Objective To evaluate a single-reaction genome amplification method, the multisegment reverse transcription-PCR （M-RTPCR）, for its sensitivity to full genome sequencing of influenza A virus, and the ability to differentiate mix-subtype virus, using the next generation sequencing （NGS） platform. Methods Virus genome copy was quantified and seria（iy diluted to different titers, followed by amplification with the M-RTPCR method and sequencing on the NGS platform. Furthermore, we manually mixed two subtype viruses to different titer rate and amplified the mixed virus with the M-RTPCR protocol, followed by whole genome sequencing on the NGS platform. We also used clinical samples to test the method performance. Results The M-RTPCR method obtained complete genome of testing virus at 125 copies/reaction and determined the virus subtype at titer of 25 copies/reaction. Moreover, the two subtypes in the mixed virus could be discriminated, even though these two virus copies differed by 200-fold using this amplification protocol. The sensitivity of this protocol we detected using virus RNA was also confirmed with clinical samples containing Iow-titer virus. Conclusion The M-RTPCR is a robust and sensitive amplification method for whole genome sequencing of influenza A virus using NGS platform.

In this work,the formation mechanism of the droplet-to-granule was investigated in detail based on mold powder manufacturing.A specific mathematical model of two-stage spray drying was established to describe droplet and granule motion,heat and mass transfer,and granule morphology during spray ***,the relationships between spray drying parameters(inlet temperature,atomization pres-sure,slurry mass flow rate)and the properties of the drying tower(temperature and velocity fields)and mold powder granules(temperature,evaporation rates,moisture content,and diameter)were simulated and calculated using ANSYS/Fluent *** ensure that the granule size of mold powder was controlled within the ideal range(0.2-0.6 mm)for producing granules with appropriate mechanical and metallurgical properties,the following optimum spray drying parameters were chosen based on the results of the numerical simulation:inlet temperatures,873 K;slurry atomization pressure,1.8 MPa;slurry mass flow rate,0.05 kg *** these parameters,the slurry mass flow rate has the most significant effect on granule size.

In some military application scenarios,Unmanned Aerial Vehicles(UAVs)need to perform missions with the assistance of on-board cameras when radar is not available and communication is interrupted,which brings challenges for UAV autonomous navigation and collision *** this paper,an improved deep-reinforcement-learning algorithm,Deep Q-Network with a Faster R-CNN model and a Data Deposit Mechanism(FRDDM-DQN),is proposed.A Faster R-CNN model(FR)is introduced and optimized to obtain the ability to extract obstacle information from images,and a new replay memory Data Deposit Mechanism(DDM)is designed to train an agent with a better *** training,a two-part training approach is used to reduce the time spent on training as well as retraining when the scenario *** order to verify the performance of the proposed method,a series of experiments,including training experiments,test experiments,and typical episodes experiments,is conducted in a 3D simulation *** results show that the agent trained by the proposed FRDDM-DQN has the ability to navigate autonomously and avoid collisions,and performs better compared to the FRDQN,FR-DDQN,FR-Dueling DQN,YOLO-based YDDM-DQN,and original FR outputbased FR-ODQN.

Construction of heterojunctions is a normal and effective strategy to improve the photocatalytic performances of semiconductors,through which both the lifetime and the redox ability of electrons/holes can be improved,as co mpared to the respective *** this basis,we constructed Z-scheme CdS/LaFeO_(3) heterojunctions(CdS/LFO),by in-situ growing different amounts of CdS on the surface of LaFeO_(3),for photocatalytic degradation of tetracycline hydrochloride(TC)in aqueous solution at room *** crystal structure,surface morphology and optoelectronic properties of the COS/LFO heterojunctions were systemically characterized to correlate the reaction *** tests indicate that the CdS/LFO heterojunctions exhibit promising activity for TC degradation under visible light irradiation(λ≥420 nm),with 85%TC conversion obtained at reaction time of 45 min,which is 3.4 and 1.9 times higher than that of CdS and LaFeO_(3).The CdS/LFO heterojunctions are also stable in the reaction and can be reused for four cycles with no appreciable activity *** applicability of CdS/LFO to photocatalytic degradation of organic dyes,as well as the reaction mechanism,was also explored.

As an important factor that directly affects agricultural production, the social economy, and policy implementation,observed changes in dry/wet conditions have become a matter of widespread concern. However, previous research has mainly focused on the long-term linear changes of dry/wet conditions, while the detection and evolution of the non-linear trends related to dry/wet changes have received less attention. The non-linear trends of the annual aridity index, obtained by the Ensemble Empirical Mode Decomposition(EEMD) method, reveal that changes in dry/wet conditions in China are asymmetric and can be characterized by contrasting features in both time and space in China. Spatially, most areas in western China have experienced transitions from drying to wetting, while opposite changes have occurred in most areas of eastern China. Temporally, the transitions occurred earlier in western China compared to eastern China. Research into the asymmetric spatial characteristics of dry/wet conditions compensates for the inadequacies of previous studies, which focused solely on temporal evolution;at the same time, it remedies the inadequacies of traditional research on linear trends over centennial timescales. Analyzing the non-linear trend also provides for a more comprehensive understanding of the drying/wetting changes in China.

