Development and Application of a Validated HPLC Method for the Determination of Clindamycin Palmitate Hydrochloride in Marketed Drug Products: An Optimization of the Current USP Methodology for Assay

A simple efficient isocratic reversed-phase HPLC method was developed and validated for the determination of clindamycin palmitate hydrochloride (CPH) and its commercially available oral solution products. Separation was achieved on a Phenomenex Zorbax (Luna) cyano column (150 × 4.6 mm, 5 μm) with a Phenomenex cyano guard cartridge (4 × 3.0 mm) on Agilent 1050 series HPLC system. CPH and its resolution standard lincomycin were eluted isocratically at a flow rate of 1 mL/min with a simplified mobile phase (potassium phosphate buffer (5 mM, pH 3.0)—acetonitrile—tetrahydrofuran (20:75:5, v/v/v)) and detected at 210 nm. The column was maintained at 25?C. The method was validated according to USP category I requirements. Robustness and forced degradation studies were also conducted. CPH marketed drug products were obtained from a drug distributor and assayed for potency using the validated method. Validation acceptance criteria were met in all cases. The analytical range for CPH was 15 - 500 μg/mL and the linearity was r2 > 0.999 over three days. The method was determined to be specific and robust. Both accuracy (92.0% - 103.8%) and precision (0.67% - 1.52%) were established across the analytical range for low, intermediate and high QC concentrations. Method applicability was demonstrated by analyzing two marketed products of CPH, in which results showed potency >98%. The method was determined to be an enhancement over the current USP methodology for assay as a result of increased efficiency, reduced organic solvents and the elimination of matrix modifiers. This method was successfully applied for the quality assessment of: 1) currently marketed drug products and 2) will in future assess the product quality of novel dosage forms of CPH for pediatric use.

Heat and Mass Transfer in Visco-Elastic Fluid through Rotating Porous Channel with Hall Effect

This paper examined the hydromagnetic boundary layer flow of viscoelastic fluid with heat and mass transfer in a vertical channel with rotation and Hall current. A constant suction and injection is applied to the plates. A strong magnetic field is applied in the direction normal to the plates. The entire system rotates with uniform angular velocity (Ω), about the axis perpendicular to the plates. The governing equations are solved by perturbation technique to obtain an analytical result for velocity, temperature, concentration distributions and results are presented graphically for various values of viscoelastic parameter (k2), Prandtl number (Pr), Schmidt number (Sc), radiation parameter (R), heat generation parameter (Qh) and Hall parameter (m).

Microtexture and Distribution of Minerals in Hydrothermal Barite-Silica Chimney from the Franklin Seamount, SW Pacific: Constraints on Mode of Formation

An extinct hydrothermal barite-silica chimney from the Franklin Seamount of the Woodlark Basin, in the southwestern Pacific Ocean, was investigated for mineral distribution and geochemical composition. Six layers on either side of the orifice of a chimney show significant disparity in color, mineral assemblage and major element composition. Electron microscope（SEM） images reveal that the peripheral wall of the chimney is composed of colloform silica, suggesting that incipient precipitation of silica-saturated hydrothermal fluid initiated the development of the chimney wall. Intermediate layers, between the exterior wall and the inner fluid-orifice, dominate with barite and sulfides. Low Sr-to-Ba ratios（SrO/BaO = 0.015–0.017） indicate restricted fluid-seawater mixing, which causes relatively high-temperature formation of the intermediate layers. Whereas the innermost layer bordering the chimney orifice is characterized by more silica and a higher Sr-to-Ba ratio（SrO/BaO = 0.023）, could have formed due to a paragenetic shift from a high-temperature active phase to a cooler waning stage of formation. A paragenetic shift is also probably responsible for the change in mineral formation mechanism that resulted in the textural variation of barite and colloform silica developed during different growth phases of this barite-silica chimney.

