Much effort has been made to uncover the cellular heterogeneities of human hearts by single-nucleus RNA sequencing.However,the cardiac transcriptional regulation networks have not been systematically described because of the limitations in detecting transcription factors.In this study,we optimized a pipeline for isolating nuclei and conducting single-nucleus RNA sequencing targeted to detect a higher number of cell signal genes and an optimal number of transcription factors.With this unbiased protocol,we characterized the cellular composition of healthy human hearts and investigated the transcriptional regulation networks involved in determining the cellular identities and functions of the main cardiac cell subtypes.Particularly in fibroblasts,a novel regulator,PKNOX2,was identified as being associated with physiological fibroblast activation in healthy hearts.To validate the roles of these transcription factors in maintaining homeostasis,we used single-nucleus RNA-sequencing analysis of transplanted failing hearts focusing on fibroblast remodelling.The trajectory analysis suggested that PKNOX2 was abnormally decreased from fibroblast activation to pathological myofibroblast formation.Both gain-and loss-of-function in vitro experiments demonstrated the inhibitory role of PKNOX2 in pathological fibrosis remodelling.Moreover,fibroblast-specific overexpression and knockout of PKNOX2 in a heart failure mouse model induced by transverse aortic constriction surgery significantly improved and aggravated myocardial fibrosis,respectively.In summary,this study established a high-quality pipeline for single-nucleus RNA-sequencing analysis of heart muscle.With this optimized protocol,we described the transcriptional regulation networks of the main cardiac cell subtypes and identified PKNOX2 as a novel regulator in suppressing fibrosis and a potential therapeutic target for future translational studies.

Factors Associated with Voluntary Cosmetic Depigmentation in Schools in the Collines Department (Benin)

Skin depigmentation is a worrying practice that is gaining popularity, particularly among young girls. However, this practice poses health risks. It also reflects a negative view of black skin color. This was a cross-sectional study carried out between April and May 2023 which involved 1039 female students from schools and universities in the Collines department selected by stratified sampling. Data was collected during a face-to-face interview using a questionnaire providing information on the demographic, socio-cultural, and economic characteristics of the girls. The depigmentation products used were identified as well as the complications caused by the use of these products. Statistical analysis made it possible to calculate the frequencies and logistic regression made it possible to identify the factors associated with depigmentation. The prevalence of depigmentation among the girls surveyed was 78.2%. The main products used were soaps based on mercurial derivative and hydroquinone (21.6%) and lotions based on hydroquinone and corticosteroids (75.7%). The factors associated with the practice of depigmentation were the ethnicity of the respondents (OR = 2.52;95% CI = [0.47 - 13.33], p = 0.001);the average monthly income of the parents (OR = 3.26;95% CI = [1.71 - 6.09], p = 0.003);the opinion of the respondents on depigmentation (OR = 2.48;95% CI = [0.84 - 7.3], p < 0.001) and the value given to light skin versus black skin (OR = 3.41;95% CI = [2.32 - 5.01], p < 0.001). In conclusion, the prevalence of depigmentation among the girls surveyed is high. Consequently, reinforced awareness measures and stricter control of bleaching products are imperative to address this high prevalence of the phenomenon.

Nasopharyngeal liposarcoma is an extremely rare clinical entity. Less than ten cases have been reported in the literature. It mainly occurs in humans. The non-specificity of the clinical and radiological signs explains the difficulty of the diagnosis confirmed by anatomopathological and immunohistochemical examinations. Its pathogenesis remains obscure and often correlates with genetic abnormalities. Pleomorphic liposarcoma is its rarest subtype, remarkable for its aggressiveness and rapidity of growth. Complete surgical excision remains the treatment of choice. We report a case of liposarcoma whose diagnosis was made during histological examination coupled with immunohistochemistry. .

Using Metaheuristic OFA Algorithm for Service Placement in Fog Computing

The use of fog computing in the Internet of Things(IoT)has emerged as a crucial solution,bringing cloud services closer to end users to process large amounts of data generated within the system.Despite its advantages,the increasing task demands from IoT objects often overload fog devices with limited resources,resulting in system delays,high network usage,and increased energy consumption.One of the major challenges in fog computing for IoT applications is the efficient deployment of services between fog clouds.To address this challenge,we propose a novel Optimal Foraging Algorithm(OFA)for task placement on appropriate fog devices,taking into account the limited resources of each fog node.The OFA algorithm optimizes task sharing between fog devices by evaluating incoming task requests based on their types and allocating the services to the most suitable fog nodes.In our study,we compare the performance of the OFA algorithm with two other popular algorithms:Genetic Algorithm(GA)and Randomized Search Algorithm(RA).Through extensive simulation experiments,our findings demonstrate significant improvements achieved by the OFA algorithm.Specifically,it leads to up to 39.06%reduction in energy consumption for the Elektroensefalografi(EEG)application,up to 25.86%decrease in CPU utilization for the Intelligent surveillance through distributed camera networks(DCNS)application,up to 57.94%reduction in network utilization,and up to 23.83%improvement in runtime,outperforming other algorithms.As a result,the proposed OFA algorithm enhances the system’s efficiency by effectively allocating incoming task requests to the appropriate fog devices,mitigating the challenges posed by resource limitations and contributing to a more optimized IoT ecosystem.

