Neuroprotective effects of combined lead and cadmium,as well as N-acetylcysteine,on cerebral cortical neurons following lipid peroxidation injury

BACKGROUND： Studies have reported the antioxidant effects of lead and cadmium in the central nervous system, but very few have addressed the combined toxicity of lead and cadmium. The mechanisms by which these combined heavy metals are toxic, as well as how to protect cells from these agents, remains poorly understood. OBJECTIVE： Primary cultured rat cortical neurons were used to determine the effects of combined lead and cadmium on levels of glutathione peroxidase （GSH-Px）, superoxide dismutase （SOD）, catalase （CAT）, and acetylcholinesterase （ACHE）, as well as malondialdehyde （MDA）, and to evaluate the neuroprotective effects of N-acetylcysteine （NAC）. DESIGN, TIME AND SETTING An in vitro toxicological observation was performed at the Comparative Medicine Center of Yangzhou University from August 2007 to April 2008. MATERIALS： Lead acetate, cadmium acetate, and NAC were purchased from Sigma-Aldrich （St. Louis, USA）. Commercial kits of GSH-Px, SOD, CAT, ACHE, and MDA were purchased from Nanjing Jiancheng Bioengineering Institute, Nanjing, China. METHODS： The cerebral cortical neurons were isolated from newborn Sprague dawley rats at 24 hours after birth and primary cultured for 6 days. Thereafter, the cells were treated with a range of cadmium doses （0, 5.0, and 10.0μmol/L）, lead doses （0, 1.0, and 2.0 μmol/L）, or a combination of the two for 12 hours at 37℃in a 5% CO2 incubator, respectively. In addition, the cells were incubated with different doses of cadmium and/or lead and （0 and 50 μmol/L） NAC for 12 hours to assess the protective effects on cell survival. MAIN OUTCOME MEASURES： The activity of SOD, GSH-Px, CAT, and ACHE, as well as MDA content, in the cell lysates was detected using commercial kits. RESULTS： At 12 hours after treatment, compared to the control group, activity of GSH-Px, SOD, and AChE in the lead, cadmium, or combined treated cells was significantly decreased with increasing doses of cadmium/or lead （P 〈 0.05）, but CAT activity and MDA levels

Zinc alleviates the heat stress of primary cultured hepatocytes of broiler embryos via enhancing the antioxidant ability and attenuating the heat shock responses

Zinc(Zn)has been shown to attenuate the adverse effects of heat stress on broilers,but the mechanisms involving this process remain *** aimed to investigate possible protective mechanisms of Zn on primary cultured hepatocytes of broiler embryos subjected to heat *** experiments were *** Exp.1,hepatocytes were treated with 0,50,100,200,or 400μmol/L added Zn as inorganic Zn sulfate(iZn)for 12,24 or 48 *** Exp.2,cells were exposed to 40℃(a normal temperature[NT])and 44℃(a high temperature[HT])for 1,2,4,6,or 8 *** Exp.3,cells were preincubated with 0 or 50μmol/L Zn as iZn or organic Zn lysine chelate(oZn)for 8 h under NT,and then incubated with the same Zn treatments under NT or HT for 4 or 6 *** biomarkers of antioxidative status and heat stress in cells were *** results in Exp.1 indicated that 50μmol/L Zn and 12 h incubation were the optimal conditions for increasing antioxidant ability of *** Exp.2,the 4 or 6 h incubation under HT was effective in inducing heat shock responses of *** Exp.3,HT elevated(P<0.01)malondialdehyde content and expressions of heat shock protein 70(HSP70)mRNA and protein,as well as HSP90 ***,Zn supplementation increased(P<0.05)copper zinc superoxide dismutase(CuZnSOD)activity and metallothionein mRNA expression,and effectively decreased(P<0.05)the ex-pressions of HSP70 mRNA and protein,as well as HSP90 ***,oZn was more effective(P<0.05)than iZn in enhancing CuZnSOD activity of hepatocytes under *** was concluded that Zn(especially oZn)could alleviate heat stress of broiler hepatocytes via enhancing their antioxidant ability and attenuating heat shock responses.

