Formation potential and analysis of 32 regulated and unregulated disinfection by-products: Two new simplified methods

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic matter naturally present in water, leading to the formation of disinfection by-products(DBPs). multi-analyte methods based on mass spectrometry(mS) are preferred to quantify multiple DBP classes at once however, most require extensive sample pre-treatment and significant resources. In this study, two analytical methods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap(P&T) coupled with gas chromatography mass spectrometry(GC-mS) method was optimized that automated sample pre-treatment and analyzed volatile and semi-volatile compounds, including trihalomethanes(THms), iodinated trihalomethanes(I-THms), haloacetonitriles(HANs), haloketones(HKTs) and halonitromethanes(HNms). LOQs were between 0.02-0.4 μg/L for most DBPs except for 8 analytes that were in the low μg/L range. A second method with liquid chromatography(LC) tandem mass spectrometry(mS/mS) was developed for the quantification of 10 haloacetic acids(HAAs) with a simple clean-up and direct injection. The LC-mS/mS direct injection method has the lowest detection limits reported(0.2-0.5 μg/L). Both methods have a simple sample pre-treatment, which make it possible for routine analysis. Hyperchlorination and uniform formation conditions(UFC) formation potential tests with chlorine were evaluated with water samples containing high and low TOC. Hyperchlorination formation potential test maximized THms and HAAs while UFC maximized HANs. Ascorbic acid was found to be an appropriate quencher for both analytical methods. Disinfected drinking water from four water utilities in Alberta, Canada were also evaluated.

目的利用生物信息学方法筛选湿性年龄相关性黄斑变性(wet age-related macular degeneration,w AmD)的氧化应激相关差异表达基因及靶向中药,以期为wAmD的预防和治疗提供新思路。方法从GEO数据库中筛选数据集,利用R软件limma包筛选差异基因,从Genecard数据库筛选氧化应激差异基因,并利用韦恩图得到共有基因。应用metascape对氧化应激差异基因进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)分析,并通过String数据库构建蛋白质相互作用(protein-protein interaction,PPI)网络,再通过Cytoscape软件筛选出wAmD中氧化应激的关键(Hub)基因。采用GSE103060数据集验证Hub基因的差异表达。利用Coremine medical预测靶向中药。结果经筛选共得到差异表达基因1874个,其中上调基因747个,下调基因1127个。KEGG富集分析表明,差异表达基因主要富集于细胞外基质受体相互作用、黏附斑激酶通路、河马(Hippo)信号通路、晚期糖基化终末产物-晚期糖基化终末产物受体(advanced glycation end products-receptor for advanced glycation end products,AGE-RAGE)信号通路、钙信号通路和磷脂酰肌醇-3-羟激酶(phosphatidylinositol-3-hydroxykinase,PI3K)/蛋白激酶B(protein kinase B,Akt)信号通路。通过PPI网络筛选得到与w AmD氧化应激密切相关的8个Hub基因,分别为IL6、EGFR、HIF1A、APOE、FN1、PPARG、EDN1和NGF。最后在GSE103060数据集中验证了以上Hub基因的显著差异表达。利用Coremine medical预测人参、黄芪、枸杞子、丹参、银杏叶等可作为预防或治疗wAmD的靶向中药。结论通过对w AmD芯片的挖掘和生物信息分析,差异基因主要富集于氧化应激的相关通路与wAmD的氧化应激相关基因引起的。防治wAmD的首选中药为人参、黄芪、枸杞子、丹参和银杏叶等26种。这将对理解和探索wAmD的中医药防治策略有重要意义。

Objective: Impower210(NCT02813785) explored the efficacy and safety of single-agent atezolizumab *** as second-line treatment for advanced non-small cell lung cancer(NSCLC) in East Asian ***: Key eligibility criteria for this phase Ⅲ, open-label, randomized study included age ≥18 years;histologically documented advanced NSCLC per the Union for International Cancer Control/American Joint Committee on Cancer staging system(7th edition);Eastern Cooperative Oncology Group performance status of 0 or 1;and disease progression following platinum-based chemotherapy for advanced or metastatic NSCLC. Patients were randomized 2:1 to receive either atezolizumab(1,200 mg) or docetaxel(75 mg/m^(2)). The primary study endpoint was overall survival(OS) in the intention-to-treat(ITT) population with wild-type epidermal growth factor receptor expression(ITT EGFR-WT) and in the overall ITT ***: median OS in the ITT EGFR-WT population(n=467) was 12.3 [95% confidence interval(95% CI),10.3-13.8] months in the atezolizumab arm(n=312) and 9.9(95% CI, 7.8-13.9) months in the docetaxel arm[n=155;stratified hazard ratio(HR), 0.82;95% CI, 0.66-1.03]. median OS in the overall ITT population was 12.5(95% CI, 10.8-13.8) months with atezolizumab treatment and 11.1(95% CI, 8.4-14.2) months(n=377) with docetaxel treatment(n=188;stratified HR, 0.87;95% CI, 0.71-1.08). Grade 3/4 treatment-related adverse events(TRAEs) occurred in 18.4% of patients in the atezolizumab arm and 50.0% of patients in the docetaxel ***: Impower210 did not meet its primary efficacy endpoint of OS in the ITT EGFR-WT or overall ITT populations. Atezolizumab was comparatively more tolerable than docetaxel, with a lower incidence of grade3/4 TRAEs.

