Hierarchical Hydrogels with Ordered Micro-Nano Structures for Cancer-on-a-Chip Construction

In the drug therapy of tumor,efficient and stable drug screening platforms are required since the drug efficacy varies individually.Here,inspired by the microstructures of hepatic lobules,in which hepatocytes obtain nutrients from both capillary vessel and the central vein,we present a novel hierarchical hydrogel system with ordered micro-nano structure for liver cancer-on-a-chip construction and drug screening.The hierarchical hydrogel system was fabricated by using pregel to fill and replicate selfassembled colloidal crystal arrays and microcolumn array template.Due to the synergistic effect of its interconnected micro-nano structures,the resultant system could not only precisely control the size of cell spheroids but also realize adequate nutrient supply of cell spheroids.We have demonstrated that by integrating the hierarchical hydrogel system into a multichannel concentration gradients microfluidic chip,a functional liver cancer-on-a-chip could be constructed for high-throughput drug screening with good repeatability and high accuracy.These results indicated that the hierarchical hydrogel system and its derived liver cancer-on-a-chip are ideal platforms for drug screening and have great application potential in the field of personalized medicine.

Sex-specific participation in inter-group conflicts within a multilevel society:the first evidence at the individual level

Inter-group conflicts are common among many group-living animals and involve potentially complex motivations and interactions.Mammals living in multilevel societies offer a good opportunity to study inter-group conflicts.This study is the first to explore the function of sex-specific participation during inter-group conflicts within a multilevel society at the individual level.The Sichuan snub-nosed monkey(Rhinopithecus roxellana)is an endangered seasonal breeding species living in a multilevel society.From Sep 2007 to May 2008 we recorded 290 inter-group conflicts of a free-ranging provisioned band of R.roxellana in the Qinling Mountains of China to investigate the function of individual aggression during inter-group encounters.Our findings show that adult males were the main participants in inter-group conflicts,while females took part in them only occasionally.The male participation rate during the mating season,when adult females were estrous,was significantly higher than that during the non-mating season.Furthermore,males directed their aggression to other males,and directed more intense aggression towards bachelor males than towards other resident males.For both sexes,the participation rate as initiators was higher in the winter than that in the spring;and there was a significant positive correlation between group size and the participation rate as initiators.Our results suggest that inter-group aggression in Sichuan snub-nosed monkeys is linked to both mate defense and resource defense.

Sex differences in anogenital distances and digit ratios in wild David＇s rock squirrels Sciurotamias davidianus

The prenatal hormonal environment plays an important role in organizing sex differences in the morphology, physiology and behavior of humans as well as other vertebrates. Currently, all related research on rodents has been focused on Myo-morpha. This study presents data on sex differences in the anogenital distance (AGD) and the ratio of the second to the fourth digit length (2D:4D) from 44 wild David’s rock squirrelsSciurotamias davidianuscaptured from the Qinling Mountains, China. This was the first study of a species from Sciuromorpha. The AGD as well as body mass are sexually dimorphic. There are no sex differences or lateral asymmetry in 2D:4D. Significant correlation was not found between AGD and 2D:4D for any paw. The findings indicate that 2D:4D may not be fixed or influenced by the prenatal steroidal environment in David’s rock squirrels.

Recently,organ-on-chips have become a fast-growing research field with the widespread development of microfluidic chips and synthetic materials in tissue engineering.Due to the existing cardiotoxicity of many cardiovascular drugs,heart-onchips which are promising to replace traditional animal models have been extensively researched and developed to mimic human organ functions in vitro.The heart-on-chips mainly focus on cardiac mechanics,which is regarded as the central indicator of in vitro heart models and drug testing.Traditional methods for the detection of myocardial mechanics have been demonstrated complex and inefficient in heart-on-chips.Therefore,photonic crystal materials with unique optical properties have attracted interests and have been introduced into the heart-on-chips,developing a visualized self-reporting system for cardiomyocytes activity monitoring.In this review,photonic crystal-based heart-on-chips for biosensing are introduced,as well as the fabricationmethods and design criteria of them.The characterizations of the photonic crystal materials are classified into optical properties and structural properties,and their applications in cell culture and biosensing are further discussed.Then,several representative examples and developments of the integration of photonic crystal materials into microfluidic chips are described in detail.Finally,potentials and limitations are put forward to promote the development of the photonic crystal-based intelligent heart-on-chips.

Elastin-like polypeptide modified silk fibroin porous scaffold promotes osteochondral repair

Silk fibroin(SF)is considered biocompatible and biodegradable for osteochondral repair.However,it lacks a bioactive domain for cell adhesion,proliferation and differentiation,limiting its therapeutic efficacy.To revamp SF as a biomimicking and bioactive microenvironment to regulate cell behaviours,we engineered an elastin-like polypeptide(ELP,Val-Pro-Gly-Xaa-Gly)to modify SF fibers via simple and green dehydrothermal(DHT)treatment.Our results demonstrated that the ELP successfully bound to SF,and the scaffold was reinforced by the fusion of the silk fiber intersections with ELP(S-ELP-DHT)via the DHT treatment.Both bone mesenchymal stem cells(BMSCs)and chondrocytes exhibited improved spreading and proliferation on the S-ELP-DHT scaffolds.The ex vivo and in vivo experiments further demonstrated enhanced mature bone and cartilage tissue formation using the S-ELP-DHT scaffolds compared to the naked SF scaffolds.These results indicated that a recombinant ELPmodified silk scaffold can mimic three-dimensional(3D)cell microenvironment,and improve bone and cartilage regeneration.We envision that our scaffolds have huge clinical potential for osteochondral repair.

