Study on Antibody Dynamics against Porcine Contagious Pleuropneumonia in Piglets

In order to provide basis for establishing a scientific and reasonable immune procedure against porcine contagious pleuropneumonia in pig farms, antibody dynamics in piglets inoculated with two vaccines against porcine contagious pleuropneumonia （ APP-1,2, 7 ） were detected. The result showed that average antibody blocking rate and antibody positive rate in piglets inoculated with two vaccines against porcine contagious pleuropneumonia exhibited similar variation trend. At 21 day after primary vaccination, average antibody blocking rate and antibody positive rate against porcine contagious pleuropneumonia in piglets reached the peak and then declined gradually. After secondary vaccination, average antibody blocking rate and antibody positive rate against porcine contagious pleuropneumonia in piglets increased gradually, which reached the second peak at 30 day after secondary vaccination （60 day after primary vaccination） ; subsequently, average antibody blocking rate declined gradually but still remained at a high level; antibody positive rate had little change. Average antibody blocking rate against APP-1 was slightly lower than the critical level of antibody protection at 44 day after primary vaccination ; antibody positive rate was slightly higher than the qualification rate at 37 day after primary vaccination; average antibody blocking rate against APP-7 was close to the critical level of antibody protection at 30 day after primary vaccina- tion, suggesting that piglets should be inoculated twice with polyvaccine against porcine contagious pleuropneumonia during the entire growth period, and the secondary vaccination should be performed at 30 -37 day after primary vaccination.

In order to establish the immune procedure for Actinobacillus pleuropneumonia and to determine the date for the first immunization on piglets, the growth and decline law of A. pleuropneumonia material antibodies in the piglets borne by cows inoculated with A. pleuropneumonia vaccines （type I, II, and VII） before delivery was detected. The results showed that type I, II, and VII maternal antibodies in piglets decreased gradually with the age growing overall, and was at the critical protection value at the ages of 42-50 days （type I） and 28 days （type VII）, lower than the quantification rate; and the antibodies all turned to be negative until the ages of 70 days （type I）, 60 days （type VII） and 35 days （type II）. The first immunization should be carried out at the age of 42-50 days using type I A. pleuropneumonia vaccine, and at the age of 28 days using type VII A. pleuropneumonia vaccine. However, type II A. pleuropneumonia maternal antibody had lower level and positive rate and could not well protect piglets, so the various A. pleuropneumonia vaccines differed in the date for the first immunization. In order to achieve a better immunization effect, A. pleuropneumonia vaccines with different valences should be further researched and developed.

Isolation and Sequence Analysis of NSP2, ORF3 and ORF5 of PRRSV XH-GD Strain from Guangdong Province

The diseased samples collected from Guangdong Province were inoculated to Marc-145 cells, PRRSV-specific cytopathogenic effects （CPE） such as cell aggregation, cell shrinkage and cell exfoliation were observed and a PRRSV strain XH-GD was obtained. By using primers designed in accordance with the pub- lished gene se^lences of PRRSV in GenBank, NSP2, ORF3 and ORF5 fragments were amplified by reverse transcription polymerase chain reaction and analyzed by sequence aligment. The results demonstrated that PRRSV strain XH-GD belonged to the North American genotype and the NSP2 gene contained discontinuous de- letions of 30 amino acids. NSP2, ORF3 and ORF5 genes of XH-GD shared 83.3% -98.9%, 88.6% -99.2% and 88.1% -99.2% nucleotide homology, and 76.8% -98.3%, 83.7% -98.8% and 88.1% -99.2% amino acid homology with HUB1, NX06, BJsy06, VR-2332, HB-1（sh）2002, HB-2（sh）2002, CH-1a and RespPRRS MLV, respectively; however, NSP2, ORb3 and ORF5 genes of XH-GD shared only 52.9%, 65.0% and 64. 3% nucleotide homology, and 31.3%, 57.5% and 58.9% amino acid homology with LV, respectively. Phylogenetic analysis revealed that XH-GD had a closer relationship with HUB1, NX06, BJsy06, VR-2332, HB-1 （sh） 2002, HB-2 （sh） 2002, CH-la and RespPRRS MLV strains compared with LV strain.

