Integration of Cytological Features with Molecular and Epigenetic Properties of Rice Chromosome 4

It has been reported that rice chromosome 4 has eight major heterochromatic knobs within the heterochromatic half and that this organization correlates with chromosomal-level transcriptional activity. To better understand this chromosomal organization, we created a model based on the statistical distribution of various types of gene models to divide chromosome 4 into 17 euchromatic and heterochromatic regions that correspond with the cytological staining. Fluorescence in-situ hybridization （FISH） experiments using a set of bacterial artificial chromosome （BAC） clones from chromosome 4 placed all 18 clones in the region predicted by the model. Elevated levels of H3K4 di- and tri-methylation detected by chromatin-immunoprecipitation （CHIP） on chip were correlated with euchromatic regions whereas lower levels of these two modifications were detected in heterochromatic regions. Small RNAs were more abundant in the heterochromatic regions. To validate these findings, H3K4 trimethylation, H3K9 acetylation, H4K12 acetylation, and H3K9 di- and tri-methylation of 19 individual genes were measured by ChlP-PCR. Genes in heterochromatic regions had elevated H3K9 di- and tri-methylation while genes in euchromatic regions had elevated levels of the other three modifications. We also assayed cytosine methylation of these genes using the restriction enzymes McrBC, Hapll, and Msp I. This analysis indicated that cytosines of transposable elements and some genes located in heterochromatic regions were methylated while cytosines of the other genes were unmethylated. These results suggest that local transcriptional activity may reflect the organization of the corresponding part of the chromosome. They also indicate that epigenetic regulation plays an important role in correlating chromosomal organization with transcriptional activity.

Rice OsGL 1-1 Is Involved in Leaf Cuticular Wax and Cuticle Membrane

Cuticular wax forms a hydrophobic barrier on aerial plant organs; it plays an important role in protecting a plant from damage caused by many forms of environmental stress. In the present study, we characterized a rice leaf wax-deficient mutant osgll-1 derived from a spontaneous mutation, which exhibited a wax-deficient and highly hydro- philic leaf phenotype. We cloned the OsGLI-1 gene by the map-based cloning method and performed a complementation test to confirm the function of the candidate gene. Molecular studies revealed that OsGLI-1 was a member of the OsGL1 family, and contained regions that were homologous to some regions in sterol desaturases and short-chain dehydro- genases/reductases. Compared to the wild-type, the osgll-1 mutant showed decreased cuticular wax deposition, thinner cuticular membrane, decreased chlorophyll leaching, increased rate of water loss, and enhanced sensitivity to drought. OsGL 1-1 is expressed ubiquitously in rice. The transient expression of OsGLl-l-green fluorescent protein fusion protein indicated that OsGLI-1 is localized in the cytoplasm, plasma membrane, and nucleus.

Analysis of impact pressure,rock-breaking effect,and ground vibration induced by the disposable CO_(2)fracturing tube

The technology of expansion fracturing with liquid CO_(2)(EFLCO_(2))has attracted increasing attention due to reduced vibration and *** disposable fracturing tube has been developed and is gradually replacing the Cardox ***,there is a lack of impact pressure testing of disposable tubes under real working conditions,selection of gas explosion design parameters,and systematic analysis of blasting *** limitation has constrained the widespread application of disposable fracturing tubes in engineering.A joint monitoring of the pressure-time curves in the disposable tubes and boreholes was *** rock-breaking effect of varying hole spacing parameters in the EFLCO_(2)design was analyzed,and a systematic study was carried out on the vibration peak value,frequency,and energy *** results show that(1)the pressure distribution characteristics,stress peak value,and duration in the disposable tubes are different from those of Cardox tubes,which show multi-peak distribution,low-pressure peak value,and short *** correlation between the pressure in the disposable tube,filling pressure,and liquid CO_(2)weight is established,and a theoretical calculation method for the borehole wall pressure is proposed;(2)The hole spacing in rocks of different hardness is suggested;and(3)At the same scale distance,the peak particle velocity(PPV)caused by EFLCO_(2)(PPVCO_(2))is significantly smaller than that caused by blasting(PPVexplosive).The ratio of PPVexplosive to PPVCO_(2)is a power function related to scale distance,and a distance-related zonality exist in this *** frequency composition of the vibration signal caused by EFLCO_(2)is relatively simple with a narrow frequency *** PPV and energy are mainly concentrated in the low-frequency *** research contributes to the optimization of disposable fracturing tubes,gas explosion design,and vibration hazard control.

Determining an optimal control strategy for anthropogenic VOC emissions in China based on source emissions and reactivity

Effects of content and particle size of TiH2 powders on the energy output rules of RDX composite explosives

In order to improve the detonation characteristics of RDX,a RDX-based composite explosive with TiH_(2)powders was *** effects of content and particle size of TiH_(2)powders on thermal safety,shock wave parameters and thermal damage effects of RDX-based composite explosive were studied with the C80 microcalorimeter,air blast experiment system and colorimetric thermometry *** results showed that TiH_(2)powders could enhance the thermal stability of RDX-based composite explosive and increase its ultimate decomposition *** content and particle size of TiH_(2)powders also had significant effects on the thermal safety,detonation velocity,shock wave parameters,fireball temperature and duration of RDX-based composite ***,the differences of TiH_(2)and Ti powders on the detonation energy output rules of RDX-based composite explosives were also compared,showing that TiH_(2)powders had better influences on improving the explosion power and thermal damage effect of RDX-based composite explosives than Ti powders,for the participation of free H_(2)released by TiH_(2)powders in the detonation ***_(2)powders have important research values as a novel energetic additive in the field of military composite explosives.

Clinical analysis of multiple primary gastrointestinal malignant tumors:A 10-year case review of a single-center
