Background Parkinson’s disease(PD)is the second most common neurodegenerative disease.The pathologic characteristic feature is the loss of dopaminergic neurons in the substantia nigra(SN).However,the biochemical mechanisms are unclear.A large number of studies have shown that oxidative damage is the primary cause of PD.Hence,antioxidants could become a suitable option to treat PD.The thioredoxin(Trx)system represents a useful,potentially disease-relevant oxidation-reduction system.Thioredoxin reductase 1(TR1)is a significant component of the Trx system.Methods The overexpression lentivirus(LV)or LV-TR1 in the TR1-A53T model of PD by the stereotactic brain,and successful overexpression of LV or LV-TR1 in the MPP^(+)-induced cellular model by LV or LV-TR1 transfection.Results We confirmed that interleukin-7 mRNA levels increased in MPP^(+)compared to that in the control and MPP^(+)-TR1 groups using quantitative polymerase chain reaction.Theγ-H_(2)AX level was increased in the Tg-A53T group compared to that in the TR1-A53T group by western blotting.The expression of Na^(+)-K^(+)-ATP was decreased in the MPP^(+)group compared to that in the control and MPP^(+)-TR1 groups by high content screening.Tg-A53T(the C57BL/6 mice transferred with mutant human a-syn);TR1-A53T(A53T mice which were injected TR1-LV 2µl in SNc on two sides with minipump).The mice were fed for 10 months.control(the N2a cells cultivated with DMEM);MPP^(+)(the N2a cells dealt with MPP^(+)(1 mM)48 h),MPP^(+)-LV(the N2a cells over-expressed LV for 24 h then dealt with MPP^(+)(1 mM)48 h).MPP^(+)-TR1(the N2a cell over-expressed TR1-LV for 24 h then dealt with MPP^(+)(1 mM)48 h).From the Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis,we confirmed that the overexpression of TR1 in SN pars compacta cells decreased oxidative stress,apoptosis,DNA damage,and inflammatory response and increased NADPH,Na^(+)-K^(+)-ATP,and immune response in this PD model.Conclusions Our study shows that overexpressed TR1 can be developed as a neuroprotective agent for PD.

A novel defluorinative ring-opening of gem-difluorocyclopropenes is presented,providing a concise and efficient method for accessing 2-fluoropropenals and 2-fluorobuta-1,3-dienes in moderate to good yields with excellent regio-and stereoselectivities.The reaction is performed under mild conditions with no need of using an excess amount of nucleophilic reagents.Water plays a crucial role in this transformation.

Because neurons are susceptible to oxidative damage and thioredoxin reductase 1 is extensively distributed in the central nervous system and has antioxidant properties, we speculated that the enzyme may be involved in the pathogenesis of Parkinson＇s disease. A Parkinson＇s disease model was produced by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine into C57BL/6 mice. Real-time reverse transcription-PCR, western blot analysis and colorimetric assay showed that the levels of thioredoxin reductase 1 mRNA and protein were decreased, along with a significant reduction in thioredoxin reductase activity, in the midbrain of Parkinson＇s disease mice compared with normal mice. Immunohistochemical staining revealed that the number of thioredoxin reductase 1-positive neurons in the substantia nigra pars compacta of Parkinson＇s disease mice was significantly decreased compared with normal mice. These experimental findings suggest that the expression of thioredoxin reductase 1 in the substantia nigra pars compacta of Parkinson＇s disease mice is significantly decreased, and that the enzyme may be associated with disease onset.

20世纪人类活动加剧全球氮沉降,而氮沉降已经导致全球生态系统功能退化和生物多样性丧失。该文综述大气氮沉降通过改变土壤微生物和植被组成影响昆虫种群及群落功能,重点论述外来物种和土著物种对氮添加的响应。外源氮添加主要通过食物网上行效应对昆虫产生级联效应,短期低氮添加对土著物种种群数量产生正效应,而长期高氮添加会抑制土著种群增长,降低生物群落多样性,破坏生态系统稳定,进而导致生态系统功能变化。外来昆虫对氮添加的响应更敏感,氮添加下外来昆虫种群数量增长更快,加速了外来物种的入侵过程。

