目的探讨肺结核(TB)患者外周血低密度粒细胞(LDGs)的比例变化及其意义。方法采用流式细胞术分析80例TB患者及25例健康体检者外周血单个核细胞(PBMCs)层中CD14^(low)CD15^+LDGs的比例及变化情况,检测LDGs表面CD15、CD16、CD33、CD66b、CD62L的表达水平、凋亡坏死率及ROS水平。分析LDGs与TB发生发展以及转归的相关性。结果 TB患者外周血PBMCs层中LDGs的比例显著高于健康对照组(P<0.01);痰涂片抗酸阳性的TB患者LDGs的比例显著高于抗酸阴性者(P<0.01)。与TB患者正常密度粒细胞(NDGs)比较,TB患者LDGs表面CD15、CD33、CD66b表达量显著升高,CD62L的表达量显著下降,凋亡率上调,ROS水平上调。LDGs的比例在肺部有无空洞及空洞数量分组之间差异有统计学意义(P<0.05),即LDGs的比例随空洞累及范围的增加而升高。TB患者经抗痨治疗后,外周血LDGs的比例随治疗时间延长而下降,治疗3个月后,LDGs的比例显著降低,治疗6个月后与健康对照组无明显差异。结论 LDGs水平在TB患者外周血中显著上调,TB患者外周血中的LDGs为一群发生脱颗粒的活化中性粒细胞,且与TB的严重程度相关。

背景与目的 ANXA2在癌症进展中起着非常重要的作用,趋化因子18(chemokine ligand 18, CCL18)与肺腺癌(lung adenocarcinoma, LUAD)的侵袭、迁移、转移及预后不良有关。本研究旨在探究CCL18是否通过ANXA2促进LUAD侵袭以及其在LUAD侵袭中的作用和分子机制。方法 Western blot检测LUAD组织与癌周正常组织中ANXA2表达量,并检测转染效率及ANXA2作为上游调节剂在AKT/cofilin信号通路中的作用。细胞趋化实验、Transwell侵袭实验等实验探讨ANXA2对LUAD的作用机理。F-actin聚合实验和Western blot检测转染Si RNA的A549细胞侵袭是否与F-actin相关。结果与相邻非肿瘤组织相比,癌组织中A NX A2的蛋白表达水平升高(P<0.05)。敲低A NX A2组的LUA D细胞中CCL18诱导的趋化运动能力和侵袭能力下降(P<0.05)。与对照组相比,敲低ANXA2的LUAD细胞的F-actin聚合明显降低,而敲低ANXA2的LUAD细胞中AKT在Ser473和Thr308位点的磷酸化和cofilin、LIMK的磷酸化水平均降低(P<0.05)。结论敲低ANXA2可以通过降低AKT及下游通路磷酸化,进而降低CCL18对LUAD细胞的侵袭性的影响。

目的探讨基因预测软件Targetscan预测到的微小RNA-130a如何调节GTP酶激活蛋白SH3功能区结合蛋白2(GTPase activating protein SH3 binding protein 2,G3BP2)的表达,进而影响乳腺癌细胞的侵袭。方法应用实时荧光定量PCR(qRT-PCR)检测正常乳腺上皮及乳腺癌细胞系中miR-130a的表达水平;蛋白质印迹法(Western blot)检测改变miR-130a表达水平对乳腺癌细胞株MCF-7和MDA-MB-231中G3BP2及上皮间质转化(epithelial-to-mesenchymal transition,EMT)相关蛋白的影响;双荧光素酶报告基因实验检测miR-130a是否能与G3BP2靶向结合,以及Transwell侵袭实验检测miR-130a表达水平与MCF-7和MDA-MB-231细胞侵袭能力的关系。结果qRT-PCR显示miR-130a表达量在高侵袭乳腺癌细胞株明显高于低侵袭乳腺癌细胞株MCF-7及正常乳腺上皮细胞;Western blot检测显示,miR-130a负向调控G3BP2的表达并促进乳腺癌细胞发生EMT;qRT-PCR显示改变乳腺癌细胞内miR-130a表达后G3BP2 mRNA基本没有变化;双荧光素酶报告基因结果显示,miR-130a能与G3BP2 mRNA的3’UTR结合;Transwell侵袭实验显示,miR-130a促进乳腺癌细胞的体外侵袭。结论miR-130a通过靶向结合G3BP2 mRNA的3’UTR区,在翻译水平抑制G3BP2表达后促进乳腺癌细胞发生EMT,从而促进乳腺癌细胞的侵袭。

The Phototropism of Jurassic Petrified Wood in North China Plate

Trees on the side directly exposed to sunlight generally grow faster than on the opposite side, a phenomenon termed plant phototropism. There are in situ vertical trunks of silicified wood in the Xiadelongwan area of Yanqing County, north Beijing, where the first National Geologic Park of Petrified Wood of China has been built since 2002. A few trunks have well-preserved growth rings. One petrified stump from the formation shows a positive phototropism direction of SW230°. As compared with the modern normal growth stumps in Beijing plain area, which have a positive phototropism direction of SW210 °± 5°, the evidence of wood phototropism supports the conclusion of previous palaeomagnetic studies that the North China Plate has rotated clockwise since the Late Jurassic. The known petrified wood stumps in the Yanshan-Liaoning area are mainly found from the strata of 165-136 Ma, which corresponds to the main stage of the Yanshanian Movement.

