The polybasic region of cytohesin-2 determines paxillin binding specificity to mediate cell migration

The intracellular signaling pathways through ADP-ribosylation factors (Arfs) of the small GTPase family control cell morphological changes by regulating membrane components and/or cytoskeletal protein dynamics. We previously reported that cytohesin-2 (CYTH2), an Arf-guanine-nucleotide exchange factor (GEF), binds to the cytoskeletal scaffold protein paxillin through C-terminal region of CYTH2 and promotes the migration of mouse 3T3-L1 fibroblasts. In mammals, CYTH family GEFs are composed of four subfamilies. Among them, CYTH2 and CYTH3 are widely expressed in tissues and it remains to be clarified to determine whether they have specific biochemical and cellular functions or are redundant. Here, we show that the C-terminal short polybasic region of CYTH2 is necessary and sufficient for binding to paxillin to mediate cell migration. Although 3T3-L1 cells primarily express CYTH2 and CYTH3 of four CYTH family members, neither knockdown of CYTH3 by the specific siRNA nor expression of its C-terminal region inhibits migration. Importantly, replacing the C-terminal region of CYTH3 with that of CYTH2 adds the ability of paxillinbinding and mediating migration to CYTH3. Conversely, replacing the C- terminal region of CYTH2 with that of CYTH3 leads to loss of these abilities of CYTH2. These results reveal that paxillin is a unique binding partner with CYTH2 in migrating cells, presenting the first CYTH family GEF’s region that is involved in the selectivity of the binding protein.

Evaluation of Sawdust and Rice Husks as Fillers for Phenolic Resin Based Wood-Polymer Composites

We produced Wood-Polymer Composites (WPCs) with phenolic resin (PR) filled with saw dust (SD) and rice husks (RH) in a PR:fillerratio of 60:40 wt.%. RH and SD were grinded and sieved into particles μm. The aim of this research work was to evaluate sawdust and rice husks as fillers for sustainable phenolic resin based WPCs. Therefore, we investigated the thermal stability of PR/RH and PR/SD WPCs then we studied and compared the tensile, flexural properties of PR/SD and PR/RH WPCs samples, as well as their dimensional stability after water absorption test. Furthermore, through ultraviolet light exposure, we evaluated the effects of photo-oxidation on the water stability and mechanical properties of PR/RH and PR/SD WPCs samples compared to unexposed ones. PR filled with SD presented better mechanical properties compared to PR/RH WPCs samples. However, PR/RH WPCs showed good mechanical properties, and better thermal resistance and better water repulsion capabilities compared to PR/SD WPCs samples. Although, long time UV exposure ended up lowering considerably the mechanical properties and water resistance of PR/SD and PR/RH WPCs, both RH and SD offer great added value as fillers for PR based WPCs;SD having better interactions with PR matrix compared to RH.

Determination of <i>α</i>-1,3-Linked Mannose Residue in the Cell Wall Mannan of <i>Candida tropicalis</i>NBRC 1400 Strain

To investigate the chemical structure of cell wall mannan obtained from pathogenic yeast, Candida tropicalis NBRC 1400 (former antigenic standard strain, IFO 1400). As a result of two-dimensional NMR analysis, it was shown that the mannan of this strain is composed of α-1,6-, α-1,3-, α-1,2- and β-1,2-linked mannose residues. In this research, the mannan was subjected to three degradation procedures, acid-treatment, α-mannosidase, and acetolysis under two conditions in order to determine the chemical structure of the antigenic oligomannosyl side chains in this molecule. The 1H-nuclear magnetic resonance spectra of resultant oligosaccharides, pentaose and hexaose, demonstrated the existence of the oligomannosyl side chains corresponding to Manα1-3Manα1-2Manα1-2Manα1-2Man and Manα1-3Manα1-2Manα1-2Manα1-2Manα1-2Man, respectively, which have previously also been found in Candida albicans serotype A strain mannans. These findings indicate that C. tropicalis and C. albicans serotype A have no significant difference in the chemical structure of these cell wall mannans. Therefore, it can be interpreted that it is extremely difficult to distinguish both species by targeting the antigenic group in these mannans.

Contrast enhanced ultrasound of hepatocellular carcinoma

Sonazoid(Daiichi Sankyo,Tokyo,Japan),a secondgeneration of a lipid-stabilized suspension of a perfluorobutane gas microbubble contrast agent,has been used clinically in patients with liver tumors and for harmonic gray-scale ultrasonography(US)in Japan since January ***-enhanced US has two phases of contrast enhancement:vascular and *** the late phase of Sonazoid-enhanced US,we scanned the whole liver using this modality at a low mechanical index(MI)without destroying the microbubbles, and this method allows detection of small viable hepatocellular carcinoma(HCC)lesions which cannot be detected by conventional US as perfusion defects in the late ***-injection of Sonazoid into an HCC lesion which previously showed a perfusion defect in the late phase is useful for confirming blood flow intothe *** MI intermittent imaging at 2 frames per second in the late phase is also helpful in differentiation between necrosis and viable hypervascular HCC ***-enhanced US by the coded harmonic angio mode at a high MI not only allows clear observation of tumor vessels and tumor enhancement, but also permits automatic scanning with Sonazoidenhanced three dimensional(3D)*** images combining US with contrast-enhanced CT or contrastenhanced MRI have made it easy to detect typical or atypical HCC *** these methods,Sonazoidenhanced US can characterize liver tumors,grade HCC lesions histologically,recognize HCC dedifferentiation, evaluate the efficacy of ablation therapy or transcatheter arterial embolization,and guide ablation therapy for unresectable *** article reviews the current developments and applications of Sonazoid-enhanced US and Sonazoid-enhanced 3D US for diagnosing and treating hepatic lesions,especially HCC.

