The bone marrow contains a heterogeneous milieu of cells, including macrophages, which are key cellular mediators for resolving infection and inflammation. Macrophages are most well known for their ability to phagocytose foreign bodies or apoptotic cells to maintain homeostasis; however, little is known about their function in the bone microenvironment. In the current study, we investigated the in vitro interaction of murine macrophages and bone marrow stromal cells （BMSCs）, with focus on the juxtacrine induction of IL-6 signaling and the resultant effect on BMSC migration and growth. The juxtacrine interaction of primary mouse macrophages and BMSCs activated IL-6 signaling in the co-cultures, which subsequently enhanced BMSC migration and increased BMSC numbers. BMSCs and macrophages harvested from IL-6 knockout mice revealed that IL-6 signaling was essential for enhancement of BMSC migration and increased BMSC numbers via juxtacrine interactions. BMSCs were the main contributor of IL-6 signaling~ and hence activation of the IL-6/ gp130/STAT3 pathway. Meanwhile, macrophage derived IL-6 remained important for the overall production of IL-6 protein in the co-cultures. Taken together, these findings show the function of macmphages as co-inducers of migration and growth of BMSCs, which could directly influence bone formation and turnover.

Introduction: Infections are additional factors of morbidity and mortality in multiple myeloma (MM), and the current recommendation is antibiotic prophylaxis. In sub-Saharan Africa, few data on infectious complications of MM are available. We aim to describe the microbiological features of infections in MM, and their impact on survival in Senegalese patients. Methods: A retrospective (January 2005-January 2022), analytic, multicenter study on infections in patients followed for MM (IMWG criteria) in Senegalese clinical hematology services. The socio-epidemiological, diagnostic, microbiological, evolutionary and survival aspects were analyzed. Results: The study included 106 patients with multiple myeloma who had an infection at admission or during the treatment. Ten patients have the comorbidity (hypertension, lupus, type 2 diabetes). These patients had 136 infectious events identified at diagnosis (79.2%) or during chemotherapy (20.8%). The sites of infection are lung (42.6%), urinary (29.4%), dermatological (6.6%), digestive (5.2%), osteoarticular (4.4%), ear, nose and throat (3.7%), central nervous system (1.5%), or without site. We recorded 26.4% of patients with multi-site infections. The causal pathogens are bacteria (Gram-negative bacilli: 22.1%;Gram positive bacilli: 9.5%, Mycobacterium tuberculosis: 13.3%), parasitique (plasmodium falciparum 6.6%), viruses (SARS-COV2: 2.9%, VZV: 2.2%) and fungal (2.9%). Survival was reduced in patients who had an infection at the time of multiple myeloma diagnosis (p: 0.189) and those who had multiple infectious foci (p: 0.011). Conclusion: Infections in multiple myeloma are more frequent at diagnosis. The germs are varied and mostly bacteria, particularly gram-negative bacteria, and kochs bacillus. Our study reveals that multiple infectious foci are a poor prognosis factor. It is necessary to evaluate the infectious risk early, and to adopt an antibiotic prophylaxis based on our tropical environment.

In this study,we demonstrated the label-free continuous separation and enrichment of Bacillus subtilis populations based on length using viscoelastic ***,a gram-positive,rod-shaped bacterium,has been widely used as a model organism and an industrial *** can be arranged in different morphological forms,such as single rods,chains,and clumps,which reflect differences in cell types,phases of growth,genetic variation,and changing environmental *** ability to prepare *** populations with a uniform length is important for basic biological studies and efficient industrial ***,we systematically investigated how flow rate ratio,poly(ethylene oxide)(PEO)concentration,and channel length affected the length-based separation of *** *** lateral positions of *** cells with varying morphologies in a straight rectangular microchannel were found to be dependent on cell length under the co-flow of viscoelastic and Newtonian ***,we evaluated the ability of the viscoelastic microfluidic device to separate the two groups of *** cells by length(i.e.,1-5μm and>5μm)in terms of extraction purity(EP),extraction yield(EY),and enrichment factor(EF)and confirmed that the device could separate heterogeneous populations of bacteria using elasto-inertial effects.

Esophageal and gastric cancers are both common and deadly. Patients present most often after disease progression and survival is therefore poor. Due to demographic variability and recent changes in disease incidence, much emphasis has been placed on studying risk factors for both esophageal and gastric cancers. However, with increasing understanding of these diseases, low survival rates persist and continued intensive studies are necessary to optimize treatment plans. This review article discusses updates in the evolving epidemiology, clinical presentation, risk factors, and diagnostic and treatment modalities of esophageal and gastric cancers.

