Synergistic effects of interferon-alpha in combination with chemoradiation on human pancreatic adenocarcinoma

AIM: To determine whether IFN-α is the agent that turns a slightly effective treatment (radiochemotherapy) into a potent therapy, we tested IFN-α for its synergistic ***: Eight pancreatic carcinoma cell lines were treated with the single agents and combinations of *** role of IFN-α regarding a) direct inhibitory effects; b)radio and chemosensitizing effects; c) anti-angiogenic properties and d) enhancement of immunogenicity was ***: Our results show that IFN-α has direct inhibitory properties and some synergistic influence as determined by AnnexinV/PI stain and cell count. IFN-α is also able to prevent the increase in proliferation rate and VEGF secretion of CDDP resistant cells. Having taken the results from immunogenicity experiments together, we found cells that can be influenced by IFN-α but were less susceptible against T cells. Furthermore, high expression of MHC molecules, CD118, EGF-R and Fas was predictive for a good ***: In conclusion, IFN-α has direct cytotoxic effects, acts as a radiosensitizer and circumvents tumor cell-regrowth after CDDP treatment. These mechanisms may be responsible for the good clinical outcome of CapRI.

Male infertility might be clearly associated with aberrant DNA methylation patterns in human spermatozoa. An association between oxidative stress and the global methylation status of the sperm genome has also been suggested. The aim of the present study was to determine whether the global sperm DNA methylation status was affected in the spermatozoa of carriers of chromosome structural aberrations. The relationships between the 5-methylcytosine （msC） levels in spermatozoa and chromatin integrity status were evaluated. The study patients comprised male carriers of chromosome structural aberrations with reproductive failure （n = 24）, and the controls comprised normozoospermic sperm volunteers （n = 23）. The global msC level was measured using thin-layer chromatography （TLC） and immunofluorescence （IF） techniques. The sperm chromatin integrity was assessed using aniline blue （Ab） staining and TUNEL assay. The mean msC levels were similar between the investigated chromosome structural aberrations carriers （P） and controls （K）. However, sperm chromatin integrity tests revealed significantly higher values in chromosomal rearrangement carriers than in controls （P 〈 0.05）. Although the potential relationship between sperm chromatin integrity status and sperm DNA fragmentation and the msC level juxtaposed in both analyzed groups （P vs K） was represented in a clearly opposite manner, the low chromatin integrity might be associated with the high hypomethylation status of the sperm DNA observed in carriers of chromosome structural aberrations.

Controlling Membrane Phase Separation of Polymersomes for Programmed Drug Release

Transposons are effective mutagens alternative to T-DNA for the generation of insertional mutants in many plant species including those whose transformation is inefficient. The current strategies of transposon tagging are usually slow and labor-intensive and yield low frequency of tagged lines. We have constructed a series of transposon tagging vectors based on three approaches：（i） AcTPase controlled by glucocorticoid binding domain/VP16 acidic activation domain/Gal4 DNA-binding domain （GVG） chemical-inducible expression system; （ii） deletion of AcTPase via Cre-lox site-specific recombination that was initially triggered by Ds excision; and （iii） suppression of early transposition events in transformed rice callus through a dual-functional hygromycin resistance gene in a novel Ds element （HPT-Ds）, We tested these vectors in transgenic rice and characterized the transposition events. Nuclear migration and positioning are crucial for the morphogenesis of plant cells. We addressed the potential role of nuclear positioning for polarity induction using an experimental system based on regenerating protoplasts, where the induction of a cell axis de novo can be followed by quantification of specific regeneration stages. Using overexpression of fluorescently tagged extranuclear （perinu- clear actin basket, kinesins with a calponin homology domain （KCH）） as well as intranuclear （histone H2b） factors of nuclear positioning and time-lapse series of the early stages of regeneration, we found that nuclear position is no prerequi- site for polarity formation. However, polarity formation and nuclear migration were both modulated in the transgenic lines, indicating that both phenomena depend on factors affecting cytoskeletal tensegrity and chromatin structure. We integrated these findings into a model where retrograde signals are required for polarity induction. These signals travel via the cytoskeleton from the nucleus toward targets at the plasma membrane.

Dear Editor, Plasma membrane （PM）-Iocalized PIN-FORMED （PIN） auxin efflux carriers were shown to cycle rapidly and con- tinuously between PM and the endomembrane system and this cycling is affected by many exogenous factors and endogenous programs （Grunewald and Friml, 2010）. The fungal lactone metabolite brefeldin A （bFA） is believed to interrupt the continuous cycling of PINs and other PM proteins by inhibiting their re-secretion （Kleine-Vehn and Friml, 2008）.

The paper presents rather some conclusions from large investigations over dynamic behaviour of bridges under travelling loads. There, as basic tool was applied the 3D-Time Space Method (3D-TSM) in edition proposed by present author. The method uses four-dimensional space, where besides of usual 3D space, the time is the fourth dimension. The bridge with simply supported steel girder is here modelled by means of theory for thin-walled bars (TWbs). In final calculations, solutions are obtained here on numerical way applying well known and simple Finite Differences Method (FDM). In consequence the task is brought to trivial determination of unknowns from set of linear algebraic equations. There, essential part of these equations is so called dynamical stiffness matrix (DSM). The last is additionally tested by Uniform Criterion (...) for evaluation of bridges Critical States (CrS).
