人体基因编辑技术是生物技术发展的热点和重点,但随之也引起了技术风险、基因生物安全、人类社会伦理尊严和应用合法性的争议,引发了人们对应用该技术的担忧。国际上普遍禁止以生殖为目的基因编辑,各国依据本国情况建立法律规范体系来规范基因编辑技术的应用。我国应当建立基因编辑技术安全风险准入制度,保障国家生物安全,建立健全科研伦理规范体系,完善立法规范化建设,为研究和应用基因编辑技术提供法治保障。

随着数字经济迅猛发展,国际贸易行业也面临数字化转型升级。跨境电商产业作为发展速度最快、潜力最大、带动作用最强的外贸新业态,近年来显示出巨大的市场活力和增长韧性,成为外贸增长新引擎。广东省作为我国外贸大省,面对跨境电商转型升级加快以及国际贸易人才需求的持续增加,培养数字经济时代外贸行业所需的复合型人才,进而实现与企业人才需求的高效对接很有必要。文章通过分析广东省跨境电商政策及跨境电商发展现状,从我国数字经济发展规划、数字经济发展需求、外贸国际竞争力等方面阐述了优化国际贸易人才培养模式的必要性,并针对广东省高职院校国际贸易人才培养中现存的主要问题提出了若干优化建议。

背景:青蛙活性肽有丰富活性,如抗菌、抗肿瘤,有望解决抗生素耐药性问题。目的:从红眼树蛙皮肤分泌物中发现活性肽QUB2984,采用生物信息学手段模拟其结构和性质,合成、纯化、鉴定该肽并考察其生物学功能。方法:采用电刺激得到红眼树蛙皮肤分泌物,分离出mRNA构建cDNA文库,测序得到肽序列。用生物大分子序列搜索工具进行肽序列比对,蛋白质结构模型和功能预测工具开展蛋白二级结构模拟。通过固相合成法合成多肽、反向高效液相进行纯化、基质辅助激光解吸电离飞行时间质谱进行验证。所获得的纯肽用于生物活性评价,采用最小抑菌浓度法评价其抗菌作用,MTT法检测其细胞毒作用,溶血实验考察其安全性。结果与结论:①所得活性肽QUB2984的结构预测为α-螺旋,有完整的疏水面,对生物膜有一定破坏能力,其序列上第3个氨基酸位置由W组成,并具有G-X-G结构;②活性肽QUB2984对革兰阳性金黄色葡萄球菌的最小抑菌浓度为2μmol/L,对革兰阴性大肠杆菌的最小抑菌浓度为2μmol/L,对真菌白色念珠菌的最小抑菌浓度为8μmol/L;③活性肽QUB2984在10^(-5)mol/L浓度下对人非小细胞肺癌细胞NCI-H838具有明显抑制作用,在64μmol/L浓度下对马红细胞的溶血作用为50%;④结果显示,QUB2984具有抗菌抑癌活性,其带+3正电荷,有利于与细菌或细胞接触。

Stability-Considered Lane Keeping Control of Commercial Vehicles Based on Improved APF Algorithm

Regarding the lane keeping system,path tracking accuracy and lateral stability at high speeds need to be taken into account especially for commercial vehicles due to the characteristics of larger mass,longer wheelbase and higher mass center.To improve the performance mentioned above comprehensively,the control strategy based on improved artificial potential field(APF)algorithm is proposed.In the paper,time to lane crossing(TLC)is introduced into the potential field function to enhance the accuracy of path tracking,meanwhile the vehicle dynamics parameters including yaw rate and lateral acceleration are chosen as the repulsive force field source.The lane keeping controller based on improved APF algorithm is designed and the stability of the control system is proved based on Lyapunov theory.In addition,adaptive inertial weight particle swarm optimization algorithm(AIWPSO)is applied to optimize the gain of each potential field function.The co-simulation results indicate that the comprehensive evaluation index respecting lane tracking accuracy and lateral stability is reduced remarkably.Finally,the proposed control strategy is verified by the HiL test.It provides a beneficial reference for dynamics control of commercial vehicles and enriches the theoretical development and practical application of artificial potential field method in the field of intelligent driving.

