The olfactory system of Pieris brassicae caterpllars:from receptors to glomeruli

Tolfactory system of adult lepidopterans is among the best described neuronal circuits.However,comparatively little is known about the organization of the olfactory system in the larval stage of these insects.Here,we explore the expression of olfactory receptors and the organization of olfactory sensory neurons in caterpillars of Pieris brassicae,a significant pest species in Europe and a well-studied species for its chemical ecology.To describe the larval olfactory system in this species,we first analyzed the head transcriptome of third-instar larvae(L3)and identified 16 odorant receptors(ORs)including the OR coreceptor(Orco),13 ionotropic receptors(IRs),and 8 gustatory receptors(GRs).We then quantified the expression of these 16 ORs in different life stages,using qPCR,and found that the majority of ORs had significantly higher expression in the L4 stage than in the L3 and L5 stages,indicating that the larval olfactory system is not static throughout caterpillar development.Using an Orco-specific antibody,we identified all olfactory receptor neurons(ORNs)expressing the Orco protein in L3,L4,and L5 caterpillars and found a total of 34 Orco-positive ORNs,distributed among three sensilla on the antenna.The number of Orco-positive ORNs did not differ among the three larval instars.Finally,we used retrograde axon tracing of the antennal nerve and identified a mean of 15 glomeruli in the larval antennal center(LAC),suggesting that the caterpillar olfactory system follows a similar design as the adult olfactory system,although with a lower numerical redundancy.Taken together,our results provide a detailed analysis of the larval olfactory neurons in P brassicae,highlighting both the differences as well as the commonalities with the adult olfactory system.These findings contribute to a better understanding of the development of the olfactory system in insects and its life-stage-specific adaptations.

Enhanced osteoclastogenesis and osteoclast activity contribute to the development of osteoporosis,which is characterized by increased bone resorption and inadequate bone formation.As novel antiosteoporotic therapeutics are needed,understanding the genetic regulation of human osteoclastogenesis could help identify potential treatment targets.This study aimed to provide an overview of transcriptional reprogramming during human osteoclast differentiation.Osteoclasts were differentiated from CD14+monocytes from eight female donors.RNA sequencing during differentiation revealed 8980 differentially expressed genes grouped into eight temporal patterns conserved across donors.These patterns revealed distinct molecular functions associated with postmenopausal osteoporosis susceptibility genes based on RNA from iliac crest biopsies and bone mineral density SNPs.Network analyses revealed mutual dependencies between temporal expression patterns and provided insight into subtype-specific transcriptional networks.The donor-specific expression patterns revealed genes at the monocyte stage,such as filamin B(FLNB)and oxidized low-density lipoprotein receptor 1(OLR1,encoding LOX-1),that are predictive of the resorptive activity of mature osteoclasts.The expression of differentially expressed G-protein coupled receptors was strong during osteoclast differentiation,and these receptors are associated with bone mineral density SNPs,suggesting that they play a pivotal role in osteoclast differentiation and activity.The regulatory effects of three differentially expressed G-protein coupled receptors were exemplified by in vitro pharmacological modulation of complement 5 A receptor 1(C5AR1),somatostatin receptor 2(SSTR2),and free fatty acid receptor 4(FFAR4/GPR120).Activating C5AR1 enhanced osteoclast formation,while activating SSTR2 decreased the resorptive activity of mature osteoclasts,and activating FFAR4 decreased both the number and resorptive activity of mature osteoclasts.In conclusion,we report the occurrence of transcriptional

