Drought-responsive genes expressed predominantly in root tissues are enriched with homotypic cis-regulatory clusters in promoters of major cereal crops

The root appears to be the most relevant organ for breeding drought stress ***, our knowledge about temporal and spatial regulation of drought-associated genes in the root remains fragmented, especially in crop plants. We performed a meta-analysis of expression divergence of essential drought-inducible genes and analyzed their association with cis-elements in model crops and major cereal crops. Our analysis of42 selected drought-inducible genes revealed that these are expressed primarily in roots,followed by shoot, leaf, and inflorescence tissues, especially in wheat. Quantitative real-time RT-PCR analysis confirmed higher expression of TaDREB2 and TaAQP7 in roots,correlated with extensive rooting and drought-stress tolerance in wheat. A promoter scan up to 2 kb upstream of the translation start site using phylogenetic footprinting revealed708 transcription factor binding sites, including drought response elements(DREs), auxin response elements(Aux REs), MYCREs/MYBREs, ABAREs, and ERD1 in 19 selected ***, these elements were organized into clusters of overlapping transcription factor binding sites known as homotypic clusters(HCTs), which modulate drought physiology in plants. Taken together, these results revealed the expression preeminence of major drought-inducible genes in the root, suggesting its crucial role in drought adaptation. The occurrence of HCTs in drought-inducible genes highlights the putative evolutionary modifications of crop plants in developing drought adaptation. We propose that these DNA motifs can be used as molecular markers for breeding drought-resilient cultivars, particularly in the cereal crops.

Reabsorption of iron into acutely damaged rat liver:A role for ferritins

AIM To studied iron metabolism in liver, spleen, and serum after acute liver-damage, in relation to surrogate markers for liver-damage and *** Rats received intraperitoneal injection of the hepatotoxin thioacetamide(TAA), and were sacrificed regularly between 1 and 96 h thereafter. Serum levels of transaminases and iron were measured using conventional laboratory assays. Liver tissue was used for conventional histology, immunohistology, and iron staining. The expression of acute-phase cytokines, ferritin light chain(FTL), and ferritin heavy chain(FTH)was investigated in the liver by q RT-PCR. Western blotting was used to investigate FTL and FTH in liver tissue and serum. Liver and spleen tissue was also used to determine iron *** After a short initial decrease, iron serum concentrations increased in parallel with serum transaminase(aspartate aminotransferase and alanine aminotransferase) levels, which reached a maximum at 48 h, and decreased thereafter. Similarly, after 48 h a significant increase in FTL, and after 72 h in FTH was detected in serum. While earliest morphological signs of inflammation in liver were visible after 6 h, increased expression of the two acute-phase cytokines IFN-γ(1 h) and IL-1β(3 h) was detectable earlier, with maximum values after 12-24 h. Iron concentrations in liver tissue increased steadily between 1 h and 48 h, and remained high at 96 h. In contrast, spleen iron concentrations remained unchanged until 48 h, and increased mildly thereafter(96 h). Although tissue iron staining was negative, hepatic FTL and FTH protein levels were strongly elevated. Our results reveal effects on hepatic iron concentrations after direct liver injury by TAA. The increase of liver iron concentrations may be due to the uptake of a significant proportion of the metal by healthy hepatocytes, and only to a minor extent by macrophages, as spleen iron concentrations do not increase in parallel. Three new alkyl substituted anthraquinone derivatives, trivially named as symploquinones A-C(Compounds 1-3) were isolated from Symplocos racemosa. The structures of these compounds were determined on the basis of extensive spectroscopic analyses(UV, IR, Mass, ~1 H-and ^(13)C-NMR, and two-dimensional(2D) NMR techniques). The resulting data were also compared with the reported literature. These compounds were then subjected to antibacterial or antibiofilm testing. Compounds 1 and 3 exhibited good antibacterial activity in the concentration range of 160-83 μg·m L^(-1) against Streptococcus mutans, methicillin resistant Staphylococcus aureus and Proteus mirabilis. Both compounds were further screened for anti-biofilm activity, which revealed promising activities at sub-MIC concentrations. None of the compounds were found to be active against Klebsiella pneumoniae.

