With the size of the biopharmaceutical market exponentially increasing,there is an aligned growth in the importance of data-rich analyses,not only to assess drug product safety but also to assist drug development driven by the deeper understanding of structure/function *** monoclonal antibodies,many functions are regulated by N-glycans present in the constant region of the heavy chains and their mechanisms of action are not completely *** importance of their function focuses analytical research efforts on the development of robust,accurate and fast methods to support drug development and quality *** N-glycan analysis is considered as the gold standard for glycosylation characterisation;however,it is not the only method for quantitative analysis of glycoform *** this study,ten different analytical workflows for N-glycan analysis were compared using four monoclonal *** observing good comparability between the quantitative results generated,it was possible to appreciate the advantages and disadvantages of each technique and to summarise all the observations to guide the choice of the most appropriate analytical workflow according to application and the desired depth of data generated.

BACKGROUND:emergency physicians(ePs)often care for patients with acute small bowel *** some patients require exploratory laparotomy,others are managed successfully with supportive *** aimed to determine features that predict the need for operative management in emergency department(eD)patients with small bowel obstruction(SBO).MeTHODS:We performed a retrospective chart review of 370 consecutive patients admitted to a large urban academic teaching hospital with a diagnosis of SBO over a two-year *** evaluated demographic characters(prior SBO,prior abdominal surgery,active malignancy)and clinical findings(leukocytosis and lactic acid)to determine features associated with the need for urgent operative ***:Patients with a prior SBO were less likely to undergo operative intervention[20.3%(42/207)]compared to those without a prior SBO[35.2%(57/162)].Abnormal bloodwork was not associated with need for operative intervention.68%of patients with CT scan findings of both an SBO and a hernia,however,were operatively ***:Patients with a history of SBO were less likely to require operative intervention at any point during their *** bloodwork was not associated with operative *** CT finding of a hernia,however,predicted the need for operative intervention,while other findings(ascites,duodenal thickening)did *** research would be helpful to construct a prediction rule,which could help community ePs determine which patients may benefit from expedited transfer for operative management,and which patients could be safely managed conservatively as an initial treatment strategy.

Complex role of matrix metalloproteinases in angiogenesis

Matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMP) play a significant role in regulating angiogenesis, the process of new blood vessel formation. Interstitial collagenase (MMP-1), 72 kDa gelatinase A/type IV collagenase (MMP-2), and 92 kDa gelatinase B/type IV collagenase (MMP-9) dissolve extracellular matrix (eCM) and may initiate and Promote angiogenesis. TIMP-1, TIMP-2, TIMP-3, and possibly,TIMP-4 inhibit neovascularisation. A new paradigm is emerging that matrilysin (MMP-7), MMP-9, and metalloelastase (MMP-12) may block angiogehesis by converting plasndnogen to angiostatin, which is one of the most potent angiogenesis antagonists. MMPs and TIMPs play a complex role in regulating angiogenesis. An understanding of the biochemical and cellular pathways and mechanisms of angiogenesis will provide importal information to allow the control of angiogenesis, e.g. the stimulation of angiogenesis for coronary collateral circulation formation; while the inhibition for treating arthritis and cancer.

OBJeCTIVe Over 30% of all new psychoactive substances identified by the UN Office on Drugs and Crime in 2016 were synthetic *** recent emergence of MAM-2201 on the illicit market is troubling because this drug has no precedent in either the scientific or patent literature,and appears to be a novel compound developed specifically as a "graymarket" drug of abuse bystructurally combining the known synthetic cannabinoids JWH-122 and *** is currently no published information regarding the pharmacology of *** The present studies characterized cannabinoid-like effects of MAM-2201 in vitro(interactions with cannabinoid type 1 receptors[CB1 Rs]) and in vivo(in mice and rats).ReSULTS In a radioligand binding assay using [3 H]CP55,940 in HeK cell membranes transfected with the CB1 R,MAM-2201(K i=5.4 nmol·L^(-1)),had higher binding affinity than WIN 55,212-2(K i=80 nmol·L^(-1)),and D9-THC(K i=8.3 nmol·L^(-1)).The e max values for MAM-2201 and WIN 55,212-2 in an assay of agonist inhibition of forskolin-stimulated c AMP were 85%(eC50=0.45 nmol·L^(-1)) and 95%,respectively,as compared with the D9-THC e max of 74%.In mice,MAM-2201(0.003-1.0 mg·kg^(-1),IP) produced dose-dependent cannabimimetic effects which were both more potent and more effective than those of ***-2201 and D9-THC dose-dependently produced hypothermia:eD50=0.287 and 25.4 mg·kg^(-1),analgesia:eD50=0.125 and 29.4 mg·kg^(-1),and catalepsy:eD50=0.301 and18.9 mg·kg^(-1) in adult male CD1 ***,MAM-2201 also elicited convulsant effects at a dose of 1.0 mg·kg^(-1) in 8/8 murine *** rats,MAM-2201 produced dose-dependent D9-THC-like interoceptive effects in subjects trained to discriminate 3.0 mg·kg^(-1)(IP) D9-THC from *** MAM-2201 binds CB1 Rs with high affinity and agonist efficacy,and functions as a potent cannabinoid agonist in vivo across several complementary measures of cannabinoid activity in two rodent species.

