Antibacterial activity of Lawsonia inermis Linn(Henna) against Pseudomonas aeruginosa

Objective:To investigate the antibacterial activity of henna(Lawsonia inermis Linn) obtained from different regions of Oman against a wide array of ***:fresh henna samples were obtained from different regions of Oman as leaves and seeds,100 g fresh and dry leaves and SO g of fresh and dry seeds were separately soaked in 500 mL of ethanol for three days,respectively,with frequent *** mixture was filtered,and the crude extract was *** crude extract was then heated,at 48 ℃ in a water bath to evaporate its liquid *** dry crude henna extract was then tested for its antibacterial activity using well-diffusion antibiotic susceptibility *** extracts were investigated for their antibacterial activity at different concentrations against a wide array of different micro-organisms including a laboratory standard bacterial strain of Pseudomonas aeruginosa(NCTC 10662)(A aeruginosa) and eleven fresh clinical isolates of *** obtained from patients attending the Sultan Qaboos University Hospital(SQUH).2-Hydroxy-p-Nathoqinone-Tech(2-HPNT, MW=174.16,C_(10)H_40_3) was included as control(at 50%concentration) along with the henna samples ***:Henna samples demonstrated antibacterial activity against all isolates but the highest susceptibility was against *** with henna samples obtained from al-sharqyia ***:Omani henna from al-sharqyia region demonstrates high in vitro anti-P. aeruginosa activity compared with many henna samples from different regions of Oman.

Studies have suggested that the nucleus accumbens(NAc)is implicated in the pathophysiology of major depression;however,the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been ***,we identified a specific reduction of cyclic adenosine monophosphate(cAMP)in the subset of dopamine D1 receptor medium spiny neurons(D1-MSNs)in the NAc that promoted stress susceptibility,while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like *** treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons(D2-MSNs)of depressed mice,however,the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc *** discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration,but not a lower *** fast onset property of crocin was verified through multicenter ***,crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on *** findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc,and provide a potential rapid antidepressant drug candidate,crocin.

Influence of different cultivars of Phoenix dactylifera L-date fruits on blood clotting and wound healing

Objective: To investigate different types of dates and medical properties of influencing blood clotting and wound healing in an animal model. Methods: Three different cultivars of dates(Ajwa, Khalas, and Fardh) were examined in-vivo, for blood clotting and wound healing using CD1 mice of both sexes. Study of toxicity to animals was performed accordingly prior to further investigations. The ethanolic extracts were given orally to animals as a constituent in their daily water. Blood samples were obtained from the mice inferior vena cava to carry out the prothrombin time(PT) assay using the manual method and confirmed using a semi-automated machine. The bleeding time(BT) assay was performed using the cutting technique. In the wound healing analysis, a small cut(5-10 mm) in the skin overlying the thigh was conducted in all mice under anesthesia. The diameter of the cut and healing status were measured on a daily basis throughout the time of the experiment using a roller. Results: Ajwa was able to elevate both PT and BT(Palas date(Palas date showed a potential property to enhance wound healing in contrast to other dates and the control groups in this study. Conclusions: Omani Khalas date fruit has both antithrombotic as well as wound healing properties. The results open a new gate with these fruits for exploring the potential component(s) that may play an important role in antithrombotic as well as wound healing process.

Rapid isolation and identification of Mycobacterium tuberculosis from pulmonary tuberculosis patients

Objective:The Study aimed to develop a rapid inexpensive and simple method for preliminary isolation and detection of *** from clinical *** to probe the impact of deteriorating health measures on the pulmonary tuberculosis control program in Basra ***:A simple monophasic-diphasic setup (MDCS) was used for the isolation and preliminary identification of *** setup consists of a slanted Lowenstein-Jensen medium,the bottom of it is covered with 1 mL of tuberculosis broth thus establishing a diphasic solid liquid environment at the bottom of the screw capped test tube and above them a monophasic ***:During 7 months period from the beginning of December 2003 to the end of June 2004,1 295 cases were managed in Tuberculosis and Chest Diseases Clinic in Basra *** 348 cases were diagnosed as active pulmonary *** consist of 232 new cases and 116 previously registered *** MDCS method showed higher recovery of isolation rate of ***(92.3%) than Lowenstein-Jensen,and pyruvate methods with 78.1%and 51.9%*** this method revealed more rapid appearance of results of 12 days than Lowenstein-Jensen,and pyruvate media with 22 days and 23 days respectively by the traditional culturing *** the study revealed that drug resistance against one or more anti tuberculosis drugs was 23.1%for new cases and 70.8%for the old *** drug resistance accounts for 20%of total isolates from old *** was based on using the critical concentration and 1%proportional *** was more common among males than females with 69.4%and 30.6%respectively. A significant correlation(P<0.05) was found between the productive age group(25-54) and incidence of tuberculosis. 目的研究达立通颗粒主要活性成分在正常和功能性消化不良(FD)模型大鼠胃肠道组织中的分布差异。方法采用夹尾刺激结合不规律饮食双因素干预方法建立FD模型。12只大鼠(对照、模型各6只)用于FD模型验证,进行胃、小肠排空实验,HE染色后进行胃肠组织病理学观察;36只大鼠(对照、模型各18只)用于胃肠道组织差异分布研究,禁食不禁水18 h,按22.4 g‧kg^(−1)剂量(8倍临床等效剂量)ig给予达立通颗粒1次,于给药后1、3、6 h收集各组大鼠的胃、十二指肠、空肠、回肠和结肠。应用超高效液相色谱-串联四级杆/线性离子阱质谱(UPLC-QTRAP-MS/MS)法检测各组织牡荆素-4''-O-葡糖苷、原阿片碱、黄连碱、小檗碱、甜橙黄酮、川陈皮素、桔皮素、去甲基川陈皮素、木香烃内酯、去氢木香内酯、原儿茶酸、伞形花内酯、木犀草苷、橙皮苷、木犀草素、柚皮素、橙皮素、白杨素含量,采用ACQUITY HPLCTM HSS T3(50 mm×2.1 mm,1.8μm)色谱柱,柱温为40℃,0.1%甲酸水-乙腈为流动相,梯度洗脱,体积流量为0.3 mL‧min^(−1),进样量为2μL。结果与对照组比较,模型组大鼠体质量显著降低(P<0.001),胃、小肠排空率显著降低(P<0.001),十二指肠肠黏膜的紧密连接被破坏,但未出现胃肠组织的器质性病变,符合FD的病理特征。建立的UPLC-QTRAP-MS/MS方法中,各化合物线性关系良好,R^(2)≥0.9900。对建立的检测方法进行专属性、精密度与准确度、提取回收率、稳定性和基质效应等方面的考察,均符合要求。各成分在对照和模型组大鼠胃肠道组织中浓度差异明显,且在各组织中达到最高浓度的时间点差异较大,模型组出现达峰时间延后的现象;与对照组比较,模型组大鼠胃组织中化合物的暴露水平降低,可能与FD引起胃肠道蠕动减慢有关。橙皮苷、橙皮素、柚皮素、原儿茶酸、去氢木香内酯、川陈皮素、去甲基川陈皮素、小檗碱和桔皮素在胃、十二指肠、空肠、回肠和结肠中暴露水平较高,且在对照组和模型组中各时间点下的浓度差异较显著。结论经方法学验证,所建立的UPLC-QTRAP-MS/MS法灵敏、专属性强、准确性高,适用于大鼠胃肠道组织中达立通颗粒主要活性成分的胃肠组织分布研究。达立通颗粒主要活性成分在正常和FD模型大鼠胃肠组织分布具有显著性差异。

