Aldosterone-induced mesangial cell proliferation is mediated by EGF receptor transactivation

First published April 1, 2009; doi: 10.1152 / ajprenal. 90428.2008. -Aldosterone (Aldo) stimulates glomerular mesangial cell (mC) proliferation, in part, through an ERK1 / 2-dependent pathway. In this study, we examined whether Aldo activation of ERK1 / 2 in mC is mediated through redox-dependent EGF receptor (EGFR) transactivation, as well as the involvement of other signaling mechanisms in Aldo-induced mC proliferation. Aldo increased human mC proliferation, as determined by [H-3]thymidine incorporation and cell counts. This increase in proliferation was blocked by inhibition of the mineralocorticoid receptor (mR). Continuing our observations downstream in the signaling pathway, we examined the ability of Aldo to activate both the Ras / mAPK and the PI3K signaling pathways. Aldo increased Ki-RasA and Ki-RasA:GTP levels, and sequentially phosphorylated c-Raf, mAPK kinase (mEK1 / 2), and ERK1 / 2. Ki-RasA small interfering RNA (siRNA), the c-Raf inhibitor GW5074, and the mEK1 / 2 inhibitor PD98059 reduced Aldo-induced cell proliferation by similar to 65%. Aldo also increased phosphorylation of PI3K, Akt, the mammalian target of rapamycin (mTOR), and the 70-kda ribosomal S6 kinase (p70S6K1). Inhibition of the PI3K pathways by the selective PI3K inhibitor LY 294002, an Akt inhibitor, or the mTOR inhibitor rapamycin reduced cell proliferation by 51% . Combining LY 294002 and PD98059 completely blocked Aldo-induced mC proliferation. Next, we confirmed that Aldo exerts its effect on mAPK and PI3K activation, as well as on cell proliferation, by activating the EGFR. Pretreatment with the EGFR antagonist AG1478 inhibited mC proliferation, as well as the activation of Ras / mAPK and PI3K / Akt, suggesting that Ras / mAPK and PI3K / Akt activation occur downstream of EGFR activation. Role of colectomy in preventing recurrent primary sclerosing cholangitis in liver transplant recipients

AIm To study the published evidence on the impact of colectomy in preventing recurrent primary sclerosing cholangitis(r PSC). mETHODS An unrestricted systematic literature search in Pub med, EmBASE, medline Ovid SP, ISI Web of Science, Lista(EBSCO) and the Cochrane library was performed on clinical studies investigating colectomy in liver transplantation(LT) recipients with and without r PSC in the liver allograft. Study quality was evaluated according to a modification of the methodological index for non-randomized studies(mINORS) criteria. Primary endpoints were the impact of presence, timing and type of colectomy on r PSC. Overall presence of inflammatory bowel disease(IBD), time of IBD diagnosis, posttransplant IBD and immunosuppressive regimen were investigated as secondary *** The literature search yielded a total of 180 publications. No randomized controlled trial was identified. Six retrospective studies met the inclusion criteria of which 5 studies were graded as high quality articles. Reporting of IBD was heterogenous but in four publications, either inflammatory bowel disease, ulcerative colitis or in particular active colitis post-LT significantly increased the risk of r PSC. The presence of an intact(i.e., retained) colon at LT was identified as risk factor for r PSC in two of the high quality studies while four studies found no effect. Type of colectomy was not associated with r PSC but this endpoint was underreported(only in 33% of included studies).Neither tacrolimus nor cyclosporine A yielded a significant benefit in disease recurrence of primary sclerosing cholangitis(PSC).CONCLUSION The data favours a protective role of pre-/peri-LT colectomy in r PSC but the current evidence is not strong enough to recommend routine colectomy for r PSC prevention.

