Arabidopsis Protein Phosphatase 2C ABI1 Interacts with Type I ACC Synthases and Is Involved in the Regulation of Ozone-Induced Ethylene Biosynthesis

Ethylene plays a crucial role in various biological processes and therefore its biosynthesis is strictly regu- lated by multiple mechanisms. Posttranslational regulation, which is pivotal in controlling ethylene biosynthesis, impacts 1-aminocyclopropane 1-carboxylate synthase （ACS） protein stability via the complex interplay of specific factors. Here, we show that the Arabidopsis thaliana protein phosphatase type 2C, ABI1, a negative regulator of abscisic acid signaling, is involved in the regulation of ethylene biosynthesis under oxidative stress conditions. We found that ABI1 interacts with ACS6 and dephosphorylates its C-terminal fragment, a target of the stress-responsive mitogen-activated protein kinase, MPK6. In addition, ABI1 controls MPK6 activity directly and by this means also affects the ACS6 phosphorylation level. Consistently with this, ozone-induced ethylene production was significantly higher in an ABI1 knockout strain （abiltd） than in wild-type plants. Importantly, an increase in stress-induced ethylene production in the abiltd mutant was compen- sated by a higher ascorbate redox state and elevated antioxidant activities. Overall, the results of this study provide evi- dence that ABI1 restricts ethylene synthesis by affecting the activity of ACS6. The ABI1 contribution to stress phenotype underpins its role in the interplay between the abscisic acid （ABA） and ethylene signaling pathways.

Enzyme Substrates Protective Encapsulation within Polymeric Microspheres

Determination of enzymes activity is an important task of analytical and biomedical oriented fluorimetry. Despite of a long track record of application, there is still some room for improvements in this field. In the case of hydrolases, spontaneous decomposition of substrate leads to substantial errors in determination of enzyme activity. An innovative and effective approach is proposed allowing protection of enzyme substrate within the lipophilic moiety of polyacrylate microspheres. It is shown that the introduction of substrate into microspheres is not only an effective method of prevention of unwanted spontaneous process, but also does not disturb the availability of substrate for enzymatic hydrolysis. The effect of presence of proteins in the sample on fluorimetric responses was studied. In contrary to previous studies related to application of lipophilic polymers in biomedical analysis, it is shown that the presence of bovine serum albumins enhances the sensitivity of fluorimetric determination. It is shown that this surprising effect is related to adsorption of proteins on the microspheres surface and change of surface charge of polymer.

This paper describes a formation of hybrid coatings on a Ti-2Ta-3Zr-36Nb *** is accomplished by plasma electrolytic oxidation and a dip-coating technique with poly(adipic anhydride)((C6H8O3)n)that is loaded with drugs:amoxicillin(C16H19N3O5S),cefazolin(C14H14N8O4S3)or vancomycin(C66H75Cl2N9O24·xHCl).The characteristic microstructure of the polymer was evaluated using scanning electron microscopy and confocal *** on the surface treatment,the surface roughness varied(between 1.53μm and 2.06μm),and the wettability was change with the over of time.X-ray photoelectron spectroscopy analysis showed that the oxide layer did not affect the polymer layer or loaded ***,the drugs lose their stability in a phosphate-buffered saline solution after 6.5 h of exposure,and its decrease was greater than 7%(HPLC analysis).The stability,drug release and concentration of the drug loaded into the material were precisely analyzed by high-performance liquid *** results correlated with the degradation of the polymer in which the addition of drugs caused the percent of degraded polymer to be between 35.5%and 49.4%after 1 h of material immersion,depending on the mass of the loaded drug and various biological responses that were ***,all of the coatings were cytocompatible with MG-63 osteoblast-like *** drug concentrations released from the coatings were sufficient to inhibit adhesion of reference and clinical bacterial strains(***).The coatings with amoxicillin showed the best results in the bacterial inhibition zone,whereas coatings with cefazolin inhibited adhesion of the above bacteria on the surface.

Antimicrobial susceptibility of strains of Enterobacteriaceae isolated from bloodstream infections using current CLSI and EUCAST breakpoints

Understanding local susceptibility patterns is important when selecting antimicrobials for initial empirical antibiotic-therapy of bloodstream infections. Because the determination of susceptibility is dependent on the breakpoints used, the aim of the study was to compare the antimicrobial susceptibility results to different classes of antibiotics of 512 strains of Enterobacteriaceae (200 ESβL positive) isolated from bloodstream using CLSI 2013 and current EUCAST 2013 guidelines to evaluate the impact of break-point discrepancies. The results of the study showed that statistically significant discrepancies (p ≤ 0.001) were found for amoxicillin/clavulanic acid, piperacillin alone or with tazobac-tam, imipenem, meropenem, cefepime (only ESβL negative isolates), amikacin and gentamicin using current CLSI or EUCAST interpretive criteria. Further harmonization of CLSI and EUCAST breakpoints is warranted. This study could give useful information to physicians for managing bloodstream infections caused by Enterobacteriaceae.

