Transcriptomic analysis identifies the neuropeptide cortistatin (CORT) as an inhibitor of temozolomide (TMZ) resistance by suppressing the NF-kB-MGMT signaling axis in human glioma

Glioma is a common tumor originating in the brain that has a high mortality ***(TMZ)is the first-line treatment for high-grade ***,a large pro-portion of gliomas are resistant to TMZ,posing a great challenge to their *** the study,the specific functions and mechanism(s)by which cortistatin(CORT)regulates TMZ resis-tance and glioma progression were *** decreased expression of CoRT was detected in glioma tissues,and highly expressed CORT was associated with a better survival rate in pa-tients with *** overexpression notably decreased the capacity of glioma cells to pro-liferate and migrate in vitro and to form tumors in *** overexpression also markedly suppressed the viability and enhanced the apoptosis of TMZ-resistant U251 cells by regulating MGMT,p21,and Puma ***,CORT overexpression reduced the resistance of gliomas to TMZ in *** expression Was negatively correlated with MGMT expression in both glioma tissues and cells,and it was found that CORT inhibited NF-kB pathway activation in glioma cells,thereby inhibiting MGMT *** conclusion,CORT regulates glioma cell growth,migration,apoptosis,and TMZ resistance by weakening the activity of NF-kB/p65 and thereby regulating MGMT *** CORT/NF-kB/MGMT axis might be regarded as a molecular mechanism contributing to the resistance of glioma to *** data also suggest that CORT regulates the viability and metastatic potential of glioma cells,independent of its effects on TMZ resistance,providing evidence of novel therapeutic targets for glioma that should be evaluated infurther studies.

Gliomas are the most prevalent and aggressive malignancies of the nervous *** bioinformatic studies have revealed the crucial role of the secretory pathway kinase FAM20C in the prediction of glioma invasion and ***,little is known about the pathogenesis of FAM20C in the regulation of ***,we construct the full-length transcriptome atlas in paired gliomas and observe that 22 genes are upregulated by full-length transcriptome and differential APA *** of ATAC-seq data reveals that both FAM20C and NPTN are the hub genes with chromatin openness and differential ***,in vitro and in vivo studies suggest that FAM20C stimulates the proliferation and metastasis of glioma ***,NPTN,a novel cancer suppressor gene,counteracts the function of FAM20C by inhibiting both the proliferation and migration of *** blockade of FAM20C by neutralizing antibodies results in the regression of xenograft ***,MAX,BRD4,MYC,and REST are found to be the potential trans-active factors for the regulation of *** together,our results uncover the oncogenic role of FAM20C in glioma and shed new light on the treatment of glioma by abolishing FAM20C.

Dear Editor,This letter concerns the parameter tuning problem for nonlinear satellite buffer networks with communication delays, aiming to optimize their stability properties under L_(1)-gain. We first model the satellite buffer networks by a nonlinear time-delay positive system and propose a novel characterization under which the nonlinear system is asymptotically stable with a prescribed L_(1)-induced performance.

Damage evolution and removal behaviors of GaN crystals involved in double-grits grinding

Elucidating the complex interactions between the work material and abrasives during grinding of gallium nitride(GaN)single crystals is an active and challenging research *** this study,molecular dynamics simulations were performed on double-grits interacted grinding of GaN crystals;and the grinding force,coefficient of friction,stress distribution,plastic damage behaviors,and abrasive damage were systematically *** results demonstrated that the interacted distance in both radial and transverse directions achieved better grinding quality than that in only one *** grinding force,grinding induced stress,subsurface damage depth,and abrasive wear increase as the transverse interacted distance ***,there was no clear correlation between the interaction distance and the number of atoms in the phase transition and dislocation *** interacted distances between abrasives can decrease grinding force,coefficient of friction,grinding induced stress,subsurface damage depth,and abrasive wear during the grinding *** results of grinding tests combined with cross-sectional transmission electron micrographs validated the simulated damage results,*** atoms,high-pressure phase transition,dislocations,stacking faults,and lattice *** results of this study will deepen our understanding of damage accumulation and material removal resulting from coupling between abrasives during grinding and can be used to develop a feasible approach to the wheel design of ordered abrasives.

A resonant high-pressure microsensor based on a composite pressure-sensitive mechanism of diaphragm bending and volume compression

In this paper,a composite pressure-sensitive mechanism combining diaphragm bending and volume compression was developed for resonant pressure microsensors to achieve high-pressure measurements with excellent *** composite mechanism was explained,and the sensor structure was designed based on theoretical analysis and finite element *** all-silicon resonant high-pressure microsensor with multiple miniaturized cavities and dual resonators was developed,where dual resonators positioned in two resonant cavities with suitably different widths are used to perform opposite characteristics in pressure and the same characteristics at different temperatures,which can improve pressure sensitivities and realize temperature self-compensation by differential frequency *** microsensor was fabricated by microfabrication,and the experimental results showed that the sensor had an accuracy of±0.015%full scale(FS)in a pressure range of 0.1~100 MPa and a temperature range of−10~50℃.The pressure sensitivity of the differential frequency was 261.10 Hz/MPa(~2523 ppm/MPa)at a temperature of 20℃,and the temperature sensitivities of the dual resonators were−1.54 Hz/℃(~−14.5 ppm/℃)and−1.57 Hz/℃(~−15.6 ppm/℃)at a pressure of 2 *** differential output had an outstanding stability within±0.02 Hz under constant temperature and ***,this research provides a convenient solution for high-pressure measurements because of its advantages,namely,large range,excellent accuracy and stability.

