The changes in arachidonic acid (AA) and fatty acids profiles along the growth curve of Parietochloris incisa, a coccoid snow green alga, were studied in a 2.8 cm light-path flat photobioreactor, exposed to strong photon flux density [PFD, 2400 μEmol/(m 2·s)]. Sixteen fatty acids were identified by gas chromatography showing that AA was the dominant fatty acid (33%-41%) followed by linoleic acid (17%-21%). AA content was closely investigated with respect to total fatty acids (TFA), ash free dry weight (AFDW) of cell mass as well as total culture content. These parameters were influenced significantly in a similar manner by culture growth phase, i.e., slightly decreasing in the lag period, gradually increasing in the logarithmic phase, becoming maximal at the early stationary phase, starting to decrease at the late stationary phase, sharply dropping at the decline phase. The increase in AA per culture volume during the logarithmic phase was not only associated with the increase in AFDW but also connected with a corresponding increase in AA/TFA, TFA/AFDW as well as AA/AFDW. The sharp decrease in AA content of the culture during the decline phase was mainly due to the decrease in AA/TFA, TFA/AFDW and AA/AFDW, although AFDW declined only a small extent. Maximal AA concentration, obtained at the early stationary phase, was 900 mg/L culture volume, and the average daily net increase of AA during 9 days logarithmic growth was 1.7 g/(m 2·day). Therefore, harvesting prior to the decline phase in a batch culture, or at steady state in continuous culture mode seems best for high AA production. The latter possibility was also further confirmed by continuous culture with 5 gradients of harvesting rate.

AIM: To determine whether adding vitamin D, a potent immunomodulator, improves the hepatitis C virus (HCV) response to antiviral therapy. METHODS: Seventy-two consecutive patients with chronic HCV genotype 1 were randomized into two groups: the treatment group (n = 36, 50% male, mean age 47 ± 11 years) received Peg-α-2b interferon (1.5 μg/kg per week) plus ribavirin (1000-1200 mg/d) together with vitamin D3 (2000 IU/d, target serum level > 32 ng/mL), and the control group (n = 36, 60% male, mean age 49 ± 7 years) received identical therapy without vitamin D. HCV-RNA was assessed by realtime polymerase chain reaction (sensitivity, 10 IU/mL). The sustained virologic response (SVR) was defined as undetectable HCV-RNA at 24 wk post-treatment. RESULTS: Clinical characteristics were similar in both groups. The treatment group had a higher mean bodymass index (27 ± 4 kg/m2 vs 24 ± 3 kg/m2, P F2: 42% vs 19%, P < 0.001) than the controls. At week 4, 16 (44%) treated patients and 6 (17%) controls were HCV-RNA negative (P < 0.001). At week 12, 34 (94%) treated patients and 17 (48%) controls were HCV-RNA negative (P < 0.001). At 24 wk post-treatment (SVR), 31 (86%) treated patients and 15 (42%) controls were HCV-RNA negative (P < 0.001). Viral load, advanced fibrosis and vitamin D supplementation were strongly and independently associated with SVR (multivariate analysis). Adverse events were mild and typical of Peg-α-2b/ribavirin. CONCLUSION: Adding vitamin D to conventional Peg-α-2b/ribavirin therapy for treatment-na■ve patients with chronic HCV genotype 1 infection significantly improves the viral response.

AIM: To compare the effect of oral erythromycin vs no preparation with prokinetics on the transit time and the image quality of capsule endoscopy (CE) in evaluating small bowel (SB) pathology. METHODS: We conducted a retrospective, blinded (to the type of preparation) review of 100 CE studies, 50 with no preparation with prokinetics from one medical center (Group A) and 50 from another center with administration of a single dose of 200 mg oral erythromycin 1 h prior to CE (Group B). Gastric, SB and total transit times were calculated, the presence of bile in the duodenum was scored, as was cleanliness within the proximal, middle and distal intestine. RESULTS: The erythromycin group had a slightly shorter gastric transit time (21 min vs 28 min, with no statistical significance). SB transit time was similar for both groups (all P > 0.05). Total transit time was almost identical in both groups. The rate of incomplete examination was 16% for Group A and 10% for Group B (P = 0.37). Bile and cleanliness scores in different parts of the intestine were similar for the two groups (P > 0.05). CONCLUSION: Preparation for capsule endoscopy with erythromycin does not affect SB or total transit time. It tends to reduce gastric transit time, but it does not increase the cecum-reaching rate. Erythromycin does not adversely affect image quality. We consider the routine use of oral erythromycin preparation as being unjustified, although it might be considered in patients with known prolonged gastric emptying time.

Modeling and Stability Analysis of a Communication Network System

In this work, the modeling and stability problem for a communication network system is addressed. The communication network system consists of a transmitter which sends messages to a receiver. The proposed model considers two possibilities. The first one, that messages are successfully received, while in the second one, during the sending process the transmitter breaks down and as a result the message does not reach the receiver. Timed Petrinets is the mathematical and graphical modeling technique utilized. Lyapunov stability theory provides the required tools needed to aboard the stability problem. Employing Lyapunov methods, a sufficient condition for stabilization is obtained. It is shown that it is possible to restrict the communication network system state space in such a way that boundedness is guaranteed. However, this restriction results to be vague. This inconvenience is overcome by considering a specific recurrence equation, in the max-plus algebra, which is assigned to the timed Petri net graphical model.

The Almighty created man for contentment, and not for suffering. Creation of the universe was a leap from absolute entropy to substance and the vital world. According to our hypothesis, the goal of creating the universe and humans was to create enjoyment and pleasure by depressing the level of entropy. Current definitions do not adequately express the nature and origin of happiness or suffering, nor do they suggest how to treat, relieve, and prevent the torment of a healthy or sick individual. Our team developed and proposed the entropy theory as a new definition for human happiness and suffering. Entropy is a lack of order or a state of chaos. Human happiness may be defined as the complex of positive sensations, perceptions, emotions, or thoughts. These arise due to the depressed level of entropy of the individual＇s organism, empathy for others, or germane surroundings in the past, present, or in the future. Human suffering is the complex of negative sensations, perceptions, emotions, or thoughts that arise due to an increased level of entropy of a person＇s organism, empathy for others, or germane surroundings in the past, present, or future. A source of happiness and pleasure denotes depression of entropy in humans. Therefore, in order to treat suffering, one should ensure that the entropy level is reduced by complementing it with that which is missing.

A discrete event system is a dynamical system whose state evolves in time by the occurrence of events at possibly irregular time intervals. Timed Petri nets are a graphical and mathematical modeling tool applicable to discrete event systems in order to represent its states evolution where the timing at which the state changes is taken into consideration. One of the most important performance issues to be considered in a discrete event system is its stability. Lyapunov theory provides the required tools needed to aboard the stability and stabilization problems for discrete event systems modeled with timed Petri nets whose mathematical model is given in terms of difference equations. By proving stability one guarantees a bound on the discrete event systems state dynamics. When the system is unstable, a sufficient condition to stabilize the system is given. It is shown that it is possible to restrict the discrete event systems state space in such a way that boundedness is achieved. However, the restriction is not numerically precisely known. This inconvenience is overcome by considering a specific recurrence equation, in the max-plus algebra, which is assigned to the timed Petri net graphical model.
