Assimilation of GOES-R Geostationary Lightning Mapper Flash Extent Density Data in GSI 3DVar, EnKF, and Hybrid En3DVar for the Analysis and Short-Term Forecast of a Supercell Storm Case

Capabilities to assimilate Geostationary Operational Environmental Satellite “R-series ”(GOES-R) Geostationary Lightning Mapper(GLM) flash extent density(FED) data within the operational Gridpoint Statistical Interpolation ensemble Kalman filter(GSI-EnKF) framework were previously developed and tested with a mesoscale convective system(MCS) case. In this study, such capabilities are further developed to assimilate GOES GLM FED data within the GSI ensemble-variational(EnVar) hybrid data assimilation(DA) framework. The results of assimilating the GLM FED data using 3DVar, and pure En3DVar(PEn3DVar, using 100% ensemble covariance and no static covariance) are compared with those of EnKF/DfEnKF for a supercell storm case. The focus of this study is to validate the correctness and evaluate the performance of the new implementation rather than comparing the performance of FED DA among different DA schemes. Only the results of 3DVar and pEn3DVar are examined and compared with EnKF/DfEnKF. Assimilation of a single FED observation shows that the magnitude and horizontal extent of the analysis increments from PEn3DVar are generally larger than from EnKF, which is mainly caused by using different localization strategies in EnFK/DfEnKF and PEn3DVar as well as the integration limits of the graupel mass in the observation operator. Overall, the forecast performance of PEn3DVar is comparable to EnKF/DfEnKF, suggesting correct implementation.

A Novel Approach to Measuring Spatiotemporal Changes in Social Vulnerability at the Local Level in Portugal

Combining infrared and mode synthesizing atomic force microscopy： Application to the study of lipid vesicles inside Streptomyces bacteria

We propose a new analytical approach combining vibrational spectroscopy and acoustic tomography for the detection and characterization of vesicles inside Streptomyces bacteria. Using atomic force microscopy and infrared spectroscopy （AFM-IR）, we detect the presence of triglyceride vesicles. Their sizes in depth are measured with high accuracy using mode synthesizing atomic force microscopy （MS-AFM）. We conducted a comparative study of AFM-IR and MS-AFM, and highlighted the advantages of the coupling of these techniques in having a full characterization （chemical, topographical, and volumetric） of a biological sample. With these complementary techniques, a complete access to the vesicle size distribution has been achieved with an accuracy of less than 50 nm. A 3D reconstruction of bacteria showing the in-depth distribution of vesicles is given to underline the great potential of the acoustic method.

Biochar Increases Diuron Sorption and Reduces the Potential Contamination of Subsurface Water with Diuron in a Sandy Soil

The herbicide diuron is widely used in agricultural areas in Brazil, whereas it has high potential for subsurface water contamination due to its physicochemical characteristics. Recent studies have demonstrated the potential of biochar as a sorbent and possible pesticide leaching mitigation. The objective of this study was to investigate the long-term effect of biochar application on the kinetics of sorption and desorption of diuron in a Cerrado Haplic Plinthosol. Samples were collected in an experiment conducted in the field in a randomized block design consisting of the combination of two levels of fertilizer application (0 and 300 kg ha −1 of 5-25-15 formula of NPK fertilizers) and three doses of biochar (0, 16, and 32 Mg ha −1 ). The Freundlich isotherm accurately described the sorption of diuron in all treatments. Biochar application increased the sorption and reduced the desorption of diuron. This effect was attributed to the contribution of biochar to total organic carbon (C) and C in the humin fraction and to the increase in the reactivity of the humic acid and humin fractions, which was significantly highly correlated with the sorption coefficient ( K f ). A positive correlation between the partition coefficient of organic C and K f confirmed the importance of the soil organic compartment for the sorption of diuron. The higher diuron sorption and lower diuron desorption capacities of sandy soils after biochar application could reduce the potential risk of diuron leaching and contamination of subsurface water.

Neuronal protein-tyrosine phosphatase 1B hinders sensory-motor functional recovery and causes affective disorders in two different focal ischemic stroke models

Ischemic brain injury causes neuronal death and *** activates protein-tyrosine phosphatase 1B(PTP1B).Here,we tested the significance of PTP1B activation in glutamatergic projection neurons on functional recovery in two models of stroke:by photothrombosis,focal ischemic lesions were induced in the sensorimotor cortex(SM stroke)or in the peri-prefrontal cortex(peri-PFC stroke).Elevated PTP1B expression was detected at 4 days and up to 6 weeks after *** ablation of PTP1B in neurons of neuronal knockout(NKO)mice had no effect on the volume or resorption of ischemic lesions,markedly different effects on functional recovery were *** stroke caused severe sensory and motor deficits(adhesive removal test)in wild type and NKO mice at 4 days,but NKO mice showed drastically improved sensory and motor functional recovery at 8 *** addition,peri-PFC stroke caused anxiety-like behaviors(elevated plus maze and open field tests),and depression-like behaviors(forced swimming and tail suspension tests)in wild type mice 9 and 28 days after stroke,respectively,with minimal effect on sensory and motor ***-PFC stroke-induced affective disorders were associated with fewer active(FosB+)neurons in the PFC and nucleus accumbens but more FosB+neurons in the basolateral amygdala,compared to sham-operated *** contrast,mice with neuronal ablation of PTP1B were protected from anxiety-like and depression-like behaviors and showed no change in FosB+neurons after peri-PFC *** together,our study identifies neuronal PTP1B as a key component that hinders sensory and motor functional recovery and also contributes to the development of anxiety-like and depression-like behaviors after ***,PTP1B may represent a novel therapeutic target to improve stroke *** procedures for animal use were approved by the Animal Care and Use Committee of the University of Ottawa Animal Care and Veterinary Service(protocol 1806)on July 27,2018.

