Sophoralines A–C,three novel[2+2]cycloaddition dimers of matrine-based alkaloids with an unprecedented 6/6/6/6/4/6/6/6/6 nonacyclic skeleton containing 11 stereogenic centers,were isolated from Sophora alopecuroides.Their structures were determined by spectroscopic methods,and the absolute configurations were further determined by single-crystal X-ray diffraction analysis for 1 and quantum chemical calculations of electronic circular dichroism(ECD)spectra for 2 and 3.Moreover,1 exhibited excellent hepatoprotective activities in acetaminophen-induced liver injury in vitro and in vivo.

Antibiotic fermentation residue(AFR)is nutrient-rich solid waste generated from fermentative antibiotic production process.It is demonstrated that AFR contains high-concentration of remaining antibiotics,and thus may promote antibiotic resistance development in receiving environment or feeding farmed animals.However,the dominate microorganisms and antibiotic resistance genes(ARGs)in AFRs have not been adequately explored,hampering understanding on the potential antibiotic resistance risk development caused by AFRs.Herein,seven kinds of representative AFRs along their production,storage,and treatment processes were collected,and multiple methods including amplicon sequencing,metagenomic sequencing,and bioinformatic approaches were adopted to explore the biological characteristics of AFRs.As expected,antibiotic fermentation producer was found as the predominant species in raw AFRs,which were collected at the outlet of fermentation tanks.However,except for producer species,more environment-derived species persisted in stored AFRs,which were temporarily stored at a semi-open space.Lactobacillus genus,classified as Firmicutes phylum and Bacilli class,became predominant bacterial taxa in stored AFRs,which might attribute to its tolerance to high concentration of antibiotics.Results from metagenomic sequencing together with assembly and binning approaches showed that these newly-colonizing species(e.g.,Lactobacillus genus)tended to carry ARGs conferring resistance to the remaining antibiotic.However,after thermal treatment,remaining antibiotic could be efficiently removed from AFRs,and microorganisms together with DNA could be strongly destroyed.In sum,the main risk from the AFRs was the remaining antibiotic,while environment-derived bacteria which tolerate extreme environment,survived in ARFs with high content antibiotics,and may carry ARGs.Thus,hydrothermal or other harmless treatment technologies are recommended to remove antibiotic content and inactivate bacteria before recycling of AFRs in pharmaceutical industry.

Three matrine-derived alkaloids, alopecuroidine A(1), alopecuroidine B(2 a) and alopecuroidine C(2 b)were isolated from the seeds of Sophora alopecuroides. Their structures were elucidated by extensive spectroscopic analyses and X-ray diffraction. Three compounds possess an unprecedented rearranged fused7/6/5/6 tetracyclic skeleton with a diazacycloheptane structure. Their plausible biosynthetic pathway was also proposed. The anti-proliferative activities of compounds 1 and 2 a were examined by the MTT assay.Compound 1 inhibited the viability of human lung cancer A549 cells, having a half maximal inhibitory concentration(IC50) of 7.58 ± 2.47 μmol/L at 72 h. The flow cytometric analysis suggested that 1 inhibited A549 cell growth by inducing apoptosis and cell-cycle arrest. Additionally, 1 induced the loss of mitochondrial membrane potential, elevated intracellular reactive oxygen species, increased the Bax/Bcl-2 ratio, stimulated cleaved-caspase-3 and P53 protein levels, and suppressed the pro-caspase-3 level. Thus,1 appeared to induce A549 cells apoptosis through a mitochondria-mediated apoptotic pathway.

The 1,2-dibromoethane-and Kl-mediatedα-acyloxylation of ketones is reported in moderate to good yield without the use of transition metals and strong oxidants.Various acids are well tolerated with wide functional group compatibility.An 1,2-dibromoethane-and Kl-catalysed reaction mechanism is proposed based on the results of control experiments.

