A universal vaccine is in high demand to address the uncertainties of antigenic drift and the reduced effectiveness of current influenza *** this study,a strategy called computationally optimized broadly reactive antigen(COBRA)was used to generate a consensus sequence of the hemagglutinin globular head portion(HA1)of influenza virus samples collected from 1918 to 2021 to trace evolutionary changes and incorporate them into the designed *** carrying different HA1regions were delivered into eukaryotic cells by Salmonella-mediated bactofection using a Semliki Forest virus RnA-dependent RnApolymerase(RdRp)-based eukaryotic expression system,*** protein expression was confirmed by Western blot and immunofluorescence *** immunized with the designed constructs produced a humoral response,with a significant increase in immunoglobulin G(IgG)levels,and a cell-mediated immune response,including a 1.5-fold increase in CD4^(+) and CD8^(+)T ***,constructs #1 and #5 increased the production of interferon-γ(IFn-γ)producing CD4^(+)and CD8^(+)T cells,skewing the response toward the T helper type 1 cell(Th1)***,interleukin-4(IL-4)-producing T cells were upregulated *** efficacy was demonstrated,with up to 4-fold higher production of neutralizing antibodies and a hemagglutination inhibition titer>40 against the selected viral *** designed constructs conferred a broadly protective immune response,resulting in a notable reduction in viral titer and minimal inflammation in the lungs of mice challenged with the influenza A/PR8/34,A/Brisbane/59/2007,A/California/07/2009,KBPV VR-92,and nCCP 43021 *** discovery revolutionizes influenza vaccine design and delivery;Salmonella-mediated COBRA-HA1 is a highly effective in vivo antigen presentation *** approach can effectively combat seasonal H1n1 influenza strains and potential pandemic outbreaks.

Memory loss and dementia are major public health concerns with a substantial economic *** stress has been shown to play a crucial role in the pathophysiology of hippocampal damage-induced memory *** investigate whether the antioxidant and anti-inflammatory compound vanillyla cetone(zingerone) can protect against hippocampal damage and memory loss induced by cadmium chloride(CdCl_(2)) administration in rats,we explo red the potential involvement of the nuclear factor erythroid 2-related factor 2(nrf2) signaling pathway,which is known to modulate oxidative stress and *** healt hy male Wistar rats were divided into five groups:vehicle-treated(control),vanillylacetone,CdCl_(2),vanillylacetone+ CdCl_(2),vanillylacetone+ CdCl_(2)+ brusatol(a selective pharmacological n rf2inhibitor) *** effectively attenuated CdCl_(2)-induced damage in the dental gyrus of the hippocampus and improved the memory function assessed by the Morris Water Maze ***,vanillylacetone markedly decreased the hippocampal tissue levels of inflammatory biomarkers(interleukin-6,tumor necrosis factor-α,intracellular cell adhesive molecules) and apoptosis biomarkers(Bax and cleaved caspase-3).The control and CdCl_(2)-treated groups treated with va nillylacetone showed reduced generation of reactive oxygen species,decreased malondialdehyde levels,and increased superoxide dismutase and glutathione activities,along with significant elevation of nuclear nrf2 mRnA and protein expression in hippocampal *** the protective effects of vanillylacetone we re substantially blocked by the co-administration of brusatol(a selective n rf2 inhibitor).Va nillylacetone mitigated hippocampal damage and memory loss induced by CdCl_(2),at least in part, by activating the nuclear transcription factor ***,vanillylacetone exerted its potent antioxidant and antiinflammatory actions.

A homogeneous layer of Bi_2O_3-Bi_(14)WO_(24) composite(BWO/Bi_2O_3) thin film was fabricated using a combination of electrodeposition and thermal treatment. The evenly distributed Bi14 WO24 component within the Bi_2O_3 layer was found to be important in stabilising the photoelectrochemical performances of Bi_2O_3 photoanode by promoting the photoelectron transport. The unmodified Bi_2O_3 suffered from severe photocorrosion as proven by X-ray diffraction(XRD) and inductively coupled plasma(ICP) analyses while the composite thin film was active without noticeable activity decay for at least 3 h of illumination. This strategy might be applicable to other photocatalysts with stability issues.

AIM： To observe the imbalance between T helper cell Th1 and Th2 cytokines in several chronic hepatitis disease at different stages of disease progression. METHODS： We measured the cytokine levels of Thl （IL-2 and IL-2R）, Th2 （IL-10） and the pro-inflammatory cytokines （IL-6 and IL-6R and TnF and TnF-RI and II） by the ELISA technique in the sera of 33 hepatocellular carcinoma （HCC） patients and 20 chronic liver disease （CLD） patients. In addition, 20 asymptomatic hepatitis C virus carriers and 20 healthy subjects negative for hepatitis C virus（HCV） markers served as controls. RESULTS： Anti-HCV antibodies were found to be positive in 94% of HCC cases and 75% of CLD cases. On the other hand, HCV viremia was detected using RT- PCR in 67% of HCC cases and 65% of CLD cases. HBsAg was positive in 9% of HCC cases and 30% of CLD cases. Also bilharziaI-Ab was positive in 55% of HCC cases, 65% of CLD cases and in 70% of asymptomatic carriers （ASC）. HCC patients had significantly higher values of IL- 2R, TnF-RII （P〈0.001）, and TnF-RI （P〉0.05）, but lower TnFα （P〈0.001） and IL-6 （P = 0.032） in comparison to ASC. But, in comparison to non-cancer controls, HCC patients had higher values of IL-2R, IL-6R, TnF-RI and TnF-RII, but lower TnF-α （P〈0.001）. CLD patients had higher IL-2R, TnF-RI, and TnF-RII （P〈0.001） than ASC. But, in comparison to non-cancer controls, CLD patients had higher values of IL-2R, TnF-RI and TnF-RII, but lower TnF-α （P〈0.001）. IL-10 was higher （though not significantly） in HCC and CLD patients than in symptomatic carriers and non-cancer controls. COnCLUSIOn： Liver disease progression from CLD to HCC due to HCV genotype-4 infection is associated with an imbalance between Th1 and Th2 cytokines. IL-2R, TnF-RI, and TnF-RII could be used as potential markers.