采用基于力平衡理论的漂移流模型,对竖直圆管道内气体-液态铅铋合金(LBE)两相流的空泡份额进行预测。通过数值计算得到气体-液态LBE两相流在不同流道半径、Bankoff指数、Galileo数下的液相流速分布、切应力分布和空泡份额分布规律,分析了漂移流模型分布参数与上述宏观参数的内在联系。研究结果表明,预测得到的空泡份额及分布参数演化规律都与实验结果符合较好。本研究建立的数值计算方法能够用于圆管内气体-液态LBE两相流流动特性研究,为铅铋快堆蒸汽发生器传热管破裂(SGTR)事故中空泡份额等关键两相流参数的快速测算提供参考。

尖峰放电的脉冲神经网络(SNN)具有接近大脑皮层的信号处理模式,被认为是实现大脑启发计算的重要途径。但是,目前对于深度脉冲神经网络的学习仍缺乏有效的监督学习算法。受尖峰放电速率标识的时空反向传播算法的启发,该文提出一种面向深度脉冲神经网络训练的基于时间脉冲序列标识的监督学习算法,通过定义突触后电位和膜电位反传迭代因子分别分析脉冲神经元的空间和时间依赖关系,使用替代梯度的方法解决反传过程中不连续可微的问题。不同于现有基于尖峰放电速率标识的学习算法,该算法能够充分反映脉冲神经网络输出的时间脉冲序列的动态特性。因此,所提算法非常适合应用于需要较长时间序列标识的计算任务,例如行为的时间脉冲序列控制。该文在静态图像数据集CIFAR10和神经形态数据集NMNIST上验证了所提算法的有效性,在所有这些数据集上都显示出良好的性能,这有助于进一步研究基于时间脉冲序列应用的大脑启发计算。

A biosensor based on single-fiber optical tweezers is proposed,which can detect the motion trajectory of cells based on the stable capture and transmission of silica microspheres as well as biological yeast cells by using a tapered optical fiber as a sensing element.The interference cavity is formed by using the fiber tip and the target particle,the detected interference signal is demodulated using Hilbert transform,and the displacement curve of the particle is plotted to realize the particle motion trajectory tracking.This method provides potential technical support for process monitoring of targeted drug delivery in biomedicine.

Background: The brain is a common metastatic site in patients with non-small cell lung cancer (NSCLC), resulting in a relatively poor prognosis. Systemic therapy with epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitors (TKIs) is recommended as the first-line treatment forEGFR-mutated, advanced NSCLC patients. However, intracranial activity varies in different drugs. Thus, brain metastasis (BM) should be considered when choosing the treatment regimens. We conducted this network meta-analysis to explore the optimal first-line therapeutic schedule for advancedEGFR-mutated NSCLC patients with different BM statuses.Methods: Randomized controlled trials focusing on EGFR-TKIs (alone or in combination) in advanced andEGFR-mutant NSCLC patients, who have not received systematic treatment, were systematically searched up to December 2021. We extracted and analyzed progression-free survival (PFS) and overall survival (OS). A network meta-analysis was performed with the Bayesian statistical model to determine the survival outcomes of all included therapy regimens using the R software. Hazard ratios (HRs) and 95% confidence intervals (CIs) were used to compare intervention measures, and overall rankings of therapies were estimated under the Bayesian framework.Results: This analysis included 17 RCTs with 5077 patients and 12 therapies, including osimertinib + bevacizumab, aumolertinib, osimertinib, afatinib, dacomitinib, standards of care (SoC, including gefitinib, erlotinib, or icotinib), SoC + apatinib, SoC + bevacizumab, SoC + ramucirumab, SoC + pemetrexed based chemotherapy (PbCT), PbCT, and pemetrexed free chemotherapy (PfCT). For patients with BM, SoC + PbCT improved PFS compared with SoC (HR = 0.40, 95% CI: 0.17-0.95), and osimertinib + bevacizumab was most likely to rank first in PFS, with a cumulative probability of 34.5%, followed by aumolertinib, with a cumulative probability of 28.3%.