【目的】建立永生化猪视网膜色素上皮细胞系,为病毒学研究提供一种新的细胞材料。【方法】合成猪腺病毒3型E1基因,将其克隆至真核表达质粒pCI-neo中,构建E1基因真核表达质粒pCI-neo-E1,进行PCR和测序鉴定。提取pCI-neo-E1质粒,转染原代猪视网膜色素上皮细胞,用新霉素G418筛选猪永生化视网膜色素上皮细胞系(RPECs)。检测RPECs的角蛋白18和19,通过细胞计数测定其生长曲线,流式细胞仪检测细胞周期,并对其进行染色体核型分析。分别将猪流行性腹泻病毒(PEDV)CV77毒株、猪圆环病毒2型(PCV2)DBN-SX07株和猪伪狂犬病毒(PRV)Bartha-K61株接种RPECs,同时将PEDV CV77毒株接种绿猴肾细胞(Vero细胞),将PCV2 DBN-SX07株接种猪肾细胞(PK15细胞),将PRV Bartha-K61株接种仓鼠肾成纤维细胞(BHK细胞),观察细胞病变效应(CPE),并采用间接免疫荧光法测定病毒的滴度。【结果】PCR和测序结果显示,真核表达质粒pCI-neo-E1构建成功。将pCI-neo-E1转染原代猪视网膜色素上皮细胞,经G418筛选得到永生化细胞株,该细胞株表达角蛋白18和19,传代50代仍保持上皮样细胞形态,增殖活性良好,细胞周期及染色体核型特征与正常的二倍体细胞一致。PEDV CV77毒株感染猪RPECs 24 h后,CPE明显,滴度为10^(5.25)TCID_(50)/mL,低于其在Vero细胞中的滴度(10^(8.125) TCID_(50)/mL)。PCV2 DBN-SX07毒株感染猪RPECs后可产生明显CPE,病毒滴度为10^(6.125)TCID_(50)/mL,略高于其在PK15细胞上的滴度(106.0 TCID_(50)/mL)。PRV Bartha-K61毒株感染猪RPECs后,CPE明显,病毒滴度为10^(8.75)TCID_(50)/mL,略低于其在BHK上的滴度(108.875 TCID_(50)/mL)。【结论】成功构建了永生化猪视网膜细胞系,可用于猪源病毒的培养,尤其是培养PCV2可产生明显CPE,易于观察,为后续开展疫苗工艺的提升提供了一种新的细胞材料。

Phylogenetic Relationship Analysis of the Complete Genomes of Porcine Circovirus Type 2( PCV2) Strains Isolated from Hainan Province

[ Objective] This study aimed to investigate the molecular characteristics of porcine circovirus type 2 （PCV2） strains isolated from Hainan Province. [ Method] The complete genome of PCV2 was amplified from PMWS-suspected samples by PCR for sequence analysis. [ Result] A total of eight PCV2 strains were isolated and identified. All the eight isolates belonged to genotype PCV2b, among which seven isolates belonged to subgenotype PCV2b-1 C, and one isolate be- longed to subgenotype PCV2b-IA/1B. ORF2 gene of PCV2 isolates from Hainan Province was 705 bp in length, encoding 234 amino acids. Antigenic epitopes of Cap protein exhibited certain changes. Nucleotide sequences and deduced amino acid sequences of ORF2 gene shared 95.3% -99.7% and 93.6% - 100% simi- larities among eight PCV2 isolates from Hainan Province, respectively. Moreover, nucleotide sequences and deduced amino acid sequences of ORF2 gene of PCV2 isolates from Hainan Province shared 91.0% -99.9% and 91.0% -99.6% similarities with other PCV2 strain isolated from China （AY682994, AF381175, JX945577, JX682407, AY180397 ）, respectively; nucleotide sequences and deduced amino acid sequences of ORF2 gene of PCV2 isolates from Hainan Province shared 90.0% - 97.0% and 88.0% -97.9% similarities with PCV2 isolates from other countries （ NC_005148, JQ994268, KJ187306, AF201307, AF454546, AY＂/Sd020）, respectively; nucleotide sequences and deduced amino acid sequences of ORF2 gene of PCV2 isolates from Hainan Province shared 98.2% -100% and 94.9% -100% similarities with vaccine strain SH, respectively; nucleotide sequences and deduced amino acid sequences of ORF2 gene of PCV2 isolates from Hainan Province shared 90.6% -91.7% and 89.7% - 91.0% similarities with vaccine strain LG, respectively. [ Conclusion] This study provided theoretical hasis for the prevention and control of PCV2 and selection of vaccine strains in Hainan Province.

