Dissecting the effect of soil on plant phenology and berry transcriptional plasticity in two Italian grapevine varieties (Vitis vinifera L.).

Grapevine embodies a fascinating species as regards phenotypic plasticity and genotype-per-environment *** terroir,namely the set of agri-environmental factors to which a variety is subjected,can influence the phenotype at the physiological,molecular,and biochemical level,representing an important phenomenon connected to the typicality of *** investigated the determinants of plasticity by conducting a field-experiment where all terroir variables,except soil,were kept as constant as *** isolated the effect of soils collected from different areas,on phenology,physiology,and transcriptional responses of skin and flesh of a red and a white variety of great economic value:Corvina and *** results,together with physio-phenological parameters,suggest a specific effect of soil on grapevine plastic response,highlighting a higher transcriptional plasticity of Glera in respect to Corvina and a marked response of skin compared to *** a novel statistical approach,we identified clusters of plastic genes subjected to the specific influence of *** findings could represent an issue of applicative value,posing the basis for targeted agricultural practices to enhance the desired characteristics for any soil/cultivar combination,to improve vineyards management for a better resource usage and to valorize vineyards uniqueness maximizing the terroir-effect.

The comprehension of molecular processes underlying the development and progression of flowering in plants is a hot topic,not only because that often the products of interest for human and animal nutrition are linked to the development of fruits or seeds,but also because the processes of gametes formation occurring in sexual organs are at the basis of recombination and genetic variability which constitutes the matter on which evolution acts,whether understood as natural or human *** the present study,we used an NGS approach to produce a grapevine flower transcriptome snapshot in different whorls and tissues including calyx,calyptra,filament,anther,stigma,ovary,and embryo in both pre-and post-anthesis *** investigation aimed at identifying hub genes that unequivocally distinguish the different tissues providing insights into the molecular mechanisms that are at the basis of floral whorls and tissue *** this end we have used different analytical approaches,some now consolidated in transcriptomic studies on plants,such as pairwise comparison and weighted-gene coexpression network analysis,others used mainly in studies on animals or human's genomics,such as the tau(t)analysis aimed at isolating highly and absolutely tissue-specific *** intersection of data obtained by these analyses allowed us to gradually narrow the field,providing evidence about the molecular mechanisms occurring in those whorls directly involved in reproductive processes,such as anther and stigma,and giving insights into the role of other whorls not directly related to reproduction,such as calyptra and *** believe this work could represent an important genomic resource for functional analyses of grapevine floral organ growth and fruit development shading light on molecular networks underlying grapevine reproductive organ determination.

The plant WRKY gene family represents an ancient and complex class of zinc-finger transcription factors(TFs)that are involved in the regulation of various physiological processes,such as development and senescence,and in plant response to many biotic and abiotic *** the growing number of studies on the genomic organisation of WRKY gene family in different species,little information is available about this family in grapevine(Vitis vinifera L.).In the present study,a total number of 59 putative grapevine WRKY transcription factors(VvWRKYs)were identified based on the analysis of various genomic and proteomic grapevine *** to their structural and phylogentic features,the identified grapevine WRKY transcription factors were classified into three main *** order to shed light into their regulatory roles in growth and development as well as in response to biotic and abiotic stress in grapevine,the VvWRKYs expression profiles were examined in publicly available microarray *** analysis of these data revealed distinct temporal and spatial expression patterns of VvWRKYs in various tissues,organs and developmental stages,as well as in response to biotic and abiotic *** also extend our analysis to situations not covered by the arrays and to validate our results,the expression profiles of selected VvWRKYs in response to drought stress,Erysiphe necator(powdery mildew)infection,and hormone treatments(salicilic acid and ethylene),were investigated by quantitative real-time reverse transcription PCR(qRT-PCR).The present study provides a foundation for further comparative genomics and functional studies of this important class of transcriptional regulators in grapevine.

Cu/Ta sheaths for iron-based superconductors:First experimental findings in Ca/K-1144 wires

Iron based superconducting wires(IBSCs)produced by the Powder in Tube(PIT)method rely on the use of silver sheaths as chemical buffer between the outer metal and the superconducting *** adoption of silver entails however some limitations,such as the viable temperature range when coupled with copper,and the incompatibility with calcium-based IBSCs already at 600℃,driving the research towards other wires *** inspiration from the low temperature superconductors field,we decided to evaluate the adoption of tantalum as diffusion barrier in a layered Cu/Ta architecture,choosing a Ca/K-1144 IBSC as case study considering the high reactivity issues already reported in the case of silver sheaths for this *** wires were produced through a groove rolling lamination process coupled with a thermal treatment at 800℃.The microstructural analyses show the absence of interdiffusion between the different parts of the wire,and the magnetic characterization shows performance in line with similar polycrystalline manufacts,with margin of enhancement to be pursued via the optimization of the mechanical process and other experimental *** reported results suggest thus the effectiveness of tantalum as diffusion barrier for Ca/K-1144 PIT wires.
