Alkaloids Production from Callus of Hyoscyamus niger L, in Vitro

Callus cultures of Hyoscyamus niger L. were initiated from leaf segments cultured on Murashige and Skoog （MS） medium supplemented with 0.5 mg/L Benzyl Adenine （BA） and 0, 1, 2 and 3 mg/L Naphthalene acetic acid （ NAA ）. Half of cultures were incubated under light of 16 hr/day, while the other half was incubated under complete darkness. The incubation temperature was 25 ± 1 ℃in both incubation conditions. The fresh and dry weight of the produced callus was obtained after six weeks of incubation. Callus produced were recultured on medium that gave the highest production of callus. Constant weight （300 mg） of callus was cultured in each of these medium supplemented with abiotic elicitor of 50 g/L sucrose, 200 mg/L NaCI, 50 or 100 mg/L proline and 2 mg/L BA each one added separately and incubated under complete darkness. The fresh and dry weights of callus were measured after six weeks. HPLC was used to determine the tropane alkaloids （Hyoscyamine and Scopolamine）. The results showed that the significant highest average of fresh and dry weight of callus （112 and 89.6 mg） achieved using the medium contained 0.5 mg/L BA and 2 mg/L NAA under dark condition. The amount of fresh and dry weight of callus produced under dark condition was significantly higher than that produced under light condition, with increase in percentage of 51.3 and 37.62% respectively. The addition of abiotic elicitors caused reduction in both fresh and dry weight of callus, therefore the highest fresh weight average was 1,727 mg using 100 mg/L proline. The results indicated that addition of 50 or 100 mg/L proline led to increase in Hyoscyamine concentration of 58.03 and 21.37% respectively compared with the control. While other abiotic elicitors were found to cause reduction in Hyoscyamine concentration. The major awkwardness when joining AA2014 lies in the presence of creation of oxide films and brittle intermetallic in the bond region. However, diffusion bonding can be used to join these alloys without much difficulty. This paper reveals the characterization of diffusion connection of aluminum alloy AA 2014 with and without applying copper powder. Joining was achieved in vacuum up to (1 × 10-5 mbar) by means of vacuum system. Aluminum alloy AA2014 samples were done as cylinder form and interlayer thickness was equal 100 μm, under joining conditions of (325°C - 475°C) and 4 MPa and period of (1 hour). The ultimate strength was found equal to 188.36 MPa. Bonding joint was examined using light microscope, (SEM) with (EDS) for determining microstructure and depth of diffusion and microhardness was also used at connection zone. XRD outcomes display that the main intermetallic compound phase formula was Al2C. The maximum depth of diffusion obtained was 19.1 μm.

Validation of High Performance Liquid Chromatography with Fluorescence Detector Methods for Determination of Aflatoxins in Different Food and Animal Feed Samples

Method validation for quantitative analysis of aflatoxins, AFB1, AFB2, AFG1 and AFG2 in sorghum, peanut butter, groundnut and animals feed is presented. Aflatoxins were extracted with a mixture of methanol: acetonitrile: water (60:30:10) and cleaned with Alfa test IAC chromatography before analysis by High Performance Liquid Chromatography coupled with fluorescence detection (HPLC-FLD) by adopting an isocratic chromatographic system using a mobile phase consisting acetic acid: acetonitrile: methanol (59:14:27), the separation of the four aflatoxins was achieved in less than 15 minutes. Calibration curves were linear over the concentration range from 2 - 18 ng/mL for AFB1 and AFG1, 0.4 - 3.6 ng/mL for AFB2 and AFG2, respectively. The LOD and LOQ in spiked samples were found to be 0.02 and 0.05 μg/kg for both AFB1 and AFG1, 0.01 and 0.03 μg/kg for both AFB2 and AFG2. The mean recovery values were in range from 84.2% to 96.9% was obtained. Five samples were found to be contaminated with aflatoxins and the total aflatoxins ranged from 0.02 to 3.26 μg/kg were obtained. Nineteen different samples were found to be contaminated with aflatoxins;the total aflatoxins ranged from 0.27 to 10.48 μg/kg were obtained. The highest total aflatoxins value was obtained in animal feeds.

Porous silicon has been produced in this work by photochemical etching process (PC).The irradiation has been achieved using ordinary light source (150δ250 W) power and (875 nm) wavelength. The influence of various irradiation times and HF concentration on porosity of PSi material was investigated by depending on gravimetric measurements. The I-V and C-V characteristics for CdS/PSi structure have been investigated in this work too.