Benefits of the use of blood conservation in scoliosis surgery

AIM To investigate whether autologous blood transfusion(ABT) drains and intra-operative cell salvage reduced donor blood transfusion requirements during scoliosis *** Retrospective data collection on transfusion requirements of patients undergoing scoliosis surgery is between January 2006 and March 2010. There were three distinct phases of transfusion practice over this time: Group A received "traditional treatment" with allogeneic red cell transfusion(ARCT) in response to an intra- or postoperative anaemia(Hb < 8 g/d L or a symptomatic anaemia); Group B received intra-operative cell salvage in addition to "traditional treatment". In group C,ABT wound drains were used together with both intra-operative cell salvage and "traditional treatment".RESULTS Data from 97 procedures on 77 patients,there was no difference in mean preoperative haemoglobin levels between the groups(A: 13.1 g/d L; B: 13.49 g/d L; C: 13.66 g/d L). Allogeneic red cell transfusion was required for 22 of the 37 procedures(59%) in group A,17 of 30(57%) in group B and 16 of 30(53%) in group C. There was an overall 6% reduction in the proportion of patients requiring an ARCT between groups A and C but this was not statistically significant(χ2 = 0.398). Patientsin group C received fewer units(mean 2.19) than group B(mean 2.94)(P = 0.984) and significantly fewer than those in group A(mean 3.82)(P = 0.0322). Mean length of inpatient stay was lower in group C(8.65 d) than in groups B(12.83) or A(12.62).CONCLUSION When used alongside measures to minimise blood loss during surgery,ABT drains and intra-operative cell salvage leads to a reduced need for donor blood transfusion in patients undergoing scoliosis surgery.

The gut microbiome plays a key role in the pathogenesis and disease activity of inflammatory bowel disease(IBD).While research has focused on the bacterial microbiome,recent studies have shifted towards host genetics and host-fungal *** mycobiota is a vital component of the gastrointestinal microbial community and plays a significant role in immune *** fungi,Candida species,particularly Candida albicans(***),have been extensively studied due to their dual role as gut commensals and invasive *** findings indicate that various strains of *** exhibit consid-erable differences in virulence factors,impacting IBD's *** fungal dysbiosis and antifungal mucosal immunity may be associated to IBD,especially Crohn's disease(CD).This article discusses intestinal fungal dysbiosis and antifungal immunity in healthy individuals and CD *** discusses factors influencing the mycobiome's role in IBD pathogenesis and highlights significant contributions from the scientific community aimed at enhancing understanding of the mycobiome and encouraging further research and targeted intervention studies on specific fungal *** article also provided insights into a recent study by Wu et al in the World Journal of Gastroenterology regarding the role of the gut microbiota in the pathogenesis of CD.

目的:证实西印度醋栗(Phyllanthus acidus(L.)Skeels)的果实在传统医学中作为保肝药使用的药用用途。方法:连续5d分别给予各组大鼠西印度醋栗果实的70%乙醇提取物(100,200和400mg/kg口服)及对照药水飞蓟素(100mg/kg口服)并于第2天及第3天皮下注射四氯化碳(2mL/kg)。测定血清天冬氨酸氨基转移酶(aspartate transaminase,AST)、丙氨酸氨基转移酶(alanine transaminase,ALT)、碱性磷酸酶(alkaline phosphatase,ALP)、总胆红素以及总蛋白并进行肝脏组织病理学检测。检测大鼠肝组织匀浆中的氧化应激标记物如脂质过氧化反应(lipid peroxidation,LPO)、还原型谷胱甘肽(reduced glutathione,GSH)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)以及谷胱甘肽过氧化物酶(glutathione peroxidase,GPx)以衡量西印度醋栗的体外抗氧化作用。此外,本实验还记录了伊维巴诱导的给药后小鼠睡眠时间,使用2,2-二苯基-1-苦基肼(2,2-diphenyl-1-picrylhydrazil,DPPH)法测定小鼠服药后的自由基清除率。结果:与对照组相比,西印度醋栗果实的乙醇提取物及水飞蓟素均显著降低了模型大鼠血清AST、ALT和ALP水平及LPO并升高了TP、GSH、SOD、CAT和GPx的水平(P<0.01或P<0.05)。大鼠肝脏组织的组织病理学切片结果也证实了以上实验结果。在小鼠实验中,与对照组相比,西印度醋栗果实的乙醇提取物显著缩短了伊维巴诱导的小鼠睡眠时间(P<0.01),并具有较高的DPPH清除率。结论:本研究的实验结果证实了西印度醋栗果实对四氯化碳引起的大鼠急性过氧化肝损伤的保护作用,机制可能与其抗氧化作用及自由基清除功能有关。

Enhancement of Phase Matching Ability in One-Dimensional Photonic Crystal through Plasma Materials

We have theoretically studied the modal dispersion equation and effective refractive index of one-dimensional plasma photonic crystals (1-D PPCs) having different materials in one unit cell. The dispersion relations related for such structure is derived by solving Maxwell’s equation using the transfer matrix method. It is found that the presence of plasma in a unit cell enhanced the phase matching ability and provides additional degree of freedom to control phase matching condition compared to the conventional one-dimensional photonic crystals (1-D PCs).
