AIM To determine tissue expression(mRNA, protein) of two types of mucins [mucin 1(MUC1) and mucin 2(MUC2)] in patients with colorectal cancer(CRC).METHODS Expression of membrane-bound mucin(MUC1) and secretory mucin(MUC2) in CRC(mRNA, protein) were analyzed in tissue material including fragments of tumorsobtained from CRC patients(n = 34), and fragments of normal colorectal tissue from the same patients(control). The analysis was conducted using real-time quantitative polymerase chain reaction(RT-qPCR)(transcripts), immunohistochemistry(IHC)(apomucins), and the modern approach for morphometric analysis of IHC reaction(HSV filter software). Results on tissue expression of both mucins(mRNA, protein) were compared to histological alterations in colorectal cancer samples and correlated with selected clinical data in the patients. The statistical analysis was conducted using Statistica PL v. 12.0 *** Significantly higher expression of the MUC1 mRNA in the CRC, compared with the control and the borderline correlation of mRNA expression with MUC1 protein levels in colorectal samples was observed. The expression of apomucins concerned cell membranes(MUC1) and cytoplasm(MUC2) and occurred both in control tissues and in most cancerous samples. There were no significant relationships between MUC1(mRNA, protein) and the clinicopathological data of patients. MUC2 protein expression was significantly lower as compared to the control, while MUC2 mRNA expression was comparable in both groups. The MUC1/MUC2 ratio was significantly higher in CRC tissues than in the control. The higher expression of MUC2 was a feature of mucinous CRC subtypes, and characterized higher histological stage of tumors. Electron microscopy (EM) is widely used for nanoparticle (NP) sizing. Following an initial assessment of two sample preparation protocols described in the current literature as"unperturbed", we found that neither could accurately measure the size of NPs featuring a broad size distribution, e.g., aggregates. Because many real-world NP samples consist of aggregates, this finding was of considerable concern. The data showed that the protocols introduced errors into the measurement by either inducing agglomeration artefacts or providing a skewed size distribution towards small particles (skewing artefact). The focus of this work was to develop and apply a mathematical refinement to correct the skewing artefact. This refinement provided a much improved agreement between EM and a reference methodology, when applied to the measurement of synthetic amorphous silica NPs. Further investigation, highlighted the influence of NP chemistry on the refinement. This study emphasised the urgent need for greater and more detailed consideration regarding the sample preparation of NP aggregates to routinely achieve accurate measurements by EM. This study also provided a novel refinement solution applicable to the size characterisation of silica and c让rate-coated gold NPs featuring broad size distributions. With further research, this approach could be extended to other NP types.

Celiac disease(CD) is a chronic immune-mediated disorder triggered by the ingestion of gluten in genetically predisposed individuals. Before activating the immune system, gluten peptides are transferred by the epithelial barrier to the mucosal lamina propria, where they are deamidated by intestinal tissue transglutaminase 2. As a result, they strongly bind to human leucocyte antigens(HLAs), especially HLA-DQ2 and HLA-DQ8, expressed on antigen-presenting cells. This induces an inflammatory response, which results in small bowel enteropathy. Although gluten is the main external trigger activating both innate and adaptive(specific) immunity, its presence in the intestinal lumen does not fully explain CD pathogenesis. It has been hypothesized that an early disruption of the gut barrier in genetically susceptible individuals, which would result in an increased intestinal permeability, could precede the onset of gluten-induced immune events. The intestinal barrier is a complex functional structure, whose functioning is dependent on intestinal microbiotahomeostasis, epithelial layer integrity, and the gutassociated lymphoid tissue with its intraepithelial lymphocytes(IELs). The aim of this paper was to review the current literature and summarize the role of the gut microbiota, epithelial cells and their intercellular junctions, and IELs in CD development.

Laser drivers are an enabling factor to inertial confinement fusion, because laser diodes must be used instead of flash lamps. We discuss the limitations of laser diode arrays and show what steps the industry is taking. The pump power requirements of large-scale projects such as LIFE or Hi PER are within reach of semiconductor laser diode *** light output powers per laser bars have been around 300 W per bar, as in the Jenoptik 940 nm bars previously used for pumping the Yb:YAG slabs in the Di POLE project. By redesigning the semiconductor laser structures 500 W per bar is now commercially available for 808, 880 and 940 nm pump wavelengths. The construction of one inertial fusion power plant will require an amount of semiconductor laser chips in excess of the current annual production by two orders of magnitude. This adds to the engineering task of improving the device characteristics a challenge to production *** the industry benefits from the recent boost in solid-state lighting that acts as a technology driver, cooperation between manufacturers will be imperative, and to this end we propose standardization efforts.

Erythroprotein-producing human hepatocellular carcinoma receptors （Eph receptors） compose a subfamily of transmembrane protein-tyrosine kinases receptors that takes part in numerous physiological and pathological processes. Eph family receptor-interacting proteins （Ephrins） are ligands for those receptors. Eph/ephrin system is responsible for the cytoskeleton activity, cell adhesion, intercellular connection, cellular shape as well as cell motility. It affects neuron development and functioning, bone and glucose homeostasis, immune system and correct function of enterocytes. Moreover Eph/ephrin system is one of the crucial ones in angiogenesis and lymphangJogenesis. With such a wide range of impact it is clear that disturbed function of this system leads to pathology. Eph/ephrin system is involved in carcinogenesis and cancer progression. Although the idea of participation of ephrin in carcinogenesis is obvious, the exact way remains unclear because of complex bi-directional signaling and cross-talks with other pathways. Further studies are necessary to find a new target for treatment.

Second harmonic generation and sum frequency generation(SHG and SFG)provide effective means to realize coherent light at desired frequencies when lasing is not easily *** have found applications from sensing to quantum optics and are of particular interest for integrated photonics at communication *** the footprints of nonlinear components while maintaining their high up-conversion efficiency remains a challenge in the miniaturization of integrated *** we explore lithographically defined AlGaInP nano(micro)structures/Al_(2)O_(3)/Ag as a versatile platform to achieve efficient SHG/SFG in both waveguide and resonant cavity configurations in both narrow-and broadband infrared(IR)wavelength regimes(1300-1600 nm).The effective excitation of highly confined hybrid plasmonic modes at fundamental wavelengths allows efficient SHG/SFG to be achieved in a waveguide of a cross-section of 113 nm×250 nm,with a mode area on the deep subwavelength scale(λ2/135)at fundamental ***,we demonstrate direct visualization of SHG/SFG phase-matching evolution in the *** together with mode analysis highlights the origin of the improved SHG/SFG *** also demonstrate strongly enhanced SFG with a broadband IR source by exploiting multiple coherent SFG processes on 1μm diameter AlGaInP disks/Al_(2)O_(3)/Ag with a conversion efficiency of 14.8%MW^(−1) which is five times the SHG value using the narrowband IR *** both configurations,the hybrid plasmonic structures exhibit>1000 enhancement in the nonlinear conversion efficiency compared to their photonic *** results manifest the potential of developing such nanoscale hybrid plasmonic devices for state-of-the-art on-chip nonlinear optics applications.
