Effect of hydro-alcoholic extract of Olea europaea on apoptosis-related genes and oxidative stress in a rat model of torsion/detorsion-induced ovarian damage

Objective: To evaluate the impact of Olea (O.) europaea extract on markers of oxidative stress and apoptosis of ovarian tissues in a rat model of torsion/detorsion-induced ovarian damage. Methods: A total of 28 Wistar female rats were randomly assigned into 4 groups, with 7 rats in each group. The sham group received a 2.5 cm longitudinal incision in the midline part of the abdomen which was then sutured with 5-0 nylon thread;the torsion/detorsion group underwent torsion induction for 3 h followed by reperfusion for 10 days;the torsion/detorsion+O. europaea group received 300 mg/kg hydro-alcoholic extract of O. europaea 30 min before detorsion, followed by reperfusion for 10 days;and the O. europaea group only received 300 mg/kg hydro-alcoholic extract of O. europaea for 10 days. After the treatment period, blood samples were taken;the levels of estrogen, glutathione peroxidase, superoxide dismutase, and malondialdehyde were assayed. The histological changes, as well as the rate of apoptosis in ovarian tissues, were also carried out by histomorphometric analysis at day 10 post-procedure. Results: Histological comparisons demonstrated a significant detrimental change in the torsion/detorsion group as compared with other groups. The number of pre-antral and antral follicles and corpus luteum was significantly decreased in the torsion/detorsion group compared with the sham group, while treatment with O. europaea could enhance their numbers (P<0.05). The index of apoptosis and the number of atretic body in the ovarian tissue were significantly higher in the torsion/detorsion group compared with the sham group (P<0.05). The concentrations of glutathione peroxidase, estrogen, and superoxide dismutase as well as the mRNA expression of Bcl-2 were considerably diminished in the torsion/detorsion group while they were elevated in the torsion/detorsion+O. europaea group (P<0.05) compared with the torsion/detorsion group. The present study investigated the potential probiotic effect of Enterococcus faecalis against a pathogenic microorganism,Vibrio harveyi occurring in the giant freshwater prawn(Macrobrachium rosenbergii).*** was isolated from the intestine of *** through biochemical tests and PCR-based molecular *** in-vitro challenge was conducted by mixing isolated probiotic(5.12 log CFU/g)and ***(3.09 log CFU/g).Feeds incorporated with ***(9.02 log CFU/g)were applied to the treatment group during in vivo test in the aquarium and earthen pond(70 m^(2),1 m depth).Pathogenic bacteria were found to be reduced after 8 h(from 5.02 to 3.62 log CFU/g)and 60 days(from 5.44 to 2.15 log CFU/g)of probiotic administration in the in vitro and in vivo test,*** *** mixed feed also showed higher weight gain(WG%,481.22%±17.71%;SGR%,2.93%±0.18%)and digestive enzymes activities(amylase,1.26±0.08 unit/mg;protease,2.78±0.02 unit/mg)compared to control group(WG%,371.31%±13.35%;SGR%,2.30%±0.32%;amylase,0.7±0.03 unit/mg;protease,1.82±0.02 unit/mg).Furthermore,the probiotic also improved the immune response by augmenting NGH(from 73.33%±0.23%to 74.63%±0.11%)and SGH(from 22.24%±0.06%to 23.13%±0.08%)in the treatment ***,*** could be recommended to use against bacterial infections of the ***.