Applying Dairy Cow Behavior in Management Practice

Applying dairy cow behavior in management practice is an effective way of improving cow health, welfare and performance. This paper first reviewed daily time budget and normal patterns of dairy cow behavior, and then discussed the influence of major management conditions and practices （such as competitive environments, stocking density, grouping strategies） on cow＇s feeding, lying and social behavior. Finally, new findings of using feeding behavior to predict disorders in transition period were addressed. It was suggested that dairy researchers and farmers should take advantage of related knowledge of dairy cow behavior to improve dairy cow health and welfare. More research is required to further study dairy cow behavior so as to better apply it in practical management and meet the needs of production.

Layer-structured Ruddlesden–Popper(RP)perovskites(RPPs)with decent stability have captured the imagination of the photovoltaic research community and bring hope for boosting the development of perovskite solar cell(PSC)***,two-dimensional(2D)or quasi-2D RP PSCs are encountered with some challenges of the large exciton binding energy,blocked charge transport and poor film quality,which restrict their photovoltaic ***,these issues can be readily resolved by rationally designing spacer cations of *** review mainly focuses on how to design the molecular structures of organic spacers and aims to endow RPPs with outstanding photovoltaic *** firstly elucidated the important roles of organic spacers in impacting crystallization kinetics,charge transporting ability and stability of *** we brought three aspects to attention for designing organic ***,we presented the specific molecular structure design strategies for organic spacers of RPPs aiming to improve photovoltaic performance of RP *** proposed strategies in this review will provide new avenues to develop novel organic spacers for RPPs and advance the development of RPP photovoltaic technology for future applications.

Expression of VFA Receptors GPR41 and GPR43 in Cow Abomasums

With the enlargement of intensive feeding scale of dairy cows and improvement of milk yield, the incidence rate of displacement of abomasum in dairy cows increases gradually in recent years. Previous researches have shown that VFA in abomasums can significantly inhibit the shrinkage of smooth muscles of abomasums and lead to atony of abomasums, but the specific regulatory mechanisms are still unclear. In this study, the expression of VFA receptors GPR41 and GPR43 in cow abomasums were detected by PCR method, and the fragments were consistent with the excepted sizes. This study provided theoretical basis for the pathologic mechanism investigation, prevention and treatment of displacement of abomasum in dairy cows.

Construction and Identification of Mammary Gland-specific Expression Vector of Bovine Tracheal Antimicrobial Peptide (TAP)

[ Objective] This study aimed to construct nmnmm_ry gland-specific expression vector of bovine tracheal antimicrobial peptide （TAP） gene. [ Method] TAP gene of dairy cattle was amplified from the mammary gland tissue by RT-PCR using a pair of primers which were designed according to bovine TAP cDNA se- quence （NM_174776） in GenBank, and then cloned into pMD19-T Simple vector for sequencing. The recombinant plasmid was digested using EcoRI and KpnI, the target gene fragment was recovered and inserted into general mammary gland-specific expression vector pBLG-EGFP harboring enhanced green fluorescent protein （ EGFP）, and transfected into bovine mammary epithelial cells （bMEC）, COS-7 cells and lactating rabbit mmmnary gland tissue by lipofectin transfection. The ex- pression of green fluorescent protein in transfected cells was detected under fluorescence microscopy, and the expression of TAP mRNA in rabbit mammary gland tis- sue was detected by semi-quantity RT-PCR. [ Result] The constructed mammary gland-specific expression vector pBLG-EGFP-TAP specifically expressed EGFP in transfected bMECs. In addition, semi-quantitative RT-PCR result showed that the expression level of TAP mRNA in rabbit mammary gland tissue was significantly enhanced after transfeeted with pBLG-EGFP-TAP. [ Conclusion] The mammary gland-specific expression vector pBLG-EGFP-TAP was successfully constructed, which provided important materials for further investigation of expression characteristics of TAP gene and prevention of bovine mastitis by using genetic engineering technology.

Application of CD45/SSC Gating Multiparameter Flow Cytometry in the Classification of Acute Leukemia——An Analysis of 139 Cases

In order to study the significance of flow cytometry immunophenotyping in the diagnosis of acute leukemia, CD45/SSC gating multiparameter flow cytometry (FCM) was utilized to analyze the immunophenotypes of 139 cases of acute leukemia. 139 cases of acute leukemia were enrolled in our hospital from April 1998 to April 2000. Morphological analysis and FCM immunophenotypic tests were conducted on all cases. Our results showed that CD45/SSC gating multiparameter flow cytometry immunophenotyping could reflect the origin of leukemic cells specifically. It is one of the important methods for the diagnosis of ALL, AML, and HAL. CD45/SSC gating multiparameter FCM analysis is a good technique for immunophenotyping. FCM immunophenotypic analysis can help improve the diagnosis and classification of acute leukemia, and extend the use of FCM in clinical practice.

