A total of 288 grapevine samples of 61 different grapevine cultivars,collected from 22 provinces and regions,were analyzed by reverse transcription-PCR(RT-PCR) for the presence of grapevine fabavirus(GFabV).PCR detection results showed the incidences of GFabV were 12.8%(30/235) and 48.1%(25/52) in the asymptomatic and symptomatic vines,*** genetic diversity of GFabV isolates was analyzed based on partial nucleotide and encoded amino acid sequences of the RNA1 and RNA2 polyprotein *** analyses of the RNA1 and RNA2 gene sequences divided the GFabV isolates into five well-defined *** 1,2,and 4 comprised only Chinese *** article represents the first report for the prevalence and genetic diversity of GFabV in grapevines grown in China.

Vitis vinifera 87-1 plants infected by grapevine fleck virus(GFkV) and grapevine rupestris stem pitting-associated virus(GRSPaV) were used as the plant materials for virus elimination treatment. This study evaluated the effects of ribavirin at different concentrations(15 and 25 μg mL^(–1);R15 and R25, respectively), thermotherapy(37°C;T), and the combination of ribavirin and thermotherapy(R15+T and R25+T) on eliminating viruses from grapevine plants in vitro. Both R15 and R25 had phytotoxic effects and weakened plant growth. Thermotherapy positively affected the growth of grapevine plants. Plant height was significantly greater in T, R15+T, and R25+T than in CK, R15 and R25. The proportion of dead plants after T, R15+T, and R25+T was 51.4, 11.4, and 8.6%, respectively. The survival rates of regenerated plants after all treatments were >68.0%. Ribavirin concentration and treatment time were related to the regeneration of shoot tips and elimination efficiencies of the two viruses. The survival rates of plants after R15+T for 30, 40, and 50 days were 97.3, 90.7, and 74.4%, respectively. The elimination rates of GRSPaV from plants in the three time quantum were 55.6, 84.6, and 93.8%, respectively. The elimination rate of GFkV was 23.9% higher in R25(35/44) than in R15(25/45), and that of GRSPaV was 7.0% higher in R25 than in R15. The combination of thermotherapy and chemotherapy was found to have a positive effect on the eradication of GFkV and GRSPaV, and R25+T for 50 days was able to completely eliminate the two viruses from in vitro grapevines.

AIM To investigate the hepatic differentiation potential of human umbilical cord-derived mesenchymal stem cells(h UC-MSCs) and to evaluate their therapeutic effect on liver fibrosis/*** A CCl4-induced liver fibrotic/cirrhotic rat model was used to assess the effect of h UC-MSCs. Histopathology was assessed by hematoxylin and eosin(H&E), Masson trichrome and Sirius red staining. The liver biochemical profile was measured using a Beckman Coulter analyzer. Expression analysis was performed using immunofluorescent staining, immunohistochemistry, Western blot, and real-time *** We demonstrated that the infused h UC-MSCs could differentiate into hepatocytes in vivo. Functionally, the transplantation of h UC-MSCs to CCl4-treated rats improved liver transaminases and synthetic function, reduced liver histopathology and reversed hepatobiliary fibrosis. The reversal of hepatobiliary fibrosis was likely due to the reduced activation state of hepatic stellate cells, decreased collagen deposition, and enhanced extracellular matrix remodeling via the up-regulation of MMP-13 and down-regulation of TIMP-1. CONCLUSION Transplanted h UC-MSCs could differentiate into functional hepatocytes that improved both the biochemical and histopathologic changes in a CCl4-induced rat liver fibrosis model. h UC-MSCs may offer therapeutic opportunities for treating hepatobiliary diseases, including cirrhosis.

In this work,a force measurement system is proposed to measure the thrust of plasma microthruster with thrust magnitude ranging from sub-micro-Newtons to hundreds *** thrust measurement system uses an elastic torsional pendulum structure with a capacitance sensor to measure the displacement,which can reflect the position change caused by the applied force perpendicular to the pendulum *** the open-loop mode,the steady-state thrust or the impulse of the plasma micro-thruster can be obtained from the swing of the pendulum,and in the closed-loop mode the steady-state thrust can be obtained from the feedback force that keeps the pendulum at a specific *** thrust respond of the system was calibrated using an electrostatic weak force generation *** results show that the system can measure a thrust range from 0 to 200μN in both open-loop mode and closed-loop mode with a thrust resolution of 0.1μN,and the system can response to a pulse bit at the magnitude of 0.1 m N s generated by a micro cathode arc *** background noise of the closed-loop mode is lower than that of the open-loop mode,both less than 0.1 m N/Hz in the range of 10 mHz to 5 Hz.

With advances in imaging technology and surgical instruments,hepatectomy can be perfectly performed with technical precision for hepatocellular carcinoma(HCC).However,the 5-year tumor recurrence rates remain greater than 70%.Thus,the strategy for hepatectomy needs to be reappraised based on insights of scientific *** evidence has suggested that the main causes of recurrence after hepatectomy for HCC are mainly related to underlying cirrhosis and the vascular spread of tumor cells that basically cannot be eradicated by *** transplantation and systemic therapy could be the solution to prevent postoperative recurrence in this ***,determining the severity of liver cirrhosis for choosing the appropriate surgical modality,such as liver transplantation or hepatectomy,for HCC and integrating newly emerging immune-related adjuvant and/or neoadjuvant therapy into the strategy of hepatectomy for HCC have become new aspects of exploration to optimize the strategy of *** this new area,hepatectomy for HCC has evolved from a pure technical concept emphasizing anatomic resection into a scientific concept embracing technical considerations and scientific advances in underlying liver cirrhosis,vascular invasion,and systemic *** introducing the concept of scientific hepatectomy,the indications,timing,and surgical techniques of hepatectomy will be further scientifically optimized for individual patients,and recurrence rates will be decreased and long-term survival will be further prolonged.

