Memory CD8 T cells can provide long-term protection against tumors,which depends on their enhanced proliferative capacity,self-renewal and unique metabolic rewiring to sustain cellular ***,memory CD8 T cells engage oxidative phosphorylation and fatty acid oxidation to fulfill their metabolic *** contrast,tumor-infiltrating lymphocytes(TILs)display severe metabolic defects,which may underlie their functional ***,we show that overexpression of proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),the master regulator of mitochondrial biogenesis(MB),favors CD8 T cell central memory formation rather than resident memory ***-1α-overexpressing CD8 T cells persist and mediate more robust recall responses to bacterial infection or peptide ***,CD8 T cells with enhanced PGC-1αexpression provide stronger antitumor immunity in a mouse melanoma ***,TILs overexpressing PGC-1αmaintain higher mitochondrial activity and improved expansion when rechallenged in a tumor-free ***,our findings indicate that enforcing mitochondrial biogenesis promotes CD8 T cell memory formation,metabolic fitness,and antitumor immunity in vivo.

Tetrahedral amorphous carbon coatings have the potential to significantly reduce friction and wear between sliding ***,we provide atomistic insights into the evolution of the sliding interface between naked and hydrogen-passivated ta-C sliding partners under dry and lubricated *** reactive classical atomistic simulations we show that sliding induces a sp3 to sp2 rehybridization and that the shear resistance is reduced by hydrogen-passivation and hexadecane-lubrication-despite our finding that nanoscale hexadecane layers are not always able to separate and protect ta-C counter surfaces during *** asperities deform,carbon atoms within the hexadecane lubricant bind to the ta-C sliding partners resulting in degradation of the hexadecane molecules and in increased material intermixing at the sliding *** atoms from the passivation layer and from the hexadecane chains continue to be mixed within a sp2 rich sliding interface eventually generating a tribo-layer that resembles an a-C:H type of *** separation of the sliding partners,the tribo-couple splits within the newly formed sp2 rich a-C:H mixed layer with significant material transfer across the sliding *** leaves behind a-C:H coated ta-C surfaces with dangling C bonds,linear C chains and hydrocarbon fragments.

Diabetic retinopathy (DR) is a complication of diabetesmellitus thatappears in the retina. Clinitians use retina images to detect DR pathologicalsigns related to the occlusion of tiny blood vessels. Such occlusion brings adegenerative cycle between the breaking off and the new generation of thinnerand weaker blood vessels. This research aims to develop a suitable retinalvasculature segmentation method for improving retinal screening proceduresby means of computer-aided diagnosis systems. The blood vessel segmentationmethodology relies on an effective feature selection based on SequentialForward Selection, using the error rate of a decision tree classifier in theevaluation function. Subsequently, the classification process is performed bythree alternative approaches: artificial neural networks, decision trees andsupport vector machines. The proposed methodology is validated on threepublicly accessible datasets and a private one provided by Hospital Sant Joanof Reus. In all cases we obtain an average accuracy above 96% with a sensitivityof 72% in the blood vessel segmentation process. Compared with the state-ofthe-art, our approach achieves the same performance as other methods thatneed more computational *** method significantly reduces the numberof features used in the segmentation process from 20 to 5 dimensions. Theimplementation of the three classifiers confirmed that the five selected featureshave a good effectiveness, independently of the classification algorithm.

Diabetic macular edema (DME) is the leading cause of blindness in young adults in developed countries, affecting 12% of type 1 and 28% of type 2 diabetic patients. The gold standard DME treatment should be based on a good control of glycemia along with control of lipids and renal function. However, despite the systemic metabolic control values being essential for patients with diabetic retinopathy (DR), it has proven to be insufficient for DME if it appears. With these patients, additional measures are needed in order to avoid the subsequent loss of vision. While laser treatment of DME has been the only valid treatment so far, it has been inadequate in chronic cases. The introduction of new treatments, such as intravitreal corticosteroids or anti-VEGF drugs, have recently shown their safety and efficacy and together with laser photocoagulation are becoming the treatments of choice in the management of DME.

Diabetes is a serious chronic condition,which increase the risk of cardiovascular diseases,kidney failure and nerve damage leading to *** the ocular complications include diabetic macular edema,is the leading cause of blindness among adults in the industrialized ***,blindness from diabetic macular edema is largely preventable with timely detection and appropriate interventional *** treatment should include an optimized control of glycemia,arterial tension,lipids and renal *** photocoagulation laser is currently restricted to focal macular edema in some countries,but due the high cost of intravitreal drugs,the use of laser treatment for focal and diffuse diabetic macular edema(DME),can be valid as gold standard in many *** intravitreal anti vascular endothelial growth factor drugs(ranibizumab and bevacizumab),are indicated in the treatment of all types of DME,but the correct protocol for administration should be defined for the different Retina Scientific *** corticosteroids for diffuse DME,has a place in pseudophakic patients,but its complications restricted the use of these drugs for some *** the intravitreal interface plays an important role and its exploration is mandatory in all DME patients.
