目的:⑴探究Halicin(SU3327)对E.faecalis的抗菌和抗生物膜活性。⑵探究Halicin与其他常规抗生素是否具有协同作用。⑶探究Halicin在体内的抗菌活性和对人类细胞系的细胞毒性作用。方法:⑴使用标准微量稀释法确定最低抑菌浓度(MIC),该方法美国临床和实验室标准协会(CLSI)制定。首先将菌振摇过夜生长至对数期,然后将菌悬液调至0.5麦氏浊度,再以MH培养基稀释100倍。将50μL梯度浓度的Halicin加入到96孔板中,再加入50μL的菌悬液。轻轻混匀,放入培养箱中,37℃培养16-18小时,最后通过酶标仪测量OD630。肉眼未见混浊的最低浓度的孔对应MIC。同时我们也测定了组合抗菌药物对E.faecalis ATCC29212和E.faecium ATCC19434的MIC。用MH培养基对药物进行连续稀释,得到梯度浓度的药物。两种药物的稀释液各50μL分别加入96孔板的列和行中,使每孔得到两种药物不同浓度组合的混合物,37℃孵育16-18小时,使用微孔板分光光度计测定OD 630。⑵为了确定时间-杀伤曲线,我们选择单个菌落的E.faecalis ATCC29212和E.faecium ATCC19434,并将其接种到BHI培养基中,在37°C下生长过夜至对数期。随后我们取菌液5 m L,分别置于不同的离心管中。4℃6000 rmp离心10 min,然后用生理盐水洗涤一次,收集细菌沉淀,并用含不同浓度Halicn的BHI培养基重悬,以DMSO作为对照。然后将菌悬液放入恒温摇床上,分别于0、2、4、8、12、24 h吸取菌液100μL倍比稀释后点样于血琼脂平37℃24 h⑶为了研究细菌膜和细胞形态,将ATCC29212在BHI培养基中培养过夜。然后用新鲜的BHI培基稀释过夜培养物并在37℃下培养3小时,使其处于对数生长期。然后,将细菌重新悬浮在MH培基中,菌液终浓度约为1×108 CFU/m L,并在37℃和200 rpm下,用Halicin处理1小时,再用4,000 rpm离心10分钟收集细菌沉淀,并用1 m L 2.5%戊二醛固定24小时。对于TEM,用0.1 m L二甲胂酸钠缓冲液洗涤固定细菌,然后用1%四氧化锇进行二次固定。最后,将样品用2%乙酸双氧铀染色并用TEM观察。对于SEM,样品溅射镀上一层薄金后通过SEM观察。⑸为了确定Halicin对E.faecalis ATCC29212细菌内膜的影响,我们使用SYTOX Green和Di SC3染色。细菌在BHI培基中生长过夜至对数期。然后将样品用PBS洗涤两次,细菌重悬于5 m M HEPES钠缓冲液中,使OD630=0.2。我们分别将10、5和2μM的SYTOX Green和Di SC3的荧光染料添加到细菌悬浮液中,然后在室温下避光孵育30分钟。最后,将50μL悬浮液添加到黑色96孔板50μL Halicin⑹最低生物膜抑制浓度(MBIC)是可以抑制细菌生物膜形成的最低药物浓度。我们将4μL过夜培养的菌液加入到含有196μL倍比稀释Halicin药物的96孔板中,同时不添加药物的BHI培养基和4μL细菌培养物作为阳性对照,200μL纯BHI培养基作为空白对照/阴性对照。然后将板在37℃孵育24小时,弃去培养基,用生理盐水洗涤两次以洗去浮游菌。XTT用PBS稀释至终浓度为0.2 mg/m L并与PMS混合。然后每孔加入200μL混合物,37℃避光孵育3h后,在OD 490处检测其吸光度,并与阴性和阳性对照的OD值进行MBIC⑺结晶紫(CV)染色生物膜。E.faecalis ATCC29212过夜培养,96孔板中加入196μL梯度浓度Halicin药物和4μL过夜培养的菌液,同时不添加药物的BHI培养基和4μL细菌培养物作为阳性对照,200μL纯BHI培养基作为空白对照/阴性对照。37℃下静态孵育24小时后,用盐水洗涤生物膜3次以去除浮游细胞。风干后,生物膜在室温下用结晶紫染色15分钟。然后,用盐水洗涤生物膜以去除未结合的染料,并将板在50℃下干燥30分钟。然后加入100μL95%乙醇溶解染料20分钟。最后在570 nm(A 570)处测量吸光度。⑻为了确定Halicin和DOX单独或联合使用分散生物膜的效果,我们在6孔板中添加了1960μL的BHI培养基,并在2 mm×2mm无菌载玻片存在下添加了40μL过夜细菌培养物。在此之前,我们将无菌盖玻片浸泡在75%酒精中,然后用镊子夹起盖玻片,将盖玻片上多余的酒精用火焰燃烧,达到消毒杀菌的目的。冷却后,将盖玻片放入6孔板中,随后37℃孵育24 h形成生物膜,同时使用含有0.1%DMSO的BHI培养基作为对照组。然后在镊子的帮助下取下盖玻片并用生理盐水轻轻冲洗。随后,我们将含有生物膜的盖玻片放入另一个6孔板中,该板中含有2m L新鲜BHI培养基,并含有不同浓度的Halicin和DOX,37℃下再孵