Geographical Information Science (GIS) technologies have been used increasingly for ecology and epidemiology of water-borne diseases, providing approach for animal health issues. This study was set up to investigate the geographical distribution of Bovine that was affected by Leptospira hardijo, in River Nile state, on October 2012. Locations of targeted cattle were delimited using GPS. Fifty three (53) of blood samples were collected, and screened in the laboratory for Leptospira hardijo specific antibodies using indirect ELISA. 15.09% had evidence of infection as determined by the presence of anti-leptospira antibodies. It was inferred that no incidences were recorded in 45 locations out of the 53 selected locations in the state. Leptospirosis risk area for transmission was mapped using 5 km buffer distance. Animals’ movements routes were mapped with their contacts area and positive samples locations, hafair locations where animals contacts were mapped. This study demonstrated the value of GIS and GPS in disease mapping for animals’ health, and this might help veterinary authorities to implement strategic interventions for animal disease control.

The present study focuses on determination of alpha particles concentration in different samples of toothpaste by using nuclear track detector (CR-39). Nine samples of toothpaste have been collected from local Iraqi markets in Babylon Province. These samples have been chosen according to their most commonly use in Babylon Province. The samples were kept with CR-39 detector for six weeks period and then etched by 6.25 N of NaOH solution at temperature of 60°C for 5 hours. The results of the present work show that Sample 3 (Frosty) found to have a high alpha concentration about four times compared with Sample 4 (Aquafresh) and about 2 - 3 times compared with other samples.

Background: The local population in developing countries is at higher risk of contracting hepatitis B (HBV) due to some of the factors they practice which promote its establishment and distribution within their communities and may constitute a burden for them. Objective: This study aimed to determine the prevalence and risk factors associated with the hepatitis B virus in populations of the Algamosi region, Gezira State, central Sudan. Method: A cross-sectional study was conducted among 492 people, populations of the Algamosi region, Gezira state in central Sudan. A questionnaire was used to collect information on socio-demographic factors, transmission, and prevention of hepatitis B infections. As well, blood samples were taken from each participant, and serum was used for rapid tests for HBsAg. Positive samples were then tested by the ELISA method for confirmation. Data were obtained using SPSS version 21 and a P value of less than 0.05 was considered statistically significant. Results: The prevalence of hepatitis B was 16.10% and was higher in men than in women (05.7 - 1.00). People aged 31 to 45 had the highest prevalence of 27 (20,600%) for HBV infection. There was a significant association between HBV and age groups (X2 = 7.816 and P value = 0.05). This study also found that there was a significant association between viral hepatitis and knowledge about transmission and prevention (P value Conclusion: Although most people who live in developing countries are at high risk for transmission of hepatitis B, this indicates the need to implement the screening policy and integrate it with other health services and create awareness such as proper treatment and surveillance for hepatitis infection in all nations.

Hepatitis C (HCV) remains a global health challenge and is transmitted via contact with contaminated blood and body fluids. This study aimed To find the factors influencing hepatitis c viral infections and to identify its prevalence among populations of the Algamosi area, Gezira state central Sudan. This cross-sectional community-based study was conducted. A total of 492 participants were randomly selected from among 4 villages and were interviewed to find out the factors that lead to transmission of infection and tested for markers of hepatitis C infection. ELISA confirmed all the positive card tests. The results showed that the individuals within the age groups > 45 and 31 - 45 years are more affected by HCV the percentage was 3.37% and 0.6% respectively, while the age groups of 15 - 30 and p 2 = 14.588 and p value = 0.001) The study concluded that the prevalence of hepatitis C viruses was only detected in three areas. This study explored many possible causes associated with viral hepatitis, tattooing is the common cause of hepatitis C virus. Based on the finding the study recommends, the crucial intervention program to screen most of the Algamosi population who have been not screened for HCV, proper treatment for HCV, and the health education program is strongly recommended to create awareness among the general population.

The aim of this study was to assess Acacia senegal trees’ characteristics as well as evaluate the carbon stock under a variety of ages in the El Demokeya forest in Sudan, where the Gum Arabic belt is located. 12 sample plots, in 2021 were randomly distributed to represent the entire area of the forest prior to the required measurements. The sample was designed as squire plots with one hectare. In each sample plot, all trees were counted, their height (m), and Diameters Breast Height (DBH in cm), respectively. The results showed the highest number of trees per ha at age 20 years old and the lowest number at age 47 years, while the highest values of DBH and volume were found at age 47 years old. As a result, the maximum and minimum values of the aboveground biomass were found in the age 47 years old and 16 years, accounting for 19.87 tons and 1.9 tons respectively. Thus, the amount of carbon stock was 11.92 tons/ha in the 35-years-old and 1.19 tons/ha in the 21-year stands. Furthermore, the average carbon stock in all plots was estimated as 18.70 tons/ha and hence the total carbon stock in the El Demokeya forest is equal to 620.11 tons. Conclusively, the characteristics of trees, amount of aboveground biomass and carbon stock in the El Demokeya forest varied among the uneven-aged plantation groups. The study recommends and encourages the protection of A. senegal in order to increase the carbon sink as well as protect the environment in the era of climatic changes.