Background:Shunt systems are used to provide cerebrospinal fluid drainage in the treatment of hydrocephalus.Recently,antibiotic-impregnated shunt systems are used to prevent colonization in the ventriculoperitoneal catheters.Methicillin-resistant Staphylococcus aureus(MRSA)is the most common causative microorganism of shunt infections.The aim of the study is to investigate effects of several substances on MRSA biofilms in the ventriculoperitoneal catheters.Methods:The present study consists of mainly eight groups(each has two subgroups as antibiotic-impregnated and nonantibiotic-impregnated catheters).In addition,each group contains six molds using MRSA strains.In this study,daptomycin(DAPT)(2 mg/ml),vancomycin(VAN)(10 mg/ml),linezolid(LIN)(2 mg/ml),N-acetylcysteine(NAC)(6 mg/ml),and various combinations of these substances were used to evaluate the treatment against MRSA using scanning electron microscope(SEM)images and microbiological enumeration.Results:The colony count in the antibiotic-impregnated samples significantly decreased compared to nonantibiotic-impregnated samples in the MRSA,MRSA+DAPT,and MRSA+LIN groups(p<0.01),respectively.Conversely,the colony count in antibiotic-impregnated samples significantly increased compared to nonantibiotic-impregnated samples in NAC+DAPT and NAC+VAN groups(p<0.01),respectively.Conclusions:The results showed that the use of antibiotic-impregnated catheters has a significant impact on the prevention of infection whereas the combination of NAC and DAPT showed better antibiofilm and antibacterial effects than other combinations on the prevention and treatment of nonantibiotic-impregnated catheter infections.

Investigation of Single-Wall MoS_2 Monolayer Flakes Grown by Chemical Vapor Deposition

Recently, two-dimensional monolayer molybdenum disulfide(MoS_2), a transition metal dichalcogenide, has received considerable attention due to its direct bandgap, which does not exist in its bulk form, enabling applications in optoelectronics and also thanks to its enhanced catalytic activity which allows it to be used for energy harvesting. However,growth of controllable and high-quality monolayers is still a matter of research and the parameters determining growth mechanism are not completely clear. In this work, chemical vapor deposition is utilized to grow monolayer MoS_2 flakes while deposition duration and temperature effect have been systematically varied to develop a better understanding of the MoS_2 film formation and the influence of these parameters on the quality of the monolayer flakes. Different from previous studies, SEM results show that single-layer MoS_2 flakes do not necessarily grow flat on the surface, but rather they can stay erect and inclined at different angles on the surface, indicating possible gas-phase reactions allowing for monolayer film formation. We have also revealed that process duration influences the amount of MoO_3/MoO_2 within the film network. The homogeneity and the number of layers depend on the change in the desorption–adsorption of radicals together with sulfurization rates, and, inasmuch, a careful optimization of parameters is crucial. Therefore, distinct from the general trend of MoS_2 monolayer formation, our films are rough and heterogeneous with monolayer MoS_2 nanowalls. Despite this roughness and the heterogeneity, we observe a strong photoluminescence located around 675 nm.

A survey on communication protocols and performance evaluations for Internet of Things

The Internet of Things (IoT) is a large-scale network of devices capable of sensing, data processing, and communicating with each other through different communication protocols. In today's technology ecosystem, IoT interacts with many application areas such as smart city, smart building, security, traffic, remote monitoring, health, energy, disaster, agriculture, industry. The IoT network in these scenarios comprises tiny devices, gateways, and cloud platforms. An IoT network is able to keep these fundamental components in transmission under many conditions with lightweight communication protocols taking into account the limited hardware features (memory, processor, energy, etc.) of tiny devices. These lightweight communication protocols affect the network traffic, reliability, bandwidth, and energy consumption of the IoT application. Therefore, determining the most proper communication protocol for application developers emerges as an important engineering problem. This paper presents a straightforward overview of the lightweight communication protocols, technological advancements in application layer for the IoT ecosystem. The survey then analyzes various recent lightweight communication protocols and reviews their strengths and limitations. In addition, the paper explains the experimental comparison of Constrained Applications Protocol (CoAP), Message Queuing Telemetry (MQTT), and WebSocket protocols, more convenient for tiny IoT devices. Finally, we discuss future research directions of communication protocols for IoT.