Gradients in predation risk in a tropical river system

The importance of predation risk as a key driver of evolutionary change is exemplified by the Northern Range in Trinidad, where research on guppies living in multiple parallel streams has pro- vided invaluable insights into the process of evolution by natural selection. Although Trinidadian guppies are now a textbook example of evolution in action, studies have generally categorized predation as a dichotomous variable, representing high or low risk. Yet, ecologists appreciate that community structure and the attendant predation risk vary substantially over space and time. Here, we use data from a longitudinal study of fish assemblages at 16 different sites in the Northern Range to quantify temporal and spatial variation in predation risk. Specifically we ask： 1） Is there evidence for a gradient in predation risk？ 2） Does the ranking of sites （by risk） change with the defi- nition of the predator community （in terms of species composition and abundance currency）, and 3） Are site rankings consistent over time？ We find compelling evidence that sites lie along a contin- uum of risk. However, site rankings along this gradient depend on how predation is quantified in terms of the species considered to be predators and the abundance currency is used. Nonetheless, for a given categorization and currency, rankings are relatively consistent over time. Our study sug- gests that consideration of predation gradients will lead to a more nuanced understanding of the role of predation risk in behavioral and evolutionary ecology. It also emphasizes the need to justify and report the definition of predation risk being used.

Assessing disease activity using the pediatric Crohn’s disease activity index:Can we use subjective or objective parameters alone?

BACKGROUND The pediatric Crohn’s disease activity index(PCDAI)is used as a standard tool to assess disease activity in clinical trials for pediatric Crohn’s *** To examine which items on the PCDAI drive assessment of disease activity,and how subgroups of subjective and objective items reflect change in disease state over *** Selective raw data from three prospectively collected datasets were combined,including 703 children with full PCDAI data at baseline,at 3-mo(Q1,n=670),and 1-year(Q4,n=474).Change in individual PCDAI scores from baseline to Q1 and to Q4 were examined using the non-weighted *** Abdominal pain,well-being,weight,and stooling had the highest change scores over *** indicators including albumin,abdominal exam,and height velocity *** scores for well-being and abdominal exam did not explain significant variance at Q1 but were significant predictors at Q4(Pms predicted less variance(18%and 22%)on total PCDAI scores at Q1 and Q4 compared to the full PCDAI,or a composite scale(both 32%)containing significant *** Although subjective items on the PCDAI change the most over time,the full PCDAI or a smaller composite of items including a combination of subjective and objective components classifies disease activity better than a subgroup of either subjective or objective items *** on subjective or objective items as stand-alone proxies for disease activity measurement could result in misclassification of disease state.

Non-steroidal anti-inflammatory drugs and statins in relation to colorectal cancer risk

AIm： To investigate the association between individual or combined use of non-steroidal anti-inflammatory drugs （NSAIDs） or statins and colorectal cancer ***： In a population-based case-control study in women, we examined the association between NSAIDs and statin use and the risk of colorectal cancers. We further investigated whether the use of statins modifies the protective effect of NSAIDs. Female cases （n = 669）of colorectal cancer aged 50-74 years were identified from a storewide registry in Wisconsin during 1999-2001. Community control women （n = 1375） were randomly selected from lists of licensed drivers and medicare beneficiaries. medication use and risk factor information were gathered during a structured telephone interview. A multivariable logistic regression model was used to calculate odds ratio （OR） and 95% confidence interval （CI）.RESULTS： Overall, NSAIDs users had a 30% reduction in risk of colorectal cancer （95% CI： 0.56-0.88）. Statin use was not associated with colorectal cancer risk （OR = 1.17, 95% CI： 0.74-1.85）, regardless of structural type （lipophilic or hydrophilic）, duration of use, or recency. There was no evidence of an interaction between NSAIDs and statins and colorectal cancer risk （P-interaction = 0.28）.CONCLUSION： Although our results confirm the inverse association between NSAIDs use and colorectal cancer risk, they do not support a risk reduction in statin users, or an interaction effect of combined NSAIDs and statin use.

The complexities of female mate choice and male polymorphisms： Elucidating the role of genetics, age, and mate-choice copying

Genetic, life history, and environmental factors dictate patterns of variation in sexual traits within and across popula- tions, and thus the action and outcome of sexual selection. This study explores patterns of inheritance, diet, age, and mate-choice copying on the expression of male sexual signals and associated female mate choice in a phenotypically diverse group of Schizo- cosa wolf spiders. Focal spiders exhibit one of two male phenotypes：＇omamented＇ males possess large black brushes on their fo- relegs, and ＇non-ornamented＇ males possess no brushes. Using a quantitative genetics breeding design in a mixed population of ornamented/non-ornamented males, we found a strong genetic basis to male phenotype and female choice. We also found that some ornamented males produced some sons with large brushes and others with barely visible brushes. Results of diet manipula- tions and behavioral mating trials showed no influence of diet on male phenotype or female mate choice. Age post maturation, however, influenced mate choice, with younger females being more likely to mate with ornamented males. A mate-choice copy- ing experiment found that, following observations of another female＇s mate choice/copulation, virgin mature females tended to match the mate choice （ornamented vs. non-ornamented males） of the females they observed. Finally, analyses of genetic varia- tion across phenotypically pure （only one male phenotype present） vs. mixed （both phenotypes present） populations revealed ge- netic distinction between phenotypes in phenotypically-pure populations, but no distinctionin phenotypically-mixed populations. The difference in patterns of genetic differentiation and mating across geographic locations suggests a complex network of fac- tors contributing to the outcome of sexual selection .