Osteoprotegerin disrupts peripheral adhesive structures of osteoclasts by modulating Pyk2 and Src activities

Osteoprotegerin has previously been shown to modulate bone mass by blocking osteoclast maturation and *** detailed mechanisms of osteoprotegerin-induced disassembly of podosomes,disruption of adhesive structures and modulation of adhesion-related proteins in osteoclasts,however,are not well *** this study,tartrate-resistant acidic phosphatase staining demonstrated that osteoprotegerin inhibited differentiation of *** use of scanning electron microscopy,real-time cell monitoring and confocal microscopy indicated that osteoclasts responded in a time and dose-dependent manner to osteoprotegerin treatments with retraction of peripheral adhesive structures and detachment from the extracellular *** imaging and Western blot studies showed that osteoprotegerin induced dephosphorylation of Tyr 402 in Pyk2 and decreased its labeling in peripheral adhesion *** induced increased intracellular labeling of Tyr 402 in Pyk2,Tyr416 in Src,increased dephosphorylation of Tyr 527 in Src,and increased Pyk2/Src association in the central region of *** evidence suggests that Src may function as an adaptor protein that competes for Pyk2 and relocates it from the peripheral adhesive zone to the central region of osteoclasts in response to osteoprotegerin *** may induce podosome reassembly and peripheral adhesive structure detachment by modulating phosphorylation of Pyk2 and Src and their intracellular distribution in osteoclasts.

干细胞在疾病发生、细胞治疗、药物筛选和临床应用等方面具有巨大价值。自日本科学家Shinya Yamanaka发现诱导多能干细胞以来,相关的研究立即成为研究热点并持续至今。小鼠干细胞的研究为其它动物的相关研究开辟了道路,但在药物筛选和疾病治疗等方面有很大局限性。非人灵长类与人类具有更高的基因同源性,在生理、病理、生殖、药理、神经等多方面与人类高度相似,是研究人类疾病和开发人用药物的理想动物模型。非人灵长类研究也是新药在进入临床实验前必须的一个环节。非人灵长类干细胞的研究在疾病的机理研究和药物开发中越发重要,同时避免了伦理问题。本文从体外培养系统的优化和诱导分化的方法两个方面对非人灵长类多能干细胞的相关研究进行了综述,以期为非人灵长类干细胞进一步深入研究提供参考。

目的:观察沙格列汀对老年糖尿病肾病((diabetic nephropathy,DN)患者血糖相关指标以及血清相关因子肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和白细胞介素1β(IL-1β)水平的影响,评价其临床疗效和安全性。方法:选取我院2016年5月至2018年4月期间接受治疗的老年DN患者70例作为研究对象,将所有患者采用随机数字表法分为对照组和实验组,每组各35例。对照组患者采用常规治疗,实验组患者在常规治疗基础上给予沙格列汀治疗,并对治疗前后两组患者空腹血糖(FPG)、餐后两小时血糖(2hPG)、糖化血红蛋白(Hb A1c)、24 h尿白蛋白量(24hUpro)、血肌酐(Cr)、尿白蛋白肌酐比率(UACR)、TNF-α、IL-6和IL-1β水平进行检测。结果:对照组患者FPG、2hPG、24hUpro、Cr、TNF-α、IL-6和IL-1β水平均有不同程度改善。实验组患者治疗前后FPG、2hPG、24hUpro、Cr、Hb A1c、UACR水平差异均有统计学意义(P <0. 05)。治疗后与对照组患者相比,实验组患者FPG、2hPG、24hUpro、Cr、Hb A1c、UACR、TNF-α、IL-6和IL-1β的水平差异均有统计学意义(P <0. 05)。结论:DN患者治疗过程中,沙格列汀对患者FPG、2hPG、24hUpro、Cr、Hb A1c、UACR水平均有改善效果,同时可降低患者自身TNF-α、IL-6和IL-1β的水平,且对患者的不良反应小,在DN患者治疗中效果较好。
