BACKGROUND:The incidence of hepatocellular carcinoma (HCC)in China is closely related to the population infected with hepatitis B virus(HBV).HCC cells with HBV secrete soluble HBsAg into blood but do not express it on the cell membrane This study aimed to construct and investigate a new glycosyl phosphatidylinositol(GPI)-anchored protein(GPC3+α+EGFP) as a DNA vaccine against HCC associated with HBV. METHODS:A recombinant plasmid(pcDNA3.1(+)/GPC3+ α+EGFP)was constructed and verified by restriction endo nuclease digestion and ***3.1(+)/GPC3+α+ EGFP was transfected into HepG2 cells(experimental group) using lipofectamine ***-N1-transfected HepG2 cells were used as a negative control,and non-transfected HepG2 cells sreved as a blank ***2 cells that steadily expressed the fusion protein GPC3+α+EGFP were screened by G418,propagated,and co-cultured with lymphocytes from healthy *** proliferation was measured by the classic sulforhodamine B *** was assessed by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL),and Fas gene transcription was determined by quantitative fluorescent PCR. Hugoniot curve calculation of nitromethane decomposition mixtures:A reactive force field molecular dynamics approach

We investigate the Hugoniot curve, shock-particle velocity relations, and Chapman-Jouguet conditions of the hot dense system through molecular dynamics （MD） simulations. The detailed pathways from crystal nitromethane to reacted state by shock compression are simulated. The phase transition of N2 and CO mixture is found at about 10 GPa, and the main reason is that the dissociation of the C-O bond and the formation of C-C bond start at 10.0-11.0 GPa. The unreacted state simulations of nitromethane are consistent with shock Hugoniot data. The complete pathway from unreacted to reacted state is discussed. Through chemical species analysis, we find that the C-N bond breaking is the main event of the shock-induced nitromethane decomposition.

目的探索北京市中小学生青春发育时相与父母生育年龄的关系,为进一步了解儿童青少年青春发育提供参考。方法于2012年10月,采取分层整群抽样方法对3113名来自北京市房山区16所中小学校的学生进行问卷调查,采用多分类多因素logistic回归模型分析父母生育年龄对中小学生青春发育时相的影响。结果男生青春发育时相提前和延迟检出率分别为14.73%和9.61%,女生为12.84%和10.99%。调整了超重肥胖、低出生体重和家庭经济水平等因素后,多因素分析结果表明,相较于父亲生育年龄20岁~组(对照组),父亲生育年龄≥35岁组的男生青春发育提前风险升高2.36倍(95%CI=1.05~5.32),父亲生育年龄30岁~组的男生青春发育延迟风险降低为对照组的0.42倍(95%CI=0.18~0.97)。对女生而言,调整了超重肥胖、低出生体重和家庭经济水平等因素后,多因素分析表明,父亲生育年龄30岁~组和≥35岁组、母亲生育年龄30岁~组和≥35岁组的女生青春发育提前风险分别升高1.87倍(95%CI=1.10~3.19)、5.20倍(95%CI=2.68~10.08)和2.21倍(95%CI=1.32~3.70)、4.14倍(95%CI=1.79~9.53);进一步调整了父母亲生育年龄后,父亲生育年龄≥35岁组的女生青春发育提前风险是20岁~组的4.18倍(95%CI=1.73~10.13);未发现父母亲生育年龄和女生青春发育时相延迟存在相关性。结论父亲生育年龄大是男生青春发育时相提前和延迟的独立危险因素,也是女生青春发育时相提前的独立危险因素,应鼓励和倡导适龄生育。