Reliable communication and intensive computing power cannot be provided effectively by temporary hot spots in disaster areas and complex terrain ground *** this has greatly developed the application and integration of UAV and Mobile Edge Computing(MEC)to the Internet of Things(loT).However,problems such as multi-user and huge data flow in large areas,which contradict the reality that a single UAV is constrained by limited computing power,still *** to allowing UAV collaboration to accomplish complex tasks,cooperative task offloading between multiple UAVs must meet the interdependence of tasks and realize parallel processing,which reduces the computing power consumption and endurance pressure of *** the computing requirements of the user terminal,delay constraint of a computing task,energy constraint,and safe distance of UAV,we constructed a UAV-Assisted cooperative offloading energy efficiency system for mobile edge computing to minimize user terminal energy ***,the resulting optimization problem is originally nonconvex and thus,difficult to solve *** tackle this problem,we developed an energy efficiency optimization algorithm using Block Coordinate Descent(BCD)that decomposes the problem into three convex ***,we jointly optimized the number of local computing tasks,number of computing offloaded tasks,trajectories of UAV,and offloading matching relationship between multi-UAVs and multiuser *** results show that the proposed approach is suitable for different channel conditions and significantly saves the user terminal energy consumption compared with other benchmark schemes.

Class imbalance is a common characteristic of industrial data that adversely affects industrial data mining because it leads to the biased training of machine learning *** address this issue,the augmentation of samples in minority classes based on generative adversarial networks(GANs)has been demonstrated as an effective *** study proposes a novel GAN-based minority class augmentation approach named classifier-aided minority augmentation generative adversarial network(CMAGAN).In the CMAGAN framework,an outlier elimination strategy is first applied to each class to minimize the negative impacts of ***,a newly designed boundary-strengthening learning GAN(BSLGAN)is employed to generate additional samples for minority *** incorporating a supplementary classifier and innovative training mechanisms,the BSLGAN focuses on learning the distribution of samples near classification ***,it can fully capture the characteristics of the target class and generate highly realistic samples with clear ***,the new samples are filtered based on the Mahalanobis distance to ensure that they are within the desired *** evaluate the effectiveness of the proposed approach,CMAGAN was used to solve the class imbalance problem in eight real-world fault-prediction *** performance of CMAGAN was compared with that of seven other algorithms,including state-of-the-art GAN-based methods,and the results indicated that CMAGAN could provide higher-quality augmented results.

The most critical part of a neutron computed tomography(NCT) system is the image processing algorithm,which directly affects the quality and speed of the reconstructed *** types of noise in the system can degrade the quality of the reconstructed ***,to improve the quality of the reconstructed images of NCT systems,efficient image processing algorithms must be *** anisotropic diffusion filtering(ADF) algorithm can not only effectively suppress the noise in the projection data,but also preserve the image edge structure information by reducing the diffusion at the image ***,we propose the application of the ADF algorithm for NCT image *** compare the performance of different algorithms in NCT systems,we reconstructed images using the ordered subset simultaneous algebraic reconstruction technique(OS-SART) algorithm with different regular terms as image processing *** the iterative reconstruction,we selected two image processing algorithms,the Total Variation and split Bregman solved total variation algorithms,for comparison with the performance of the ADF ***,the filtered back-projection algorithm was used for comparison with an iterative *** reconstructing the projection data of the numerical and clock models,we compared and analyzed the effects of each algorithm applied in the NCT *** on the reconstruction results,OS-SART-ADF outperformed the other algorithms in terms of denoising,preserving the edge structure,and suppressing *** example,when the 3D Shepp–Logan was reconstructed at 25 views,the root mean square error of OS-SART-ADF was the smallest among the four iterative algorithms,at only *** universal quality index,mean structural similarity,and correlation coefficient of the reconstructed image were the largest among all algorithms,with values of 0.9877,0.9878,and 0.9887,respectively.

Photocatalytic CO_(2)reduction provides a promising strategy for the alleviation of greenhouse effect and energy shortage *** single atom modification is an effective method to improve the CO_(2)reduction performance of covalent organic framework(COF)photocatalysts,while the intrinsic mechanism is not revealed in ***,a Pt-COF photocatalytic system was constructed by embedding Pt single atom in TFPT-TMT-COF(TFPT:1,3,5-tris(4-formylphenyl)-triazine,TMT:2,4,6-trimethyl-1,3,5-triazine).Based on density functional theory(DFT)calculation,the geometric structure and electronic property of Pt-COF were investigated,and the CO_(2)adsorption and reduction reaction process on Pt-COF were *** results show that the Pt atom can be steadily anchored in COF via the formation of Pt-N and Pt-C ***,there is strong electronic interaction between Pt and *** incorporation of Pt atom benefits the CO_(2)reduction activity of COF from multiple aspects:(ⅰ)reducing the band gap and improving light absorption;(ⅱ)enhancing CO_(2)adsorption and activating the CO_(2)molecule;(ⅲ)decreasing the energy barrier in the hydrogenation step of CO_(2)→COOH,thus facilitating CO_(2)conversion to CO;(ⅳ)inhibiting the hydrogenation of CO,thereby increasing the selectivity of *** work brings a novel insight into the excellent CO_(2)reduction performance of Pt modified COF,and provides useful references for the design of single atom photocatalysts.
