Achillea biebersteinii extracts suppress angiogenesis and enhance sensitivity to 5-fluorouracil of human colon cancer cells via the PTEN/AKT/mTOR pathway in vitro

Objective:To investigate the antiproliferative,anti-angiogenic,and apoptotic effects of extracts of Achillea biebersteinii(ABE)and combined treatments of ABE with 5-fluorouracil(5-FU)on HT-29 ***:The effects of ABE,5-FU,and combined treatments on the viability of HT-29 cells were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide *** analysis was used to determine synergism between ABE and *** apoptotic and anti-angiogenic effects were determined by cell death detection and human vascular endothelial growth factor ELISA method,*** and translational expressions of p53,Bax,Bcl-2,p38 MAPK,Akt,PTEN,and m TOR were also evaluated by real-time PCR and Western blotting ***:ABE decreased the viability of HT-29 cells in a dosedependent *** treatment of hexane,chloroform,and methanol extracts of Achillea biebersteinii with 5-FU at IC50 doses decreased the cell viability to 26.0%,19.1%,and 14.9%,respectively(Pcantly downregulated m TOR,Akt,Bcl-2 expression,upregulated p53,Bax,PTEN,p38 MAPK expression,and exhibited anti-angiogenetic ***:Our findings indicate that ABE shows synergism with 5-FU and inhibits the proliferation of HT-29 cells by inducing apoptosis and suppressing angiogenesis,which may provide biological evidence for further use of ABE in the treatment of colorectal cancer.

Ultrasound focusing in three-dimensional(3 D)space is of crucial and enduring significance in a variety of biomedical and industrial *** ultrasound focusing based on active phase array or passive geometry of bulky size is unable to realize the 3 D arbitrary focusing with subwavelength *** metamaterial of complex deep-subwavelength microstructure has facilitated the advanced airborne-sound-focusing but is inevitably not applicable for underwater ultrasound,restricted by the law between the multi-modes coupling/thermal viscosity and the feature size of the ***,we aim to circumvent the restriction by increasing the feature size of the metamaterial while keeping the compact overall geometry,and realize the robust subwavelength ultrasound focusing with the sparse metalens of the wavelength-scale *** theoretically propose and demonstrate numerically and experimentally the broadband arbitrary ultrasound focusing in 3 D *** axial and off-axis ultrasound focusing with the subwavelength resolution(FWHMtalens within one-octave *** advantages of 3 D freewheeling focusing,subwavelength resolution,spatial sparsity,geometric simplicity,and broadband,the sparse metalens would offer more initiatives to advanced researches in ultrasound focusing and empower applications such as precise biomedical imaging and therapy,nondestructive evaluation,integrated and multiplexed ultrasound devices.

This study proposed a measurement platform for continuous blood pressure estimation based on dual photoplethysmography(PPG)sensors and a deep learning(DL)that can be used for continuous and rapid measurement of blood pressure and analysis of cardiovascular-related *** proposed platform measured the signal changes in PPG and converted them into physiological indicators,such as pulse transit time(PTT),pulse wave velocity(PWV),perfusion index(PI)and heart rate(HR);these indicators were then fed into the DL to calculate blood *** hardware of the experiment comprised 2 PPG components(i.e.,Raspberry Pi 3 Model B and analog-todigital converter[MCP3008]),which were connected using a serial peripheral *** DL algorithm converted the stable dual PPG signals acquired from the strictly standardized experimental process into various physiological indicators as input parameters and finally obtained the systolic blood pressure(SBP),diastolic blood pressure(DBP)and mean arterial pressure(MAP).To increase the robustness of the DL model,this study input data of 100 Asian participants into the training database,including those with and without cardiovascular disease,each with a proportion of approximately 50%.The experimental results revealed that the mean absolute error and standard deviation of SBP was 0.17±0.46 *** mean absolute error and standard deviation of DBP was 0.27±0.52 *** mean absolute error and standard deviation of MAP was 0.16±0.40 mmHg.

Images are generally corrupted by impulse noise during acquisition and *** deteriorates the quality of *** remove corruption noise,we propose a hybrid approach to restoring a random noisecorrupted image,including a block matching 3D(BM3D)method,an adaptive non-local mean(ANLM)scheme,and the K-singular value decomposition(K-SVD)*** the proposed method,we employ the morphological component analysis(Mca)to decompose an image into the texture,structure,and edge ***,the BM3D method,ANLM scheme,and K-SVD algorithm are utilized to eliminate noise in the texture,structure,and edge parts of the image,*** results show that the proposed approach can effectively remove interference random noise in different parts;meanwhile,the deteriorated image is able to be reconstructed well.

AIM: To analyze the prognostic factors influencing the overall survival of peripheral cholangiocarcinoma (PCC) patients undergoing surgical treatment during 25 years at a single institution. METHODS: This study retrospectively reviewed prospectively collecting data about 373 patients with historically proven PCC who underwent surgical treatment between 1977 and 2001. RESULTS: Three hundred and seventy-three PCC patients (159 men and 214 women) underwent surgical treatment from 1977 to 2001. Among them, 187 PCC patients underwent hepatectomy and 135 had curative resection (curative resectability rate: 36.2%). The follow-up duration ranged from 1.05 to 167.6 mo (mean/median = 14.1/7.2 mo). Overall cumulative survival rates at 1, 3, and 5 years were 32.5%, 9.2%, and 4.1%, respectively. Univariate log-rank analysis identified the following as adverse influences on overall survival: presence of symptoms, absence of mucobilia, elevated CEA and ca 19-9 levels, non-papillary tumor type, receiving non-hepatectomy, advanced tumor staging, lack of post-operative chemotherapy, and radiotherapy. Meanwhile, multivariate Cox's proportional hazard analysis demonstrated that absence of mucobilia, non-papillary tumor type, advanced tumor staging, non-hepatectomy, and lack of post-operative chemotherapy were the five independent prognostic factors that adversely affected overall survival. CONCLUSION: Favorable overall survival of PCC patients undergoing surgical treatment depends on early tumor stage, presence of mucobilia, papillary tumor type, hepatic resection, and post-operative chemotherapy.

Although the prevalence of chronic hepatitis B virus (HBV) infection has declined in renal transplant recipients (RTRs), it remains a relevant clinical problem with high morbidity and mortality in long-term follow up. A thorough evaluation, including liver biopsy as well as assessment of HBV replication in serum (i.e. hepatitis B e antigen and/or HBV DNA) is required before transplantation. Interferon should not be used in this setting because of low efficacy and precipitation on acute allograft rejection. The advent of effective antiviral therapies offers the opportunity to prevent the progression of liver disease after renal transplantation. However, as far as we are aware, no studies have compared prophylactic and preemptive strategies. To date, the majority of RTRs with HBV-related liver disease have had a high virological and biochemical response to lamivudine use. However, lamivudine resistance is frequent with a prolonged course of therapy. Considering long-term treatment, antiviral agents with a high genetic barrier to resistance and lack of nephrotoxicity are suggested. The optimal strategy in RTRs with HBV infection remains to be established in the near future.