PA1426 regulates Pseudomonas aeruginosa quorum sensing and virulence:an in vitro study

Objective:Pseudomonas aeruginosa(P.aeruginosa)contains a hierarchy of quorum sensing(QS)network,consisting of the las,rhl and pqs systems,which play a key role in coordinating the expression of virulence factors.PA2146 was found to be associated with P.aeruginosa pathogenicity in macrophage and host Immune response.The aim of this study was to investigate the effects of PA2146 on the virulence of P.aeruginosa and explore its mechanism.Methods:PA2146 gene knockout strain and complement strain of P.aeruginosa PAO1 were constructed.The biomass of biofilm was detected by crystal violet staining;the virulence factors were measured,including pyocyanin,rhamnolipid,LasA elastase,LasB elastase and hemolytic activity;RNA-seq and label-free relative quantitative proteomics analyses were carried out to test the influence of PA2146 on transcriptomics and proteomics.This study was approved by the Institutional Review Board of the Third Xiangya Hospital,Central South University,China(approval No.2019-S021).Results:PA2146-deficient strains showed reduced biofilm formation and increased pyocyanin,rhamnolipid,LasA elastase,LasB elastase,and hemolytic activity,as well as increased motility,compared with the wild-type strain.RNA-seq and label-free relative quantitative proteomics analyses revealed that PA2146 repressed the transcription of several genes that are integral to the pqs system and to pyocyanin biosynthesis,and increased the expression of MexEF-OprN efflux pump components at the gene and protein level.Conclusion:PA2146 gene was found to inhibit the pqs system.PA2146 may affect quorum sensing by directly inhibiting the pqs system or by enhancing the expression of MexEF-OprN efflux pump components,thereby promoting efflux of 2-heptyl-4(1H)-quinolone,a Pseudomonas quinolone signal precursor,and thus affecting P.aeruginosa virulence.

本文研究了4 ℃贮藏条件下,羊肉僵直和解僵过程中风味品质的变化。以羊背最长肌为研究对象,采用超快速气相电子鼻和气相色谱质谱联用技术(Gas Chromatography-Mass Spectrometer,GC-MS),测定宰后不同贮藏时间羊肉挥发性风味物质的差异。结果表明,宰后贮藏过程中共检测出挥发性风味物质6类51种,主要为醛类和醇类物质,随着贮藏时间的延长,熟制羊肉的醛类物质含量先增加后降低,醇类物质的含量先降低后增加,己醛、庚醛、辛醛、壬醛、1-戊醇、1-辛烯-3-醇和1-庚醇为宰后贮藏过程中羊肉的关键风味物质,宰后1 d与1 h羊肉的共有关键风味物质含量无显著差异(p>0.05),宰后 3 d 和 5 d羊肉的风味物质种类和含量降低;超快速气相电子鼻主成分分析结果与 GC-MS 结果相一致。因此,僵直前(宰后 1 h)和解僵初期(宰后 1 d)羊肉的共有关键风味物质种类和含量差异不显著,解僵后期(宰后 3 d 和 5 d)羊肉关键风味物质含量和种类明显降低。

