Transcriptional control of pancreaticβ-cell identity and plasticity during the pathogenesis of type 2 diabetes

Type 2 diabetes(T2D)is caused by insulin resistance and insufficient insulin *** has increasingly indicated that pancreaticβ-cell dysfunction is the primary determinant of T2D disease progression and *** plasticity is an important feature of pancreaticβ-*** T2D development,pancreaticβ-cells undergo dynamic ***β-cell death/apoptosis in later-stage T2D is the major cause ofβ-cell dysfunction,recent studies have revealed thatβ-cell dedifferentiation and reprogramming,which play critical roles inβ-cell functional regulation in the early and middle T2D progression stages,are characterized by(i)a loss of matureβ-cell-enriched genes;(ii)dedifferentiation to a progenitor-like state;and(iii)transdifferentiation into other cell *** roles of transcription factors(TFs)in the establishment and maintenance ofβ-cell identity during pancreatic development have been extensively ***,we summarize the roles and underlying mechanisms of TFs in the maintenance ofβ-cell identity under physiological and type 2 diabetic *** feasible approaches for restoring islet functions are also discussed.A better understanding of the transcriptional control ofβ-cell identity and plasticity will pave the way for developing more effective strategies,such asβ-cell regeneration therapy,to treat T2D and associated metabolic disorders.

Dual innervation of spinal ganglion neurons on perineum and bladder

Experiments were performed on 13 Wister rats. Fastblus (FB) was injected into subcutaneous tissue in perineum area and nuclear yellow (NY) was injected into subserous lamina of the bladder wall, respectively. FB, NY and FB+NY were mostly found in the spinal ganglions of L6, S1 ans S2 segments. The numbers of FB,NY and FB+NY labeled cells were 146, 186 and 81, in a total of 463 labeled cells and their proportion was 40%, 51% and 9%, respectively. The result indicates that the spinal ganglion neurons dually innervate both the somatic and visceral tissues, and the convergence of somato_visceral sensory pathways might occur in the spinal ganglion cells.

The Modern Approach to "Xungu Leads to(Confucian) Yili"

The proposition that "xungu tong yili"(an exegetical tradition leading to Confucian teachings) expresses the core idea of traditional Chinese philology, which threads throughout the Han and Song dynasties and reached its historical acme in the form of the Qian-Jia school. The overemphasis of scholars from this school on exegesis led to the division between Hanxue(Qing scholarship in the Han dynasty tradition) and Songxue(Qing scholarship in the Song dynasty tradition) and a disconnection between "xungu(Chinese exegetical tradition exegesis) and yili(Confucian teachings)." The modern "linguistized"shift has seen the slow death of the xunguxue tradition(the theory and practice of xungu as a discipline), under which "xungu tong yili." At the same time, modern xunguxue has clarified its own disciplinary orientation and has made great progress in both theory and practice,laying a reliable foundation for the renewed process of working out meanings through *** heights of modern xunguxue allow the scholarly development of the meanings behind the classic texts in five respects: semantic verification, context representation, validation of themes, history of xungu interpretation, and pursuit of imagery. In the cross-disciplinary collision centered on Chinese hermeneutics, the modern approach to the proposition that "xungu tong yili" has shown abundant vitality.

Can we be aware of both visceral and somatic sensations via a single neuronal pathway?

Sensory information arising from somatic and visceral area is thought to be respectively transmitted to the brain by two distinct sensory neuronal systems, the somatic and visceral sensory pathway. A novel and unique spinal sensory system, the spinosolitary tract-dorsal column postsynaptic neuronal system (SST-DCPS), was physiologically and anatomically identified. The spinal neurons project to both visceral (the solitary tract nucleus) and somatic sensory (the dorsal column nuclei) nuclei via their branched axons and receive both visceral and somatic sensory information ascending from periphery through dichotomized primary afferents and descending from their targets via their branched axons. The brain might thus be aware of both visceral and somatic sensation via a single SST-DCPS neuronal channel. The finding of SST-DCPS system, as an example, might be considered as an intersection or fuzzy set of the SST and DCPS system and the concept of dichotomy in classification of neurons and neuronal pathways

Anaerobic treatment of wastewater containing methanol in up-flow anaerobic sludge bed (UASB) reactor