Objective Human can focus attention to one sensory modality(*** or visual) and reach a biased attentional state in a top-down manner,which will facilitate information processing in that sensory *** activity in human EEG was related to this intersensory selective attention in previous *** we study the time course of intersensory selective attention by analyzing alpha-band ERD(event-related desynchronization) during the *** Ten subjects(4 females) with a mean age of(21.3±2.7) years participated in the ***-target paradigm was used,in which there were 3 kinds of cues:auditory(A),visual(V) and audiovisual(AV).In each trial,the cue was firstly presented on a display and the subject was required to direct his/ her attention to a sensory modality as *** a variable cue-target interval(CTI),a compound audiovisual stimulus appeared as the *** performed a GO/NoGo task to detect the cued sensory part of the target in A or V condition,and detect both sensory parts in AV *** EEG data were recorded from 30 scalp electrodes mounted in an elastic cap(EasyCap,BrainAmp amplifier,Brain Products GmbH,munich,Germany). This paper discusses the design of a wideband low noise amplifier（LNA） in which specific architecture decisions were made in consideration of system-on-chip implementation for radio-astronomy *** LNA design is based on a novel ultra-low noise InGaAs/InAlas/InP *** and non-linear modelling of this pHEmT has been used to design an LNA operating from 2 to 4 GHz.A common-drain in cascade with a common source inductive degeneration,broadband LNA topology is proposed for wideband *** proposed configuration achieved a maximum gain of 27 dB and a noise figure of 0.3 dB with a good input and output return loss（S11〈—10 dB,S22〈—11 dB）.This LNA exhibits an input 1-dB compression point of-18 dBm,a third order input intercept point of 0 dBm and consumes 85 mW of power from a 1.8 V supply.

Aim： Sperm chromatin/DNA integrity is essential for the accurate transmission of paternal genetic information, and normal sperm chromatin structure is important for sperm fertilizing ability. The routine examination of semen, which includes sperm concentration, motility and morphology, does not identify defects in sperm chromatin structure. The origin of sperm DNA damage and a variety of methods for its assessment are described. Evaluation of sperm DNA damage appears to be a useful tool for assessing male fertility potential both in vivo and in vitro. The possible impact of sperm DNA defects on the offspring is also discussed. （Asian J Androl 2006 Jan; 8： 11-29）

Jasmonates （JAs） are plant hormones with essential roles in plant defense and development. The basic- helix-loop-helix （bHLH） transcription factor （TF） mYC2 has recently emerged as a master regulator of most aspects of the jasmonate （JA） signaling pathway in Arabidopsis. mYC2 coordinates Ja.mediated defense responses by antagonistically regulating two different branches of the JA signaling pathway that determine resistance to pests and pathogens, respectively. mYC2 is required for induced systemic resistance （ISR） triggered by beneficial soil microbes while mYC2 function is targeted by pathogens during effector-mediated suppression of innate immunity in roots. Another notable function of mYC2 is the regulation of crosstalk between the signaling pathways of JA and those of other phytohormones such as abscisic acid （ABA）, salicylic acid （SA）, gibberellins （GAs）, and auxin （IAA）. mYC2 also regulates interactions between JA signaling and light, phytochrome signaling, and the circadian clock, mYC2 is involved in JA-regulated plant development, lateral and adventitious root formation, flowering time, and shade avoidance syndrome. Related bHLH TFs mYC3 and mYC4 also regulate both overlapping and distinct mYC2-regulated functions in Arabidopsis while mYC2 orthologs act as ＇master switches＇ that regulate Ja.mediated biosynthesis of secondary metabolites. Here, we briefly review recent studies that revealed mechanistic new insights into the mode of action of this versatile TF.

metabolic syndrome(mS)is associated with a sedentary and stressful lifestyle and affects underactive people *** is considered to be a low-impact mind-body stress-relieving exercise,and researchers are increasing their focus on the benefits of yoga for managing metabolic *** is also important for physicians and health care professionals to understand the therapeutic efficacy of yoga intervention,in terms of its type,duration and frequency on various mS risk *** present review summarizes the current scientific understanding of the effects of yoga on mS risk factors such as glucose homeostasis markers,lipid profile,adipocytokines and cardiovascular risk factors,and discusses the possible mechanisms of ***,Pub med,Scopus and Cochrane Library were searched from their inception up to December 2019,using the keywords“metabolic syndrome,”“diabetes,”“cardiovascular diseases,”“obesity”and“yoga.”The literature summarized in this review have shown mixed effects of yoga on mS risk factors and do not provide robust evidence for its *** rigorous research and well-designed trials that have a higher standard of methodology and evaluate yoga's long-term impacts on mS are *** yoga's biochemical and molecular mechanisms of action on various metabolic pathways is also needed.
