Methyl gallate isolated from Mangifera pajang kernel induces proliferation inhibition and apoptosis in MCF-7 breast cancer cells via oxidative stress

Objective:To determine the lead bioactive compound in kernel extract of Mangifera pajang and its anti-cancer activity against human breast cancer cell lines with positive estrogen receptor(MCF-7).Methods:The methanolic extract of dried powder kernel of Mangifera pajang was exposed to column chromatography for *** structural elucidation of the isolated compound was characterized using infrared,nuclear magnetic resonance,mass ***,cytotoxicity,morphological changes,flow cytometry and cell cycle arrest analyses were performed to examine the mechanism of anti-proliferation and apoptosis induced by methyl gallate against ***:One compound was isolated from the methanolic extract of Mangifera pajang kernel and identified as methyl *** flow cytometric results demonstrated induction of apoptosis in MCF-7 cells by three concentrations of methyl *** cell cycle arrest showed a significant(P<0.05)decrease in cell progression at G2/M phase of MCF-7 after treatment with 100μM of methyl *** cell percentage of early and late apoptosis was significant at 10 and 100μM of methyl ***,methyl gallate treatment induced up-regulation of reactive oxygen species levels in MCF-7 cells with a reduction in superoxide dismutase ***:These findings indicate that isolated methyl gallate from Mangifera pajang kernel extracts induces growth inhibition and apoptosis in MCF-7 cells via up-regulating oxidative stress pathway.

In situations when the precise position of a machine is unknown,localization becomes *** research focuses on improving the position prediction accuracy over long-range(LoRa)network using an optimized machine learning-based *** order to increase the prediction accuracy of the reference point position on the data collected using the fingerprinting method over LoRa technology,this study proposed an optimized machine learning(ML)based *** signal strength indicator(RSSI)data from the sensors at different positions was first gathered via an experiment through the LoRa network in a multistory round layout *** noise factor is also taken into account,and the signal-to-noise ratio(SNR)value is recorded for every RSSI *** study concludes the examination of reference point accuracy with the modified KNN method(MKNN).MKNN was created to more precisely anticipate the position of the reference *** findings showed that MKNN outperformed other algorithms in terms of accuracy and complexity.

Objective:To determine the distribution,phenotypic and genetic background of extended spectrumβ-lactamases(ESBL)-producing Klebsiella(K.)pneumoniae clinical isolates associated with K1 and K2 serotypes in two selected hospitals in ***:A total of 192 *** isolates were collected and subjected to antibiotic susceptibility,hypermucoviscosity test and multiplex PCR to detect the presence of K1-and K2-serotype associated *** sequence typing(MLST)was performed on ESBL-producing *** isolates presented with K1 and K2 serotypes,followed by phylogenetic ***:A total of 87 out of 192(45.3%)of the *** isolates collected were ESBL ***,only 8.3%(16/192)and 10.9%(21/192)of the total isolates were detected to carry K1-and K2-serotype associated genes,*** analysis showed that K1 and K2 capsular serotypes were not significantly associated with ESBL phenotype(P=0.196).However,they were significantly associated with hypervirulent,as demonstrated by the positive string test(P<0.001).MLST analysis revealed that ST23 as the predominant sequence type(ST)in the K1 serotype,while the ST in the K2 serotype is more ***:Although the occurrence of ESBL-producing isolates among the hypervirulent strains was low,their coexistence warrants the need for continuous *** showed that these isolates were genetically heterogeneous.

Fused deposition modelling (FDM) is one of rapid prototyping (RP) technologies which uses an additive fabrication *** commercially available FDM model has different types of process parameters for different *** of the desired parts require excellent surface finish as well as good *** most common parameters requiring setup are the raster angle,tool path,slice thickness,build orientation,and deposition *** purpose of this paper is to discuss the process parameters of FDM Prodigy Plus (Stratasys,Inc.,Eden Prairie,MN,USA).Various selected parameters were tested and the optimum condition was *** quality of the parts produced was accessed in terms of dimensional accuracy and surface *** optimum parameters obtained were then applied in the fabrication of the master pattern prior to silicone rubber moulding (SRM).These parameters would reduce the post processing *** dimensional accuracy and surface roughness were analyzed using coordinate measuring machine (CMM) and surface roughness tester,*** on this study,the recommended parameters will improve the quality of the FDM parts produced in terms of dimensional accuracy and surface roughness for the application of SRM.

