Binary vector Construction for Site-Directed Mutagenesis of <i>Kafirin</i>Genes in Sorghum

Sorghum (Sorghum bicolor (L.) Moench) is one of the world’s leading cereal crops in agricultural production, which has a special importance in the arid regions. However, unlike other cereals, sorghum grain has a lower nutritional value, which is caused, inter alia, by the resistance of its seed storage proteins (kafirins) to protease digestion. One of the effective approaches to improve the nutritional value of sorghum grain is to obtain mutants with partially or completely suppressed synthesis or altered amino acid composition of kafirins. The employment of genome editing may allow to solve this problem by introducing mutations into the nucleotide sequences of the α- and γ-kafirin genes. In this study, genomic target motifs (23 bp sequences) were selected for the introduction of mutations into the α- and γ-KAFIRIN genes of sorghum. The design of the gRNAs was conducted using the online tools CRISPROR and CHOPCHOP. Two most suitable targets were chosen for α-KAFIRIN (k1C5) and two for γ-KAFIRIN (gKAF1) genes. The insertion of respective sequences in the generic vector pSH121 was performed at the BsaI (Eco31I) sites. validation of the cloning procedure was performed by DNA sequencing. Subcloning of the resulting constructs was performed using the SfiI restriction sites into the compatible binary vector B479p7oUZm-LH. The correct assembly of binary vectors was confirmed by restriction analysis using the MluI and SfiI cleavage sites. The four vectors created (1C - 4C) were transferred by electroporation into the Agrobacterium tumefaciens strain AGL0. Currently, this vector series is used for stable

The Information Security of Digital Control Systems for Nuclear Power Plants

The NPP （nuclear power plant） being vital objects of an energy infrastructure must be protected against malicious actions affecting their safety, and cyber security plays a key part in attaining this goal. The paper considers, implemented by the authors within the project of advanced digital control system for NPP with the reactor vvER-1000, a system of unauthorized access protection, partially built up on the technology of AA （active audit） and expert system. The AA technology is based on response of the system on deviation of current signature of the automated process control system from stable state rather than on a certain signature of attack and relies on the estimation of the behavioral models of the particular digital control system. The advent of active audit reflects the current situation in the digital control systems where complex distributed platforms are used to construct automated process control system. The active audit allows one to make the digital control system functionally closed, provided that it is determinate. The methodology of the active audit does not give u external （barrier） and traditional （password, antivirus） methods of unauthorized access protection. These methods can be used when it is appropriate to achieve a required protection level.

Polymorphisms in the SNCA Gene: Association with the Risk of Development of the Sporadic Form of Parkinson’s Disease and the Level of SNCA Gene Expression in Peripheral Blood of Patients from Russia

Parkinson’s disease (PD) is one of the most common human neurodegenerative disorders caused by the loss of dopaminergic neurons in the brain. The α-synuclein (SNCA) gene is one of the most studied genes involved in the pathogenesis of PD. In our study, we conducted a genetic analysis of promoter and intron single-nucleotide polymorphisms (SNPs) in the SNCA gene. We also analyzed the association of genotypes of these SNPs with expression levels of SNCA mRNA. One of four SNPs in the SNCA gene, and the rs2736990 polymorphism, associates with the risk of the sporadic form of PD in Russian population. The risk of PD was increased almost twofold in carriers of allele C (odds ratios = 1.9, 95% confidence interval: 1.2-2.91, p = 0.003). However, no association was found between any of the genotypes of SNPs tested (rs2583988, rs2619363, rs2619364 and rs2736990) and alterations in SNCA levels. Our findings support the hypothesis that the rs2736990 polymorphism is associated with PD. SNPs rs2583988, rs2619363 and rs2619364 in the promoter region of the SNCA gene themselves do not significantly influence the expression of SNCA. Most likely, SNCA gene expression is a very complex process that is affected by different genetic and epigenetic factors.

Assembly of Oligoglycine Layers on Mica Surface

Assembly of [Gly7-NHCH2-]4C, [Gly7-NHCH2-]3C CH3 and [Gly4NH(CH2)5-]2 peptides on mica surface in aqueous so-lution was studied. The peptides are capable of forming atomically smooth (2.65-4.3 nm in height) layers assembled as polyglycine II. Monomers in the layers are situated normally to the surface. Formation of analogous flat 2D structures also takes place in solution but much more slowly than on mica surface, i.e. negatively charged surface plays an active role promoting the assembly.

Molecular Dynamics Simulations of Perovskites: The Effect of Potential Function Representation on Equilibrium Structural Properties

The perovskites with general formula ABX3 have been widely used as for materials with their unique properties (ferroelectric, piezoelectric, dielectric, catalytic and so on). Hybrid organolead halide perovskites are a class of semiconductors with ABX3 (X = Cl, Br, and I) structures consisting of lead cations in 6-fold coordination (B site), surrounded by an octahedron of halide anions (X site, face centered) together with the organic components in 12-fold cub octahedral coordination. These hybrid perovskites have a direct band gap, a large absorption coefficient as well as high charge carrier mobility that represent a very attractive characteristic of cost-effective solar cells. Basically, these crystals are inorganic solids of CaTiO3 type held together by bonds that are either ionic or partially ionic and partially covalent. In spite of the partially covalent character of the Ti-O bond, the system is modeled by a two-body central force interatomic potential (the form of the vashishta and Rahman interatomic potential), which has been used successfully for many materials with a perovskite structure. In the present work using molecular dynamics (MD) simulation method we investigate the dynamical and structural behavior of CaTiO3 perovskite at normal pressure and temperature conditions. The MD calculations were performed on a system of 16,000 particles (3200Ca + 3200Ti + 96,00O), initially in an orthorhombic-Pbnm structure. The orthorhombic MD box had edges Lx = 53.4 Å, Ly = 53.4 Å and Lz = 61.12 Å, which provided a density matching the experimental value of ρ = 4 g/cm3. Starting with this structure and using proposed interatomic potentials the MD system stabilizes at room temperature in its initial configuration.
