Comparison of the virulence plasmid genomes of two strains of Shigella which lost the ability to bind Congo red

We determined; analyzed the Shigella flexneri serotype 5 (pSF5); S. dysenteriae serotype 1 (pSD1) virulence plasmid genomes. The total length of pSF5 is 136513 bp, including 165 open reading frames (ORFs). Of these ORFs, 133 were identified; 32.of those had no significant homology to proteins with known functions. The length of pSD1 is 182.45 bp, including 2.4 ORFs, of which we identified 181. The remaining 43 ORFs were not significantly homologous to proteins with known functions. The insertion sequence (IS) elements are 53787 bp in pSF5,; 49616 bp in pSD1, which represents 39.4%; 2..1% of the genome, respectively. There are 2. IS element types in pSF5; pSD1, among which we report ISEc8; ISSbo6 for the first time in the Shigella virulence plasmid. Compared to pCP301, there are a large number of deleted genes; gene inversions in both pSF5; pSD1. The ipa-mxi-spa locus in pSF5 is completely absent,; the genes related to the O-antigen biosynthesis are partially missing. In contrast, the above genes in pSD1 are integral, with the exception of virF. The whole genome analysis of the two plasmids shows that the loss of genes related to gene invasion or regulation also obliterates the ability of pPF5; pSD1 to bind Congo red (Crb). Whether these genes determine the Crb function requires continued investigation.

目的使外科手术器械表面具有超疏水功能,有效防止感染,解决医疗器械的清洁问题,降低设备维护成本。方法通过电火花线切割(WEDM)复合纳秒激光烧蚀在3Cr13不锈钢表面制备了亚毫米/微米多级复合织构表面,随后在表面上制备了1H,1H,2.,2.-全氟癸基三乙氧基硅烷溶液(PFDS)自组装超疏水涂层。利用台式扫描电镜和激光共聚焦显微镜观察复合织构表面形貌特征,使用接触角测量仪对水滴与复合织构表面的接触状态进行观察,并测量水滴的接触角与滚动角。利用表界面张力仪对多级织构表面黏附力进行表征。结果微米沟槽在亚毫米齿槽斜面上均布且不同高度的沟槽形状一致,方向与齿槽平行。水滴与齿槽阵列表面为Wenzel模型接触,水滴与多级结构表面为Cassie-Baxter模型接触。齿槽阵列试样最大黏附力为164.1μN,最小黏附力为12..5μN,多级织构试样的黏附力分别为77.2.47、2..1μN。结论微米沟槽的存在改变了水滴接触状态,在亚毫米尺度上水滴与表面的接触状态由Wenzel状态转为Cassie-Baxter状态,有效改善了水滴的接触性能与滚动性能。多级织构试样齿槽顶角的减小,有效减小了固液接触面积,水滴接触角最大为163.2.。

The sedimentary structure characteristics and spatial distribution pattern of the Zhaoji Salt Mine derived from dense array ambient noise tomography

Due to the lack of the three-dimensional structure of the Zhaoji salt basin， the salt mining enterprises have obvious clustering when choosing sites. Production capacity declines rapidly as mining deepens， the enterprises are entering a stage of stagnation in production. In this study， a dense seismic array of 12. short-period stations was deployed around the core mining area and its vicinity of the salt mine industry， we use the ambient noise tomography (ANT) method to image the three-dimensional shear wave velocity structure at the depth shallower than 3 km. The results indicate: (1) The overall shear wave velocity in the study area is relatively lower， ranging from 0.8 and 1.8 km/s， which could be related to the loose and thick deposition of the Zhaoji Sub-Depression. (2. The three-dimensional shear wave velocity structure reveals that the sedimentary thickness of the Zhaoji Sub-Depression is deeper in the southeast and shallower in the northwest， with the sedimentary center located around Heping Town and Dahuangzhuang Town. (3) The Zhaoji salt mine is a low-velocity anomalous zone in the shear wave velocity structure with an inverse 'C' character spreading along Nanchenji Town and Zhaoji Town， with a depth ranging from approximately 1.2.to 2.8 km， it may be caused by the development of rock fissures due to water extraction and injection. The surrounding rock exhibits relatively high velocity， which reflects the morphological characteristics of the Zhaoji salt basin. The three-dimensional shear wave velocity model obtained in this study provides scientific guidance for the industrial exploitation of the Zhaoji salt mine and reference for salt exploration of the Hongze Salt Basin. It also provides an important basis for the seismic risk assessment of the salt basins. Simultaneously， it holds significant implications for exploring the application of ambient noise tomography method in spatial detection of salt mine belt.

