In this report, we describe morphological characteristics of a dicephalic Steppes Ratsnake （Elaphe dione） collected from the wild in 2011 in South Korea. The specimen has two heads and two long necks. Unlike normal individuals, the dicephalic snake has divided ventral scales under the necks of the bifurcated columns. The snout- vent length （SVL） and overall total length of the individual are shorter than those of normal snakes of the same age. Nevertheless, the counts of nine different scale types that are often used for classification are all within the ranges of normal individuals. As far as we know, this is the first detailed morphological description of a dicephalic E. dione in the scientific literature.

AIM:To investigate whether expressing biliary phenotype predicted poor outcome after the surgical treatment in primary liver cancers. METHODS:Out of 204 patients that underwent liver resection due to hepatocellular carcinoma (HCC), liver specimens of 70 patients with HCC were evaluated for biliary components by cytokeratin (CK) 19 immunostain (CK19 - HCC and CK19 + HCC). CK19 positivity was defined as membranous and/or cytoplasmic expression in ≥ 5% of tumor cells with moderate or strong intensity. Patients with other primary liver cancers, such as com- bined HCC and cholangiocarcinoma (cHCC-CC), intrahe- patic cholangiocarcinoma (ICC) who received curative liver resection, were also included in the study. Clinical characteristics of CK19-HCC and CK19 + HCC patients, including survival outcome after curative liver resection, were compared with that of cHCC-CC and ICC patients. RESULTS: The overall survival (OS) rate of CK19 - HCC(n = 49) after the curative surgical treatment was 90.7%, and 80.4% at 1 and 5 years after the resection. OS rate of CK19 + HCC (n = 21) was 74.3%, 28.9% and OS rate of cHCC-CC (n = 22) was 66.7%, 32.2% at 1 and 5 years after the surgery. For ICC (n = 19), 1 and 5-year-OS rate was 50.2% and 14.3% after the cura-tive resection. The OS rates of CK19 + HCC and cHCC-CC were significantly lower than that of CK19-HCC, but higher than the OS rate of ICC (P = 0.000). There was no statistically significant difference in OS rate between CK19 + HCC and cHCC-CC. The disease free survival (DFS) rate of CK19-HCC was 72.0% and 54.5% at 1 and 3 years after the surgical treatment. DFS rate of CK19 + HCC was 53.3%, 34.3% and DFS rate of cHCC- CC was 51.5%, 39.2% at 1 and 3 years after the resection. For ICC, 1 and 3-year-DFS rate was 28.0% and 14.0% after the curative resection. DFS rate of CK19-HCC was significantly higher than that of ICC (P = 0.017), but marginally higher than DFS rate of either CK19 + HCC or cHCC-CC (P = 0.097, P = 0.089, respec-tively). AIM: To verify the expression and methylation status o the MAGE-A1 and MAGE-A3 genes in colorectal cance tissues and cancer cell lines. METHODS: We evaluated promoter demethylation status of the MAGE-A1 and MAGE-A3 genes by RT-PCR analysis and methylation-specific PCR (MS-PCR), as we as sequencing analysis, after sodium bisulfite modifica tion in 32 colorectal cancer cell lines and 87 cancer tis sues. RESULTS: Of the 32 cell lines, MAGE-A1 and MAGE-A3 expressions were observed in 59% and 66%, respective ly. Subsequent to sodium bisulfite modification and MS PCR analysis, the promoter hypomethylation of MAGE-A1 and MAGE-A3 was confirmed in both at 81% each Promoter hypomethylation of MAGE-A1 and MAGE-A3 in colorectal cancer tissues was observed in 43% and 77% respectively. Hypomethylation of MAGE-A1 and MAGE-A3 genes in corresponding normal tissues were observed in 2% and 6%, respectively. CONCLUSION: The promoter hypomethylation of MAGE genes up-regulates its expression in colorectal carcino mas as well as in gastric cancers and might play a signif icant role in the development and progression of human colorectal carcinomas.

Can an Invasive Prey Species Induce Morphological and Behavioral Changes in an Endemic Predator? Evidence from a South Korean Snake(Oocatochus rufodorsatus)

Introduction of an invasive prey species into an ecosystem may affect an endemic predator＇s fitness by altering the prey-predator system. Successful adaptation may allow the endemic predator to eat and control the invasive species, while unsuccessful adaptation may result in extinction of the predator. We examine the possible effects of the invasive North American bullfrog （Rana [Lithobates] catesbeiana） on the endemic Red-backed rat snake （Oocatochus rufodorsatus） in South Korea. We do so by comparing the morphology and behavior of adult and hatchling snakes from bullfrog-exposed （Taean） and bullfrog-unexposed （Hongcheon） populations. Among the seven morphological characteristics investigated, relative tail length （tail length/snout-vent length） of both adults and hatchlings from Taean was significantly greater than that of adults and hatchlings from Hongcheon. Also, adult snakes from Taean had a signiifcantly shorter latency of ifrst tongue lfick in response to prey compared to adults from Hongcheon. This difference was not observed in hatchlings. In other snake species, a longer relative tail length and shorter latency of ifrst tongue lfick are known to improve foraging efifciency, and these characters may be adaptations ofO. rufodorsatus to prey on bullfrogs. This study provides preliminary evidence that the presence of an invasive prey species may cause morphological and behavioral changes in an endemic predator.