Interest in metal nanoparticles as nanoscale drug-carrying vehicles has been increasing.In such an aspect,lanthanide metal nanoparticles are particularly intriguing.Herein,we report praseodymium nanoparticles as a novel anticancer therapeutic transporting agent.The material was made as a composite with the supramolecular host polymer,poly-β-cyclodextrin.The praseodymium nanorods were characterized using standard analytical techniques,including X-ray diffraction（XRD）,X-ray photoelectron spectroscopy（XPS）,and thermogravimetric analysis（TGA）.In addition,they were imaged using transmission electron microscopy.The nanostructures are a long rod-shaped material with 2.0±0.1 μm in length and 230±10 nm in width.The material shows a saturation magnetization value above 30 emu/g with a coercivity value of 130 Oe.The nanocarrier is freighted with the drug 5-fluorouracil,and the encapsulation efficiency is 76.09%.The drug release is continuous from the nanocarrier（polymer-containing praseodymium nanoparticles） and proceeds for above 250 h.The in vitro cytotoxicities of the blank nanocarrier and the 5-fluorouracil-loaded one are deduced by testing them on mCF-7 and Vero（non-cancerous） cell lines.The cytotoxicity on the cancerous cell lines is observed,whereas the noncancerous cells are unaffected.The IC50values on the cell lines are reported.

Effects of site preparation methods on soil physical properties and outplanting success of coniferous seedlings in boreal forests

This study assessed the effect of patch scarification and mounding on the physical properties of the root layer and the success of tree planting in various types of forests.This study was conducted on 12 forest sites in taiga forests of the European part of Russia.A total of 54 plots were set up to assess seedling survival;root collar diameter,height,and heigh increment were measured for 240 seedlings to assess growth.In the rooting layer,240 soil samples were taken to determine physical properties.The study showed that soil treatment methods had no effect on bulk density and total porosity in Cladina sites.However,reduced soil moisture was noted,particularly in mounds,resulting in increased aeration.In myrtillus sites,there were increased bulk density,reduced soil moisture,and total porosity in the mounds.mounding treatment in Polytrichum sites resulted in reduced soil moisture and increased aeration porosity.In the myrtillus and Polytrichum sites,patch scarification had no effects on physical properties.In Polytrichum sites,survival rates,heights,and heigh increments of bareroot Norway spruce seedlings in mounds were higher than in patches;however,the same did not apply to diameter.In Cladina and myrtillus sites,there was no difference in growth for bareroot and containerised seedlings with different soil treatments.Growing conditions and soil types should be considered when applying different soil treatment methods to ensure high survival rates and successful seedling growth.

A Statistical Study of magnetic Flux Emergence in Solar Active Regions Prior to Strongest Flares

Using the data on magnetic field maps and continuum intensity for Solar Cycles 23 and 24,we explored 100 active regions(ARs)that produced m5.0 or stronger flares.We focus on the presence/absence of the emergence of magnetic flux in these ARs 2-3 days before the strong flare onset.We found that 29 ARs in the sample emerged monotonically amidst quiet-Sun.A major emergence of a new magnetic flux within a pre-existing AR yielding the formation of a complex flare-productive configuration was observed in another 24 cases.For 30 ARs,an insignificant(in terms of the total magnetic flux of pre-existing AR)emergence of a new magnetic flux within the pre-existing magnetic configuration was observed;for some of them the emergence resulted in a formation of a configuration with a small δ-sunspot;11 out of 100 ARs exhibited no signatures of magnetic flux emergence during the entire interval of observation.In six cases the emergence was in progress when the AR appeared on the Eastern limb,so that the classification and timing of emergence were not possible.We conclude that the recent flux emergence is not a necessary and/or sufficient condition for strong flaring of an AR.The flux emergence rate of flare-productive ARs analyzed here was compared with that of flare-quiet ARs analyzed in our previous studies.We revealed that the flare-productive ARs tend to display faster emergence than the flare-quiet ones do.