目的了解老年急性脑卒中患者术前家属代理决策现状并探究其影响因素。方法采用便利抽样法于2020年9月—2022年9月对本院117例老年急性脑卒中患者采取一般资料调查问卷、决策参与量表、决策冲突量表等进行问卷调查,并采用多元回归分析老年急性脑卒中患者术前家属代理决策的影响因素。结果老年急性脑卒中患者术前家属代理决策冲突得分为(40.22±11.05)分,对疾病了解程度比较,差异均无统计学意义(P>0.05);年龄、居住地、家庭月收入、与患者感情状况、以往就医体验、对疾病了解程度、期望决策类型比较,差异均有统计学意义(P<0.05);多元线性回归结果显示家庭月收入、与患者感情状况、文化程度为老年急性脑卒中患者术前家属代理决策的影响因素(P<0.05)。结论老年急性脑卒中患者术前家属代理决策冲突受到多种因素的影响,其中医护人员在其中起主要作用,应评估患者的决策现状制定针对性的干预方案从而改善患者家属代理的决策现状。

近年来,为了打破数据“壁垒”,联邦学习被广泛关注。联邦学习不需要客户端上传原始数据就能完成模型训练,保护了用户的隐私。针对客户端设备具有异构性的问题,考虑各个客户端对加速全局模型收敛的贡献程度和系统的通信开销,以最大化客户端在本地训练模型的权重变化量为优化目标,解决在一定系统训练周期下的联邦学习中的客户端选择优化问题。由此,提出了两个基于背包模型的联邦学习协议,分别是Offline KP-FL协议和Online KP-FL协议。Offline KP-FL协议基于离线背包模型选择合适的客户端参与全局模型的聚合更新。为了降低Offline KP-FL协议的复杂度,进一步基于在线背包模型选择用户提出了Online KP-FL协议。通过仿真发现,在特定情况下Offline KP-FL协议有更高的收敛速度,优于之前提出的方法。而与Offline KP-FL协议和Fed CS协议相比,Online KP-FL协议下,系统不仅每轮选择更少的用户,而且能够在Fed CS协议所需时间的64.1%内完成模型训练,使全局模型达到相同精度。

目的探讨急性胸痛患者发病后延迟就医的影响因素,为缩短延迟时间提供理论依据。方法以2018年8月至2021年8月于南华大学附属第二医院救治的178例急性胸痛患者为研究对象,按发生胸痛后就医时间>6 h和≤6 h分为延迟组(124例)、非延迟组(54例)。对所有患者均采用自制的调查问卷收集其一般资料,并采用单因素及Logistic回归分析急性胸痛患者延迟就医的影响因素。结果178例急性胸痛患者中,发病后及时就医的有54例(30.34%),延迟就医的有124例(69.66%)。单因素分析结果表示,年龄、文化程度、发病时有无人员陪同、家庭经济情况和转运方式均是急性胸痛患者延迟就医的影响因素,差异均有统计学意义(P均<0.05)。Logistic回归分析结果表示,年龄、文化程度、发病时有无人员陪同、家庭经济情况和转运方式均是急性胸痛患者延迟就医的独立影响因素,差异均有统计学意义(P均<0.05)。结论大部分急性胸痛患者普遍存在就医延迟现象,年龄、文化程度、发病时有无人员陪同、家庭经济情况和转运方式均是急性胸痛患者延迟就医的独立影响因素。

采用热交联、京尼平交联、原花青素交联和戊二醛/聚乙二醇交联4种方法制备类弹性蛋白-蚕丝纤维多孔支架材料。利用扫描电镜、傅立叶变换红外光谱和力学性能测试研究了交联类弹性蛋白-蚕丝纤维多孔材料的内部形态和结构。通过细胞毒性实验分析了不同方法交联类弹性蛋白-蚕丝纤维多孔材料的细胞相容性。结果表明,热交联和京尼平交联能制备出的类弹性蛋白-蚕丝纤维多孔支架材料具有较高和较稳定的力学性能,且具有良好的细胞相容性。

应用PSO-RBF神经网络预测太阳能PV/T系统的热、电性能

为准确预测太阳能光伏光热(Solar Photovoltaic/Thermal,PV/T)系统的热、电性能,文章利用PSO(Particle Swarm Optimization)算法优化了RBF(Radial Basis Function)神经网络,并基于此方法建立了太阳能PV/T系统性能的仿真预测模型,与基于未优化RBF神经网络建立的预测模型进行了对比分析。同时,搭建了太阳能PV/T实验平台,通过云平台采集实验数据用于上述模型。研究结果表明:使用PSO算法优化后的RBF神经网络模型相较于未优化模型预测精度提高了20%,预测稳定性提高了30%,拟合优度R值有所提升。基于PSO-RBF神经网络建立的预测模型可精确预测太阳能PV/T系统的热、电性能。