In terms of global cancer-related deaths,hepatocellular carcinoma (HCC) has the fourth highest mortality *** until 2017,treatment of advanced HCC was largely limited to sorafenib,an oral tyrosine kinase inhibitor,with little to no success in the development of alternative treatment ***,in the past two years,there has been an unprecedented increase in both the number and type of treatment options available for *** of 2019,the US FDA has approved four oral tyrosine kinase inhibitors,two immune checkpoint inhibitors,and one anti-angiogenesis antibody for the treatment of *** with this new variety,systemic treatment of advanced HCC remains largely unsatisfactory,and the median survival rate stands at approximately one *** expected breakthrough of using immune checkpoint inhibitors in advanced HCC did not materialize in *** use of immune checkpoint inhibitors in conjunction with oral tyrosine kinase inhibitors or antiangiogenesis medications is the current clinical research trend,the results of which are eagerly *** limited progress in survival,HCC research is currently experiencing a period of growth and innovation,and there is hope for significant advances in the treatment of advanced HCC as the field continues to develop.

Human ADAM19 is a recently identified member of the ADAM *** is highly expressed in human placentas,but its dynamic change and function at the human feto-maternal interface during placentation remain to be *** this present study,the spatial and temporal expression and cellular localization of ADAM19 in normal human placentas were first demonstrated,and the effects of ADAM19 on trophoblast cell adhesion and invasion were further investigated by using a human choriocarcinoma cell line(JEG-3) as an in vitro *** data demonstrated that ADAM19 was widely distributed in villous cytotrophoblast cells,syncytiotrophoblast cells,column trophoblasts,and villous capillary endothelial cells during early *** mRNA and protein level of ADAM19 in placentas was high at gestational weeks 8-9,but diminished significantly at mid-and term *** JEG-3 cells,the overexpression of ADAM19 led to diminished cell invasion,as well as increases in cell adhesiveness and the expression of E-cadherin,with no changes in β-catenin expression *** data indicate that ADAM19 may participate in the coordinated regulation of human trophoblast cell behaviors during the process of placentation.

The present study characterised the types and amounts of non-starch polysaccharides(NSP)remaining undigested along the gastrointestinal tract(GIT)of broiler chickens offered a typical wheat-or maizebased ***-day old Cobb 500 mixed-sex chicks were assigned to 24 pens,with 10 birds/pen and 12 pens/*** were offered the experimental diets in 3 phases(starter,day 0 to 10;grower,day 11 to 24 and finisher,day 25 to 35).Excreta and digesta samples from the crop,gizzard,duodenum,jejunum,ileum and caeca were collected at 12 and 35 days of age,and analysed for the NSP *** wheat-based diet contained higher levels of soluble NSP than the maize-based diet,whereas insoluble NSP levels were similar between the 2 *** analysis of NSP constituents revealed that arabinoxylans were the primary NSP in the wheat-based diet,mostly in insoluble *** were the predominant NSP in the maize-based diet,followed by ***,birds offered the wheatbased diet presented higher levels of soluble NSP remaining in all gut sections compared to birds offered the maize-based diet,at both 12 and 35 days of age(Pked differences in the amounts and types of NSP delivered to the different gut sections when feeding wheat-compared to maize-based diets,particularly in the gizzard and the lower GIT of birds.

This study was to characterise the undigested nutrients present along the gastrointestinal tract of birds offered common wheat-or maize-based diets,with the goal of optimising utilisation of enzymes to enhance digestive ***-and maize-based diets were offered to 240 mixed-sex broilers(10 birds/pen;n=12)from 1 to 35 d *** of dry matter,starch,crude protein and nonstarch polysaccharides(NSP)were measured in the crop,gizzard,duodenum,jejunum,ileum,caeca and excreta at d 12 and 35 *** of nutrient levels in the excreta presented that more than 30%of nutrients provided in the feed was wasted,irrespective of wheat or maize diet *** average,92 g/kg crude protein,92 g/kg insoluble NSP and 14 g/kg oligosaccharides were not utilised by birds at d 12 *** quantity of water-insoluble NSP in the small intestine at d 12 was lower in birds offered the wheat-based diet compared to those fed the maize-based diet(Pkg crude protein,79 g/kg insoluble NSP and 9 g/kg oligosaccharides remained in the excreta at 35 d of *** this time period,accumulation of feed in the gizzard was noted for birds offered both diets,but was more pronounced in those offered the maize-based diet(P<0.001).Birds offered the maize-based diet demonstrated improved utilisation of oligosaccharides compared to those fed the wheat-based diet at both d 12 and 35(P=0.087 and P=0.047,respectively).Protein utilisation in the jejunum and ileum was greater in birds offered the wheat-based diet compared to those fed the maize-based diet(P=0.004 and P<0.001,respectively).Thus,while both diets supported standard growth performance of birds,the degree and flow of nutrient disappearance along the gastrointestinal tract was influenced by cereal type and bird age.