Identification of a long non-coding RNA TCONS00019715 associated with polarization of macrophage

Mycobacterium tuberculosis infects approximately one-third of the world’s population. M. tuberculosis exploits macrophages to be the main host cell, which suggests that it has evolved mechanisms to counter bactericidal effectors of macrophages. Unfortunately, although extensively studied, the mechanisms of M. tuberculosis to defend against macrophages have not yet been elucidated completely. Mycobacterium bovis BCG is known to have the capacity to inhibit the positioning of iNOS on phagosomes by interfering with EBP50, a scaffolding protein that controls the recruitment of iNOS at the vicinity of phagosomes in macrophages. However, knockdown of the expression of EBP50 still facilitates the intracellular survival of BCG, which suggested that EBP50 may have some other unknown antimycobacterial properties. In this study we show that overexpression of EBP50 by a recombinant lentivirus had no effect on the iNOS recruitment to ***-containing phagosomes, but significantly promoted the elimination of intracellular ***. We revealed in the present study that the enhancement of intracellular killing to M. tuberculosis upon EBP50 overexpression was due to the increased level of apoptosis in macrophages. We showed that EBP50 overexpression significantly increased the expression of iNOS and generation of nitric oxide(NO), and EBP50-induced apoptosis was NO-dependentand mediated by Bax and caspase-3. Meanwhile, we found for the first time that *** significantly downregulated, while avirulent mycobacteria upregulated, the expression of EBP50 in macrophages. Thus, this study identified a novel role of EBP50 in increasing the apoptosis ratio of macrophages in an NO-dependent mechanism, and identified a novel mechanism of M. tuberculosis to evade immune elimination by suppressing the expression of EBP50 in macrophages.

Phototropism of Petrified Wood and Its Relation with the Rotation of Different Blocks in China and the Possibility of Application in the World

Long non-coding RNAs(lncRNAs) are non-protein coding transcripts longer than 200 nucleotides and play roles in various biological pathways. However, the role of lncRNAs in regulating Macrophages(Mφ) polarization has yet to be explored. The two states of Mφ activation, "Classical" and "Alternative" occupy two extremes of a phenotypic continuum in which Mφ respond to secreted factors to evoke distinct functional responses. In this study, we employed a lncRNA microarray-based profiling assay to document changes in the abundance of lncRNAs induced by the activation of primary monocyte-derived macrophages(MDMs) with two distinct polarizing conditions to span the spectrum of described activation patterns M(IFN-γ+LPS) and M(IL-4)). Data showed that when compared with primary MDMs, 9343 lncRNAs and 5903 mRNAs were deregulated in M(IFN-γ+LPS) group(fold change≥2.0, P<0.05), 4592 lncRNAs and 3122 mRNAs were deregulated in M(IL-4) group. There were significant difference in the expression levels of 326 lncRNAs and 275 mRNAs between three groups. RT-qPCR results were generally consistent with the microarray data. Furthermore, we found that TCONS0019715 is expressed at a higher level in M(IFN-γ+LPS) Mφ than in M(IL-4) Mφ. TCONS0019715 expression was decreased when M(IFN-γ+LPS) converted to M(IL-4) whereas increased when M(IL-4) converted to M(IFN-γ+LPS). Knockdown of TCONS0019715 following the activation of THP-1 cellls using IFN-γ and LPS diminished the expression of M(IFN-γ+LPS) markers, and elevated the expression of M(IL-4) markers.

Normally, trees on the side directly exposed to sunlight will grow faster than the opposing side. This phenomenon is termed plant phototropism. Moreover, palaeomagnetists have revealed that the Junnar Block has never rotated since the Mesozoic. The petrified woods in the Jiangjunmiao area of Qitai County show the positive phototropism direction of SSW220. By compared with the modern normal growth stumps in plain area, which have positive phototropism direction of SSW 219 ± 5, this observation supports the conclusion of palaeomagnetic researchers: the Junggar basin has never rotated since the Late Jurassic.

目的探讨TIGIT在系统性红斑狼疮(SLE)患者外周血NK细胞上的表达及临床意义,以阐明其在SLE发生和发展中的作用。方法应用流式细胞仪检测44例SLE患者和27例健康对照外周血NK细胞表面TIGIT表达水平,比较SLE组和健康对照组以及SLE稳定组与SLE活动组之间NKT细胞表面TIGIT表达水平,并分析其与实验室检查数据及治疗的相关性。2组间比较采用t检验或者非参数检验,2变量之间相关性采用Pearson相关分析。结果 SLE组NK细胞TIGIT表达百分率显著低于健康对照组(P0.05)。SLE患者中抗r RNP抗体阳性组外周血NK细胞TIGIT表达百分率显著低于对应阴性组(P<0.05)。SLE患者NK细胞TIGIT表达百分率与C3呈正相关(P<0.05),与SLE疾病活动度指数(SLEDAI)呈负相关(P<0.05)。经过治疗后SLE患者NK细胞TIGIT表达百分率可明显升高(P<0.05)。结论 SLE患者外周血NK细胞TIGIT表达异常,与SLEDAI、自身抗体产生、炎症有明确的相关性。
