Exacerbated VEGF up-regulation accompanies diabetes-aggravated hemorrhage in mice after experimental cerebral ischemia and delayed reperfusion

Reperfusion therapy is the preferred treatment for ischemic stroke,but is hindered by its short treatment window,especially in patients with diabetes whose reperfusion after prolonged ischemia is often accompanied by exacerbated hemorrhage.The mechanisms underlying exacerbated hemorrhage are not fully understood.This study aimed to identify this mechanism by inducing prolonged 2-hour transient intraluminal middle cerebral artery occlusion in diabetic Ins2Akita/+mice to mimic patients with diabetes undergoing delayed mechanical thrombectomy.The results showed that at as early as 2 hours after reperfusion,Ins2Akita/+mice exhibited rapid development of neurological deficits,increased infarct and hemorrhagic transformation,together with exacerbated down-regulation of tight-junction protein ZO-1 and upregulation of blood-brain barrier-disrupting matrix metallopeptidase 2 and matrix metallopeptidase 9 when compared with normoglycemic Ins2+/+mice.This indicated that diabetes led to the rapid compromise of vessel integrity immediately after reperfusion,and consequently earlier death and further aggravation of hemorrhagic transformation 22 hours after reperfusion.This observation was associated with earlier and stronger up-regulation of pro-angiogenic vascular endothelial growth factor(VEGF)and its downstream phospho-Erk1/2 at 2 hours after reperfusion,which was suggestive of premature angiogenesis induced by early VEGF up-regulation,resulting in rapid vessel disintegration in diabetic stroke.Endoplasmic reticulum stress-related pro-apoptotic C/EBP homologous protein was overexpressed in challenged Ins2Akita/+mice,which suggests that the exacerbated VEGF up-regulation may be caused by overwhelming endoplasmic reticulum stress under diabetic conditions.In conclusion,the results mimicked complications in patients with diabetes undergoing delayed mechanical thrombectomy,and diabetes-induced accelerated VEGF up-regulation is likely to underlie exacerbated hemorrhagic transformation.Thus,suppression of the VEGF pathway could b

High-resolution multi-scale air pollution system:Evaluation of modelling performance and emission control strategies

A high percentage of the world's population lives in areas where air pollutant concentrations exceed the World Health Organization guidelines.This work aims to develop and test,a high-resolution multi-scale air pollution modelling system by integrating a set of adequate tools.This system is able to provide detailed air pollutant concentrations in urban areas and support air quality management strategies through a better identification of different atmospheric processes.It also allows furthering the design and assessment of air pollution control measures for a specific area.To evaluate its performance and suitability,the system was applied to the Macao Special Administrative Region(SAR),China,one of the most densely populated areas on earth,during a winter period when this area is affected by high levels of Particulate Matter(PM).Although the developed system tends to underestimate the PM concentrations,it revealed a good performance in reproducing the temporal and spatial air pollution patterns.Several exceedances of the Chinese air quality standards were calculated and high population exposure to PM pollution was estimated.The tested urban atmospheric emission reduction scenarios have shown air quality improvements,indicating that emission reduction measures at urban level should focus on the domestic sector.However,it is crucial to implement joint pollution prevention strategies with neighbouring regions to improve the air quality in Macao SAR.The approach developed in this work can support policymakers in defining new strategies to reduce atmospheric pollution in urban areas.

In recent years online shopping has become an integral part of our lives.The Covid-19 pandemic has only accelerated the entire transition from offline to online shopping.However,the online sector has certain specifics and this is the fact that the online space gives customers the opportunity to express their negative information quickly and,above all,anonymously,and they have the opportunity to reaches a large number of people and is easier to spread.In addition,there is still a lack of confidence in online shopping in the Czech Republic as a whole.Therefore,business owners must learn to work and analyse negative information as quickly as possible to eliminate the impact on their business.And for this reason,the purpose of this work is to examine the influence of product negative information and its impact on the purchase intention in the Czech Republic in online shopping.The study is based on conceptual study which works with five variables-Depth,Language style,Consistency,Source and Volume of negative information.A questionnaire using a Likert scale of1 to 5 was used to collect quantitative data.SPSS software was used for data analysis.The results of the survey showed that all variables have a correlation effect on risk and intention to purchase,except gender does not play a role in perceiving risk in our research.This study is considered beneficial to allow businesses in the market to see negative information,analysing them and to help them continue to work with them and protect their business.