Background： Cytokines play an important role in occorrence and recovery of hepatitis B virus （HBV） infection. The aim of this study was to investigate the changes ofcytokines concentration and its correlation to alanine aminotransferase （ALT）, HBV deoxyribonucleic acid （HBV-DNA）, hepatitis B envelope antigen （HBeAg）, and HBV surface antigen （HBsAg） in the development of chronic hepatitis B （CHB）. Methods： Thirteen healthy individuals （HI）, 30 chronic HBV-infected patients in immune tolerant （IT） phase, and 55 CHB patients were enrolled between August 2015 and May 2017. The peripheral blood samples were collected from all individuals. Tile levels of interferon （IFN）-α2, interleukin （IL）-10, transforming growth factor （TGF）-β1, HBV-DNA, HBsAg, and HBeAg and liver function were measured. The quantitative determinations of cytokines levels, including I FN-ct2, IL-10, and TGF-[31 were performed using luminex multiplex technology. The correlation of cytokines to ALT, HBV-DNA, HBsAg, and HBeAg was analyzed by linear regression analysis. Results： IFN-ct2 levels were similar between HI and IT groups （15.35 [5.70, 67.65] pg/ml vs. 15.24 [4.07, 30.73] pg/ml, Z = -0.610, P - 0.542）, while it elevated significantly in CH B group （35.29 [ 15.94, 70.15] pg/ml vs. 15.24 [4.07, 30.73] pg/ml; Z = -2.522, P = 0.012）. Compared with HI group （3.73 [2.98, 11.92] pg/ml）, IL-10 concentrations in IT group （5.02 [2,98, 10.11] pg/ml）, and CHB group （7.48 [3. I 0, 18.00] pg/ml） slightly increased （X^2 = 2.015, P - 0.365）, and there was no significant difference between IT and CHB group （Z =- 1.419, P = 0.156）. The TGF-β1 levels among HI （3.59 ±0.20 pg/ml）, IT （3.62 ±0.55 pg/ml）, and CHB groups （3.64±0.30 pg/ml） were similar （X^2=2.739, P = 0.254）. In all chronic HBV-infected patients （including patients in IT and CHB groups）, the elevation of IFN-α2 level was significantly associated with ALT level （β = 0.389, t = 2.423, P = 0.018）, and was also negatively correlated to HBV-DNA load （β = -0.35

3D thermoelasticity solutions for functionally graded thick plates

Thermal-mechanical behavior of functionally graded thick plates, with one pair of opposite edges simply supported, is investigated based on 3D thermoelasticity. As for the arbitrary boundary conditions, a semi-analytical solution is presented via a hybrid approach combining the state space method and the technique of differential quadrature. The temperature field in the plate is determined according to the steady-state 3D thermal conduction. The Mori-Tanaka method with a power-law volume fraction profile is used to predict the effective material properties including the bulk and shear moduli, while the effective coefficient of thermal expansion and the thermal conductivity are estimated using other micromechanics-based models. To facilitate the im-plementation of state space analysis through the thickness direction, the approximate laminate model is employed to reduce the inhomogeneous plate into a homogeneous laminate that delivers a state equation with constant coefficients. The present solutions are validated by comparisons with the exact ones for both thin and thick plates. Effects of gradient indices, volume fraction of ceramics, and boundary conditions on the thermomechanical behavior of functionally graded plates are discussed.

Nomogram based on tumor-associated neutrophil-tolymphocyte ratio to predict survival of patients with gastric neuroendocrine neoplasms

AIM To assess the predictive value of the tumor-associated neutrophil-to-lymphocyte ratio in terms of the clinical outcomes of patients with gastric neuroendocrine neoplasms after radical *** Data were retrospectively collected from 142 patients who were diagnosed with gastric neuroendocrine neoplasms and who underwent radical gastrectomy at our department from March 2006 to March 2015. These data were retrospectively analyzed, and a receiver operating characteristic curve analysis was used to identify the optimal value of the tumorassociated neutrophil-to-lymphocyte ratio. Univariate and multivariate survival analyses were used to identify prognostic factors. A nomogram was then applied to predict clinical outcomes after *** The tumor-associated neutrophil-to-lymphocyte ratio was significantly associated with tumor recurrence, especially with liver metastasis and lymph node metastasis(P 0.05 for all). A multivariate Cox regression analysis identified the tumor-associatedneutrophil-to-lymphocyte ratio as an independent prognostic factor for recurrence-free survival and overall survival(P luded the tumorassociated neutrophil-to-lymphocyte ratio, Ki-67 index, and lymph node ratio, was 0.788(0.759) for recurrence-free survival(overall survival) and was higher than the concordance index of the traditional TNM staging system [0.672(0.663)].CONCluSION The tumor-associated neutrophil-to-lymphocyte ratio is an independent prognostic factor in patients with gastric neuroendocrine neoplasms. Nomograms that include the tumor-associated neutrophil-to-lymphocyte ratio, Ki-67 index, and lymph node ratio have a superior ability to predict clinical outcomes of postoperative patients.