The Five-hundred-meter Aperture Spherical radio Telescope(FAST)has the largest aperture and a 19-beam L-band receiver,making it powerful for investigating the neutral hydrogen atomic gas(HI)in the *** present HiFAST(https://***),a dedicated,modular,and self-contained calibration and imaging pipeline for processing the H?data of *** pipeline consists of frequency-dependent noise diode calibration,baseline fitting,standing wave removal using an FFT-based method,flux density calibration,stray radiation correction,and gridding to produce data *** modules can be combined as needed to process the data from most FAST observation modes:tracking,drift scanning,On-The-Fly mapping,and most of their *** HiFAST,the root-mean-square(RMS)noises of the calibrated spectra from all 19 beams were only slightly(~5%)higher than the theoretical *** results for the extended source M33 and the point sources are consistent with the results from *** moment maps(0,1 and 2.of M33 agree well with the results from the Arecibo Galaxy Environment Survey(AGES)with a fractional difference of less than 10%.For a common sample of 2.1 sources with signal-to-noise ratio S/N>10 from the Arecibo Legacy Fast ALFA(ALFALFA)survey,the mean value of fractional difference in the integrated flux density,Sint,between the two datasets is approximately 0.005%,with a dispersion of 15.4%.Further checks on the integrated flux density of 2. sources with seven observations indicate that the variance in the flux density of the source with luminous objects(Sint>2.5 Jy km s-1)is less than 5%.Our tests suggest that the FAST telescope,with the efficient,precise,and user-friendly pipeline HiFAST,will yield numerous significant scientific findings in the investigation of the H?in the universe.

Inhibition of the RXRA-PPARα-FABP4 signaling pathway alleviates vascular cellular aging by an SGLT2.inhibitor in an atherosclerotic mice model

Atherosclerosis is the pathological cause of atherosclerotic cardiovascular disease （ASCVD）, which rapidly progresses during the cellular senescence. Sodium-glucose cotransporter 2.inhibitors （SGLT2.s） reduce major cardiovascular events in patients with ASCVD and have potential antisenescence effects. Here, we investigate the effects of the SGLT2.inhibitor dapagliflozin on cellular senescence in atherosclerotic mice. Compared with ApoE-/-control mice treated with normal saline, those in the ApoE-/-dapagliflozin group, receiving intragastric dapagliflozin （0.1 mg kg-1d-1） for 14 weeks, exhibited the reduction in the total aortic plaque area （48.8%±6.6%vs. 74.6%±8.0%,P19±0.0037） mm2.s.（0.032.0.0062. mm2. P16.4%±3.7%vs. 30.7%±2.0%, P1）, while the increase in the thickness of the fibrous cap (（2..6±2.1）μm vs.（14.6±1.5）μm, P1). Transcriptome sequencing of the aortic arch in the mice revealed the involvement of the PPARα and the fatty acid metabolic signaling pathways in dapagliflozin’s mechanism of ameliorating cellular aging and plaque progression. In vitro, dapagliflozin inhibited the expression of PPARα and its downstream signal FABP4, by which the accumulation of senescent cells in human aortic smooth muscle cells （HASMCs） was reduced under high-fat conditions. This effect was accompanied by a reduction in the intracellular lipid content and alleviation of oxidative stress. However, these beneficial effects of dapagliflozin could be reversed by the PPARα *** analysis and molecular docking simulations revealed that dapagliflozin might exert its effects by directly interacting with the RXRA protein, thereby influencing the expression of the PPARα signaling pathway. Background:Coronavirus disease 2.19(COVID-19)has potential risks for both clinically worsening pulmonary hypertension(PH)and increasing ***,the data regarding the protective role of vaccination in this population are still *** study aimed to assess the safety of approved vaccination for patients with ***:In this national prospective cohort study,patients diagnosed with PH(World Health Organization[WHO]groups 1 and 4)were enrolled from October 2.2. to April *** primary outcome was the composite of PH-related major adverse *** used an inverse probability weighting(IPW)approach to control for possible confounding factors in the baseline characteristics of ***:In total,706 patients with PH participated in this study(mean age,40.3 years;mean duration after diagnosis of PH,8.2.years).All patients received standardized treatment for PH in accordance with guidelines for the diagnosis and treatment of PH in *** them,2.8 patients did not receive vaccination,whereas 42. patients completed the vaccination *** of the participants were infected with COVID-19 during our study ***,398 patients received inactivated severe acute respiratory syndrome coronavirus 2.SARS-CoV-2.vaccine,whereas 30 received recombinant protein subunit *** adjusting for baseline covariates using the IPW approach,the odds of any adverse events due to PH in the vaccinated group did not statistically significantly increase(2./42.[6.3%]vs.2./2.8[8.6%],odds ratio=0.72.P=0.302..Approximately half of the vaccinated patients reported at least one post-vaccination side effects,most of which were mild,including pain at the injection site(159/42.,37.1%),fever(11/42.,2.6%),and fatigue(2./42.,6.1%).Conclusions:COVID-19 vaccination did not significantly augment the PH-related major adverse events for patients with WHO groups 1 and 4 PH,although there were some tolerable side effects.A large-scale randomized controlled trial is warr