Update on pre-diabetes: Focus on diagnostic criteria and cardiovascular risk

Pre-diabetes, which is typically defined as blood glucose concentrations higher than normal but lower than thediabetes threshold, is a high-risk state for diabetes and cardiovascular disease development. As such, it represents three groups of individuals: Those with impaired fasting glucose(IFG), those with impaired glucose tolerance(IGT) and those with a glycated haemoglobin(HbA1c) between 39-46 mmol/mol. Several clinical trials have shown the important role of IFG, IGT and HbA1c -pre-diabetes as predictive tools for the risk of developing type 2 diabetes. Moreover, with regard to cardiovascular disease, pre-diabetes is associated with more advanced vascular damage compared with normoglycaemia, independently of confounding factors. In view of these observations, diagnosis of pre-diabetes is mandatory to prevent or delay the development of the disease and its complications; however, a number of previous studies reported that the concordance between pre-diabetes diagnoses made by IFG, IGT or HbA1c is scarce and there are conflicting data as to which of these methods best predicts cardiovascular disease. This review highlights recent studies and cur-rent controversies in the field. In consideration of the expected increased use of HbA1c as a screening tool to identify individuals with alteration of glycaemic homeo-stasis, we focused on the evidence regarding the ability of HbA1c as a diagnostic tool for pre-diabetes and as a useful marker in identifying patients who have an increased risk for cardiovascular disease. Finally, we reviewed the current evidence regarding non-traditional glycaemic biomarkers and their use as alternatives to or additions to traditional ones.

Sensitization Properties of Propolis and Balsam of Peru in Guinea Pig Maximization Test(GPMT)

Introduction: Propolis is used in Poland as an active ingredient of some drugs administered externally, dietary supplements and cosmetics. According to the literature, propolis is a non-toxic and safe substance, although it may cause allergic contact dermatitis. Aim: The aim of this study was to assess the allergenic properties of propolis and Balsam of Peru. Material and methods: The study was conducted according to the OECD Guideline for testing of chemicals-Skin sensitization with use of Guinea pig maximization test (GPMT). Guinea pigs have similar sensitivity to allergens as human body. Sensitization properties of propolis were tested in comparison with sensitization properties of Balsam of Peru because of the possibility of cross-reaction between those two substances. Results: The skin of guinea pigs in the propolis group showed no visible change compared to the control group during the first (48 h) and second observation (72 h). The skin of guinea pigs from Balsam of Peru group showed discrete erythema in only one case at the first observation (8.33% of the animals). During the second observation, no visible changes were observed compared to the control group. Conclusions: Guinea pig maximization test showed no sensitizing properties of propolis and weak sensitizing properties of Balsam of Peru in concentration of 5%.

Comparative analysis of bed density, total phenol content and protein expression pattern in <i>Posidonia oceanica</i>(L.) Delile

Posidonia oceanicameadows are experiencing a progressive decline, and monitoring their status is crucial for the maintenance of theseecosystems. We performed a comparativeanalysis of bed density, total phenol content and protein expression pattern to assess the conservation status ofPosidoniaplants from the S. Marinella (Rome, Italy) meadow. The total phenol content was inversely related to maximum beddensity, confirming the relationship betweenhigh phenol content and stressful conditions. In addition, protein expression pattern profilesshowed that the number of differentially expressed proteins was dramatically reduced in the latest years compared to previous analyses. Our results support the usefulness of integrating solid descriptors, such as phenol content, with novel biochemical/molecular approaches in the monitoring of meadows.

翻译质量评估作为翻译研究中的一个重要分支,发展十分迅速。翻译质量评估关注的是原文和译文之间的关系。近年来,不少学者试图提出一种可靠的翻译质量评价模式,翻译的本质决定了这一领域有着广阔的研究和发展空间。本文首先介绍了翻译质量评估的发展历程,描述了自20世纪50年代以来最突出的翻译质量评估方法,这些方法有着不同的视角,包括卡特·福德(John Catford)的翻译“转换”、卡特琳娜·莱斯(Katharina Reiss)的文本类型和翻译目的论(Skopos theory)等,并提出了在设计和应用翻译质量评估模式时应注意的问题。本文采用朱莉安·豪斯(Juliane House)的翻译质量评估模式,来评估斯宾塞·约翰逊(Spencer Johnson)的《谁动了我的奶酪?》中译本的翻译质量。在《翻译质量评估:过去与现在》中,豪斯提出的基本概念是“对等”和“重新语境化”。豪斯提出了两种策略:显性翻译与隐性翻译,后者需要文化过滤工作。该模式还引入了及物性系统,以及韩礼德的语场、语旨、语式的语域概念。笔者在对原文和译文进行比较的基础上,对《谁动了我的奶酪?》的中译本进行质量评价,将译文分为显性或隐性,展示译者所使用的翻译策略,比较原文与译文的不同之处。除了对翻译质量进行评价和分析,笔者还将译文置于劳伦斯韦努蒂(Lawrence Venuti)的归化翻译法和异化翻译法的“光谱”中进行分析和研究。