Vital roles of stem cells and biomaterials in skin tissue engineering

Tissue engineering essentially refers to technology for growing new human tissue and is distinct from regenerative medicine. Currently, pieces of skin are already being fabricated for clinical use and many other tissue types may be fabricated in the *** engineering was first defined in 1987 by the United States National Science Foundation which critically discussed the future targets of bioengineering research and its consequences. The principles of tissue engineering are to initiate cell cultures in vitro, grow them on scaffolds in situ and transplant the composite into a recipient in vivo. From the beginning, scaffolds have been necessary in tissue engineering applications. Regardless, the latest technology has redirected established approaches by omitting scaffolds. Currently, scientists from diverse research institutes are engineering skin without scaffolds. Due to their advantageous properties, stem cells have robustly transformed the tissue engineering field as part of an engineered bilayered skin substitute that will later be discussed in detail. Additionally, utilizing biomaterials or skin replacement products in skin tissue engineering as strategy to successfully direct cell proliferation and differentiation as well as to optimize the safety of handling during grafting is beneficial. This approach has also led to the cells' application in developing the novel skin substitute that will be briefly explained in this review.

In this study,the controlled nucleation and growth of gold nanoparticles(GNPs)were investigated using a self-repelled mist in a liquid chemical reaction *** electrospray-based chemical reduction method was conducted in the aqueous region and at room temperature to synthesize the polymeric-stabilized gold *** electrospray technique was used to atomize a hydrogen tetrachloraurate(III)(HAuCl4)precursor solution into electrostatically charged *** atomized droplets were dispersed in an aqueous reaction bath containing L-ascorbic acid as a reducing agent and polyvinylpyrrolidone(PVP)as a *** effect of the electrospray parameters,specifically the flow rate and electrospray droplet size,as well as the reaction conditions such as the concentration of reactants,pH,and stabilizer(PVP),were *** mean diameter of the GNPs increased from around 4 to 9 nm with an increase in the electrospray flow rate,droplet size,and current passing through the electrospray *** and monodispersed GNPs were synthesized at a relatively high flow rate of 2 mL/h and a moderate concentration of 2 mM of precursor *** smallest-sized GNP with a high monodispersity was obtained in the reaction bath at a high pH of 10.5 and in the presence of *** is expected that continuous and mass production of the engineered GNPs and other noble metal nanoparticles could be established for scaling up nanoparticle production via the proposed electrospray-based chemical reduction method.

Antibiotic resistance profile and RAPD analysis of Campylobacter jejuni isolated from vegetables farms and retail markets

Objective:To investigate antibiotic resistance profile and characterize Campylobacter jejuni(***) isolates using random amplified polymorphic DNA(RAPD) ***:Ninety eight *** isolates from farms and retail outlets were screened against 10 antibiotics commonly used clinically and agriculturally by using disk diffusion *** analysis was done to characterize 98 *** ***:Fifty-one percent of the isolates had multiple antibiotic resistance index 0.2 and *** indicated that the isolates in the vegetables were not from the high risk environment or extensive farming *** isolates found resistant towards penicillin G(93%),vancomycin(86%),ampicillin(35%),erythromycin(28%),gentamycin(4%),amikacin(3%),enrofloxacin(1%),norfloxacin(1%) and no resistance towards *** clustering analysis showed that the contamination of *** in vegetables was likely due to cross contamination at retail ***:*** contamination in vegetables at retail markets was due to cross *** finding proved that *** in small scale vegetables production was less expose towards antibiotic abuse.

In-vitro Cytotoxicity and Anti-microbial Evaluations of Silver Doped Titania Composite Coatings

Microbial adhesion is a main concern in the aspect of contamination especially in medical devices products. Titania coating on stainless steel substrate has been used as biomaterial application due to its non-toxicity, chemical stability and good mechanical properties. The purpose of this study is to evaluate the effect of 1-3 wt% of silver doped titania (Ag/TiO2) composite coating on; (1) cell viability using in-vitro model, L929 mouse fibroblasts cells, and (2) effect of anti-microbial activity using in-vitro model, Staphylococcus aureus. In this work, 1-3 wt% of Ag doped TiO2 coatings were deposited on stainless steel substrate using RF magnetron sputtering system. Different coating deposition times (1 h, 2 h, 4 h and 6 h) were used. The assessment on cell viability showed that all materials are non toxic on L929 cells. MTT assay study showed more than 80% of cell viable at 200 mg/ml, 100mg/ml, 50 mg/mL and 25 mg/mL. The anti-microbial activity on 6 h deposited 2 wt% of Ag doped TiO2 coating exhibit 99.9% reduction of Staphylococcus aureus after 24 h incubation. From these evaluations, 6 h deposited 2 wt% of Ag doped TiO2 coating indicate as a non-toxic material with excellent anti-bacterial properties.