Bacterial and fungal endophthalmitis in Upper Egypt:related species and risk factors

Objective:To study risk factors,contributing factors of bacterial and fungal endophthalmitis in Upper Egypt,test the isolated species sensitive to some therapeutic agents,and to investigate the air-borne bacteria and fungi in opthalmology operating ***:Thirty one cases of endophthalmitis were clinically diagnosed and microbiologically *** air-borne bacteria and fungi inside four air-conditioned operating rooms in the Ophthalmology Department at Assiut University Hospitals were also *** isolated microbes from endophthalmitis cases were tested for their ability to produce some extracellular enzymes including protease,lipase,urease,phosphatase and *** the ability of 5 fungal isolates from endophthalmitis origin to produce mycotoxins and their sensitivity to some therapeutic agents were ***:Results showed that bacteria and fungi were responsible for infection in 10 and 6 cases of endophthalmitis,respectively and only 2 cases produced a mixture of bacteria and *** was the most prevalent risk factor of endophthalmitis where 58.1%of the 31 cases were due to *** ophthalmology operating rooms,different bacterial and fungal species were isolated.8 bacterial and 5 fungal isolates showed their ability to produce enzymes while only 3 fungal isolates were able to produce *** showed the highest effect against most isolates in ***:The ability of bacterial and fungal isolates to produce extracellular enzymes and mycotoxins may be aid in the invasion and destruction of eye tissues. Microbial contamination of operating rooms with air-borne bacteria and fungi in the present work may be a source of postoperative endophthalmitis.

Studies on the anti-aging properties of bovine whey supplementation on reproductive hormonal profiles and sperm structure and function of adult and senile rats

The purpose of this study was to investigate how well whey might protect against aging related alterations in sperm structure,seminal and antioxidant enzymes and serum hormonal *** Wistar albino rats aged 8 and 30-months were divided into two primary groups with or without whey supplementation(n=8).After the experiments of whey supplementation,the rats were euthanized,blood was collected,and serum was *** ultrastructure and flow cytometry of annexin-V and mitochondrial membrane potential were carried *** antioxidant and malondialdehyde concentrations were *** hormonal profiles of testosterone,growth(GH),follicle stimulating hormone(FSH),luteinizing hormone(LH)and thyroid stimulating hormone(TSH)as well as pituitary gland ultrastructure were *** showed that whey supplementation improved the structure and function of sperm and seminal fluid as well as the serum hormonal levels of *** aged rats,whey supplementation increased the serum levels of GH,FSH,LH and TSH as well as seminal superoxide *** supplementation also caused reduction of seminal alkaline and acid phosphatases and lipid peroxidation in aged rat compared to that in adult *** also improved sperm motility and viability but reduced sperm abnormalities by increasing sperm mitochondrial membrane potential and lowering annexin v *** conclusion,whey contains antioxidant components that alleviated oxidative stress involved in damaging sperm macromolecules during aging process.

Association of E-cadherin (CDH1) gene polymorphisms and gastric cancer risk

AIM:To investigate the associations between CDH1 gene polymorphisms and gastric cancer(GC) risk ***:We analyzed four CDH1 polymorphisms(+54 T>C,-160 C>A,-616 G>C,-3159 T>C) in an Omani population,by extraction of genomic DNA from the peripheral blood of 192 patients with GC and 170 control participants and performed CDH1 genotyping using DNA ***:CDH1-160-aa genotype was associated with an increased risk of GC(OR = 3.6,95% CI:1.1-11.8)(P = 0.03).There was no significant association between the other polymorphisms and GC *** haplotype analysis of +54 T>C,-160 C>A,-616 G>C,-3159 T>C genotypes revealed that the OR of CCGC and CAGC haplotypes was 1.5(95% CI:0.7-3.5) and 1.5(95% CI:0.2-3.0),but did not reach statistical ***:The current study suggests that the-160 aa genotype was associated with an increased risk of GC in Oman.
