Antimutagenic Profile of Antioxidant Vitamins in Drosophila Mulation Test

Objective To assess the antimutagenicity of antioxidant vitamins （vitamins A, C, and E） as expressed by their efficacy to lower aflatoxin-induced mutations. Methods The Muller-5 method for mutation detection was used to assay the frequency of X-chromosome linked recessive lethal mutations （XRLMs） in Drosophila. Larvae were exposed to dietary concentration of aflatoxins and/or the human therapeutic doses of any of the three antioxidant vitamins. Absence of normal eyed males among M2 progeny gave an indication of mutation induction. Results Aflatoxin supplimentation significantly increased the incidence of XRLMs in Drosophila. Mutation frequency was also raised a little above the control level in case of vitamin treatment. However, notable mitigation in mutation frequency was registered when aflatoxin-treated larvae were concomitantly fed with any of the three antioxidant vitamins. Conclusion Aflatoxin exposure can enhance the frequency of gene mutation in Drosophila which is significantly lowered by each of the three antioxidant vitamins. The degree of amelioration produced by them is almost identical. This mitigation is based on the scavenging/trapping by antioxidant vitamins of DNA-reactive products （metabolites and radicals） emanating from aflatoxin metabolism.

The genotoxic potentiality of the crude leaf extract of Casearia tomentosa, a medicinal preparation, has been evaluated in swiss albino mice. The extract significantly induced the division_disruptive chromosomal changes in bone marrow cells as well as in primary spermatocytes; the latter also exhibited marked increase in synaptic disruptions. A significant decrease in sperm count was noted. The incidence of structural damages in chromosomes, however, remained within the range of control level frequency. This herbal preparation, therefore, appears to be primarily spindle_poisoning in its action, but not clastogenic. The probable mechanism of this differential genotoxicity is discussed.

Application of plasma chemistry for gas cleaning is gaining prominence in recent years, mainly from an energy efficiency point of view. In this paper we conducted a comparative study of NO/NOx removal using two different types of dielectric barrier discharge electrodes, wire- cylinder reactor, pipe-cylinder reactor. Investigations were first carried out with synthetic gases to obtain the baseline information on the NO/NOx removal with respect to the two geometries studied. Further, experiments were carried out with raw diesel exhaust under loaded condition. A high NOx removal efficiency of 90% was observed for the pipe-cylinder reactor as compared to that of 53.4% for the wire-cylinder reactor. Furthermore, for the same energy consumed per NO molecule （about 73 eV/NO molecule）, the removal efficiency increased from 67% for the wire- cylinder to about 98% for the pipe-cylinder which was quite appreciable.

An attempt was made to determine the qualitative changes in basmati rice （Pusa Basmati 1121, PBl121） during soaking at 40 ℃ to 80 ℃. soaking temperature had significant effect （a = 0.01） on chemical composition, glycemic index and starch characteristics of rice. starch content, apparent amylose content, crude protein content and crude fat content in un-soaked rice were found to be 73.24%, 27.26%, 8.79% and 2.56%, respectively, but differences in these traits were observed after soaking. Amylose to amylopectin ratio （Am/Ap） decreased from 0.59 to 0.52 （soaked at 80 ℃）. Crude fibre and crude ash contents increased after soaking. The mineral composition （k, P, s, Ca, Mg, Mn, Fe, Cu and Zn） in soaked rice was found to be 16.46% higher than un-soaked rice at the same degree of polishing. Glycemic index of un-soaked rice was found to be 58.41, but decreased to 54.31 after soaking at 80 ℃. Pasting properties, scanning electron microscope images, and X-ray diffractograms suggested partial gelatinization of starch in the temperature range of 60 ℃ to 80 ℃. Based on qualitative changes in rice （apparent amylose content, Am/Ap ratio and crystallinity rate）, it was concluded that intermediate soakincl temperatures （60 ℃ to 70 ℃） would be useful for soaking of PB1121.

