Plasma Proteome Profiling with Monoclonal Antibody Libraries: A Pilot Biomarker Analysis for nanomedicine-Induced Complement Activation

Complement (C) activation-related hypersensitivity reactions (HSRs) represent an unsolved adverse immune effect of many i.v. administered “nanomedicines”, such as liposomal doxorubicin (Doxil/Caelyx). Because these pseudoallergic reactions can be severe or even lethal, it is an important clinical objective to find biomarkers for proneness for C activation by reactogenic nanoparticles that will allow the prediction of in vivo reactions by in vitro assays. With this goal in mind we identified a normal human blood donor who consistently showed high sensitivity to Caelyx-induced C activation in vitro, whose plasma (Caelyx sensitive plasma, CSP) was subjected to proteome profiling with a library of human plasma proteome specific mAbs. The chip (PlasmaScan-380TM) contained 380 non redundant (with respect to epitopes) mAbs. The analysis revealed 8 proteins that were differentially represented in CSP in comparison with Caelyx-insensitive control plasma. These proteins were identified by mass spectrometry and Western blot analyses to represent factor H (decreased in CSP), factor H related protein, serum amyloid P component, fibronectin, complement component C4, Apo B100, prothrombin and alpha-2-HS glycoprotein (all increased in CSP). Some of these protein changes are consistent with proneness for increased C activation, suggesting the potential use of this method in the search for biomarkers for liposome-induced or other types of nanomedicine-induced HSRs.

Mg-Y-Zn-Al alloys processed by rapidly solidified ribbon consolidation(RSRC)technique exhibit an exceptional mechanical performance indicating promising application *** material has a bimodal microstructure consisting of fine recrystallized and coarse non-recrystallized grains with solute-rich stacking faults forming cluster arranged layers(CALs)and nanoplates(CAnaPs),or complete long period stacking ordered(LPSO)*** order to reveal the deformation mechanisms,in-situ synchrotron X-ray diffraction line profile analysis was employed for a detailed study of the dislocation arrangement created during tension in Mg-0.9%Zn-2.05%Y-0.15%Al(at%)*** uncovering the effect of the initial microstructure on the mechanical performance,additional samples were obtained by annealing of the as-consolidated specimen at 300 and 400℃ for 2 *** heat treatment at 300℃ had no significant effect on the initial microstructure,its evolution during tension and,thus,the overall deformation behavior under tensile *** the other hand,annealing at 400℃ resulted in a significant increase of the recrystallized grains fraction and a decrease of the dislocation density,leading to only minor degradation of the mechanical *** maximum dislocation density at the failure of the samples corresponding to the plastic strain of 10-25% was estimated to be about 16-20×10^(14)m^(-2).The diffraction profile analysis indicated that most dislocations formed during tension were of non-basal and pyramidal types,what was also in agreement with the Schmid factor values revealed independently from orientation *** was also shown that the dislocation-induced Taylor hardening was much lower below the plastic strain of 3% than above this value,which was explained by a model of the interaction between prismatic dislocations and CAnaPs/LPSO plates.

Methionine sources and genotype affect embryonic intestinal development, antioxidants, tight junctions, and growth-related gene expression in chickens

Methionine(Met) is an essential and first limiting amino acid in the poultry diet that plays a significant role in chicken embryonic development and growth. The present study examined the effect of in ovo injection of DL-Met and L-Met sources and genotypes on chicken embryonic-intestinal development and health. Fertilized eggs of the two genotypes, TETRA-SL layer hybrid(TSL)—commercial layer hybrid and Hungarian Partridge colored hen breed(HPC)—a native genotype, were randomly distributed into four treatments for each genotype. The treatment groups include the following: 1) control non-injected eggs(noIn); 2) saline-injected(SaIn); 3) DL-Met injected(DLM); and 4) L-Met injected(LM). The in ovo injection was carried out on 17.5 d of embryonic development; after hatching, eight chicks per group were sacrificed, and the jejunum was extracted for analysis. The results showed that both DLM and LM groups had enhanced intestinal development as evidenced by increased villus width, villus height, and villus area(P ntrol. The DLM group had significantly reduced crypt depth, glutathione(GSH) content, glutathione S-transferase 3 alpha(GST3), occludin(OCLn) gene expression and increased villus height to crypt depth ratio in the TSL genotype than the LM group(P notype has overexpressed insulin-like growth factor 1(IGF1) gene, tricellulin(MD2), occludin(OCLn),superoxide dismutase 1(SOD1), and GST3 genes than the TSL genotype(P n conclusion, these findings showed that in ovo injection of Met enhanced intestinal development, and function, with genotypes responding differently under normal conditions. Genotypes also influenced the expression of intestinal antioxidants, tight junction, and growth-related genes.