Three new thionic compounds,(S)-2-(2-carboxyl-2-hydroxyethylthio)-ferulic acid(1),(E)-2-methoxy-4-(3-(methylsulfonyl)prop-1-en-1-yl)phenol(2), and thiosenkyunolide C(3), together with two new aromatic glycosides(4 and 5) were isolated from the rhizome of Ligusticum chuanxiong Hort. Two known compounds(6 and 7) were also obtained. Their structures were elucidated based on extensive spectroscopic data(UV, IR, 1D and 2D NMR, and HR-ESI-MS). Furthermore the absolute configurations were established by comparison of their calculated and experimental circular dichroism spectra and by a dimolybdenum tetraacetate [Mo_2(AcO)_4]-induced circular dichroism procedure. All compounds were evaluated against lipopolysaccharide(LPS)-induced NO production in BV2 cells, and compounds 4 and 5 showed strong inhibitory activities with IC_(50) values of 2.03 and 3.09 μmol/L, respectively(positive control curcumin, IC_(50)=6.17 μmol/L). In addition, compound 1 showed weak proteintyrosine phosphatase-1 B(PTP1B) inhibitory activity.

Tea(Camellia sinensis[L.]O.Kuntze.)is an important cash crop,which mainly uses tender shoots and young leaves for manufacturing.Due to the marketing characteristic that earlier made tea has higher price,the time of the breaking of winter dormancy buds in spring is extremely important in tea industry.Strigolactones are a group of carotenoids-derived metabolites which regulates bud outgrowth,shoot branching,tiller angle and environmental stress responses.The role of strigolactones in tea plant was briefly summarized in the current review,with an emphasis of the association of strigolactones on bud ecodormancy and shoot branching.The involvement of strigolactones on the biosynthesis of the tea characteristic metabolites flavonoids,caffeine and theanine were also discussed.Moreover,recent advances on the biosynthesis of strigolactones and its regulation by microRNAs and environmental stresses were also presented.This review provides a basis for future investigations underlying the mechanisms of strigolactones on bud winter dormancy and tea secondary metabolism.

OBJECTIVE:To observe and explore the effect of Fuling(Poria) in alleviating the spleen deficiency symptom pattern(SDSP).METHODS:We established an animal model of SDS in Sprague-Dawley(SD) rats by treating them with deficiency-inducing factors,including irregular feeding and tail clamping.Mice were administered Fuling(Poria) and its extracts(raw/cooked powder,aqueous/alcohol extract) by gavage once a day for 21 d.The body weight,rectal temperature,and spleen and thymus organ coefficients were calculated.The levels of motilin(MTL),gastrin(GAS),aquaporin 2(AQP2),interleukin 2(IL-2),IL-4,and 5-hydroxytryptamine(5-HT) in the serum and the level of AQP2 in the kidneys were evaluated by enzyme-linked immunosorbent assay.RESULTS:Fuling(Poria) and its extracts did not change the body weight,rectal temperature,and organ coefficients of the spleen and thymus.However,it reduced the levels of MTL and GAS and increased the levels of IL-2 and AQP2.In addition,the levels of IL-4 and 5-HT showed no significant alteration.CONCLUSIONS:These results suggested the crucial function of Fuling(Poria) in SDSP,especially promoting digestive function and water metabolism.

Two new compounds, named lyciumlignan D(1) and lyciumphenyl propanoid A(2), along with seven known compounds, were isolated from the root bark of Lycium chinense. Their structures were elucidated using spectroscopic data(UV, IR, HR-ESI-MS, 1D and 2D NMR, CD), as well as by comparison with those of the literature. Compounds 3–9 were isolated from this genus for the first time. In the in vitro assay, compounds 3, 6, and 7 exhibited stronger anti-inflammatory effects than the positive control curcumin at a concentration of 10 μmol/L.

以重庆市万州区为研究区,以生物多样性为切入点,综合运用形态学空间格局分析(MSPA)和最小累积阻力模型(MCR),识别GI网络中心(hubs)和连接廊道(links),结合图论、电路理论及几何形态学划分重要性等级,提取廊道上的生态夹点区域,并针对山地城镇的典型生态问题提出对应的精细化保护与修复策略。结果表明:1)万州区内网络中心42处,总面积996.35 km^(2),占研究区总面积的28.83%,总体“东密西疏”,形成“一屏三带、散点分布”的格局;2)连接廊道80条,总长度292.97 km,呈现“东强西弱、东短西长”的分布特征;3)生态夹点142处,占廊道长度的9.60%,主要分布于东部。据此,将网络中心分为连绵涵养区、重点保护区和独立修复区制定分区修复策略;将连接廊道分为关键廊道、重要廊道和一般廊道制定分级修复策略;对不同用地类型组合方式的生态夹点区域制定精准修复策略。