All-parameters Rayleigh wave inversion

Since S-wave velocity of the subsurface is an important parameter in near surface applications,many studies have been conducted for its estimation.Among the various methods that use surface waves or body waves,Rayleigh wave inversion is the most popular.In practice,the densities and P-wave velocities of different layers are usually assumed to be known to avoid ill-posed problems,as they have less influence on the dispersion curves.However,improper assignment of these two groups of parameters leads to inaccurate estimation of the S-wave velocity profile.In order to address this problem,the all-parameters Rayleigh wave inversion strategy is proposed in which the S-wave velocities,layer thicknesses,densities and P-wave velocities of different layers are included as the unknown parameters for inversion.Meanwhile,the transitional Markov Chain Monte Carlo(TMCMC)algorithm is applied for the implementation of all-parameters Rayleigh wave inversion.One simulated example and two real-test applications are demonstrated to verify the capability of the proposed method in the estimation of the S-wave velocity profile,the densities and the P-wave velocities.Furthermore,it is verified that the proposed method achieved more accurate S-wave velocity profile estimation than the traditional approach.

Multi-exponential model to describe pressure-dependent P-and S-wave velocities and its use to estimate the crack aspect ratio

We present new quantitative model describing the pressure dependence of acoustic P-and S-wave velocities.Assuming that a variety of individual mechanisms or defects(such as cracks,pore collapse and grain crushing)can contribute to the pressure-dependent change of the wave velocity,we order a characteristic pressure to all of them and allow a series of exponential terms in the description of the(Pand S-waves)velocity-pressure function.We estimate the parameters of the multi-exponential rock physical model in inversion procedures using laboratory measured P-and S-wave velocity data.As is known,the conventional damped least squares method gives acceptable results only when one or two individual mechanisms are assumed.Increasing the number of exponential terms leads to highly nonlinear ill-posed inverse problem.Due to this reason,we develop the spectral inversion method(SIM)in which the velocity amplitudes(the spectral lines in the characteristic pressure spectrum)are only considered as unknowns.The characteristic pressures(belonging to the velocity amplitudes)are excluded from the set of inversion unknowns,instead,they are defined in a set of fixed positions equidistantly distributed in the actual interval of the independent variable(pressure).Through this novel linear inversion method,we estimate the parameters of the multi-exponential rock physical model using laboratory measured P-and S-wave velocity data.The characteristic pressures are related to the closing pressures of cracks which are described by well-known rock mechanical relationships depending on the aspect ratio of elliptical cracks.This gives the possibility to estimate the aspect ratios in terms of the characteristic pressures.

Combining acellular nerve allografts with brainderived neurotrophic factor transfected bone marrow mesenchymal stem cells restores sciatic nerve injury better than either intervention alone

In this study, we chemically extracted acellular nerve allografts from bilateral sciatic nerves, and repaired 10-mm sciatic nerve defects in rats using these grafts and brain-derived neurotrophic factor transfected bone marrow mesenchymal stem cells. Experiments were performed in three groups： the acellular nerve allograft bridging group, acellular nerve allograft ＋ bone marrow mesenchymal stem cells group, and the acellular nerve allograft ＋ brain-derived neurotrophic factor transfected bone marrow mesenchyrnal stem cells group. Results showed that at 8 weeks after bridging, sciatic functional index, triceps wet weight recovery rate, myelin thickness, and number of myelinated nerve fibers were significantly changed in the three groups. Variations were the largest in the acellular nerve allograft ＋ brain-derived neurotrophic factor transfected bone marrow mesenchymal stem cells group compared with the other two groups. Experimental findings suggest that chemically extracted acellular nerve allograft combined nerve factor and mesenchymal stem cells can promote the restoration of sciatic nerve defects. The repair effect seen is better than the single application of acellular nerve allograft or acellular nerve allograft combined mesenchymal stem cell transplantation.