Efficacy of Xylooligosaccharides and Probiotics Additive to the Diets of Sows and/or Piglets on Performance,Milk Composition and Appearance of Diarrhea in Sows and Their Offspring

The present study investigated the effects of dietary supplementation of xylooligosaecharides （XOS） and probiotics （PR） on performance, milk compo- sition and diarrhea incidence of sows or their offspring. From day 90 of gestation until weaning, a total of 60 muhiparous sows were randomly assigned to three groups and given three different diets that included either the control diet （ sCON, n = 20 ）, the control diet containing 175 mg/kg XOS （ sXOS, n = 20 ） or 175 mg/kg XOS and 300 mg/kg probiotics （sXOS ＋ PR, n =20）. On day 21 of age, offspring were assigned within sow treatments to the piglet basal diet （pCON） or the piglet basal, diet supplemented with 175 mg/kg XOS （pXOS） for 28 d. Sow feed intake was recorded daily. Body weight and feed consumption of the pigs in each replicate were determined biweekly （on day 0, 14 and 28） to calculate the average daily gain （ADG） of body weight, average daily feed intake （ADFI） and the gain-to-feed ratio （ G： F）. The colostrum and milk samples were obtained on day 0 and 14 of lactation, respectively. Sow XOS or XOS ＋ PR dietary had no effect on the number of total piglets born and born alive and the piglet weight at birth. Milk composition in sow colostrum and milk were not affected by sow diet XOS or （ XOS ＋ PR） supplementation （ P 〉 0.05 ）. No maternal dietary effect was observed on the weaning weight and pre-weaning weight gains of piglets. However, there were interaction effects between sow and piglet dietary treatments on diarrhea rate and diarrhea index during day 35 -49 （P = 0.03 ） and day 21 -49 （P =0.02）. Although sow diet XOS and （XOS ＋ PR ） supplementation did not influence growth performance of weanling piglets, such as ADG and ADFI （ P 〉0.05 ）, piglets XOS supplementation significantly decreased ADFI during the whole experiment period and increased G： F during day 35 -49 and day 21 -49 （ P 〈0.05 ）. 本文基于开发区优势特征评价指标体系,测度开发区与县区经济耦合度变化。结果表明:第一,开发区优势特征可划分为规格、结构、经济、人口四类,武汉城市圈内具有经济优势开发区的数量最少,有悖于开发区传统发展定位,而四类优势特征开发区均与县区经济达到高度耦合水平;第二,具有优势特征的开发区多沿“开发区优势轴线”分布,优势轴线处县区的耦合度波动变化明显,高耦合度县区集聚,但同时易出现耦合度降低和退级现象;第三,开发区与县区经济耦合度空间相关性逐渐降低,其中HH型先增后减,LH型长期存在且有增加趋势,县区间的协作关系减弱。

针对点对点(P2P)文件搜索技术存在网络带宽消耗大和查询速度慢等问题,为专用的P2P系统设计一种基于标签的文件搜索方案.该方案给出了将系统底层每个节点所控制的相关文件上传到中间层子服务器,及将顶层中央服务器接收到的文件查询转发到相关子服务器的方法,并运用标签优先顺序技术实现了查询的快速转发.性能评估结果表明,基于标签的文件搜索方法在转发查询过程中,必须检测的标签个数由一个很小的常数界定,从而节省了系统的网络带宽,提高了文件的搜索速度.

高速铁路车站道岔选型对咽喉长度及追踪间隔的影响

针对高速铁路车站采用大号码道岔造成的咽喉长度过长、列车走行距离远等问题,在明确车站咽喉长度和列车间隔时间计算方法的基础上,依托某尽端式高铁车站进行仿真计算,先设计不同道岔选型优化方案,再探究道岔选型、列车进站前速度和第一离去闭塞分区(简称“一离去”)长度对车站咽喉长度及列车间隔时间的影响。结果表明:若车站全部或部分采用12号道岔,与全部采用18号道岔相比,尽管列车追踪间隔时间会小幅增加,但车站咽喉长度可分别缩短30.66%和13.40%;在站外线路受到一定地形条件制约的情况下,若全部采用12号道岔时列车区间限行速度为260 km·h^(-1)、部分采用12号道岔时限行速度为280 km·h^(-1),则可同时压缩车站咽喉长度与到达追踪间隔;若全部采用12号道岔时限制一离去的无岔区长度不超过1000 m,部分采用12号道岔时限制长度不超过1400 m,则可同时压缩车站咽喉长度与出发追踪间隔。

云南是中药种植、生产大省,中医药对云南省发展起着至关重要的作用。发展中医药服务贸易不仅充分利用了云南省的特有资源,而且也是促进民族团结、走向共同富裕的重要途径。通过文献分析、经验总结、实证分析等方法对云南省中医药服务贸易出口现状进行分析,发现存在对中医药行业缺乏重视与监管、中医药国际文化差异影响服务贸易出口、国际标准对接不强且缺乏技术创新等问题。在此基础上提出加强人才培养,提高文化认同度,规范标准,强化政策支持力度、创新力度等建议。
