New minor cyclic peptides, named arenariphilin C 1 and arenariphilin D 2, were isolated from the whole plants of Arenaria oreophila （Caryophyllaceae）. Their structures were determined as cyclo（Pro^1-Pro2-Leu^1-Leu2-Phe-Ser-Gly-Thr） and cyclo （Ser-Cys） on the basis of spectroscopic data, especially by two-dimension NMR （2D NMR） technologies.

Quorum sensing(QS)plays an essential role in virulence factor production,biofilm formation,and antimicrobial *** a potent QS inhibitor,hordenine can inhibit both QS and biofilm formation in Pseudomonas aeruginosa and Serratia *** this work,we tested the QS inhibitory potential of 27 hordenine analogs against QS and biofilm formation in *** and *** the tested analogs,seven(12,28,27,26,2,23,and 7)exhibited strong QS inhibitory activity against ***,five of which(12,28,27,26,and 2)showed better inhibitory activity than *** addition,seven analogs(28,12,23,7,26,2,and 27)exhibited better biofilm inhibition against *** than *** analogs(7,28,2,and 12)showed QS inhibitory activity against ***,two of which(7 and 28)demonstrated better inhibitory activity than ***,analog 7 showed similar biofilm inhibition against *** as ***-activity relationship(SAR)analysis indicated that the inhibitory activities of the analogs were related to four factors,i.e.,carbon chain length,presence or absence of anα,β-C=C bond,amino group with/without lipophilic group,such as methyl group,and hydroxyl group in benzene ring.

Quorum sensing(Qs)inhibition has emerged as a promising target for directed drug design,providing an appealing strategy for developing antimicrobials,particularly against infections caused by drug-resistant *** this study,we designed and synthesized a total of 33β-nitrostyrene derivatives using 1-nitro-2-phenylethane(NPe)as the lead compound,to target the facultative anaerobic bacterial pathogen Serratia *** QS-inhibitory effects of these compounds were evaluated using *** NJ01 and the reporter strain Chromobacterium violaceum *** the 33 newβ-nitrostyrene derivatives,(E)-1-methyl-4-(2-nitrovinyl)benzene(m-NPe,compound 28)was proven to be a potent inhibitor that reduced biofilm formation of *** NJ01 by 79%.Scanning electron microscopy(SEM)and confocal laser scanning microscopy(CLSM)results revealed that treatment with m-NPe(50μg/ml)not only enhanced the susceptibility of the formed biofilms but also disrupted the architecture of biofilms by 84%.m-NPe(50μg/ml)decreased virulence factors in *** NJ01,reducing the activity of protease,prodigiosin,and extracellular polysaccharide(EPs)by 36%,72%,and 52%,*** *** 4547,the activities of hemolysin and EPs were reduced by 28%and 40%,respectively,outperforming the positive control,vanillic acid(VAN).The study also found that the expression levels of QS-and biofilm-related genes(flhD,fimA,fimC,sodB,bsmB,pigA,pigC,and shlA)were downregulated by 1.21-to *** dynamics analysis showed that m-NPe could bind stably to SmaR,Rhll,RhiR,LasR,and CviR proteins in a 0.1 M sodium chloride ***,a microscale thermophoresis(MST)test revealed that SmaR could be a target protein for the screening of a quorum sensing inhibitor(QSl)against ***,this study highlights the efficacy of m-NPe in suppressing the virulence factors of ***,identifying it as a new potential Qsl and antibiofilm agent capable of restoring or improv

Chemical Constituents from Leaves of Camellia nitidissima and Their Potential Cytotoxicity on SGC7901 Cells

Objective To isolate and identify the bioactive phytochemicals from the leaves of Camellia nitidissima. Methods The chemical constituents were isolated and purified by repeated silica gel, Sephadex LH-20, MCI gel columns, recrystallization, and semi-preparative HPLC techniques. The chemicl structures of these compounds were identified on the basis of spectral data including NMR and MS. Then quorum sensing inhibition （QSI） activities of these compounds were tested using Chromobacterium violaceum CV026 as the bioindicator strain. The antitumor activities of these compounds were measured using SGC7901 as cell proliferation and cytotoxicity. Results cx-Spinasteryl-I^-D-glucopyranoside （1）, stigmasta-7,22-diene-3-O-[c^-L-arabinopyranosyl （1 -2）]-β-D-galactopyranoside （2）, kaempferol 3-O-[2-O-（trans-p-coumaroyl）-3-O-α -D-glucopyranosyl]-α-D-glucopyranoside （3）, aromadendrin （4）, catechin （5）, phlorizin 4＇-O-β-D-glucopyranoside （6）, （3R,6R,7Lg-3-hydroxy-4,7-megastigmadien- 9-one （7）, dodecanoic acid （8）, 3α-acetoxy-20-1upanol （9）, and 3β,6α- trihydroxyolean- 7-one （1 0） were successively isolated from the leaves of C. nitidissima. Unfortunately, these compounds had no QSI activity. Based on Cell Counting Kit-8 （CCK-8） assay, compound 10 showed the best anti-tumor activity or all compounds （ICs0 = 91.7 μg/mL）. Conclusion Apart from compounds 4 and 5, other eight compounds are reported in this plant for the first time. All compounds show no QSI activity, compound 10 shows potential cytotoxic activity on SGC7901 cells in vitro.

We propose a new narrowband speech watermarking scheme by replacing part of the speech with a scaled and spectrally shaped hidden signal. Theoretically, it is proved that if a small amount of host speech is modified, then not only an ideal channel model for hidden communication can be established, but also high imperceptibility and good intelligibility can be achieved. Furthermore, a practical system implementation is proposed. At the embedder, the power normalization criterion is first imposed on a passband watermark signal by forcing its power level to be the same as the original passband excitation of the cover speech, and a synthesis filter is then used to spectrally shape the scaled watermark signal. At the extractor, a bandpass filter is first used to get rid of the out-of-band signal, and an analysis filter is then employed to compensate for the distortion introduced by the synthesis filter. Experimental results show that the data rate is as high as 400 bits/s with better bandwidth efficiency, and good imperceptibility is achieved. Moreover, this method is robust against various attacks existing in real applications.