Assessing Fecal Contamination in Groundwater from the Tulum Region, Quintana Roo, Mexico

The Yucatan Peninsula’s groundwater is experiencing increases in degradation due to swelling population and tourism;yet little is known about sources and transport of contaminants in drinking water supplies. The karst allows for rapid transport of microbial and chemical contaminants to the subsurface, resulting in significantly increased potential for pollution of groundwater. The objective of this research is to determine the occurrence, source, and extent of fecal contamination in the Tulum region of the Peninsula. A multi-analytical approach was undertaken in impacted and unimpacted groundwater locations;measurements included physicochemical parameters, total coliform and e. coli, Bacteroides (human vs total) and caffeine. The results indicate a variation in geochemistry from impacted to protected sites. The total coliform and e. coli show fecal contamination is wide spread. However, the presence of human Bacteriodes and caffeine in the water in the Tulum well field indicates that the recent human activities next to the well field are impacting the drinking water supply. This project is an assessment of the area’s current water quality conditions and the probable impact that the aforementioned growth would have on the area’s water supply. By applying multiple source parameter measurements, including molecular microbiology and chemical indicators it was confirmed the extent of fecal contamination of human origin covered the entire sampling region.

Advanced treated municipal wastewater is an important alternative water source for agricultural ***,the possible persistence of chemical and microbiological contaminants in these waters raise potential safety concerns with regard to reusing treated wastewater for food crop *** low-cost and environmentally-friendly filter media,biochar(BC)and zero-valent iron(ZVI),have attracted great interest in terms of treating reused ***,we evaluated the efficacy of BC-,nanosilver-amended biochar-(Ag-BC)and ZVI-sand filters,in reducing contaminants of emerging concern(CeCs),escherichia coli(***)and total bacterial diversity from wastewater *** experiments were conducted with control quartz sand and sand columns containing BC,Ag-BC,ZVI,BC with ZVI,or Ag-BC with *** filtration,Ag-BC,ZVI,BC with ZVI and Ag-BC with ZVI demonstrated more than 90%(>1 log)removal of *** from wastewater samples,while BC,Ag-BC,BC with ZVI and Ag-BC with ZVI also demonstrated efficient removal of tested *** bacterial diversity was also observed after filtration;however,differences were marginally *** addition,significantly(per bacterial diversity was observed in wastewater samples collected during warmer versus colder *** of silver ions occurred from Ag-BC columns;however,this was prevented through the addition of *** conclusion,our data suggest that the BC with ZVI and Ag-BC with ZVI sand filters,which demonstrated more than 99%removal of both CeCs and *** without silver ion release,may be effective,low-cost options for decentralized treatment of reused wastewater.

Tubeufiaceae is based on the generic type Tubeufia,which is characterized by superficial,oval and bright ascomata,bitunicate asci,mostly long fusiform to filiform,transeptate ascospores and hyphomycetous asexual states with helicosporous *** species in this family are saprobic on terrestrial woody substrates and some are *** distinct morphology as well as combined LSU,SSU and TeF1 sequence analysis show that Tubeufiaceae should be accommodated in a new order Tubeufiales,which is introduced in this *** analyses of combined LSU and ITS sequences were used to resolve genera and species within the family *** this study,we examine and incorporate sexual and asexual states of genera in Tubeufiales to provide a modern treatment,based on single *** epitype for Tubeufia javanica,the type species of Tubeufia,is designated and represents Tubeufia sensu *** genera Acanthophiobolus,Acanthostigma,Boerlagiomyces,Chlamydotubeufia,Kamalomyces,Podonectria,Thaxteriella and Thaxteriellopsis are accepted,Acanthostigmina is reinstated,and the asexual genera Aquaphila,Helicoma,Helicomyces,Helicosporium and Tamhinispora are accepted in *** new genera Acanthohelicospora,Helicangiospora and Neoacanthostigma are *** genus Bifrontia is added to the family based on morphological *** incongruous morphological genera Acanthostigmella,Amphinectria,Chaetocrea,Chaetosphaerulina,Glaxoa,Malacaria,Melioliphila,Paranectriella,Puttemansia,Rebentischia and Uredinophila are excluded from Tubeufiaceae despite having characteristic ascomata with setae and multiseptate long spores.A key to genera accepted in Tubeufiaceae is provided.

Investigation on the epidemiology and Pathogenesis Characteristics of Staphylococcus pseudintermedius-Associated Veterinary Hospital Infections

A retrospective analysis of the diagnostic laboratory submissions from 2016 to 2020 was performed to assess the antibiotic resistance of Staphylococcus pseudintermedius and determine locations in the hospital which might be acting as environmental source(s). Previous studies have identified a significant increase in infections with S. pseudintermedius. Samples were taken from the hospital environment by swabbing areas in the intensive care unit and anesthesia preparation room and bacterial species identified. Isolates obtained from patients were then examined regarding the ability to form biofilm, an important phenotype on hospital-related infection. In addition, veterinary hospital associated strains of S. pseudintermedius were used to determine the bactericidal effect of the used disinfectant, applying the hospital current protocol, by comparing the efficacy against S. pseudintermediius and a strain of Staphylococcus aureus from a dog. The isolates identified were resistant to commonly used antibiotics such as enrofloxacin and cephalosporins, and 45% percent of those were methicillin resistant. The environmental survey in the hospital identified S. pseudintermedius in the pre-anesthesia area, although the isolate was killed by the current used disinfection protocol. A few disease associated bacteria were evaluated for biofilm formation in comparison to a dog isolate of Staphylococcus aureus, demonstrating strong ability to form biofilms.