目的探讨硕通镜联合输尿管软镜治疗2~3 cm肾下盏结石疗效及影响因素分析。方法回顾性选取2019年2月—2022年12月于玉林市第二人民医院接受治疗的肾下盏结石患者102例,根据治疗方法不同将其分为观察组和对照组,每组各51例。观察组患者采用硕通镜联合输尿管软镜治疗,对照组患者采用单纯输尿管软镜治疗。依据术后结石清除情况将患者分为结石未清除组(n=13)和结石清除组(n=89)。比较观察组和对照组患者的手术时间、住院时间、碎石时间、术中出血量、并发症发生率、结石清除率。广义估计方程分析评价治疗时间、治疗方法及两者的交互作用对视觉模拟评分(VAS)、白细胞(WBC)、血尿素氮(BUN)、血肌酐(Cr)、血红蛋白(HGB)、降钙素原(PCT)的影响。采用单因素及多因素Logistic回归分析评价影响一次性术后清石率的影响因素。基于危险因素构建列线图模型并对模型进行评价。结果与对照组相比,观察组患者的手术时间[(118.72±9.61) min比(136.65±11.27) min]、住院时间[(6.43±1.12) d比(10.29±2.23) d]及碎石时间[(51.23±10.38) min比(56.62±11.43) min]明显较短,术中出血量[(128.52±10.20) mL比(157.53±15.31) mL]明显较少,差异均具有统计学意义(P<0.05)。广义估计方程分析评价结果显示,治疗时间、治疗方法及两者的交互作用对WBC、HGB、BUN、Cr、PCT、VAS有明显影响(P<0.05)。与对照组比较,观察组患者的并发症发生率(5.88%比19.61%)明显降低,一次性术后清石率(94.12%比80.39%)明显增加,差异均具有统计学意义(P<0.05)。手术方式、下盏肾盂角(IPA)、肾盂下盏高度(CPH)、结石最大径均是2~3 cm肾下盏结石患者一次性术后清石率的影响因素(P<0.05)。本研究构建的列线图模型的区分度、校准度及临床实用性均较好,且可较好地识别2~3 cm肾下盏结石清除不彻底的高危患者。结论硕通镜联合输尿管软镜处理2~3 cm肾下盏结石是一种安全、有效的手术方式,具有操作简单、术中出血少,术后并发症少、一次性术后清石率高的优势。IPA、CPH、结石最大径、手术方式均是2~3 cm肾下盏结石患者一次性术后清石率的独立影响因素。本研究构建的列线图模型可较好地识别2~3 cm肾下盏结石清除不彻底的高危患者。

Lipogenesis is often highly upregulated in breast cancer brain metastases to adapt to intracranial low lipid microenvironments.Lipase inhibitors hold therapeutic potential but their intra-tumoral distribution is often blocked by the blood-tumor barrier(BTB).BTB activates its Wnt signaling to maintain barrier properties,e.g.,Mfsd2a-mediated BTB low transcytosis.Here,we reported VCAM-1-targeting nano-wogonin(W@V-NPs)as an adjuvant of nano-orlistat(O@V-NPs)to intensify drug delivery and inhibit lipogenesis of brain metastases.W@V-NPs were proven to be able to inactivate BTB Wnt signaling,downregulate BTB Mfsd2a,accelerate BTB vesicular transport,and enhance tumor accumulation of O@V-NPs.With the ability to specifically kill cancer cells in a lipid-deprived environment with IC_(50) at 48 ng/mL,W@V-NPs plus O@V-NPs inhibited the progression of brain metastases with prolonged survival of model mice.The combination did not induce brain edema,cognitive impairment,and systemic toxicity in healthy mice.Targeting Wnt signaling could safely modulate the BTB to improve drug delivery and metabolic therapy against brain metastases.

With over a million cases detected each year,skin disease is a global public health problem that diminishes the quality of life due to its difficulty to eradicate,propensity for recurrence,and potential for post-treatment scarring.Photodynamic therapy(PDT)is a treatment with minimal invasiveness or scarring and few side effects,making it well tolerated by patients.However,this treatment requires further research and development to improve its effective clinical use.Here,a piezoelectric-driven microneedle(PDMN)platform that achieves high efficiency,safety,and non-invasiveness for enhanced PDT is proposed.This platform induces deep tissue cavitation,increasing the level of protoporphyrin IX and significantly enhancing drug penetration.A clinical trial involving 25 patients with skin disease was conducted to investigate the timeliness and efficacy of PDMN-assisted PDT(PDMN-PDT).Our findings suggested that PDMN-PDT boosted treatment effectiveness and reduced the required incubation time and drug concentration by 25%and 50%,respectively,without any anesthesia compared to traditional PDT.These findings suggest that PDMN-PDT is a safe and minimally invasive approach for skin disease treatment,which may improve the therapeutic efficacy of topical medications and enable translation for future clinical applications.

Protective effect of marine animals hydrolysis-peptides on adjuvant-induced arthritis in mice by gut microbiota and SCFAs

Marine fauna provides a plentiful repository of peptides and bioactive proteins. Peptides and proteins isolated from marine animals have been studied and applied in the development of food supplements， drugs， and cosmeceutical products because of their special bioactivities， such as anti-inflammatory and antioxidant effects This study focused on exploring the alleviating effects of five major marine animal-derived peptides (Apostichopus japonicus， Acaudina leucoprocta， Melanogrammus aeglefinus， Phascolosoma esculenta and Rhopilema esculentum) on adjuvant-induced arthritis (AIA). The treatment with five marine animals-derived peptides downregulated the expression levels of pro-inflammatory cytokines of IL-1β， IL-17 and TNF-α in the bones of the mice with AIA and alleviated the rough surface of bone tissues significantly. A. japonicus-treatment ameliorates inflammation by restoring NF-κB pathway in AIA mice. High-throughput sequencing of the gut microbiota based on 16S rRNA sequencing revealed that A. japonicus peptide-treated AIA mice showed alterations and imbalance of intestinal flora and an increased abundance of Lactobacillus and Clostridium. Furthermore， metabolomic analysis showed that the level of SCFAs in the feces was enhanced to different degrees in mice treated with five major marine animal-derived peptides. Taken together， we propose that major marine animal-derived peptides can alleviate arthritis by improving the imbalance in the gut flora and increasing SCFA production to varying degrees.