Mungbean （Vigna radiata （L.） Wilczek） is a unique species in its ability to fix atmospheric nitrogen, with early maturity, and relatively good drought resistance. We used 454 sequencing technology for transcriptome sequencing. A total of 150 159 and 142 993 reads produced 5 254 and 6 374 large contigs （〉_500 bp） with an average length of 833 and 853 for Sunhwa and jangan, respectively. Functional annotation to known sequences yielded 41.34% and 41.74% unigenes for jangan and Sunhwa. A higher number of simple sequence repeat （SSR） motifs was identified in jangan （1 630） compared with that of Sunhwa （1 334）. A similar SSR distribution pattern was observed in both varieties. A total of 8 249 single nucleotide polymorphisms （SNPs） and indels with 2 098 high-confidence candidates were identified in the two mungbean varieties. The average distance between individual SNPs was -860 bp. Our report demonstrates the utility of transcriptomic data for implementing a functional annotation and development of genetic markers. We also provide large resource sequence data for mungbean improvement programs.

Cumulative incidence and risk factors for pouch adenomas associated with familial adenomatous polyposis following restorative proctocolectomy

BACKGROUND The emergence of restorative total proctocolectomy has significantly reduced the lifetime colorectal cancer risk associated with familial adenomatous polyposis(FAP).However,adenomas may develop in the ileal pouch over time and may even progress to carcinoma.We evaluated the cumulative incidence,time to development,and risk factors associated with ileal pouch adenoma.AIM To evaluate the cumulative incidence,time to development,and risk factors associated with pouch adenoma.METHODS In this retrospective,observational study conducted at a tertiary center,95 patients with FAP who underwent restorative proctocolectomy at our center between 1989 and 2018 were consecutively included.The mean follow-up period was 88 mo.RESULTS Pouch adenomas were found in 24(25.3%)patients,with a median time of 52 mo to their first formation.Tubular adenomas were detected in most patients(95.9%).There were no high-grade dysplasia or malignancies.Of the 24 patients with pouch adenomas,13 had all detected adenomas removed.Among the 13 patients who underwent complete adenoma removal,four(38.5%)developed recurrence.Among 11(45.8%)patients with numerous polyps within the pouch,seven(63.6%)exhibited progression of pouch adenoma.The cumulative risks of pouch adenoma development at 5,10,and 15 years after pouch surgery were 15.2%,29.6%,and 44.1%,respectively.Severe colorectal polyposis(with more than 1000 polyps)was a significant risk factor for pouch adenoma development(hazard ratio,2.49;95% confidence interval:1.04-5.96;P=0.041).CONCLUSION Pouch adenomas occur at a fairly high rate in association with FAP after restorative proctocolectomy,and a high colorectal polyp count is associated with pouch adenoma development.

Objective:To investigate the phytochemical properties and the anticonvulsant potential of the ethyl acetate soluble fraction of ethanol leaf extract of Globimetida braunii,a plant used in ethnomedicine for the treatment of epilepsy.Methods:The phytochemical screening was carried out using standard protocol while the anticonvulsant activity was studied using maximal electroshock test in chicks,pentylenetetrazole and 4-aminopyridine—induced seizures in mice.Results:The preliminary phytochemical screening carried out on the crude ethanol extract revealed the presence of saponins,carbohydrates,flavonoids,tannins,anthraquinones and steroids.Similarly,tannins,flavonoids and steroids/terpenes were found to be present in the ethyl acetate fraction,In the pharmacological screening,150 mg/kg of the fraction protected 83.33%of animals against pentylenetetrazole-induced seizure in mice whereas sodium valproate a standard anti-epileptic drug offered 100%protection.In the 4-aminopyridine-induced seizure model,the fraction produced a significant(Pilepsy and lend credence to the ethnomedical use of the plant in the management of epilepsy.

Epidermal growth factor upregulates Skp2/Cks1 and p27^(kip1) in human extrahepatic cholangiocarcinoma cells

AIM:To evaluate the expression status of S-phase kinase-associated protein 2(Skp2)/cyclin-dependent kinases regulatory subunit 1(Cks1)and p27kip1,and assess the prognostic significance of Skp2/Cks1 expression with p27kip1in patients with extrahepatic cholangiocarcinoma.METHODS:Seventy-six patients who underwent curative resection for histologically confirmed extrahepatic cholangiocarcinoma at our institution from December1994 to March 2008 were enrolled.Immunohistochemical staining for Skp2,Cks1,p27kip1,and Ki67,along with other relevant molecular biologic experiments,were performed.RESULTS:By Cox regression analyses,advanced age(>65 years),advanced AJCC tumor stage,poorly differentiated histology,and higher immunostaining intensity of Skp2 were identified as independent prognostic factors in patients with extrahepatic cholangiocarcinoma.Exogenous epidermal growth factor(EGF,especially 0.1-10 ng/mL)significantly increased the proliferation indices by MTT assay and the mRNA levels of Skp2/Cks1 and p27kip1in SNU-1196,SNU-1079,and SNU-245 cells.The protein levels of Skp2/Cks1(from nuclear lysates)and p27kip1(from cytosolic lysate)were also significantly increased in these cells.There were significant reductions in the protein levels of Skp2/Cks1and p27kip1(from nuclear lysate)after the treatment of LY294002.By chromatin immunoprecipitation assay,we found that E2F1 transcription factor directly binds to the promoter site of Skp2.CONCLUSION:Higher immunostaining intensity of Skp2/Cks1 was an independent prognostic factor for patients with extrahepatic cholangiocarcinoma.EGF upregulates the mRNA and protein levels of Skp2/Cks1and p27kip1via the PI3K/Akt pathway and direct binding of E2F1 transcription factor with the Skp2 promoter.
