Schizophrenia-associated MicroRNA–Gene Interactions in the Dorsolateral prefrontal Cortex

Schizophrenia-associated anomalies in gene expression in postmortem brain can be attributed to a combination of genetic and environmental influences. Given the small effect size of common variants, it is likely that we may only see the combined impact of some of these at the pathway level in small postmortem studies. At the gene level, however, there may be more impact from common environmental exposures mediated by influential epigenomic modifiers, such as microRNA(miRNA). We hypothesise that dysregulation of miRNAs and their alteration of gene expression have significant implications in the pathophysiology of schizophrenia. In this study, we integrate changes in cortical gene and miRNA expression to identify regulatory interactions and networks associated with the disorder. Gene expression analysis in post-mortem prefrontal dorsolateral cortex(BA 46)(n = 74 matched pairs of schizophrenia, schizoaffective, and control samples)was integrated with miRNA expression in the same cohort to identify gene–miRNA regulatory networks. A significant gene–miRNA interaction network was identified, including miR-92 a, miR-495,and miR-134, which converged with differentially expressed genes in pathways involved in neurodevelopment and oligodendrocyte function. The capacity for miRNA to directly regulate gene expression through respective binding sites in BCL11 A, pLp1, and SYT11 was also confirmed to support the biological relevance of this integrated network model. The observations in this study support the hypothesis that mi RNA dysregulation is an important factor in the complex pathophysiology of schizophrenia.

Adult duodenal intussusception associated with congenital malrotation

Enteroenteric intussusception is a condition in which full-thickness bowel wall becomes telescoped into the lumen of distal bowel. In adults, there is usually an abnormality acting as a lead point, usually a Meckels＇ diverticulum, a hamartoma or a tumour. Duodeno- duodenal intussusception is exceptionally rare because the retroperitoneal situation fixes the duodenal wall. The aim of this report is to describe the first published case of this condition. A patient with duodeno-duodenal intussusception secondary to an ampullary lesion is reported. A 66 year-old lady presented with intermittent abdominal pain, weight loss and anaemia. Ultrasound scanning showed dilated bile and pancreatic ducts. CT scanning revealed intussusception involving the full-thickness duodenal wall. The lead point was an ampullary villous adenoma. Congenital partial （type r[） malrotation was found at operation and this abnormality permitted excessive mobility of the duodenal wall such that intussusception was possible. This condition can be diagnosed using enhanced CT. Intussusception can be complicated by bowel obstruction, ischaemia or bleeding, and therefore the underlying cause should be treated as soon as possible.

Numerous reports have identified a dysbiosis in the intestinal microbiota in patients suffering from inflammatory bowel diseases(IBD),yet the mechanism(s)in which this complex microbial community initiates or perpetuates inflammation remains *** purpose of this review is to present evidence for one such mechanism that implicates enteric microbial derived proteases in the pathogenesis of *** highlight and discuss studies demonstrating that proteases and protease receptors are abundant in the digestive ***,we investigate studies demonstrating an association between increased luminal protease activity and activation of protease receptors,ultimately resulting in increased intestinal permeability and exacerbation of colitis in animal models as well as in human *** are essential for the normal functioning of bacteria and in some cases can serve as virulence factors for pathogenic *** not classified as traditional virulence factors,proteases originating from commensal enteric bacteria also have a potential association with intestinal inflammation via increased enteric *** of increased protease activity in stools from IBD patients support a possible mechanism for a dysbiotic enteric microbiota in IBD.A better understanding of these pathways and characterization of the enteric bacteria involved,their proteases,and protease receptors may pave the way for new therapeutic approaches for these diseases.

HDpairFinder:A data processing platform for hydrogen/deuterium isotopic labeling-based nontargeted analysis of trace-level amino-containing chemicals in environmental water