BACKGROUND Hallux valgus(HV)is a common foot deformity that manifests with increasing age,especially in women.The associated foot pain causes impaired gait and decreases quality of life.moderate and severe HV is a deformity that is charac-terized by the involvement of lesser rays and requires complex surgical treatment.In this study,we attempted to develop a procedure for this condition.AIm To analyse the treatment results of patients who underwent simultaneous surgical correction of all parts of a static forefoot deformity.mETHODS We conducted a prospective clinical trial between 2016 and 2021 in which 30 feet with moderate or severe HV associated with Tailor’s bunion and metatarsalgia were surgically treated via a new method involving surgical correction of all associated problems.This method included a modified Lapidus procedure,m2m3 tarsometatarsal arthrodesis,intermetatarsal fusion of the m4 and m5 bases,and the use of an original external fixation apparatus to enhance correction power.Preoperative,postoperative,and final follow-up radiographic data and American Orthopaedic Foot and Ankle Society(AOFAS)scores were compared,and P valuesmales(93.3%)and 2 males feet(6.7%),20(66.7%)of whom had a moderate degree of HV and 10(33.3%)of whom had severe deformity.m2 and m3 metatarsalgia was observed in 21 feet,and 9 feet experienced pain only at m2.The mean follow-up duration was 11 months.All patients had good correction of the HV angle[preoperative median,36.5 degrees,interquartile range(IQR):30-45;postoperative median,10 degrees,IQR:8.8-10;follow-up median,11.5 degrees,IQR:10-14;Pmetatarsalgia was resolved in most patients(30 vs 5).There was a clinically negligible decrease in the corrected angles at the final follow-up,and the overall AOFAS score was significantly better(median,65 points,IQR:53.8-70;vs 80 points,IQR:75-85;Pmethod showed good sustainability of correction po

multi-wavelength-band transmission technology based on the exploitation of the extended spectral region is considered as a potential approach to increase the transmission capacity in the deployed fiber-optic communication infrastructure.The development of optical amplifiers operating in the O-,E-,S-,and U-telecom bands is an extremely important challenge for the successful implementation of this technology.Bismuth-doped fibers are of increasing interest as gain materials,which can be used to provide broadband amplification in the mentioned telecom bands.This is due to the ability of Bi ions incorporated into glass network to form bismuth active centers(BACs)with specific optical properties,which are primarily determined by the glass modifiers.In this work,the impact of the doping profiles of both Ge atoms as glass modifiers and Bi ions on the BACs formation is studied using a series of bismuth-doped fibers fabricated by the modified chemical vapor deposition(mCVD)technique.The Bi-to-BACs conversion efficiency in various spatial regions of the studied samples is presented.It is turned out that for high-Bi concentration regions,the conversion efficiency is very low(less than 10%).In addition,the relationship of the conversion efficiency to the distribution of Bi ions and/or Ge atoms is discussed.Finally,a continuous-wave laser at 1.46μm with a record slope efficiency of 80%is demonstrated using a Bi-doped fiber with confined doping profile,where the Bi-to-BACs conversion efficiency is 35%.This paper provides new information which might help to facilitate understanding of the features of Bi-doped fibers and their potentially achievable characteristics.

microglia are present throughout the central nervous system and are vital in neural repair,nutrition,phagocytosis,immunological regulation,and maintaining neuronal function.In a healthy spinal cord,microglia are accountable for immune surveillance,however,when a spinal cord injury occurs,the microenvironment drastically changes,leading to glial scars and failed axonal regeneration.In this context,microglia vary their gene and protein expression during activation,and proliferation in reaction to the injury,influencing injury responses both favorably and unfavorably.A dynamic and multifaceted injury response is mediated by microglia,which interact directly with neurons,astrocytes,oligodendrocytes,and neural stem/progenitor cells.Despite a clear understanding of their essential nature and origin,the mechanisms of action and new functions of microglia in spinal cord injury require extensive research.This review summarizes current studies on microglial genesis,physiological function,and pathological state,highlights their crucial roles in spinal cord injury,and proposes microglia as a therapeutic target.

Gastric cancer(GC) remains one of the most common lethal diseases. Every year, about 1 million people fall ill and almost 800 thousand people die worldwide~1. Surgical treatment has reached its limit. The current advances in immunology and detection of epidermal growth factor molecules(EGFR) in gastric cancer cells allowed to improve the treatment results;however, the metastatic form of this disease is still incurable.
