Plasma Proteome Profiling with Monoclonal Antibody Libraries: A Pilot Biomarker Analysis for Nanomedicine-Induced Complement Activation

Complement (C) activation-related hypersensitivity reactions (HSrs) represent an unsolved adverse immune effect of many i.v. administered “nanomedicines”, such as liposomal doxorubicin (Doxil/Caelyx). Because these pseudoallergic reactions can be severe or even lethal, it is an important clinical objective to find biomarkers for proneness for C activation by reactogenic nanoparticles that will allow the prediction of in vivo reactions by in vitro assays. With this goal in mind we identified a normal human blood donor who consistently showed high sensitivity to Caelyx-induced C activation in vitro, whose plasma (Caelyx sensitive plasma, CSP) was subjected to proteome profiling with a library of human plasma proteome specific mAbs. The chip (PlasmaScan-380TM) contained 380 non redundant (with respect to epitopes) mAbs. The analysis revealed 8 proteins that were differentially represented in CSP in comparison with Caelyx-insensitive control plasma. These proteins were identified by mass spectrometry and Western blot analyses to represent factor H (decreased in CSP), factor H related protein, serum amyloid P component, fibronectin, complement component C4, Apo B100, prothrombin and alpha-2-HS glycoprotein (all increased in CSP). Some of these protein changes are consistent with proneness for increased C activation, suggesting the potential use of this method in the search for biomarkers for liposome-induced or other types of nanomedicine-induced HSrs.

On the functions of astrocyte-mediated neuronal slow inward currents

Integrated analysis of public datasets for the discovery and validation of survival-associated genes in solid tumors

Identifying genes with prognostic significance that can act as biomarkers in solid tumors can help stratify patients and uncover novel therapy ***,our goal was to expand our previous ranking analysis of survival-associated genes in various solid tumors to include colon cancer specimens with available transcriptomic and clinical data.A Gene Expression Omnibus search was performed to identify available datasets with clinical data and raw gene expression measurements.A combined database was set up and integrated into our kaplan-Meier plotter,making it possible to identify genes with expression changes linked to altered *** a demonstration of the utility of the platform,the most powerful genes linked to overall survival in colon cancer were identified using uni-and multivariate Cox regression *** combined colon cancer database includes 2,137 tumor samples from 17 independent *** most significant genes associated with relapse-free survival with a false discovery rate below 1%in colon cancer carcinoma were rBPMS(hazard rate[Hr]=2.52),TIMP1(Hr=2.44),and COL4A2(Hr=2.36).The three strongest genes associated with shorter survival in stage II colon cancer include CSF1r(Hr=2.86),FLNA(Hr=2.88),and TPBG(Hr=2.65).In summary,a new integrated database for colon cancer is presented.A colon cancer analysis subsystem was integrated into our kaplan-Meier plotter that can be used to mine the entire database(https://***).The portal has the potential to be employed for the identification and prioritization of promising biomarkers and therapeutic target candidates in multiple solid tumors including,among others,breast,lung,ovarian,gastric,pancreatic,and colon cancers.

High-resolution multi-scale air pollution system:Evaluation of modelling performance and emission control strategies

Anthropogenic noise interacts with the predation risk assessment in a free-ranging bird

Anthropogenic noise can affect a number of behavioral,physiological,and ecological aspects of animals from major taxonomic groups,raising serious conservation *** example,noise pollution impacts communicative behavior and perception of signals,movements and distribution,as well as predator–prey interactions,such as hunting success or predator detection and predation risk *** have carried out an experimental playback study,in which we investigated whether exposure to anthropogenic noise(sound of a tractor)distracts free-ranging barn swallows Hirundo rustica from paying attention to an approaching human“predator”(the“cognitive distraction”hypothesis),or whether noise leads to increased responsiveness to this“predator”(the“increased threat”hypothesis).The subjects were male barn swallows attending their breeding territories during the time when the females were *** found that barn swallow males initiated fight at signifcantly greater distances to the approaching human“predator”in the noise treatment than during the quiet control *** results suggest that anthropogenic noise causes increased vigilance and reactivity rather than a distraction,enabling birds to avoid the“predator”more *** further discuss the mechanism behind the increased alertness in response to noise and contrast the“increased threat”mechanism,usually tested in previous studies,with an alternative“cognitive sensitization”mechanism.

Selective deposition of a MOF at the spikes of Au nanostars for SErS detection

In the pursuit of advancing molecular sensing through surface-enhanced raman spectroscopy(SErS),the combination of plasmonic nanoparticles and metal-organic frameworks(MOFs)has emerged as a highly effective approach to enhance the sensitivity and selectivity of SErS ***,most prior investigations have predominantly focused on MOF-coated plasmonic nanoparticles in core@shell or layer-by-layer configurations,leaving a notable knowledge gap in exploring alternative *** we present a facile method to construct a particle-on-mirror architecture by selectively coating a MOF,zeolitic imidazolate framework-8(ZIF-8),onto the tips of Au nanostars and subsequently depositing the resultant nanoparticles onto a Au *** design integrates the electric field enhancement at the sharp tips and nanogaps,along with the molecular enrichment function within the porous MOF immobilized at the tips and nanogaps,leading to a substantial boost in the SErS signal *** a unique SErS platform enables consistent and outstanding SErS performance for analytes of different *** work opens up a promising strategy for constructing multifunctional nanostructures for sensitive SErS detection in real-life scenarios.

The commercial viability of thermoelectric(TE)devices relies heavily on two factors:cost reduction and efficiency *** this study,we first produce p-type Cu_(12)Sb_(4)S_(16-x)(x=0,3,4)using a low-temperature bottom-up approach and demonstrate Cu_(12)Sb_(4)S_(13)to show the best TE performance among the three tested ***,the TE energy conversion efficiency of Cu_(12)Sb_(4)S_(13)is further enhanced by optimizing its electronic band structure through the incorporation of small amounts of *** an optimal Te content of 5 mol%,more than a twofold increase in the TE figure of merit(zT)is *** gain insight into the mechanism of improvement on the transport properties of the mate-rial,we compare the interphase transport mechanism by incorporating nanodomains of different metals(Ag and Cu)into the Cu_(12)Sb_(4)S_(13)*** improved electrical conductivity obtained with Cu_(12)Sb_(4)S_(13)-Te nanocomposites is attributed to a charge flooding of the Cu_(12)Sb_(4)S_(13)*** contrast,excessive down-ward band-bending at the interphases of Ag/Cu metal-semiconductor drastically reduces the electrical ***,a weighted mobility(μw)analysis shows a dominant thermal activation of carri-ers in Cu_(12)Sb_(4)S_(13)-Te *** this material,a strong decrease in lattice thermal conductivity is also found,which is associated with a phonon-carrier scattering *** work shows the impor-tance of proper band-engineering in TE nanocomposites to decouple electrical and thermal transport to enhance TE performance,and the efficacy ofμw for electrical and thermal transport analysis.

Functionalized selenium nanoparticles ameliorated acetaminophen-induced hepatotoxicity through synergistically triggering PKCδ/Nrf2 signaling pathway and inhibiting CYP 2E1