The combination of hydrogen/deuterium(H/D)formaldehyde-based isotopic methyl labeling with solid-phase extraction and high-performance liquid chromatography–high resolution mass spectrometry(HpLC-HRMS)is a powerful analytical solution for nontargeted analysis of trace-level amino-containing chemicals in water *** the huge amount of chemical information generated in HpLC-HRMS analysis,identifying all possible H/Dlabeled amino chemicals presents a significant challenge in data *** address this,we designed a streamlined data processing pipeline that can automatically extract H/D-labeled amino chemicals from the raw HpLC-HRMS data with high accuracy and ***,we developed a cross-correlation algorithm to correct the retention time shift resulting from deuterium isotopic effects,which enables reliable pairing of H-and D-labeled ***,we implemented several bioinformatic solutions to remove false chemical features generated by in-source fragmentation,salt adduction,and natural13C ***,we used a data mining strategy to construct the AMINES library that consists of over 38,000 structure-disjointed primary and secondary amines to facilitate putative compound ***,we integrated these modules into a freely available R program,*** rationale of each module was justified and its performance tested using experimental H/D-labeled chemical standards and authentic water *** further demonstrated the application of HDpairFinder to effectively extract N-containing contaminants,thus enabling the monitoring of changes of primary and secondary N-compounds in authentic water *** is a reliable bioinformatic tool for rapid processing of H/D isotopic methyl labeling-based nontargeted analysis of water samples,and will facilitate a better understanding of N-containing chemical compounds in water.

Integration of solid phase extraction with HILIC-MS/MS for analysis of free amino acids in source water

Amino acids(AAs) are prevalent in source water, particularly during spring run-off. Monitoring of amino acids in source water is desirable for water treatment plants(WTp) to indicate changes in source water quality. The objective of this study was to establish analytical procedures for reliable monitoring of amino acids in source water. Therefore, we examined two different methods, large volume inject(LVI) and solid phase extraction(SpE), for sample preparation prior to HILIC-MS/MS. The LVI-HILIC-MS/MS method can provide fast and sensitive detection for clean samples, but suffers from matrix effects, resulting in irreproducible separation and shortening column lifetime. We have demonstrated that SpE was necessary prior to HILIC-MS/MS to achieve reproducible and reliable quantification of AAs in source water. A natural heterocyclic amine 1-methyl-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(MTCCA) was also included in the method to indicate changes in other natural nitrogenous compounds in source water. The SpE-HILIC-MS/MS method was able to achieve limits of detection from 2.6-3400 ng/L for the amino acids and MTCCA with RSDs( n = 3) of 1.1%-4.8%. As well, retention times(RT) of the analytes were reproducible with variation less than 0.01 min( n = 3) through the entire project. We further applied the SpE-HILICMS/MS method to determine AAs in authentic source water samples collected from two drinking water treatment plants(WTps) during the 2021 spring run-off season. The results support that the SpE-HILIC-MS/MS method does not require derivatization and can provide reliable, accurate, and robust analysis of AAs and MTCCA in source water, supporting future monitoring of source water quality.

A new global raster water mask at 250 m resolution

Accurate depiction of the land and water is critical for the production of land surface parameters from remote sensing data *** parameters,including the land surface temperature,active fires and surface reflectance,can be processed differently when the underlying surface is water as compared with *** errors in the underlying water mask can then pervade into these products and any products created from *** many global databases have been created to depict global surface *** databases still fall short of the current needs of the terrestrial remote sensing community working at 250 m spatial *** most recent attempt to address the problem uses the Shuttle Radar Topography Mission(SRTM)data set to create the SRTM Water Body Data set(SWBD 2005).The SWBD represents a good first step but still requires additional work to expand the spatial coverage to include the whole globe and to address some erroneous discontinuities in major river *** address this issue a new water mask product has been created using the SWBD in combination with MODIS 250 m data to create a complete global map of surface water at 250 m spatial *** effort is automated and intended to produce a dataset for use in processing of raster data(MODIS and future instruments)and for masking out water in final terrestrial raster data *** new global dataset is produced from remotely sensed data and provided to the public in digital format,free of *** data set can be found on the Global Land Cover Facility(GLCF)website at http://*** dataset is expected to be a base set of information to describe the surface of Earth as either land or water which is a fundamental distinction upon which other descriptions can be made.

Dear Editor, Approximately every fifth Arabidopsis T-DNA inser- tion line contains chromosomal translocations caused by the inserted T-DNA （Clark and Krysan, 2010）. Albeit broad use of T-DNA lines for mutant analysis, little is known about the consequences of these chromosomal rearrangements and only a few studies describe chromosomal aberrations in the mutant lines. While plant growth in general is not affected by such chromosomal translocations, defects in gametophyte development have been observed in lines that are heterozygous for the T-DNA insertion （Ray et al., 1997; Curtis et al., 2009）.