Investigation of Color Change of Different Restoration Thickness, Background Color and Resin Cement Shade on CAD/CAM Glass Ceramic Materials

The aim of this study is to investigate the color change of different restoration thicknesses, backgrounds and resin cement colors on lithium disilicate and zirconium reinforced lithium silicate materials in vitro. In this study, IPS emax CAD (LT C14) and Celtra Duo (LT C14) are used as full ceramic materials, and Variolink Esthetic LC (warm, neutral) used as resin cement and Tokuyama Estelite Sigma Quick (A3, A2) is used as composite materials. A total of 160 samples in the form of 40 pieces of 5 × 5 0.4 mm thick 40 pieces of 5 × 5 0.6 mm thick square discs from each of the all-ceramic materials in block form were obtained using a water jet device (DWJ1525-FA;Dardi International Corporation, Nanjing, China). Glass ceramic samples produced in 2 different thicknesses were cemented on 2 different backgrounds with 2 different resin types of cement. Color measurements of the samples before and after cementation were performed on a grey background with spectrophotometer Vita EasyShade V (Vita Zahnfabrik, Bad Sackingen, Germany) and color parameters (L*, a*, b*, ΔE) were calculated according to the CIE Lab (Commission Internationale de L’Eclairage) system. Average values for each group (ΔE) were not affected by ceramic type, material thickness, background color, resin cement color, and the interaction of these four variables (p > 0.05). When the triple interactions between the groups were examined, there were no statistically significant differences (p > 0.05). In the evaluation of pairwise interactions between two groups (material type-material thickness, material type-background color, and thickness of material-background interactions) statistically significant differences (p Implications: The material type, thickness, background and cement color used did not cause any statistically significant color change in lithium disilicate and zirconium-reinforced lithium silicate glass ceramic materials (p > 0.05).

Genomic resources for the brown planthopper, Nilaparvata lugens： Transcriptome pyrosequencing and microarray design

The brown planthopper, Nilaparvata lugens is a pest of cultivated rice through- out Asia and is controlled using insecticides and/or resistant rice varieties. This species has developed resistance to many classes of insecticide and biotypes have developed that are vir- ulent against formerly resistant rice cultivars. Insects use a suite of detoxification enzymes, including cytochrome P450s, glutathione S-transferases and carboxyl/cholinesterases to defend themselves against plant secondary metabolites and pesticides. Pyrosequencing on the Roche 454-FLX platform was used to produce a substantial expressed sequence tag （EST） dataset to complement the existing Sanger sequenced ESTs in GenBank. A total of 78 959 reads were combined with the 37 392 publically available Sanger ESTs; these assembled into 8 911 contigs and 10 620 singletons. Analysis of the distribution of tenta- tive unique genes （TUGs） with the gene ontology for biological processes and molecular functions suggests that the 454 and Sanger EST assembly is broadly representative of the N. lugens transcriptome. The brown planthopper transcriptome was found to contain 31 TUGs encoding P450s, nine encoding glutathione S-transferases and 26 encoding car- boxyl/cholinesterases and many of these are putatively involved in the detoxification of xenobiotics. The Agilent eArray platform was used to construct an oligonucleotide mi- croarray populated with probes for ~ 19 000 unigene sequences, including all those known to encode detoxification enzymes. The genomic resources developed in this study will be useful to the community studying this crop pest and will help elucidate the molecular mechanism underlying insecticide resistance and planthopper adaptation to resistant rice cultivars.

Stress Distributions Created by Short and Regular Implants Placed in the Anterior Maxilla at Different Angles: A Finite Element Analysis

Although short implants are seen as alternative treatments that require additional surgical techniques in posterior region, they can be applied to anterior maxilla and various studies are required on this subject. The purpose of this study was to examine and compare the peak von Mises stress distributions in the crown, implant and abutment by using finite element analysis (FEA). Besides, a comparison of the implant-abutment connection types in the short implant with the FEA method was established. A short implant (4 × 5 mm) with a taper-lock connection and a regular implant (4 × 9 mm) with a screw connection were used in maxillary central incisor tooth area. Three different titanium abutments with 0?, 15? and 25? angles were used for abutments. In addition, in order to determine whether the stress change in short implants is due to the length of the implant-abutment connection, a screw was designed for a short implant and it was also evaluated in the same three angles. A total of three groups and nine models were generated. 114.6N load was applied to the cingulum area of the crown at an angle of 135? to the long axis of the crowns. A torque load of 25 Ncm was applied to the regular and short implant screw. Von Mises stress distributions of implants, abutments and crowns were evaluated by using FEA. Increased angle in implants increased von Mises stress values of implant, abutment and crown. Screw connection was found higher at all angles in short implants. Close values were found at different angles in taper-lock short implant crowns. The length and the angle in the bone of implant with the type of implant-abutment connection results in the accumulated stress values. Clinical Implications Taper implant-abutment connection system was found to be more promising in terms of stress accumulation in crowns. Although the amount of stress on the abutment increased due to the length of the implant in short implants, taper implant-abutment connection system slightly reduced related to this increase.