利用 12对AFLP引物 ,以饭豆标准品系M 0 0作对照 ,对来自中国、日本、韩国、尼泊尔、印度、不丹的 14 6份小豆栽培种 (***)和野生种 (***)种质的基因组DNA进行扩增 ,得到 313条多态性带。据AFLP多态性数据绘制的聚类图 ,可区分其中的 14 3份种质 ,表明小豆物种 (Vi gnaangularis)存在足够的遗传多样性 ,可用于资源材料的准确鉴别与分类。鉴于此 ,采用新开发的利用AFLP数据揭示核苷多样性的Innan’s进化树分析方法 ,进一步将 14 6份小豆资源分成 7个明显不同的地理演化群 ,即中国栽培种、日本栽培种、日本综合群韩国栽培种、中国台湾野生种、中国野生种、尼泊尔不丹栽培种和喜马拉雅野生种演化群。就上述地理演化群的遗传多样性、地理分布以及野生种与栽培种之间可能的演化关系进行了分析 ,初步认为栽培小豆至少应当有 4个不同类型的野生祖先和 3个不同的地理起源。

The liver-to-spleen ratio is a risk factor predicting oxygen demand in COVID-19 patients

Background:We aimed to investigate risk factors predicting oxygen demand in COVID-19 ***:Patients admitted to Shizuoka General Hospital with COVID-19 from August 2020 to August 2021 were ***,we divided patients into groups with and without oxygen ***,we compared patients’clinical characteristics and laboratory and radiological findings to determine factors predicting oxygen ***:One hundred seventy patients with COVID-19(aged 58±15 years,57 females)were *** comorbidities were cardiovascular diseases(47.6%),diabetes mellitus(28.8%),and dyslipidemia(26.5%).Elder age,higher body mass index,cardiovascular diseases,diabetes mellitus,lower lymphocyte count,albumin,hep-atic attenuation value,and the liver-to-spleen ratio(L/S),higher D-dimer,aspartate aminotransferase,lactate dehydrogenase,troponin-T,C-reactive protein,KL-6,chest and abdominal circumference,and visceral fat were found in patients with oxygen *** to the multivariate logistic regression analysis,L/S,lympho-cyte count,D-dimer,and abdominal circumference under the diaphragm were independent risk factors predicting oxygen demand in COVID-19 ***:On admission,L/S,lymphocyte count,D-dimer,and abdominal circumference were predictive factors for oxygen *** factors may help in the appropriate triage of COVID-19 patients in the decision to admit them to the hospital.

BACKGROUND It is generally accepted that the incidence of hepatocellular carcinoma(HCC)in hepatitis C virus(HCV)-associated patients is higher than that in hepatitis B virus(HBV)-associated *** reason why this difference in the incidence of HCC occurs in patients with HBV and HCV infections remains *** report the possibility that the contributing power of inflammation,which is the main risk factor for developing HCC,may be different with HBV and HCV *** To investigate this,we surveyed the hazard ratio of inflammation for HCC development which was identified by serum alanine aminotransferase(ALT)levels between patients with HBV and HCV *** The PubMed database was searched(2001-2021)for studies published in English regarding the incidence of HCC identifying 8924 HBV-and 7376 HCV-infected *** these studies,interferon-treated patients with both HBV and HCV infections were ***,in HBV patients,those administered nucleos(t)ide analogues were excluded,and in HCV patients,those administered direct acting antivirals were also *** citing hazard ratios of HCC regarding inflammation(serum elevated alanine aminotransferase levels)were ***,there were 14 studies of HBV-infected patients and 8 studies of HCV-infected *** calculated the hazard ratio in patients in an inflammatory state(serum ALT levels were above the normal range).RESULTS In the 14 studies of HBV patients,the average hazard ratio(HR)of elevated ALT for developing HCC was 2.74[1.98-3.77]and that in the 8 studies of HCV-infected patients was 5.51[3.08-9.83].The HR of inflammation for HCC development in HCV-associated liver diseases is about twice that in HBV-associated liver *** in HCV-infected patients was significantly(P=0.0391)higher than that in HBV-infected *** hepatitis B patients,the abnormal range adopted was 28-45 IU/L,and in hepatitis C patients,it was 20-50 IU/*** was demonstrated that the