Limbal Stem Cell Deficiency(LSCD)is an eye disease that can cause corneal opacity and *** its advanced stage it can lead to a degree of visual *** involves the changing in the semispherical shape of the cornea to a drooping shape to downwards *** is hard to be diagnosed at early *** color and texture of the cornea surface can provide significant information about the cornea affected by *** such as shape and texture are very crucial to differentiate normal from LSCD *** several medical approaches exist,most of them requires complicated procedure and medical ***,in this paper,we pursued the development of a LSCD detection technique(LDT)utilizing image processing *** diagnosis of LSCD is very crucial for physicians to arrange for effective *** the proposed technique,we developed a method for LSCD detection utilizing frontal eye images.A dataset of 280 eye images of frontal and lateral LSCD and normal patients were used in this ***,the cornea region of both frontal and lateral images is segmented,and the geometric features are extracted through the automated active contour model and the spline *** the texture features are extracted using the feature selection *** experimental results exhibited that the combined features of the geometric and texture will exhibit accuracy of 95.95%,sensitivity of 97.91% and specificity of 94.05% with the random forest classifier of n=*** a result,this research developed a Limbal stem cell deficiency detection system utilizing features’fusion using image processing techniques for frontal and lateral digital images of the eyes.

BACKGROUnD Little is known about inflammatory bowel disease(IBD)burden and its impact on bone mineral density(BMD)am.ng adult patients in Saudi *** the best of our knowledge,our study is the only study to give an update about this health problem in adult Saudi patients with *** is a great risk factor for reduced BMD due to its associated chronic inflammation,malabsorption,weight loss and medication side ***,screening for reduced BMD am.ng patients with IBD is of utmost importance to curb and control anticipated morbidity and mortality am.ng those *** To assess the relationship between IBD and BMD in a sample of adult Saudi patients with *** ninety adult patients with IBD-62 Crohn’s disease(CD)and 28 ulcerative colitis(UC)-were recruited from King Fahad Specialist Hospital gastroenterology clinics in Buraidah,*** enrolled patients were interviewed for their demographic information and for IBD-and BMD-related clinical *** patients had the necessary laboratory markers and dual-energy x-ray absorptiometry scans to evaluate their BMD *** were divided into two groups(CD and UC)to explore their clinical characteristics and possible risk factors for reduced *** The CD group was significantly more prone to osteopenia and osteoporosis compared to the UC group;44%of the CD patients had normal BMD,19%had osteopenia,and 37%had osteoporosis,while 78%of the UC patients had normal BMD,7%had osteopenia,and 25%had osteoporosis(P valuen the CD group,the lowest t-score showed a statistically significant correlation with body mass index(BMI)(r=0.45,Pnd femur z-score(r=0.85,Pn the UC group,the lowest t-score showed only statistically significant correlation with the lumbar z-score(r=0.82,Pnd femur z-score(r=0.80,Pn the CD group with the best cut-off value at≤23.43(m/kg2);area

Solid state reaction method was used to prepare high temperature superconductor with a nominal composition Tl2-xHgxSr2Ca2Cu3O10+δ (0 ≤ x ≤ 1). The effect of quenching on the superconducting properties has been investigated by preparing two sets of samples at 1133 K for 3 hrs. The superconducting properties of the samples have been exam.ned by electrical measurements. It is found that a partial substitution for Tl by Hg increases the formation of high-Tc phase and increases its Tc. X-ray diffraction (XRD) pattern analyses have shown a tetragonal structure, and there are at least two superconducting phases.

Optimization of Chromate Reduction by Whole Cells of Arthrobactersp. SUK 1205 Isolated from Metalliferous Chromite Mine Environment

Arthrobacter sp. SUK 1205 isolated from metalliferous chromite mine environment of Orissa, India showed wide degree of tolerance to heavy metals including Cr(VI), variety of antibiotics and was also capable of reducing Cr(VI) during growth. Freshly grown whole cells of this bacterium were evaluated for chromate reduction under batch culture using Vogel Bonner (V. B.) broth as the base. Cells of SUK 1205 were capable of completely reducing 100 μM Cr(VI) in V. B. broth within 48 h of incubation. Reduction of chromate increased with increase in cell density which attained maximum at 1010 cells/ml, however, reverse was the phenomenon when the concentration of Cr(VI) increased gradually. Glycerol, glycine and glucose promoted chromate reduction efficiency of the cells when used as electron donors. Optimum pH and temperature were found to be 7.0 and 35°C respectively. The process of reduction was inhibited by ni(II), Mn(II), Zn(II) and Co(II), but Cu(II) and Fe(III) was promotive in nature. On the other hand, 2, 4-dinitrophenol was found to be neither promotive nor inhibitory for the reduction process, but carbonyl cyanide-m-chloro phenyl hydrazone, sodium azide, sodium fluoride and n,n,-dicyclohexyl carboiimide were inhibitory. Cells of SUK 1205 when permeabilized with toluene, triton X-100 and tween 80 showed an enhancement of the process and thereby indicated that reduction of Cr(VI) was mainly associated with soluble component of the cells. Arthrobacter sp. SUK 1205, therefore, showed great promise for use in Cr(VI) detoxification under a wide range of environmental conditions.