Apple necrotic mosaic virus(ApNMV) was identified in crabapple trees with mosaic symptoms from Zaozhuang, Shandong Province, China, by reverse transcription polymerase chain reaction(RT-PCR) analysis. The complete nucleotide sequences of one isolate from crabapple(ApNMV-Hai) and two isolates from apple(ApNMV-Hua and-Qu) were determined. The sizes of genomic RNA1, 2 and 3 of the three isolates differed from those of the previously reported isolate ApNMV-P126 from Japanese apple, especially RNA3. Compared with the nucleotide(nt) sequence of RNA3 in isolate P126, those in the Hai and Qu isolates were 7 and 33 nt shorter, respectively, and that of isolate Hua was 7 nt longer. Alignment analyses showed that these differences in size were mainly due to differences in the lengths of the 5′ untranslated region(UTR) and the UTR region between the ORFs encoding the movement protein and the coat protein. In the phylogenetic trees constructed using the full genomic sequences of RNA1, 2 and 3, the isolate Hai clustered into a group with the isolate Qu in the RNA1 tree, but formed an individual branch in the RNA2 and 3 trees. Three recombination events were identified in the nucleotide sequences of RNA1 and 2 among the isolates ApNMV-Hai,-Hua, and-Qu. This is the first report of the full genome sequence of ApNMV in crabapple.

BACKGROUND As one of the subsets of CD8+T cells,Tc17 cells have recently been identified and are characterized by the secretion of interleukin(IL)-17,which is related to inflammatory *** To assess the status of Tc17 cells in cervical cancer and investigate the biological function of Tc17 cells in cervical cancer *** Flow cytometry assay,immunohistochemistry,and immunofluorescence were performed to detect the levels and phenotype of Tc17 cells in blood and tumor samples from patients with cervical *** to cell suspension culture,ELISA was carried out to measure the production of IL-6,IL-1β,IL-23,CXCL12,and IL-17 in tumor tissue supernatant and co-cultured supernatant of patients with cervical *** addition,multivariate analysis was performed to identify factors associated with overall survival using the Cox proportional hazards *** Compared with normal tissues,Tc17 cells specifically accumulated in tumor tissues of cervical cancer *** cells produced a greater amount of IL-6,IL-1β,and IL-23,which in turn promoted Tc17 cell *** the traditional cytotoxic CD8^+T cells,Tc17 cells secreted IL-17,which subsequently promoted CXCL12 expression in tumor cells,eventually enhancing the proliferation and migration of tumor ***,the ratio of tumor-infiltrating Tc17 cells was highly correlated with poor clinical outcome in patients with cervical *** Our data identified the oncogenic role of Tc17 cells in the development of cervical *** propose that the ratio of Tc17 cells may be a useful index in the prognosis of patients with cervical cancer.

Phyllanthus Urinaria L.(PUL) is a traditional Chinese medicine used to treat hepatic and renal disorders. However, the mechanism of its hepatoprotective action is not fully understood. In the present study, blood biochemical indexes and liver histopathological changes were used to estimate the extent of hepatic injury. GC/MS and LC/MS-based untargeted metabolomics were used in combination to characterize the potential biomarkers associated with the protective activity of PUL against CCl_4-induced liver injury in rats. PUL treatment could reverse the increase in ALT, AST and ALP induced by CCl_4 and attenuate the pathological changes in rat liver. Significant changes in liver metabolic profiling were observed in PUL-treated group compared with liver injury model group. Seventeen biomarkers related to the hepatoprotective effects of PUL against CCl_4-induced liver injury were screened out using nonparametric test and Pearson's correlation analysis(OPLS-DA). The results suggested that the potential hepatoprotective effects of PUL in attenuating CCl_4-induced hepatotoxicity could be partially attributed to regulating L-carnitine, taurocholic acid, and amino acids metabolism, which may become promising targets for treatment of liver toxicity. In conclusion, this study provides new insights into the mechanism of the hepatoprotection of Phyllanthus Urinaria.

We present the axial profiles of argon helicon plasma measured by a local optical emission spectroscope(OES) and Langmuir RF-compensated probe. The results show that the emission intensity of the argon atom lines(750 nm, 811 nm) is proportional to the plasma density determined by the Langmuir probe. The axial profile of helicon plasma depends on the discharge mode which changes with the RF power. Excited by helical antenna, the axial distribution of plasma density is similar to that of the external magnetic field in the capacitive coupled mode(E-mode). As the discharge mode changes into the inductively coupled mode(H-mode), the axial distribution of plasma density in the downstream can still be similar to that of the external magnetic field, but becomes more uniform in the upstream. When the discharge entered wave coupled mode(W-mode), the plasma becomes nearly uniform along the axis, showing a completely different profile from the magnetic field. The W-mode is expected to be a mixed pattern of helicon(H) and Trivelpiece-Gould(TG) waves.
