Transcription activator-like effectors （TALEs） from Xanthomonas sp. have been used as customizable DNA- binding modules for genome-engineering applications, Ralstonia solanacearum TALE-like proteins （RTLs） exhibit similar structural features to TALEs, including a central DNA-binding domain composed of 35 amino acid-long repeats. Here, we characterize the RTLs and show that they localize in the plant cell nucleus, mediate DNA binding, and might function as transcriptional activators. RTLs have a unique DNA-binding architecture and are enriched in repeat variable di-residues （RVDs）, which determine repeat DNA-binding specificities. We determined the DNA-binding specificities for the RVD sequences ND, HN, NP, and NT. The RVD ND mediates highly specific interactions with C nucleotide, HN interacts spe- cifically with A and G nucleotides, and NP binds to C, A, and G nucleotides. Moreover, we developed a highly efficient repeat assembly approach for engineering RTL effectors. Taken together, our data demonstrate that RTLs are unique DNA-targeting modules that are excellent alternatives to be tailored to bind to user-selected DNA sequences for targeted genomic and epigenomic modifications. These findings will facilitate research concerning RTL molecular biology and RTL roles in the pathogenicity of Ralstonia spp.

Intestinal parameters of oxidative imbalance in celiac adults with extraintestinal manifestations

AIM To evaluate selected intestinal parameters of oxidative stress, and antioxidant capacity in adult celiac disease patients with extraintestinal *** The study involved 85 adult patients divided into the following subgroups:(1) patients with newly diagnosed celiac disease(CD)(n = 7);(2) celiac patients not adhering to a gluten-free diet(GFD)(n = 22);(3) patients with CD on the GFD(n = 31); and(4) patients with functional disorders of the gastrointestinal tract, serving as controls(n = 25). Celiac patients presented with non-classic symptoms or extraintestinal manifestations. Standard blood tests including serum antioxidant levels(uric acid, bilirubin, and vitamin D), celiac antibody levels, and histopathological status of duodenal biopsy specimens have been determined. The expression of m RNA for tumor necrosis factor α(TNF-α), interleukin 1β(IL-1β), interleukin 10(IL-10), superoxide dismutase(SOD), heat-shock protein 70(HSP-70), hypoxia-inducible factor 1(HIF-1α), and BAX in the duodenal mucosa of patients was analyzed by reverse transcriptase-polymerase chain reaction. RESULTS The mean plasma uric acid level in patients with active CD(newly diagnosed and nonadherent patients) and treated celiac patients was significantly higher than in controls(260.17 ± 53.65 vs 190.8 ± 22.98, P < 0.001, and 261.7 ± 51.79 vs 190.8 ± 22.98, P < 0.001, respectively). The mean bilirubin concentration in active and treated celiac patients was significantly lower than in controls(8.23 ± 5.04 vs 10.48 ± 4.08, P < 0.05 and 8.06 ± 3.31 vs 10.48 ± 4.08, P < 0.05, respectively). The mean plasma vitamin D level was significantly lower in active celiac patients than in treated celiac patients and controls(19.37 ± 9.03 vs 25.15 ± 11.2, P < 0.05 and 19.37 ± 9.03 vs 29.67 ± 5.12, P < 0.001, respectively). The expression of TNF-α, IL-10, and HSP-70 m RNAs was significantly elevated in the celiac groups regardless of the diet when compared with controls. Patients on the GFD pre

Halpern Iteration in CAT（k） Spaces

In this paper we show that an iterative sequence generated by the Halpern algorithm converges to a fixed point in the case of complete CAT（k） spaces. Similar results for Hadamard manifolds were obtained in [Li, C., L6pez, G., Martfn-Marquez, V.： Iterative algorithms for nonexpansive mappings on Hadamard manifolds. Taiwan Residents J. Math., 14, 541-559 （2010）], but we study a much more general case. Moreover, we discuss the Halpern iteration procedure for set-valued mappings.

Article 59.1,of the International Code of Nomenclature for Algae,Fungi,and Plants(ICN;Melbourne Code),which addresses the nomenclature of pleomorphic fungi,became effective from 30 July *** that date,each fungal species can have one nomenclaturally correct name in a particular *** other previously used names for this species will be considered as *** older generic epithet takes priority over the younger *** widely used younger names proposed for use,must comply with Art.57.2 and their usage should be approved by the Nomenclature Committee for Fungi(NCF).In this paper,we list all genera currently accepted by us in Dothideomycetes(belonging to 23 orders and 110 families),including pleomorphic and nonpleomorphic *** the case of pleomorphic genera,we follow the rulings of the current ICN and propose single generic names for future *** taxonomic placements of 1261 genera are listed as an *** names and suppressed names for 34 pleomorphic genera are listed *** and justifications are provided for possible proposed names after the list of *** are also provided on recent advances in our understanding of asexual and sexual morph linkages in Dothideomycetes.A phylogenetic tree based on four gene analyses supported 23 orders and 75 families,while 35 families still lack molecular data.