Anticipating the lifespan: Predicting the durability of an anode-supported solid oxide fuel cell short stack over 50,000 h

In the present study, the operational lifetime of a solid oxide fuel(SOFC) short stack is predicted by investigating the performance degradation of both the short stack and its cells throughout 1000 h at 800°C. The short stack and integral cell voltages are continuously measured during the long-term test, with electrochemical impedance spectroscopy(EIS) conducted every 200 h. The short stack voltage decreased rapidly for the initial 200–300 h and afterwards, it decreased at a slow rate due to the increase in the Ohmic and polarization resistances in the same manner. Scanning electron microscopy results show that there is no delamination or cracking among constituent layers of the short-stack cells. The single degradation effects of the Ni coarsening in the anode, cation migration and surface segregation in cathode and oxide scale growth in metallic interconnect mesh are successfully integrated into a comprehensive lifetime prediction model. The experimentally measured voltage degradation data of the short stack fits well with the developed mathematical model and allows the successful prediction of the lifetime up to 50,000 h.

Owing to the 4.0 industrial revolution condition monitoring maintenance is widely accepted as a useful approach to avoiding plant disturbances and shutdown.Recently,Motor Current Signature Analysis(MCSA)is widely reported as a condition monitoring technique in the detection and identification of individual andmultiple Induction Motor(IM)faults.However,checking the fault detection and classification with deep learning models and its comparison among them selves or conventional approaches is rarely reported in the literature.Therefore,in this work,wepresent the detection and identification of induction motor faults with MCSA and three Deep Learning(DL)models namely MLP,LSTM,and 1D-CNN.Initially,we have developed the model of Squirrel Cage induction motor in MATLAB and simulated it for single phasing and stator winding faults(SWF)using Fast Fourier Transform(FFT),Short Time Fourier Transform(STFT),and Continuous Wavelet Transform(CWT)to detect and identify the healthy and unhealthy conditions with phase to ground,single phasing and in multiple fault conditions using Motor Current Signature Analysis.The faults impact on stator current is presented in the time and frequency domain(i.e.,power spectrum).The simulation results show that the scalogram has shown good results in time-frequency analysis for fault and showing its impact on the energy of current during individual fault and multiple fault conditions.This is further investigated with three deep learning models(i.e.,MLP,LSTM,and 1D-CNN)for checking the fault detection and identification(i.e.,classification)improvement in a three-phase induction motor.By simulating the three-phase induction motor in various healthy and unhealthy conditions in MATLAB,we have collected current signature data in the time domain,labeled them accordingly and created the 50 thousand samples dataset for DL models.All the DL models are trained and validated with a suitable number of architecture layers.By simulation,the multiclass confusion matrix,precision,recall,and F1-score are obtaine

Microbial cell factories(bacteria and fungi)are the leading producers of beneficial natural products such as lycopene,carotene,herbal medicine,and biodiesel etc.These microorganisms are considered efficient due to their effective bioprocessing strategy(monoculture-and consortial-based approach)under distinct processing conditions.Meanwhile,the advancement in genetic and process optimization techniques leads to enhanced biosynthesis of natural products that are known functional ingredients with numerous applications in the food,cosmetic and medical industries.Natural consortia and monoculture thrive in nature in a small proportion,such as wastewater,food products,and soils.In similitude to natural consortia,it is possible to engineer artificial microbial consortia and program their behaviours via synthetic biology tools.Therefore,this review summarizes the optimization of genetic and physicochemical parameters of the microbial system for improved production of natural products.Also,this review presents a brief history of natural consortium and describes the functional properties of monocultures.This review focuses on synthetic biology tools that enable new approaches to design synthetic consortia;and highlights the syntropic interactions that determine the performance and stability of synthetic consortia.In particular,the effect of processing conditions and advanced genetic techniques to improve the productibility of both monoculture and consortial based systems have been greatly emphasized.In this context,possible strategies are also discussed to give an insight into microbial engineering for improved pro-duction of natural products in the future.In summary,it is concluded that the coupling of genomic modifications with optimum physicochemical factors would be promising for producing a robust microbial cell factory that shall contribute to the increased production of natural products.