The adaptive treatment tolerance(ATT) of cancer cells is the main encumbrance to cancer chemotherapy.A potential solution to this problem is to treat cancer cells with multiple drugs using nanoparticles(NPs).In this study, we tested the co-administration of curcumin(Cur) and doxorubicin(Dox) to MCF-7 resistant breast cancer cells to block the ATT and elicit efficient cell killing. Drugs were co-administered to cells both sequentially and simultaneously. Sequential drug co-administration was carried out by pre-treating the cells with albumin nanoparticles(ANPs) loaded with Cur(Cur@ANPs) followed by treatment with Dox-loaded ANPs(Dox@ANPs). Simultaneous drug co-administration was carried out by treating the cells with ANPs loaded with both the drugs(Cur/Dox@ANPs). We found that the simultaneous drug co-administration led to a greater intra-cellular accumulation of Dox and cell killing with respect to the sequential drug co-administration. However, the simultaneous drug co-administration led to a lower intracellular accumulation of Cur with respect to the sequential drug co-administration. We showed that this result was due to the aggregation and entrapment of Cur in the lysosomes as soon as it was released from Cur@ANPs, a phenomenon called lysosomotropism. In contrast, the simultaneous release of Dox and Cur from Cur/Dox@ANPs into the lysosomes led to lysosomal p H elevation, which, in turn, avoided Cur aggregation, led to lysosome swelling and drug release in the cytosol, and finally provoked efficient cell killing. Our study shed the light on the molecular processes driving the therapeutic effects of anti-cancer drugs co-administered to cancer cells in different manners.

Somnambulism: Varenicline-Induced Sleep Driving

Background: Varenicline, which is derived from the cytisine compound, is used to assess in smoking cessation. Sleep driving (SD) is often classified as a variant of somnambulism. Although, somnambulism has been reported as a side effect of varenicline, varenicline-induced sleep driving (VISD) has not been reported. Case Report: A 56-year-old man with a history of 35 pack year smoking who presented for smoking cessation counseling. Treatment with varenicline was initiated. In the following night, the patient drove to a shopping center and woke up in his car. Varenicline was discontinued. SD has not been reported in the following nights. In a repeated attempt to stop smoking, treatment with varenicline was resumed by the patient. SD reoccurred in the following night. Varenicline was discontinued indefinitely, and SD has not been reported.

Hydrothermal synthesis and characterization of carbon-doped TiO2 nanoparticles

We report the hydrothermal growth of pure and doped TiO2 nanoparticles with different concentrations of *** microstructure of the as-synthesized samples is characterized by x-ray diffraction(XRD),field emission scanning electron microscopy(FESEM),energy dispersive x-ray spectroscopy(EDX),and Raman spectroscopy to understand the structure and *** XRD patterns confirm the formation of anatase phase of TiO2 with the average crystallite size is calculated to be in the range of 13 nm to 14.7 *** functional groups of these nanostructures are characterized by Fourier transformed infrared(FT-IR)spectroscopy,which further confirms the single anatase phase of the synthesized ***-visible absorption spectroscopy is used to understand the absorption behavior,which shows modification in the optical bandgap from 3.13 eV(pure TiO2)to 3.74 eV(1.2 mol%C-doped TiO2).Furthermore,the Ti^3+centers associated with oxygen vacancies are identified using electron paramagnetic resonance spectroscopy(EPR).

The Evolution of Iron-Related Comorbidities and Hospitalization in Patients with Hemochromatosis:An Analysis of the Nationwide Inpatient Sample

Objective:To synthesize silver nanopaticles from leaves extract of Eucalyptus chapmaniana(***)and test the antimicrobial of the nanoparticles against different pathogenic bacteria,yeast and its toxicity against human acute promyelocytic leukemia(HL-60)cell ***:Ten milliliter of leaves extract was mixed with 90 mL of 0.01 mmol/mL or 0.02 mmol/mL aqueous AgNO_3 and exposed to sun light for 1 h.A change from yellowish to reddish brown color was *** using UV-vis spectrophotometery and X-ray diffraction analysis were *** activity against six microorganisms was tested using well diffusion method and cytoxicity test using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide,a yellow tetrazole was obtained on the human leukemia cell line(HL-60).Results:UV-vis spectral analysis showed silver surface plasmon resonance band at 413 nm.X-ray diffraction showed that the particles were crystalline in nature with face centered cubic structure of the bulk silver with broad beaks at 38.50°and 44.76°.The synthesized silver nanoparticles efficiently inhibited various pathogenic organisms and reduced viability of the HL-60 cells in a dose-dependent ***:It has been demonstrated that the extract of *** leaves are capable of producing silver nanoparticles extracellularly and the Ag nanoparticles are quite stable in *** studies are needed to fully characterize the toxicity and the mechanisms involved with the antimicrobial and anticancer activity of these particles.