PRRS has brought great threats to swine industry in the world. The advances of studies on the viral proteins of PRRSV were reviewed from the genome,non-structural proteins and structural proteins of PRRSV.

Marek＇s disease（MD） is a lymphoproliferative disease of domestic chickens caused by a highly infectious,oncogenic alpha-herpesvirus known as Marek＇s disease virus（MDV）.The aim of this study was to compare the efficacy of four commercial MDV vaccines in Wenchang chicken.The 1-day old Wenchang chickens tested were injected with one of four different vaccines or not unvaccinated as control;five days later,they were then challenged by virulent MDV strain MD5.The results showed that,in comparison with HVT vaccines,the CVI988 vaccine gave the immunized chickens more potent immunities against challenges of MDV strain MD5.

提出一种在2~2.4GHz频段内工作的新型Wilkinson功率分配器,该种功率分配器主要由耦合线加载传输线和可配置电容构成。完成了该种新型Wilkinson功率分配器的仿真设计和实物加工测试。该新型Wilkinson功率分配器结构紧凑,有利于电路的小型化。同时,根据设计需求,可以通过配置不同容值的电容配置该种功分器的谐波抑制特性。测试结果显示,带内插损在3.6dB左右,带内回波损耗优于-10.5dB,输出端口隔离度优于-17dB,对二次谐波抑制优于20dB,对三次、四次和五次谐波抑制均优于15dB。

Preventive Effects of Five Drugs on Mycoplasma Pneumonia of Swine (MPS)

The paper was to explore the preventive effects of five drugs on mycoplasma pneumonia of swine （MPS） and to provide reference for clinical medication of pig farms in Hainan Province. A total of 444 health piglets were randomly divided into 6 groups, including five medication groups （72 piglets in group A, 74 pig- lets in group B, 72 piglets in group C, 76 piglets in group D, 76 piglets in group E） and one control group （74 piglets）. The piglets in experimental groups were treated drugs once a day for successive 5 days at 30, 60, 90, 120 and 150 of age. The piglets in control group were free of medication. At 70 and 140 days of age, 15 piglets of each group were randomly selected to collect their blood sermn. The Mycoplasma hyopneumoniae （M-Hyo） antibodies in serum were measured by en- zyme-linked immunosorbent assay （ELISA）. During the experiment, the incidence rates of respiratory disease, lung lesion, feed conversion rate, average daily gain （ADG）, and mortality rate of pigs were also observed and recorded. The results showed that the five drugs had significant difference in preventative effects. Group C （Zhiyuanjing group） received the best preventive effect and the highest economic benefits. Compared with control group, the ADG and feed conversion rate in group C were increased by 7.53% and 9.09%, respectively; the incidence rate of respiratory disease was reduced by 13.44% and lung lesion was alleviated by 81.43% ; and the earnings of each pig could rise by 132.70 yuan. The preventative effect and economic benefit of the drugs was sequenced by Chansu Kechuanling and Bingchan Kechuanwang. Wante Feilin and amoxicillin had weaker preventive effects against MPS but greatly influenced growth performance of pigs, so they should be used alternatively with other drugs.