Menstrual cycle characteristics as an indicator of fertility outcomes:evidence from prospective birth cohort study in China

OBJECTIVE:To evidently assess the applicability of regulate menstrual cycle(MC)characteristics in Traditional Chinese Medicine(TCM)as an indicator for fertility.METHODS:A community-based prospective cohort study was conducted in China.Between January 2010 and December 2012,women who were willing to conceive within 2 years were enrolled in the study.Other than the MC length pattern,a well-adopted MC characteristic,menstrual blood color and clots were specifically concerned for women enrolled.All participants were followed up in 2 years by trained nurses.Pregnancy rate,fecundability odds ratio(FOR)and risk of miscarriage were assessed as fertility outcomes.RESULTS:A total of 2109 women were effectively included in this cohort for analysis.Results show that women with irregular MC length were less likely to achieve conception(FOR_(irregular)=0.59;95%CI=0.45-0.77,P<0.001).Menstrual blood in bright red color was also associated with decline in likelihood of conception(FOR=0.79;95%CI=0.63-0.98,P=0.04).Women with menstrual blood in light red were at higher risk of miscarriage(OR=2.39;95%CI=0.91-6.28,P=0.08).No significant impact was found between menstrual blood clots and fertility outcomes(FOR=1.02,95%CI=0.83-1.25,P=0.88;OR=1.2695%CI=0.77-2.07,P=0.35).CONCLUSIONS:MC characteristics can be an effective and simple indicator for women’s fertility.Increasing the knowledge of MC characteristics for women in reproductive ages would bring great benefits to their preconception health conditions.

The Relationship Between Orthographic Awareness and Chinese Reading in Hong Kong’s Young CSL Learners

Purpose:This study investigates young Chinese as a second language(CSL)learners’Chinese character reading performance and its relationship with their orthographic awareness.There is a pressing need to gain a better understanding of Hong Kong’s ethnic minority students’CSL acquisition,so that more effective instruction can be provided.Design/Approach/Methods:A total of 157 Hong Kong ethnic minority CSL students in Grade 4 were evaluated using a range of assessments:orthographic awareness in identifying and utilizing character components,listening comprehension for vocabulary terms and short texts,and reading Chinese character lists with single-and two-character words.Findings:The students performed consistently across the two lists but made fewer errors when reading the two-character word list,most of which were orthographical.Multiple regression analysis showed that the students’orthographic awareness contributed significantly to their character reading.Originality/Value:The findings suggest that orthographic awareness can help ethnic minority CSL learners improve their Chinese character reading skills.Chinese classes provided for such students,and for CSL learners in general,should place greater focus on literacy training.

The synergies between electricity and other energy resources could promote energy utilization efficiency in electricity market.Within this context,this paper proposes a bi-level stochastic optimization model for the joint operation of a coordinated wind power plant(WPP)and natural gas generating(NGG)-power to gas(P2G)suppliers participating in the day-ahead(DA)market and real-time(RT)market as well as providing real-time auxiliary services.The coordinated supplier’s payoff is maximized in the upper level with consideration of the uncertainties of WPP output capacity and RT electricity price,while the social welfare of the grid is maximized in the lower level.Simulation results demonstrate the effectiveness of the proposed bidding model of the coordinated WPPs and NGG-P2G suppliers by examining its bidding behaviors and benefits with comparisons of four other bidding models.

The effect of the application of cold on hematoma, ecchymosis, and pain at the catheter site in patients undergoing percutaneous coronary intervention

Objective: This study was conducted to determine the effect of the application of cold on hematoma,ecchymosis,and pain in patients undergoing percutaneous coronary intervention.Methods: It is a quasi-experimental study with a control group.A total of 200 patients (100 in the control group and 100 in the experimental group) were included in the research.In the experimental group,a cold pack was applied to the bottom of the sandbag for the first 15 min after withdrawing the femoral arterial catheter.Both groups were assessed and followed up at the first 15th minute and 4th,48th and 72 nd h.Results: After the femoral arterial catheter was withdrawn,ecchymosis and hematoma were smaller and pain was less in the experimental group at the catheter entry site compared to the control group,and the difference was statistically significant (P < 0.01).Conclusion: It was determined that the application of cold reduced the formation of hematomas,ecchymoses and pain in patients after the withdrawal of the femoral arterial catheter.