Highly dense and ultrathin gadolinia-doped ceria interlayer for enhanced performance of large-area anode-supported solid oxide fuel cell

The fabrication of a highly dense and ultrathin Gd doped ceria(GDC) interlayer on a large-area, porous electrolyte-coated planar anode support using vacuum slurry coating is studied for application in solid oxide fuel cells(SOFCs). The electrolyte-coated planar anode support body is fabricated by tape casting and pre-sintering is performed at 1100, 1150, and 1200°C. The GDC interlayer is coated onto the pre-sintered electrolyte-coated anode support using a vacuum slurry coating process with different immersion times(30–60 s). A highly dense and ultrathin GDC interlayer with a thickness of 1.48 μm is produced after co-sintering at 1400°C. The 1200°C pre-sintered electrolyte-coated anode support showed a thin and highly dense GDC interlayer which is attributed to a lower shrinkage difference between the coated GDC interlayer and pre-sintered body. The electrochemical performance of a vacuum slurry-coated GDC interlayer cell is 38 % higher compared to that of the screen-printed GDC-coated cell at 700°C due to the greatly reduced Ohmic and electrode polarization resistances. The considerable improvement in the performance is attributed to the ultrahigh GDC density which has prevented the secondary phase formation between the cathode and electrolyte. This study presents the economical, scalable, and reproducible process for the fabrication of highly robust GDC interlayer for the large-area planar anode-supported SOFCs.

A New Generalized Weibull Model:Classical and Bayesian Estimation

Statistical distributions play a prominent role in applied sciences,particularly in biomedical sciences.The medical data sets are generally skewed to the right,and skewed distributions can be used quite effectively to model such kind of data sets.In the present study,therefore,we propose a new family of distributions suitable for modeling right-skewed medical data sets.The proposed family may be called a new generalized-X family.A special sub-model of the proposed family called a new generalized-Weibull distribution is discussed in detail.The maximum likelihood estimators of the model parameters are obtained.A brief Monte Carlo simulation study is conducted to evaluate the performance of these estimators.Finally,the proposed model is applied to the remission times of the stomach cancer patient’s data.The comparison of the goodness of fit results of the proposed model is made with the other competing models such as Weibull,Kumaraswamy Weibull,and exponentiated Weibull distributions.Certain analytical measures such as Akaike information criterion,Bayesian information criterion,Anderson Darling statistic,and Kolmogorov-Smirnov test statistic are considered to show which distribution provides the best fit to data.Based on these measures,it is showed that the proposed distribution is a reasonable candidate for modeling data in medical sciences and other related fields.

Asthenoteratozoospermia is one of the most severe types of qualitative sperm defects.Most cases are due to mutations in genes encoding the components of sperm flagella,which have an ultrastructure similar to that of motile cilia.Coiled-coil domain containing 103(CCDC103)is an outer dynein arm assembly factor,and pathogenic variants of CCDC103 cause primary ciliary dyskinesia(PCD).However,whether CCDC103 pathogenic variants cause severe asthenoteratozoospermia has yet to be determined.Whole-exome sequencing(WES)was performed for two individuals with nonsyndromic asthenoteratozoospermia in a consanguineous family.A homozygous CCDC103 variant segregating recessively with an infertility phenotype was identified(ENST00000035776.2,c.461A>C,p.His154Pro).CCDC103 p.His154Pro was previously reported as a high prevalence mutation causing PCD,though the reproductive phenotype of these PCD individuals is unknown.Transmission electron microscopy(TEM)of affected individuals’spermatozoa showed that the mid-piece was severely damaged with disorganized dynein arms,similar to the abnormal ultrastructure of respiratory ciliary of PCD individuals with the same mutation.Thus,our findings expand the phenotype spectrum of CCDC103 p.His154Pro as a novel pathogenic gene for nonsyndromic asthenospermia.