AIM： To study the expression of endothelial and inducible nitric oxide synthases （enOS and inOS） and their role in inflammatory bowel disease （IBD）. METHODS： We examined the effect of sera obtained from patients with active Crohn＇s disease （CD） and ulcerative colitis （UC） on the function and viability of human umbilical vein endothelial cells （HUVEC）. HUVECs were cultured for 0-48 h in the presence of a medium containing pooled serum of healthy controls, or serum from patients with active CD or UC. Expression of enOS and inOS was visualized by immunofluorescence, and quantified by the densitometry of Western blots. Proliferation activity was assessed by computerized image analyses of Ki-67 immunoreactive cells, and also tested in the presence of the nOS inhibitor, 10^-4 mol/L L-nAME. Apoptosis and necrosis was examined by the annexin-V-biotin method and by propidium iodide staining, respectively. RESULTS： In HUVEC immediately after exposure to UC, serum enOS was markedly induced, reaching a peak at 12 h. In contrast, a decrease in enOS was observed after incubation with CD sera and the enOS level was minimal at 20 h compared to control （18%±16% vs 23%± 15% P〈0.01）. UC or CD serum caused a significant increase in inOS compared to control （UC： 300%±21%; CD： 275% ± 27% vs 108% ± 14%, P〈0.01）. Apoptosis/necrosis characteristics did not differ significantly in either experiment. Increased proliferation activity was detected in the presence of CD serum or after treatment with L-nAME. Cultures showed tube-like formations after 24 h treatment with CD serum. COnCLUSIOn： IBD sera evoked changes in the ratio of enOS/inOS, whereas did not influence the viability of HUVEC. These involved down-regulation of enOS and up-regulation of inOS simultaneously, leading to increased proliferation activity and possibly a reduced antiinflammatory protection of endothelial cells.

Role of ultrasound in colorectal diseases

Ultrasound is an undervalued non-invasive examination in the diagnosis of colonic diseases. It has been replaced by the considerably more expensive magnetic resonance imaging and computed tomography, despite the fact that, as first examination, it can usefully supplement the diagnostic process. Transabdominal ultrasound can provide quick information about bowel status and help in the choice of adequate further examinations and treatment. Ultrasonography,as a screening imaging modality in asymptomatic patients can identify several colonic diseases such as diverticulosis, inflammatory bowel disease or cancer. In addition, it is widely available, cheap, non-invasive technique without the use of ionizing radiation, therefore it is safe to use in childhood or during pregnancy, and can be repeated at any time. new ultrasound techniques such as elastography, contrast enhanced and Doppler ultrasound, miniprobes rectal and transperineal ultrasonography have broadened the indication. It gives an overview of the methodology of various ultrasound examinations, presents the morphology of normal bowel wall and the typical changes in different colonic diseases. We will pay particular attention to rectal and transperineal ultrasound because of their outstanding significance in the diagnosis of rectal and perineal disorders. This article seeks to overview the diagnostic impact and correct indications of bowel ultrasound.

Anthropogenic Pseudokarstic Depressions on Mount Bocskor(Bakony Region, Hungary)

Surface forms above the mine(ármin mine) of Mount Bocskor(Southern Bakony, Hungary) were examined. We made contour maps, plan maps, morphological maps and atectonic grike(cave) maps of some of the forms and their surroundings. We examined the distribution of the depth, length, elongation ratio in case of some depressions, the relation between the depth and the diameter of some depressions, the relationship between depression group directions and mine cut directions, the standard deviation of the direction differences of depression groups and of their depressions. The forms of the mountain related to surface mining can be separated to open and closed. The former are trenches and stairs, the latter are circular, elongated, and complex depressions. The formation of these forms can be related to the balancing movements of the vault over the mountain’s mine. At thin vault, stairs develop by collapses, while at the atectonic fissures of thicker vault, trenches and depressions are formed at the surface. In areas bordered by sinking(subsidence through) and downwardly cohesive faults, depression groups of diverse features are arranged in the marginal bands. Elongated depressions are formed at atectonic blocks bounded by dispersing faults in non-banded distribution. Where there is a superficial deposit, atectonic fissures can also be inherited directly by collapse to the surface and form depressions. They can also form indirectly over atectonic fissures by compaction, subsequent collapse and/or suffosion of the superficial deposit. The results of the study make it possible to analyse the material loss due to mining on the vault if the atectonic structures of the vault are partly or completely covered by superficial deposit.

Systematic exploration of signal-based indicators for failure diagnosis in the context of cyber-physical systems

Malfunction or breakdown of certain mission critical systems(MCSs) may cause losses of life, damage the environments, and/or lead to high costs. Therefore, recognition of emerging failures and preventive maintenance are essential for reliable operation of MCSs. There is a practical approach for identifying and forecasting failures based on the indicators obtained from real life processes. We aim to develop means for performing active failure diagnosis and forecasting based on monitoring statistical changes of generic signal features in the specific operation modes of the system. In this paper, we present a new approach for identifying emerging failures based on their manifestations in system signals. Our approach benefits from the dynamic management of the system operation modes and from simultaneous processing and characterization of multiple heterogeneous signal sources. It improves the reliability of failure diagnosis and forecasting by investigating system performance in various operation modes, includes reasoning about failures and forming of failures using a failure indicator matrix which is composed of statistical deviation of signal characteristics between normal and failed operations, and implements a failure indicator concept that can be used as a plug and play failure diagnosis and failure forecasting feature of cyber-physical systems. We demonstrate that our method can automate failure diagnosis in the MCSs and lend the MCSs to the development of decision support systems for preventive maintenance.

Stone–Wales defects in hexagonal boron nitride as ultraviolet emitters

Many quantum emitters have been measured close or near the grain boundaries of the two-dimensional hexagonal boron nitride where various Stone–Wales defects *** show by means of first principles density functional theory calculations that the pentagon–heptagon Stone–Wales defect is an ultraviolet emitter and its optical properties closely follow the characteristics of a 4.08-eV quantum emitter,often observed in polycrystalline hexagonal boron *** also show that the square–octagon Stone–Wales line defects are optically active in the ultraviolet region with varying gaps depending on their density in hexagonal boron *** results may introduce a paradigm shift in the identification of fluorescent centres in this material.