Optimizing Yield and Quality of Canola Cultivars Using Various Potash Levels

The response of several canola cultivars to different potassium (K) levels was investigated in terms of various agronomic parameters including yields, oil and protein contents in a trial conducted at The University of Agriculture, Peshawar-Pakistan in Rabi 2010-11. Three cultivars including Bulbul-98, Abaseen-95 and Dure-NIFA, with five levels of potassium i.e. 0, 30, 60, 90 and 120 kg·ha-1 were used in Randomized Complete Block (RCB) design with four replications and factorial arrangement. A larger number of days to 50% flowering (116.6 days), plant height (203.8 cm), leaf area index (4.4), 1000 grain weight (3.5 g), biological yield (13189.3 kg·ha-1), grain yield (1799.2 kg·ha-1) and harvest index (13.9%) were obtained in K applied plots than the plots where K was not applied. The highest oil (45.1%) and protein (27.7%) was obtained in plots where K was applied at the rate of 120 and 90 kg·ha-1, respectively. Among cultivars, Bulbul-98 and Abaseen-95 had higher seed yield and oil percentage. Bulbul-98 had more protein content than Abaseen-95 and Durre-NIFA. On average, cultivars gave higher and at par grain yield at 60, 90 and 120 kg·ha-1. However, they produced higher and at par oil and protein percentage at 90 and 120 kg·ha-1. Bul-bul-98 and Abaseen-95 are recommended for higher grain yield and oil content for general culti-vation in Peshawar valley. However, for higher protein content, Bulbul-98 is better. Potassium applied at 60 kg·ha-1 is recommended for higher grain yield, however, for higher oil and protein content, K at 90 kg·ha-1 is recommended.

Non-destructive plant growth parameters measurement is an important concern in automatic-seedling ***,several image-basedmonitoring approaches have been proposed and potentially developed for several agricultural *** presented study proposed and developed a RealSense-based machine vision system for the close-shot seedling-lump integrated *** strategy was based on the close-shot depth ***,the point cloud clustering and suitable algorithms were applied to obtain the segmentation of 3D seedling *** addition,the data processing pipeline was developed to assess the differentmorphological parameter of 4 different seedling *** experiments were carried out with 4 different seedling varieties(pepper,tomato,cucumber,and lettuce)and trained under different light conditions(light and dark).Moreover,analysis results showed that therewas not significantly different(p<0.05)found towards light and dark environments due to close-shot near-infrared ***,the results revealed that the stem diameter relationship between RealSense and the manual method was found for R^2=0.68 cucumber,R^2=0.54 tomato,R^2=0.35 pepper,and R^2=0.58 lettuce ***,the seedling height relationship between RealSense and the manual methodwas found higher than R^2=0.99,0.99,0.99,and 0.99 for pepper,tomato,cucumber,and lettuce,*** on the experiment results,it was concluded that the RGB-D integrated monitoring system with the purposed method could be practiced for nursery seedlings most promisingly without high labour requirements in terms of ease of *** system revealed a good sturdiness and relevance for plant growth ***,it has the perspective for future practical value to real-time vision servo operations for transplanting robots.

The potential of the high resolution nuclear track detector (NTD) CR-39 is examined carefully for the measurement of relativistic nuclear projectile fragmentation cross sections and studies of related processes using the experience of many years of such measurements. The charge resolution and the charge resolving power of CR-39 detectors for the measurements of 158 A GeV 207Pb projectiles and their fragments are presented. Exposures of target-detector stacks, the chemical etching procedure and the nuclear track measurements are described in detail discussing precautions and possible errors. The procedures discussed are also valid for other NTDs. A comparison with electronic active detectors is also made considering important detection and measurement aspects. An experimental design proposing the co-use of NTDs with in-use active detectors is described.
