 2 mg·L-1?of silver nitrate in culture media, for the micropropagation of passion fruit plant, allowing the controlling of the atmosphere inside the culture test tubes, during the establishment of explants and other steps of the process.

Introduction: Abdominal angiostrongyliasis is an infrequently diagnosed disease because it is little known and is usually well tolerated. Methods: We carried out a systematic review with PubMed as a search engine for the MedLine database, and we analysed the clinical, epidemiological and analytical parameters of this disease. Results: In total, reports of 27 case have been found, together with a series of 116 patients, which were later augmented up to 194 patients. In the results, we have observed a predominance of male patients, and Costa Rica, Brazil and the United States as the main countries of origin. Typical symptoms include abdominal pain on the right iliac fossa, with leukocytosis and eosinophilia. Discussion: There are some serological diagnostic methods, although lack of standardization leads to most of the diagnoses being reached via pathology. Surgery is the usual treatment approach. Although there are medical options, this is a controversial subject because of the low number of cases published in the literature, which implies an absence of solid studies.

Model Study: The genus Acinetobacter is composed of opportunistic pathogens that mainly affects immunocompromised and patients during a prolonged hospitalization period. A. baumannii can survive in the hospital setting at various locations such as mechanical ventilators, dialysis machines, ventilation systems, water sources, skin and mucous membranes of health professionals and patients, drug preparations and disinfectants among others. Antimicrobials are essential for the treatment of infections, so their use has become common, and often inappropriate and abusive. This pathogen has presented multi-resistance to several classes of antimicrobials and has become a major cause of death in hospitals. Objective: To evaluate the occurrence of Acinetobacter baumannii in clinical samples from the Laboratory of Clinical analyzes, HCSL of Pouso Alegre, MG. Methods: All the samples referring to the patients of the Samuel Libanio Clinical Hospital including most of the specialties were evaluated. From the positive samples for Acinetobacter baumannii the medical records of hospitalized patients were evaluated. From the medical records, data were obtained as gender and age of the patient, place of hospitalization, together with month and year of the laboratory procedure. Result: A total of 14,859 samples of diverse materials were evaluated, being 80 (0.5%) samples positive for Acinetobacter baumannii. Of these positive samples, the largest occurrences were identified in male patients 59 (73.7%), aged over 60 years 37 (46.3%). The most affected hospital sector was Adult ICU with 44 (55.0%) of the cases. In May 2015, there were 5 (6.3%) cases registered and in May 2016, 16 (20.0%) cases. The clinical material with the highest occurrence of Acinetobacter baumannii was Mini Bal 29 (36.3%). The genetic modification of the live attenuated Mycobacterium bovis BCG to deliver a protective Corynebacterium diphtheriae antigen in vivo could be a safer and less costly alternative to the new and more expensive DTP vaccines available today, in particular to third world-countries. The stability of expression of heterologous antigens in BCG, however, is a major challenge to the use of live recombinant bacteria in vaccine development and appears to be dependent to a certain extent, on a genetic compatibility between the expression cassette within the plasmid construct and the mycobacterium host. In the quest for the best recombinant BCG transformant to express the dtb gene of C. diphtheriae we generated two new rBCG strains by transforming the Moreau substrain of BCG with the mycobacterial expression vectors pUS973 and pUS977, each one carrying a different promoter to drive the expression of the target antigen. After transformation recombinant BCG clones were selected on Middlebrook 7H10 kanamycin Agar plates, expanded in Middlebrook 7H9 kanamycin Broth and analyzed by agarose gel electrophoresis and immunoblotting. rBCGs transformed with the construct carrying the weak PAN promoter from M. paratuberculosis stably expressed the dtb gene. Conversely, rBCGs transformed with the construct carrying the strong mycobacterium hsp60 promoter were unstable and consequently unfit for the expression of the C. diphtheriae gene.

A quantitative descriptive study aimed to evaluate the physical structure accessibility of gynecological nursing clinics and their bathrooms in basic health units for users with motor disabilities. A checklist tool/instrument was applied for analyzing 16 items in the bathroom and four in the nursing clinic. 157 basic health units were evaluated. In the nursing clinic, counters and tables (79.6%) were accessible, but stretchers (93%), seats (93.6%) and doors (87.3%) were inaccessible;moreover, 17.8% of the units examined proved totally inaccessible. In the bathroom, the items with the best evaluation were the height of the toilet flusher (47%) with operation by slight pressure (44.6%) and the wall-mounted sink (42.7%). Bathroom doors and turning area/space for wheelchairs were inaccessible. No bathroom reached full accessibility, and 31.2% were entirely inaccessible. Non-compliance was evident of the standard 9050:2004 of the Brazilian Association of Technical Standards. Raising the awareness of health and engineering professionals is recommended to learn technical accessibility standards and implement them.