Both room-and-pillar and longwall mining systems develop underground excavations whose stability must be ensured over their entire service life. Even though rock bolts have been extensively used as a support element in Us coal mines for about 40 years, limited research has been conducted in quantifying its composite reinforcement effects. Recently, the authors suggested an approach to quantify the reinforcement effect of roof supports over a designated area based on critical failure strains in tension, compression and shear. This paper validates the critical strain technique(CsT) using a case study and justifies the magnitude of selected critical strain by corroborating with the Us roof fall statistics. Intersections are vulnerable to failure due to the larger exposed roof span and associated stress concentrations. Through numerical application of the CsT to a case study, it was demonstrated that modifying the opening orientation and installing reinforcement at critical locations can help to improve the overall stability of intersections.

shorea robusta Gaertn. f.（sal）is one of the important tim-ber-yielding plants in India, which dominates the vegetation of Terai landscape of Uttar Pradesh state in India forming various communities based on its associations. The present study deals with delineation, map-ping and characterization of various communities of sal （shorea robusta） forests in Terai landscape of Uttar Pradesh, India ranging across over 16 districts. Field survey and visual interpretation based forest vegetation type classification and mapping was carried out as part of the project entitled ‘Biodiversity characterization at landscape level using remote sensing and GIs’. Indian Remote sensing-P6 （Resourcesat-1） Linear Imaging self scanner-III satellite data was used during the study. The total area covered by different sal forests was found to be approximately 2256.77 km2. sal communities were identified and characterized based on their spectral properties, physiognomy and phytosociological charac-teristics. Following nine sal communities were identified, delineated and mapped with reasonable accuracyviz.,Chandar,Damar, dry plains, moist plains, western alluvium, western alluvium plains, mixed moist deciduous, mixed dry deciduous andsiwalik. It is evident from the area estimates that mixed moist deciduous sal is the most dominant commu-nity in the region covering around （1613.90 km2）, other major communi-ties were found as western alluvium plains sal （362.44 km2）, mixed dry deciduous sal （362.44 km2） and dry plains sal （107.71 km2）. The Terai landscape of Uttar Pradesh faces tremendous anthropogenic pressure leading to deterioration of the forests. Community level information could be used monitoring the status as well as for micro level conserva-tion and planning of the sal forests in Terai Landscape of Uttar Pradesh.

Incorporation of Te and Gd were done based on the stoicheometric formula PbTi_(0.8-x)Te_(0.2)Gd_(x)O_(3)(PTTeG).TG characterization of green powder revealed the completion of solid state reaction at temperature 450℃.XRD of modified PTTeG powders milled for 10 h was found most suitable as it gives pure single-phase tetragonal *** constant was found as 25 43 at curie temperature of 480℃in the case of 5 wt.%of Gd in *** Coefficient was found as 241×10^(-12)C/N at 39 kv/cm of poling *** results obtained were comparable and even better than so far reported in similar kind of materials.

Discharge Plasma Assisted Adsorbents for Exhaust Treatment: A Comparative Analysis on Enhancing NO_x Removal

An analysis has been made on the discharge plasma coupled with an adsorbent system for NOx removal. The cascaded plasma-adsorbent system may be perceived as a better alternative for the existing adsorbent-based abatement system in the industry. In this study the exhaust is sourced from a diesel generator set. It was observed that better NO removal in a plasma reactor can be made possible by achieving higher average fields and subsequent NO2 removal can be improved using an adsorbent system connected in cascade with the plasma system. The paper describes various findings pertaining to these comparative analyses.

AIM： Passive immunotherapy using antibody against hepatitis B surface antigen （HBsAg） has been advocated in certain cases of Hepatitis B infection. We had earlier reported on the cloning and expression of a high affinity scFv derived from a mouse monoclonal （5s） against HBsAg. However this mouse antibody cannot be used for therapeutic purposes as it may elicit anti-mouse immune responses. Chimerization by replacing mouse constant domains with human ones can reduce the immunogenicity of this antibody.METHODs： We cloned the VH and V, genes of this mouse antibody, and fused them with CH1 domain of human IgG1 and C, domain of human kappa chain respectively. These chimeric genes were cloned into a phagemid vector. After initial screening using the phage display system, the chimeric Fab was expressed in soluble form in E. coli.REsULTs： The chimeric Fab was purified from the bacterial periplasmic extract. We characterized the chimeric Fab using several in vitro techniques and it was observed that the chimeric molecule retained the high affinity and specificity of the original mouse monoclonal. This chimeric antibody fragment was further expressed in different strains of E. coli to increase the yield.CONCLUsION： We have generated a mouse-human chimeric Fab against HBsAg without any significant loss in binding and epitope specificity. This chimeric Fab fragment can be further modified to generate a fulllength chimeric antibody for therapeutic uses.