The direct conversion of methanol into methane is the main process in anaerobic treatment of methanol containing ***,acetic acid can also be produced from methanol theoretically,which may probably result in an abrupt pH drop and deteriorate the anaerobic ***,it is interesting to know what would really happen in an anaerobic reactor treating methanol *** this study,an up-flow anaerobic sludge bed(UASB)reactor treating methanol wastewater was *** chemical oxygen demand(COD),acetic acid and pH of the effluent were monitored at different loadings and influent *** results showed that the anaerobic reactor could be operated steadily at as low as 119 mg/L of influent alkalinity and high organic loading rate with no obvious pH *** fatty acids accumulation was not observed even at strong shock *** microorganisms in the sludge at the end of the test became homogeneous in morphology,which were mainly spherical or spheroidal in shape.

Research progress of slippage characteristic and gas film stability enhancement methods on biomimetic hydrophobic surfaces

The biomimetic hydrophobic surface is a potentially efficient underwater drag reduction method and the drag reduction mechanism of this kind of surface comes from the interfacial *** now,it is a hotspot to grasp the slippage characteristic and explore slippage enhancement *** paper not only summarizes our numerical simulation and experimental results of slippage characteristic at the solid-liquid interface(SLI)of hydrophobic surfaces(HS)and the gas-liquid interface(GLI)of superhydrophobic surfaces(SHS)in recent years,but also introduces some innovative methods that can effectively improve the gas film stability and drag reduction effect of ***,we used the molecular dynamics(MD)simulation method to figure out the effect of the solid-liquid interaction strength,the system temperature and the shear rate on the slippage of SLI,and expound their action mechanism from molecular ***,by MD and multibody dissipative particle dynamics(MDPD)method,the slippage behavior at the GLI was studied under the influence of the microstructure size and the flow driving *** proposed a new kind of hybrid slip boundary condition model to describe the slippage characteristic on *** addition,we found through experiment that a three-dimensional backflow will appear on the GLI under the interfacial adsorption of surfactants,and the backflow direction will reverse with the change of GLI ***,we put forward the wettability step structure and gas injection method to enhance the stability and drag reduction effect of the gas film on SHS.

Pancreaticβ-cell dysfunction and insulin resistance are two of the major causes of type 2 diabetes(T2D).Recent clinical and experimental studies have suggested that the functional capacity ofβ-cells,particularly in the first phase of insulin secretion,is a primary contributor to the progression of T2D and its associated ***β-cells undergo dynamic compensation and decompensation processes during the development of T2D,in which metabolic stresses such as endoplasmic reticulum stress,oxidative stress,and inflammatory signals are key regulators ofβ-cell *** and exercise interventions have been shown to be effective approaches for the treatment of obesity and T2D,especially in the early *** the targeted tissues and underlying mechanisms of dietary and exercise interventions remain somewhat vague,accumulating evidence has implicated the improvement ofβ-cell functional *** this review,we summarize recent advances in the understanding of the dynamic adaptations ofβ-cell function in T2D progression and clarify the effects and mechanisms of dietary and exercise interventions onβ-cell dysfunction in *** review provides molecular insights into the therapeutic effects of dietary and exercise interventions on T2D,and more importantly,it paves the way for future research on the related underlying mechanisms for developing precision prevention and treatment of T2D.

Metabolic syndrome has become a global epidemic that adversely affects human health. Both genetic and environmental factors contribute to the pathogenesis of metabolic disorders; however, the mechanisms that integrate these cues to regulate metabolic physiology and the development of metabolic disorders remain incompletely defined. Emerging evidence suggests that SWlISNF *** complexes are critical for directing metabolic reprogramming and adaptation in response to nutritional and other physiological sigrials. The ATP-dependent SWl/SNF ing complexes comprise up to 11 subunits, among which the BAF60 subunit serves as a key link between the core complexes and specific transcriptional factors. The BAF60 subunit has three members, BAF60a, b, and c. The distinct tissue distribution patterns and regulatory mechanisms of BAF60 proteins confer each isoform with specialized functions in different m^abolic cell types. In this review, we summarize the emerging roles and mechanisms of BAF60 proteins in the regulation of nutrient sensing and energy metabolism under physiological and disease conditions.