Epidemiology of functional gastrointestinal disorders in children and adolescents: A systematic review

To assess the prevalence of functional gastrointestinal disorders (FGIDs) in children and ***, EMBASE, and Scopus databases were searched for original articles from inception to September 2016. The literature search was made in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) recommendations. For inclusion, each study had to report epidemiological data on FGIDs in children between 4 and 18 years old and contain standardized outcome based on Rome II, III or IV criteria. The overall quality of included epidemiological studies was evaluated in accordance with Loney’s proposal for prevalence studies of health literature. Two reviewers assessed each study for data inclusion and extraction. Discrepancies were reconciled through discussion with *** total of 659 articles were identified from the databases and 16 through manual search. A total of 43 articles fulfilled the eligibility criteria for full-text reading, with 26 remaining to be included in the final analysis. All studies were written in English and published between 2005 and 2016. Eight (30.8%) articles were performed in North America, five (19.2%) in Latin America, five (19.2%) in Europe, seven (27%) in Asia, and one (3.8%) in Africa. Sample size varied between 114 and 99416 subjects, totaling 132600 individuals. Fourteen (53.9%) studies recruited their target samples from schools, 11 (42.3%) from healthcare settings and the remaining one (3.8%) from online panel community. The overall FGID prevalence rates for student samples ranged from 9.9% to 29% to as high as 87% in clinical samples. Cyclic vomiting, irritable bowel syndrome and functional constipation were the most researched conditions, with a prevalence ranging from 0.2% to 6.2%, 0% to 45.1% and 0.5% to 86.9%, respectively. We report lanthanide-based micelles integrating hypericin （Hyp） for X-ray- triggered photodynamic therapy （PDT）. The lanthanide luminescence induced by X-ray irradiation excites the photosensitizer, which leads to the generation of singlet oxygen. This versatile approach can be extended to other photosensitizers or other types of liponanoparticles and can allow for magnetic resonance imaging （MRI） guidance.

Dabrafenib,an inhibitor of RIP3 kinase-dependent necroptosis,reduces ischemic brain injury

Ischemic brain injury triggers neuronal cell death by apoptosis via caspase activation and by necroptosis through activation of the receptor-interacting protein kinases （RIPK） associated with the tumor necrosis factor-alpha （TNF-a）/death receptor. Recent evidence shows RIPK inhibitors are neuroprotective and al- leviate ischemic brain injury in a number of animal models, however, most have not yet undergone clinical trials and safety in humans remains in question. Dabrafenib, originally identified as a B-raf inhibitor that is currently used to treat melanoma, was later revealed to be a potent RIPK3 inhibitor at micromolar con- centrations. Here, we investigated whether Dabrafenib would show a similar neuroprotective effect in mice subjected to ischemic brain injury by photothrombosis. Dabrafenib administered intraperitoneally at 10 mg/ kg one hour after photothrombosis-induced focal ischemic injury significantly reduced infarct lesion size in C57BL6 mice the following day, accompanied by a markedly attenuated upregulation of TNF-u. However, subsequent lower doses （5 mg/kg/day） failed to sustain this neuroprotective effect after 4 days. Dabrafenib bl ocked lipopolysaccharides-induced activation of TNF-ct in bone marrow-derived macrophages, suggesting that Dabrafenib may attenuate TNF-ct-induced necroptotic pathway after ischemic brain injury. Since Dab- rafenib is already in clinical use for the treatment of melanoma, it might be repurposed for stroke therapy.

Arabidopsis MSI1 Is Required for Negative Regulation of the Response to Drought Stress

Arabidopsis MSI1 has fundamental functions in plant development. MSI1 is a subunit of Polycomb group protein complexes and Chromatin assembly factor 1, and it interacts with the Retinoblastoma-related protein 1. Altered levels of MSI1 result in pleiotropic phenotypes, reflecting the complexity of MSI1 protein functions. In order to uncover additional functions of MSI1, we performed transcriptional profiling of wild-type and plants with highly reduced MSI1 levels （msil-cs）. Surprisingly, the known functions of MSI1 could only account for a minor part of the transcriptional changes in msil-cs plants. One of the most striking unexpected observations was the up-regulation of a subset of ABA-responsive genes eliciting the response to drought and salt stress. We report that MSI1 can bind to the chromatin of the drought-inducible downstream target RD20 and suggest a new role for MSI1 in the negative regulation of the Arabidopsis drought-stress response.
